형질전환된 담배에서 사람 B형 간염바이러스 항체 유전자의 발현 (Expression of Antibody Genes Specific for Human Hepatitis-B Virus in Transgenic Tabacco Plants)

  • 사람 B형 간염 바이러스의 pre-S2 표면항원에 결합하는 키메라 항체 유전자(카파 및 감마사슬의 cDNA클론)를 식물체에서 발현시키기 위해 식물체 발현벡터인 pBKS-1에 XbaI 자리를 이용하여 클로닝하였다. 이들 유전자를 포함하는 대장균의 플라스미드 핵산을 추출하여 아그로박테리움에 형질전환 시켰다. 다음 담배의 조직절편과 아그로박테리움을 공동배양함으로써 담배의 형질 전환을 시도하였다. 카나마이신이 포함된 신초유기배지에서 나온 신초를 시료로 하여 Western blot을 실시함으로써 이들 유전자가 형질전환 담배에서 안정하게 발현됨을 확인하였다.

  • Based on the super elastic properties of the shape memory alloy (SMA) and the inverse piezoelectric effect of piezoelectric (PZT) ceramics, a kind of hybrid semi-active control device was designed and made, its mechanical properties test was done under different frequency and different voltage. The local search ability of genetic algorithm is poor, which would fall into the defect of prematurity easily. A kind of adaptive immune memory cloning algorithm(AIMCA) was proposed based on the simulation of clone selection and immune memory process. It can adjust the mutation probability and clone scale adaptively through the way of introducing memory cell and antibody incentive degrees. And performance indicator based on the modal controllable degree was taken as antigen-antibody affinity function, the optimization analysis of damper layout in a space truss structure was done. The structural seismic response was analyzed by applying the neural network prediction model and T-S fuzzy logic. Results show that SMA and PZT friction composite damper has a good energy dissipation capacity and stable performance, the bigger voltage, the better energy dissipation ability. Compared with genetic algorithm, the adaptive immune memory clone algorithm overcomes the problem of prematurity effectively. Besides, it has stronger global searching ability, better population diversity and faster convergence speed, makes the damper has a better arrangement position in structural dampers optimization leading to the better damping effect.

이미지 분석을 이용한 살모넬라 신속 진단키트의 측정감도 향상 (Improvement of the detection limit of rapid detection kit for Salmonella Typhimurium using image analysis system)

  • 식중독을 일으키는 대표적인 원인균인 살모넬라를 검출하기 위해 측방 유동형 간이진단 키트를 제작하였다. 또한, 제작된 간이진단 키트의 검출한계를 향상시키고 살모넬라 균수를 정량적으로 분석하기 위해 이미지 분석 시스템을 적용하였다. 그 결과 간이진단 키트의 검출한계는 $10^6\;cfu/mL$에서 $10^5\;cfu/mL$로 향상시킬 수 있었다. 살모넬라 균수의 정량적 분석을 위해 $T_{peak}/C_{peak}$$T_{area}/C_{area}$값을 이용하여 다중회귀분석을 수행하였으며 개발된 다중회귀방정식 중에서 가장 단순하고 결정계수가 높은 다중회귀방정식을 선정하였다. 추가로 제작된 간이진단 키트를 이용하여 개발된 다중회귀방정식을 검증한 결과 결정계수가 0.9393으로 우수한 선형성을 나타내었다. 본 연구에서 개발된 다중회귀방정식을 이용하여 더 정확하게 간이진단 키트에서 측정된 살모넬라 균수를 예측하는 것이 가능하였다.

나노바이오촉매 기반 효소결합면역흡착검사 (Nanobiocatalyst-Linked Immunosorbent Assay(NBC-LISA))

  • 생촉매인 효소의 기질선택성은 다양한 분야에서 유용하게 이용되고 있다. 그 중에서도 효소결합면역흡착검사(enzyme-linked immunosorbent assay, ELISA)는 항원항체의 결합을 항체와 공유결합된 효소의 반응으로 나타냄으로써 다양한 항원들의 진단을 가능케 했다. 하지만 기존의 효소결합면역흡착검사는 하나의 항체당 하나의 효소가 결합된 형태이기 때문에 감도(sensitivity)의 증가 폭에 그 한계가 있으며, 이를 극복하기 위한 방안으로 하나의 항체당 결합된 효소의 수를 증가시킴으로써 혁신적인 감도의 향상을 가져오는 연구가 진행되었다. 최근 나노바이오촉매(nanobiocatalyst, NBC) 접근방식을 이용한 효소활성의 안정화는 효소결합면역흡착검사의 감도 향상뿐만 아니라 그 성능의 안정성을 확보할 수 있는 연구결과로 이어지고 있다. 본 총설에서는 일반적인 효소결합면역흡착검사의 기본적인 원리와 감도향상을 위한 연구, 그리고 성능안정성(performance stability)을 향상시키기 위한 나노바이오촉매-결합면역흡착검사(Nanobiocatalyst-Linked Immunosorbent Assay, NBC-LISA)에 대하여 살펴보고자 한다.

