Antibacterial Effect of Fructose Laurate Synthesized by Candida antarctica B Lipase-Mediated Transesterification

  • Sugar esters are valuable compounds composed of various sugars and fatty acids that can be used as antibacterial agents and emulsifiers in toothpaste and canned foods. For example, fructose fatty acid esters suppress growth of Streptococcus mutans, a typical pathogenic bacterium causing dental caries. In this study, fructose laurate ester was chosen as a target material and was synthesized by a transesterification reaction using Candida antarctica lipase B. We performed a solvent screening experiment and found that a t-butanol/dimethyl sulfoxide mixture was the best solvent to dissolve fructose and methyl laurate. Fructose laurate was synthesized by transesterification of fructose (100 mM) with methyl laurate (30 mM) in t-butanol containing 20% dimethyl sulfoxide. The conversion yield was about 90%, which was calculated based on the quantity of methyl laurate using high-performance liquid chromatography. Fructose monolaurate (Mr 361) was detected in the reaction mixture by high-resolution mass spectrometry. The inhibitory effect of fructose laurate on growth of oral or food spoilage microorganisms, including S. mutans, Bacillus coagulans, and Geobacillus stearothermophilus, was evaluated.

  • In an attempt to regain function and aesthetics in the craniofacial region, different biomaterials, including titanium, hydroxyapatite, biodegradable polymers and composites, have been widely used as a result of the loss of craniofacial bone. Although these materials presented favorable success rates, osseointegration and antibacterial properties are often hard to achieve. Although bone-implant interactions are highly dependent on the implant's surface characteristics, infections following traumatic craniofacial injuries are common. As such, poor osseointegration and infections are two of the many causes of implant failure. Further, as increasingly complex dental repairs are attempted, the likelihood of infection in these implants has also been on the rise. For these reasons, the treatment of craniofacial bone defects and dental repairs for long-term success remains a challenge. Various approaches to reduce the rate of infection and improve osseointegration have been investigated. Furthermore, recent and planned tissue engineering developments are aimed at improving the implants' physical and biological properties by improving their surfaces in order to develop craniofacial bone substitutes that will restore, maintain and improve tissue function. In this review, the commonly used biomaterials for craniofacial bone restoration and dental repair, as well as surface modification techniques, antibacterial surfaces and coatings are discussed.

화학요법제 합성연구 II N,N'-disubstituted thiourea derivatives의 합성및 그 항균성과 항인결핵성 (Synthetic studies on chemothe-rapeutic agents (II) : synthesis and antibacterial and antitubercular activity of N,N'-disubstituted thiourea derivatives)

  • Fourteen novel N,N'-disubstitued thiourea derivatives were synthesized by Hugershof reaction. Antitubercular activities of ten compounds against Mycobacterium tuberculosis H$_{37}$ Rv were tested and was found tat 1-cyclohexyl-3-(4-benzenesulfonepiperidide)-2-thiourea was considerably active at 1mg/ml and 1-$\alpha$-naphthyl-3-(4-benzenesulfonepiperidide)-2-thiourea was slightly active at 1mg/ml. Antibacterial activity of the newly synthesized compounds against E. coli, Sta. aureus and Streptococcus hemolyticus were also tested. 1-cyclohexyl-3-(4-benzenesulfonepiperidide)-2-thiourea was makedly active against E. coli, Sta, aureus and Streptococcus hemolyticus; Phenol coefficients against E. coli, Sta, aureus and Streptococcus hemolyticus were 30,17.5 and 21.3, respectively. 1,1'-p-phenylene-3,3'bis[N-(2-thiazolyl)-sulfamylphenyl]-2,2'-dithiourea and 1-$\alpha$-naphthyl-3-(4-benzenesulfonepiperidide)-2-thiourea were considerably active against E. coli: phenol coefficients, 18.8 and 37.5 respectively. 1-(4-ethoxyphenyl)3-(4-sulfamylphenyl)-2-thiourea was active against Streptococcus hemolyticus: phenol coefficients, 22.5. 1-$\alpha$-naphthyl-3-(4-sulfamylphenyl)-2-thiourea, 1-$\betha$-naphthyl-3-[4-N-(2-thiazolyl)-sulfamylphenyl]-2-thiourea and 1-$\alpha$-naphthyl-3-(4-benzenesulfonepiperidide)-2-thiourea were active against Sta. aureus: phenol coefficients, 17.5, 20.0 and 18.8 respectively.

