Changes in SARS-CoV-2 antibody titers 6 months after the booster dose of BNT162b2 COVID-19 vaccine among health care workers

  • Purpose: In Japan, the data on severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) antibody titers after the booster dose of the coronavirus disease 2019 (COVID-19) vaccine are insufficient. The aim of this study is to evaluate changes in SARS-CoV-2 antibody titers before, 1, 3, and 6 months after the booster dose of the BNT162b2 COVID-19 vaccine among health care workers. Materials and Methods: A total of 268 participants who received the booster dose of the BNT162b2 vaccine were analyzed. SARS-CoV-2 antibody titers were measured before (baseline) and at 1, 3, and 6 months after the booster dose. Factors associated with changes in SARS-CoV-2 antibody titers at 1, 3, and 6 months were analyzed. Cutoff values at baseline were calculated to prevent infection of the omicron variant of COVID-19. Results: The SARS-CoV-2 antibody titers at baseline, and 1, 3, and 6 months were 1,018.3 AU/mL, 21,396.5 AU/mL, 13,704.6 AU/mL, and 8,155.6 AU/mL, respectively. Factors associated with changes in SARS-CoV-2 antibody titers at 1 month were age and SARS-CoV-2 antibody titers at baseline, whereas changes in SARS-CoV-2 antibody titers at 3 and 6 months were associated with the SARS-CoV-2 antibody titers at 1 month. The cutoff values of the SARS-CoV-2 antibody titers at baseline were 515.4 AU/mL and 13,602.7 AU/mL at baseline and 1 month after the booster dose, respectively. Conclusion: This study showed that SARS-CoV-2 antibody titers increase rapidly at 1 month after the booster dose of the BNT162b2 vaccine and begin to decrease from 1 to 6 months. Hence, another booster may be needed as soon as possible to prevent infection.

Serum Parvovirus Antibody Titers among Healthy Adult Dogs in Korea (국내 성견의 개 파보바이러스에 대한 항체가 조사)

  • This study was performed to monitor the level of serum canine parvovirus(CPV) antibody titers in adult dogs throughout the Korea from January 2003 to April 2004. A total of clinically healthy 885 dogs between 1 year and 17 years old were included in this study. Serum antibody titers against CPV were measured by means of hemagglutination inhibition(HI) titers at the time dogs were brought to the hospital for revaccination. Most of dogs had been primarily vaccinated or previously revaccinated. Dogs were grouped by age, breed, sex, and primary vaccination and revaccination to determine whether these factors were associated with antibody titers. Serum CPV titers ${\geq}80$ were considered protective. Protective antibodies against CPV were present in 95.0% of the population. Breed, age, and primary vaccination and revaccination were not significantly associated with serum CPV antibody titers. But sex was significantly associated with CPV antibody titers. The results of this study have shown that there is a need to reconsider the annual revaccination strategy against CPV infection.

Comparison of ELISA and HI titers in broiler chicks vaccinated with infectious bronchitis virus and Newcastle disease virus (전염성기관지염 및 뉴캣슬병 백신을 접종한육계에서 ELISA 및 HI 항체가 비교)

  • To compare of serum antibody titers using ELISA and HI, serum samples were collected from 100 breeders and their progeny 550 broilers. The breeders and broilers were vaccinated with infectious bronchitis(IB)- and Newcastle disease(ND)-viruses according to general vaccination program. The antibodies in serum samples against IB and ND viruses were detected by enzyme-linked immunosorbent assay(ELISA) using commercial ELISA kit and hemagglutination inhibition(HI) test. Geometric mean titer(GMT) of ELISA and In titers were monitored from 1-day-old to 35-day-old broilers and compared to those of breeder chickens. The antibody titers of breeders vaccinated with ]B virus showed 47,800, ELISA and 7.2, HI, respectively. Progeny chicks, 1-day-old, vaccinated with IBV showed high antibody titers than those of breed chickens. Those chicks were maintained protective antibody levels until 11-day-old. From 14-day-old, the antibody level decreased below protective levels. In ND, breeders serum antibody titers ELISA and Eiu were 30,200 GMT and 8.7 HI titer, respectively. On 1-day-old chicks, antibody levels was decreased to half in ELISA(16,270) compared with those of breeders, but In titers was 7.4. Progeny broilers, protective antibody level was maintained until 14- day-old by ELISA, but at 11-day-old by HI titers. After then, ND antibody titer was continuously decreased underdefense level. These result indicated that the ELISA method be more sensitive than HI titration to detect serum antibody level for IBV and NDV.