Detection of Multi-class Pesticide Residues Using Surface Plasmon Resonance Based on Polyclonal Antibody

  • The detection of carbamate (carbofuran, carbaryl, benfracarb, thiodicarb, and methomil) and organophosphate (diazinon, cadusafos, ethoprofos, parathion-methyl, and chlorpyrifos) pesticide residues with very low detection limits was carried out using surface plasmon resonance (SPR) based equipment. The capacity to develop a portable SPR biosensor for food safety was also investigated. The applied ligand for the immunoassays was polyclonal goat anti-rabbit immunoglobulin (IgG) peroxidase conjugate. Concentration tests using direct binding assays showed the possibility of quantitative analysis. For ligand fishing to find a proper antibody to respond to each pesticide, acetylcholinesterase (AChE), and glutathione-S-transferase (GST) were tested. The reproducibility and precision of SPR measurements were evaluated. With this approach, the limit of detection for pesticide residues was 1 ng/mL and analysis took less than 11 min. Thus, it was demonstrated that detecting multi-class pesticide residues using SPR and IgG antibodies provides enough sensitivity and speed for use in portable SPR biosensors.

Development of Surface Plasmon Resonance Immunosensor through Metal Ion Affinity and Mixed Self-Assembled Monolayer

  • An immunosensor based on surface plasmon resonance (SPR) with enhanced performance was developed through a mixed self-assembled monolayer. A mixture of 16-mercaptohexadecanic acid (16-MHA) and 1-undecanethiol with various molar ratios was self-assembled on gold (Au) surface and the carboxylic acid groups of 16-MHA were then coordinated to Zn ions by exposing the substrate to an ethanolic solution of $Zn(NO_3)_2\cdot 6H_2O$. The antibody was immobilized on the SPR surface by exposing the functionalized substrate to the desired solution of antibody in phosphate-buffered saline (PBS) molecules. The film formation in series was confirmed by SPR and atomic force microscopy (AFM). The functionalized surface was applied to develop an SPR immunosensor for detecting human serum albumin (HSA) and the estimated detection limit (DL) was 4.27 nM. The limit value concentration can be well measured between ill and healthy conditions.

Antifertility Effect of Progesterone Antibodies in Mice

  • Early embryo development and implantation were arrested in ICR mice which were passively immunized with a mouse monoclonal progesterone amtibody given as a single intraperitoneal injection at 12 hrs or 60 hrs post coitum (p. c.). Unimplanted embryos were recovered from the reproductive tract of the antibody-treated mice and none of these progressed to the blastocyst stage. The most pronounced effect was an arrest of embryonic development at a stage prior to cavitation. The plasma progesterone concentration in the blood taken by carbiac puncture increased greatly after the treatment by virtue of high affinity binding by the antibody in circulation. The result showed that passive immunization against progesterone shortly after mating interfered with early hormone dependent steps which were essential for normal embryonic development.

황색포도상구균 신속 검출용 간이 진단키트 (Rapid Detection Kit for Staphylococcus aureus)

  • This study was performed to develop a rapid test kit for pathogenic Staphylococcus in various samples. The rapid detection kit has been fabricated based on nitrocellulose lateral-flow strip. Colloidal gold and Staphylococcus antibodies were used as a tag and a receptor, respectively. Manually spotted Staphylococcus antibody and anti-mouse antibody on the surface of nitrocellulose membrane were used as test and control lines, respectively. Feasibility of the rapid kit to detect Staphylococcus aureus in samples were evaluated. The intensity of the color of the test line started to increase with the samples in which higher concentration of the cells were contained. The sensitivity of the sensor was $10^6$ cfu/mL Staphylococcus spiked in PBS. Also, the rapid test kit could detect $10^5$ cfu/mL of Staphylococcus in chicken meat extract.