충치균에 대한 생리활성 생약성분의 분리 및 약효평가(2) -호장근의 항균성분과 안전성에 대하여- (The Isolation and Evaluation of Bioactive Components from Crude Drugs Against a Cariogenic Bacterium, Streptococcus mutans OMZ 176(2) -An Antibacterial Component of Polygoni Radix and Its Safety-)

  • The isolation and identification of an antibacterial component, from Polygoni Radix against a cariogenic bacterium Streptococcus mutans OMZ 176, were carried out for development of anticariogenic agents. The bioactive component was identified to be emodin. The minimal inhibitory concentration (MIC) of emodin was $100\;{\mu}g/ml$ against S. mutans OMZ 176. The bioactive component emodin weakly inhibited ${\beta}-lactamase$ activity with the inhibition ratio of 1.7, 4.3 and 7.6% at the concentration of 50, 100, and 200 uM, respectively. Emodin exhibited slight phototoxicity when analysed by the photohemolysis method.

Antimicrobial Studies of Stem of Different Berberis Species

  • Berberis is an important medicinal plant, of the family Berberidaceae. Different Berberis species and their parts are very common in herbal drug markets of India and world over as an adulterant/substitute to 'Daruharidra' i.e. B. aristata DC. Antimicrobial activity of 50% hydroalcoholic extracts of stem of four Berberis species viz. B. aristata DC., B. asiatica Roxb. ex DC., B. chitria Lindl. and B. lycium Royle and the isolated alkaloid berberine were tested against eleven bacterial and eight fungal strains. The extracts with the strongest antibacterial activity was obtained from B. lycium followed by B. aristata, B. asiatica and B. chitria. Based on these results it is possible to conclude that the hydroalcoholic extract and alkaloid (berberine) has stronger and broader spectrum against bacterial strains as compared to fungal strains. The result obtained in the present study authenticates and support the use of these plants in folklore medicine for treatment of various infectious diseases caused by the bacterial pathogens. However, an attempt has been made to explore the possibilities of utilizing stem part rather than roots of these species with the aim to conserve this species which is over exploited due to diverse use of its root. These findings will stimulate the search for novel, natural products as new antibacterial/antifungal agents which may be useful to pharmaceutical industries.

Phytochemical Screening, Isolation, Characterization of Bioactive and Biological Activity of Bungkang, (Syzygium polyanthum) Root-bark Essential Oil

  • Bungkang (Syzygium polyanthum) is a medium to tall plant which produces medicinal root-bark, the plant is normally found along inland river bank and produces small white flowers and fruits. Essential oils are among the most interesting components of the plant extracts consisting mostly of monoterpenoid or sesquiterpenoids. They are used as therapeutic agents in ethno, conventional, and complementary alternative medicines. Investigation and evaluation of the essential oil of Syzygium polyanthum as well as the antibacterial, antioxidant and antifungal activity was ascertained. The experiment was performed. 100 chemical constituents were obtained and two pure compound was isolated as Eugenol (1) and Farnesol (2). Significant growth inhibition of Staphylococcus aureus, (ATCCⓒ25923) Klebsiellia pneumonia (ATCCⓒ19155), Salmonella typhi (ATCCⓒ14028) and Escherichia coli (ATCC©25922) and the fungal strains Aspergillus flavin, Aspergillus niger, Candida, tropicalis, and Fusarium oxysporium was observed from the essential oil at concentration of 500 ㎍/mL. Antioxidant potential was observed to be strong of 18.42 ㎍/mL when compared to the control of 15.23 ㎍/mL. The result indicated that the oil obtained from root-bark of Syzygium polyanthum can be considered as an agent for antioxidant, antibacterial and antifungal in pharmaceutical food and cosmetic industries trails.

Functional Characterizations of Extruded White Ginseng Extracts

  • The antibacterial and antioxidant potentials of extruded ginseng extract (EGE) with 60% ethanol and methanol were investigated. The inhibitory activity of the EGE in Gram-positive bacteria was significantly higher than in Gram-negative bacteria. Higher antibacterial activity was observed with methanol ginseng extract when moisture content and barrel temperature were 20% and $115^{\circ}C$, respectively, that diameter of inhibition zone at 1,500 mg/mL was $15.40{\pm}0.13\;mm$ for Bacillus subtilis and $9.31{\pm}0.05\;mm$ for Salmonella typhimurium. The amount of total phenolics was highest in extruded ginseng at 20% moisture content and $115^{\circ}C$ barrel temperature. Especially, a positive correlation was observed between the total phenolic content and antioxidant activity of the extracts. In the 2,2-diphenyl-1-picryhydrazyl radical (DPPH) system, all tested extract of extruded ginseng at 20% moisture content exhibited very strong antioxidant properties when compared to red ginseng with percent scavenging effect of 23-35% at 20mg/mL. In conclusion, it can be said that the extracts of extruded ginseng could be used as natural antimicrobial and antioxidant agents in the food preservation.

