Preparation of Poly(N-vinyl-2-pyrrolidone) Microparticles Using Supercritical Anti-solvent (초임계 반용매법을 이용한 폴리비닐피롤리돈 미세입자의 제조)

  • Poly(N-vinyl-2-pyrrolidone) (PVP) has been used as biocompatible and biodegradable polymer in cosmetics, pharmaceuticals and electronics. Micro-particles of PVP were produced using an aerosol solvent extraction system (ASES). Dichloromethane (DCM) and supercritical carbon dioxide were used as solvent and antisolvent, respectively. The mean diameter of the obtained polymer particles ranged from 0.184 to $0.249\;{\mu}m$. The relationship between particle size and initial drop size was also considered.

Anti-Oxidative and Anti-Inflammatory Effects of the Solvent Fraction from Theobroma cacao L. Extract (카카오(Theobroma cacao L.) 용매 분획물의 항산화 및 항염증 효과)

  • Solvent extracts of Theobroma cacao L. (TCL) were investigated for anti-oxidative and anti-inflammatory effects in order to consider TCL as a functional ingredient for cosmetic products. TCL(A) extract was fractioned according to polarity with $CHCl_3$, EtOAc, n-BuOH, and water. Following TCL(A) fractionation, the electron-donating ability of the n-BuOH and EtOAc solvent fractions (each 100 ${\mu}g/ml$) was about 76.2% and 53.9%, respectively. The superoxide anion radical inhibitory effect of the n-BuOH and EtOAc solvent fractions (each 50 ${\mu}g/ml$) was about 76.09% and 51.4%, respectively. Results of lipid oxidation showed that $Fe^{2+}$ had a greater chelating effect than $Cu^{2+}$. The $Fe^{2+}$ chelating effect of the EtOAc solvent fraction (50 ${\mu}g/ml$) was about 64%. Hyaluronidase inhibition related to the anti-inflammatory effect was 53.0% with EtOAc at 100 ${\mu}g/ml$, while the lipoxygenase inhibitory effect was about 51.32% at 10 ${\mu}g/ml$. The anti-inflammatory activity in the EtOAc fraction inhibited the generation of nitric oxide. Results also showed that iNOS protein expression increased in RAW264.7 cells. In contrast, at 100 ${\mu}g/ml$ EtOAc, iNOS and COX-2 protein expression significantly decreased in LPS-stimulated RAW 264.7 cells.

Drowning-out Crystallization of Calcium Lactate for Crystal Size Control (결정입자 제어를 위한 젖산칼슘 용석결정화 기술)

  • In the present study, the drowning-out crystallization of L(+)-calcium lactate was investigated in order to develop the crystallization separation process. The crystallization of the calcium lactate was induced by injecting the alcoholic anti-solvent into the aqueous solution of calcium lactate and the control of the calcium lactate crystal size during the crystallization was primarily investigated under the consideration of the anti-solvent species, anti-solvent composition and agitation speed as the key operating factors. Alcohols of methanol, ethanol, n-propanol and i-propanol were used as the anti-solvent for the drowning-out crystallization. Prior to the crystallization experiment, the solubility of calcium lactate in the water-alcohol mixture was measured along with the variation of the alcohol species and composition, which was necessary to estimate the supersaturation level of the crystallization. By the drowning-out crystallization, the calcium lactate crystals of the fabric shape were obtained. Using the ethanol as the antisolvent, the fabric crystals close to the needle shape were produced. However, the hairy crystals were obtained by using the propanol as the anti-solvent. Due to such morphological features, the crystals was highly apt to form the aggregates. The aggregation of the crystals was intensified as increasing the alcohol fraction in the water-alcohol mixture. Meanwhile, the agitation caused the breakage of crystals, resulting in the decrease of the crystal size. Therefore, the crystal size in the crystallization was predominantly determined by the competition between the crystal aggregation and breakage.