A Study on Protection of Maternal Antibody against Hantavirus in Rats

  • The etiologic agents of haemorragic fever with ranal syndrom (HFRS) in Korea are Hantaan and Seoul virus in the genus Hantavirus, family Bunyaviridae. In order to elucidate the role of maternal immunity to Hantavirus infection in rats, the protective effect of the maternal antibody were studies by using rats experimentally infected with Seoul virus strain HR80-39. Antibody titers of sera and viral antigen against Seoul virus were investigated by indirect immunofluorscence antibody technique (IFA). The dam sera had IFA antibody titers ranging from 1:128 to 1:1,024 after parturition. In fetuses, IFA antibody titers ranged from 1: 16 to 1:64 just after birth, increased to peak titers ranged from 1:256 to 1:1,024 in the 2nd week after birth. Challenged newborn rats had IFA antibody titers ranging from 1:64 to 1:1,024 after inoculation. No viral antigen was detected in lungs or other organs of the newborn rats. The maternal antibody to Seoul virus was transferred prenatally through placenta and postnatally via colostrum from immune dams to their offspring. These results demonstrated that maternal antibody to Seoul virus was quite effective in protecting newborn rats against same virus infection.

Effects of Florfenicol and Chromium (III) on Humoral Immune Response in Chicks

  • One hundred and sixty day-old Hainan chicks were randomly allotted into eight pens to investigate the effect of different dietary concentrations of chromium (Cr) in the form of chromium chloride, and different dosages of florfenicol on humoral immune responses by determining antibody titers to Newcastle disease (ND) vaccines using the hemagglutination inhibition test. The results indicated that ND antibody titers were significantly higher in chicks receiving Cr at low (5 mg/kg feed) and middle (10 mg/kg feed) dose compared with the control (p<0.01). However, ND antibody titers were significantly decreased in chicks receiving Cr at a high dosage of 500 mg/kg feed (p<0.01), though the ND antibody titers of the early days (d 21 and d 28 of age) were higher than that of the control group. It is suggested that excessive Cr intake has detrimental effects on ND antibody production in chicks. No significantly lower response was measured in chicks that received florfenicol at a low dosage of 50 mg/kg feed (p>0.05), but the ND antibody titers were significantly decreased in chicks receiving 200 and 400 mg/kg feed of the drug (p<0.01). The ND antibody titers of group receiving 200 mg/kg feed of florfenicol plus 10 mg/kg Cr were slightly higher than that of the group receiving single florfenicol of 200 mg/kg although, no significant differences were observed between these two treatments. It is suggested that the humoral immune response impaired by florfenicol (200 mg/kg feed) could not be significantly reversed by Cr (10 mg/kg feed).

Studies on the combined inactivated oil emulsion vaccine of Newcastle disease and avian infectious bronchitis in chickens (닭의 뉴캣슬·전염성 기관지염 바이러스 혼합 불활화 오일 에멀션 백신의 생산시험)

  • A single inoculation of combined vaccines of Newcastle disease and avian infectious bronchitis of chicken, in a form of gel-oil emulsion type (gel-OEV) was tested their immunogenecity in chickens. The results were summerized as follows: 1. Average minimum and maximum ELISA antibody titers of ND were recorded 2407 and 13144 respectively. In the case of IB, 1824 and 4496 were recorded as minimum and maximum titers. 2. The distribution of average proportional groups, in the lowest and the highest, were 1.6 and 7.0 in ND ELISA and 1.4 and 2.8 in IB ELISA antibody titers. 3. ND ELISA antibody titers were significantly increased upto 7th week after the vaccination. On the other hand, IB ELISA antibody titers were raised upto 4th week after the vaccination.

Seroepidemiological studies on virus-borne diseases of cattle in Kwangju and Chonnam area (광주(光州)·전남지역내(全南地域內) 소의 바이러스성 질병(疾病)에 관한 혈청학적(血淸學的) 연구(硏究))

  • Sera from 304 Holsteins or Korean native cattle were collected from slaughterhouse in Kwangju area to study the infection of major virus-borne diseases. Serum antibody titers against infectious bovine rhinotracheitis virus(IBRV), bovine viral diarrhea virus(BVDV), parainfluenza type-3 virus(PI-3V), bovine ephemeral fever virus(BEFV), bovine Ibaraki virus(BIV), bovine Akabane virus(BAKV), bovine rotavirus(BRV), bovine coronavirus(BCV) were measured by serum neutralization tests. Results which obtained were as follows. Sera from 280 cattle(92.1%) contained antibodies against BRV which rate was the highest among the 8 viruses, and serum antibodies against BCV, BVDV, BIV, BAKV, BEFV, IBRV and PI-3V were detected from 266(87.5%), 149(49%), 108(35.5%), 94(30.9%), 80(26.3%), 32(10.5%) and 24(7.9%) cattle, respectively. Prevalence of seropositives to BVDV, BIV, BAKV, BEFV were higher among Holsteins than among the Korean native cattle(P<0.05). Prevalence of antibody titers against BVDV, BIV and BEFV in Korean native cattle were higher among females than males, while more males contained antibodies to BAKV, IBRV and PI-3V than females in their blood(P<0.05). Seropositives to BVDV, BIV, BAKV, BEFV and IBRV in Holsteins were higher among females than males(P<0.05). In Korean native cattle, serum antibody titers against IBRV and PI-3V ranged from 1:2~1:32 and 1:2~1:64, respectively, while serum antibody titers against the rest 6 viruses ranged from $1:2{\sim}1:{\geq}256$. In Holsteins, serum antibody titers against IBRV and PI-3V ranged from 1:2~1:64 and 1:2~1:32, respectively, while serum antibody titers against the rest 6 viruses ranged from $1:2{\sim}1:{\geq}256$.