The Wound Healing Potential of Lignosus rhinocerus and Other Ethno-myco Wound Healing Agents

  • Wound care has become increasingly important over the years. Various synthetic products for wound care treatment have been reported to cause toxic side effects and therefore natural products are in significant demand as they have minimal side effects. The presence of bioactive compounds in medicinal mushrooms contributes to various biological activities which assist in the early inflammatory phase, keratinocyte proliferation, and its migration enhancement which are pertinent to wound rehabilitation. Lignosus rhinocerus (tiger milk mushroom) can reduce the inflammation phase in wound healing by fighting off bacterial infection and modulating pro-inflammatory cytokines expression in the early stage to avoid prolonged inflammation and tissue damage. The antibacterial, immunomodulating, and anti-inflammatory activities exhibited by most macrofungi play a key role in enhancing wound healing. Several antibacterial and antifungal compounds sourced from traditional botanicals/-products may prevent further complications and reoccurrence of injury to a wounded site. Scientific studies are actively underway to ascertain the potential use of macrofungi as a wound healing agent.

자생 및 약용 식물의 Streptococcus mutans항균 활성의 검색 (Screening of Antibacterial Agent Against Streptococcus mutans from Natural and Medicinal Plants,)

  • 식용 및 약용 식물 215 종의 천연물로부터 추출부위를 달리한 메탄올 추출물 309 종을 조제하였으며, 안전성이 우수한 천연물 충치예방 및 치료제 개발을 목표로 309종의 추출물의 S. mutans JC-2에 대한 항균 활성을 평가하였다. 항균활성은 96 well microtiter method에 의한 생육저해능 측정 및 disk paper method에 의한 생육저지환 크기를 측정하여 평가하였으며, 그 결과 가죽나무(가지), 곰딸기(열매), 독활(잎), 상수리나무(잎), 여뀌(전초), 작약(종자), 짚신나물(뿌리) 추출물에서 강한 활성을 확인하였다. 상기 7종 천연물의 최소 생육저지농도(MIC)를 측정하여 항균 활성을 비교한 결과, 여뀌, 상수리나무, 가죽나무 추출물이 $25\∼30{\mu}g/ml $의 농도에서 생육을 완전히 억제하였다 이러한 결과는 항균력이 우수한 천연물로부터 새로운 충치 예방 및 치료제 개발이 가능함을 제시한다

탄닌산을 함유한 친환경농자재(공시번호 2-4-064)의 식물병원세균 기내 억제효과 (In Vitro Screening of Tannic Acid-based Eco-friendly Farming Material (notice no. 2-4-064) against Plant Pathogenic Bacteria)

  • 지금까지 주요 식물병원세균에 대한 방제대책은 화학적 방제방법에 의존하여왔다. 그러나 항생제와 같은 화학적 방제방법은 저항성 균주의 출현으로 인해 효율성이 감소함에 따라 대체방법의 개발이 요구되어지고 있는 실정이다. 따라서 본 연구에서는 주요 7종(그램음성 - 무름병, 감귤 궤양병, 풋마름병, 가지검은마름병 및 과실썩음병; 그램 양성 - 토마토 궤양병 및 더뎅이병)의 식물병원세균에 대한 탄닌산 함유 친환경농자재(공시번호 2-4-064)의 억제효과를 고체배지 확산법과 액체배지 배양법으로 검정하였다. 고체배지 확산법에서는 가지검은마름병원세균을 제외한 6종의 병원세균에 대한 억제효과를 나타내었으며, 액체배지 검정에서는 액체배지법으로 실험가능한 6종의 병원세균의 생장을 억제하였다. 전자현미경을 통한 표면변화를 관찰한 결과, 모든 병원세균의 표면구조의 변화를 발생하여 생장저해 또는 세포사멸을 유도하는 현상이 관찰되었다.