Evaluation of Solvent Extraction on the Anti-Inflammatory Efficacy of Glycyrrhiza uralensis (감초의 추출용매별 항염증 효능 평가 연구)

  • Glycyrrhiza uralensis (Leguminosae) is a well-known herbal medicine that has long been valued as a demulcent to relieve inflammatory disorders. To compare the influence of different solvents on the anti-inflammatory efficacy of G. uralensis, we measured the inhibition of pro-inflammatory mediators such as NO, TNF-$\alpha$, and $PGE_2$ in lipopolysaccharide (LPS)-stimulated mouse macrophage RAW 264.7 cells by extracts produced using different solvents (water, methanol, ethanol, or n-hexane). The results showed that methanol was the most effective solvent for the inhibition of both NO and $PGE_2$ production in RAW 264.7 cells. However, there was no difference among the extracts for inhibition of TNF-$\alpha$. Further study must be performed for the analysis of correlation between the anti-inflammatory activity of extracts produced using different solvents and the content of major bioactive compounds in G. uralensis, such as glycyrrhizin and liquiritin. The present study suggests that methanol may be a more appropriate solvent of G. uralensis than other solvents (water, ethanol, and n-hexane) to yield the greatest anti-inflammatory activity for food additives and medicine.

The Comparison between Sabaek-san Water and 30% EtOH Extracts for Anti-inflammatory Effects (사백산 물 추출물과 30% EtOH 추출물의 항염증 효과 비교연구)

  • Sabaek-san has been used for the treatment of inflammatory diseases derived from the cold with high fever, cough, and lung dysfunction in Korea and China. There is no study for the comparison between different solvent extracts of Sabaek-san. We made two samples, one is Sabaek-san water extract (SBSW) and the other is Sabaek-san 30% EtOH extract (SBSE). Both extracts inhibited inducible nitric oxide synthase(iNOS) protein, reduced iNOS-derived nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated RAW264.7 cells. Also, they reduced tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) and interleukin-$1{\beta}$ (IL-$1{\beta}$) production. These anti-inflammatory effects caused by induction of heme oxygenase (HO)-1. HO-1 enzyme plays an important role of cellular anti-oxidant and anti-inflammatory systems. The induction of HO-1 is primarily regulated at the transcriptional level, and its induction by various inducers is related to the nuclear transcription factor-E2-related factor 2 (Nrf2). However, it is worth taking note that SBSE has more powerful anti-inflammatory effects than SBSW. In this study we suggest that different solvent extraction makes different therapeutic actions.

  • Phase behavior of the ternary systems of water-insoluble simvastatin drug, which is well known to be effective drugs for hypercholesterolemia therapy, in solvent mixtures of dichloromethane and supercritical carbon dioxide was investigated to present a guideline of establishing operating conditions in the particle formation of the drugs by a supercritical anti-solvent recrystallization process utilizing dichloromethane as a solvent and carbon dioxide as an anti-solvent. The solubilities of simvastatin in the mixtures of dichloromethane and carbon dioxide were determined as functions of temperature, pressure and solvent composition by measuring the cloud points of the ternary mixtures at various conditions using a high-pressure phase equilibrium apparatus equipped with a variable-volume view cell. The solubility of the drug increased as the dichloromethane composition in solution and the system pressure increases at a fixed temperature. A lower solubility of the drug was obtained at a higher temperature. The second half of this work is focused on the particle formation of the simvastatin drug by a supercritical anti-solvent recrystallization process in a cylindrical high-pressure vessel equipped with an impeller. Microparticles of the simvastatin drug were prepared as functions of pressure (8 MPa to 12 MPa), temperature (303.15 K, 313,15 K), feed flow rate of carbon dioxide, and stirring speed (up to 3000 rpm), in order to observe the effect of those process parameters on the size and shape of the drug microparticles recrystallized.

Anti-cariogenic Activities of Mushroom Extracted with Various Solvent Systems (용매 변화에 따른 버섯추출물의 항충치활성)

  • Various mushroom extracts were examined for their anti-Streptococcus mutans and anti-glucosyltransferase (GTase) activities. Mushrooms were extracted with chloroform, methanol, acetone, ethyl acetate, and distilled water. The chloroform, methanol, and ethyl acetate extracts showed higher antimicrobial activity than that of the others. The inhibitory effects of the chloroform, methanol, and ethyl acetate extracts of Lyopyllum ulmarium, which have high anti-microbial and GTase activities, on the growth of S. mutans were examined and the optimal ratio of solvents was also evaluated to investigate the influence of solvents using a simplex centroid design. The anti-cariogenic effects of the L. ulmarium extract on S. mutans was influenced by the solvent, and the optimum anti-cariogenic activity of the extract was obtained with the ratio of chloroform:methanol:ethyl acetate=1:1:1 (v/v/v). These results suggest that solvent selection is an important factor to extract anti-cariogenic materials effectively from mushrooms.