A comparative study on immunogenicity of the porcine epidemic diarrhea virus live-vaccine and inactivated-vaccine (돼지유행성설사병(PEDV) 생독과 사독백신의 면역형성 비교연구)

  • Porcine transmissible gastroenteritis virus (TGEV), porcine epidemic diarrhea virus (PEDV) and rotaviruses are considered as the most important causative agents of diarrhea in piglets. The study established 3 method vaccination programs to prevent PEDV. A (LL)group inoculated twice vaccinations on 2-weeks-interval during the late term of pregnant sows with PEDV live vaccine. The B (LKK) group was applied that one time single PEDV live vaccine at the pre-mate followed by the TGEV PEDV combined inactivated vaccine (twice vaccination on 2-weeks interval at the third-trimester). C (KK) group was applied to sow which inoculated twice vaccination on 2-weeks-interval during the late term of pregnant sows with by the TGEV, PEDV combined inactivated vaccine. As the result of SN test on sows in the pig farm before vaccination, antibody titers was showed 9/45 (20.0%). By comparison with the serum neutralizing antibody titers against PEDV of the vaccination programs after PEDV of the vaccination, A group and B group vaccination method was higher than those of C group in sows. In the piglets up to 2 weeks of age, A group was showed antibody titers of 17/22 (81.8%) that showed 2-128, and B group was showed antibody titers of 30/37 (81.1%) that showed 2-512, and C group was showed antibody titers of 14/28 (50.0%) that showed 2-32. On the other hand, PEDV antibody titers were tested for the survey. As the results of SN test, Aujeszky's disease survey in 54 pig farms from november 2005 to august 2006, antibody titers of 47/286 (16.4%) showed above 2. Five breeding farms were antibody titers of 38/77 (49.4%), Wanggung zone farms antibody titers of 59/85 (69.4%). In pigs farms vaccinated the first of twice PEDV live vaccine, and after 6 month, the second of twice TGEV PEDV combined inactivated vaccine (LLKK, 256-1024 titer) method was higher than those of vaccinated twice the early term of pregnant, and twice the late term of pregnant sows of PEDV live vaccine (LLLL, 32 titer).

Sero-epidemiology of transmissible gastroenteritis of pigs in Gyeongbuk province (경북지방 돼지의 전염성 위장염에 대한 혈청학적 역학조사)

  • Swine sera collected from January to December 2000 were tested for the survey on transmissible gastroenteritis(TGE) infection in Gyeongbuk province. Serum neutralization(SN) test was peformed in finishing pigs of 50 pigs industry without clinical signs of TGE and sows, gilts and growing pigs of four pigs industry with TGE. As the results of SN test in fifty industry, antibody titers of 90.6%(474/523 samples) showed below 4. Antibody titers of them showed highly in two pigs industry of western region and three pigs industry of southern region of Gyeongbuk province. The results indicated that TGE had been occurred before in five pigs industry. Most of antibody titers showed of highly in four pigs industry having had TGE. There were no significant differences of the antibody titers of TGE according to age when the survey was made. The above results indicated that the pigs of Gyeongbuk province were not almost exposed in porcine respiratory coronavirus(PRCV) by present.

Relationship of Antibodies in Egg Yolk and Serum against Major Antigens of Bacterial Agents in Porcine Atrophic Rhinitis, Pneumonic Pasteurellosis and Pleuropneumonia (돼지 위축성 비염, 파스튜렐라성 폐렴 및 흉막폐렴 원인균의 주요 항원에 대한 IgG 와 IgY 의 상관 관계 분석)

  • Swine respiratory diseases have induced severe economic lasses in swine industry worldwide. Therefore, several methods have been made and applied to prevent and control the diseases. However, these methods still have a problem and also induce side effects. Recently, the use of egg yolk antibody was introduced to control and prevent the diseases as one of new trials. As a study of using egg yolk antibody, antibody titers against several different antigens of major pathogens in swine respiratory diseases were compared in egg yolk and serum of hens immunized with those antigens. The titers were measured by ELISA using the antigens as coating antigens. The relationship in antibody titers between egg yolk and serum were identified by analysis of variance for linear regression. Almost of antigens used in this study showed the high relationship in antibody titers between egg yolk and serum (r = 0.87 ~ 0.93) even though the relationship in antibody titers against P. multocida A:3 IROMP was slightly low (r = 0.74)(P<0.01). These results indicated that antibody titer in egg yolk could be useful to predict the titer in serum of chicken.