Anti-oxidative and Immunomodulating Activities of Solvent Extracts from Broccoli (Brassica oleracea) Sprouts (브로콜리 새싹 용매 추출물의 항산화 및 면역조절 활성)

  • In order to examine the functionality of broccoli sprout (Brassica oleracea, BS), solvent extracts were prepared and their anti-oxidative and immunomodulating activities were compared with those of broccoli (B). EtOH extracts (E) were potently higher than hot-water extracts (HW) in the antioxidant contents and radical scavenging activity. In particular, the total polyphenolic contents in addition to ABTS and DPPH radical scavenging activity were significantly higher in EtOH extract of broccoli sprout (BS-E; 9.15 mg GAE/g, 4.52 mg AEAC/g, and 1.14 mg AEAC/g) compared with that of broccoli (B-E; 7.83 mg GAE/g, 3.63 mg AEAC/g, and 0.97 mg/AEAC/g). Whereas, total flavonoid content was significantly higher in B-E (1.60 mg QE/g) than BS-E (1.43 mg QE/g). Anti-inflammatory activity was investigated using LPS-induced cell line model at a concentration of $10{\sim}100{\mu}g/mL$, in which all solvent extracts of both broccoli sprouts and broccoli were not toxic to RAW 264.7 cell lines. In anti-inflammatory activity of broccoli sprouts, EtOH extracts also showed significantly more potent activity than hot-water extracts in all sample concentrations tested. In addition, BS-E ($100{\mu}g/mL$) inhibited nitric oxide (NO) and IL-6 production to 60.9% and 68.9% compared with the LPS inflammation group (without extracts), whereas B-E inhibited 49.6% and 54.9%. On the other hand, in immunostimulating activity by splenocytes and macrophages, hot-water extract showed significantly higher activity than EtOH extract. Especially, BS-HW stimulated the splenocyte proliferation (1.2-fold against saline group) and IFN-${\gamma}$ production (264.39 pg/mL) at $100{\mu}g/mL$, and the production of IL-6 (1.33-fold), IL-12 (1.09-fold) and TNF-${\alpha}$ (1.49-fold) from macrophages was also significantly enhanced over broccoli. In conclusion, broccoli sprouts showed more potent anti-oxidative, anti-inflammatory and immunostimulating activity than broccoli, suggesting the possibility of using broccoli sprouts as functional food materials.

Evaluation of Anti-oxidative, Anti-thrombin, Anti-invasive and Pro-apoptotic Activities of Paeonia japonica (백작약(Paeonia japonica)의 항산화, 트롬빈 저해, 암전이 억제 및 암세포사멸 평가)

  • Paeonia japonica is a perennial flowering plant used in traditional medicine therapy. The purpose of this study was to investigate the effect of water extract and solvent fractions obtained from P. japonica on anti-oxidative, anti-thrombin, anti-invasive and pro-apoptotic activities in YD-10B cells, human oral squamous carcinoma cell line. Water fraction revealed the highest extraction yield at 11.44% (w/w). Anti-oxidative activity was the highest in ethyl acetate fraction (85.13%). In the thrombin inhibitory activity test, ethyl fraction was the highest, with a value of 87.54%. Release and activation of MMP-2/pro-MMP-2 ratio in thrombin-treated YD-10B cells were significantly inhibited in the ethyl acetate fraction. At a concentration of $120{\mu}g/m{\ell}$ water extract and solvent fractions of P. japonica inhibited cell proliferation in YD-10B cells except water fraction. Pro-apoptotic effect on human oral squamous carcinoma cell using the Bax/Bcl-2 ratio analysis was higher in water extract than other fractions. These findings suggest that the ethyl acetate fraction of P. japonica potentiates a promising antioxidant, anti-thrombin and anti-invasive agents.

Comparative Study of Extraction Solvents on the Anti-inflammatory Effects of Scutellaria baicalensis

  • Objectives: This study was performed to evaluate the influence of different extracting solvents (water, methanol, ethanol, or n-hexane) on the anti-inflammatory efficacy of Scutellaria baicalensis (Lamiaceae), which has been used widely as a traditional herbal medicine for its anti-inflammatory properties. Methods: The ability of each extract to inhibit the production of pro-inflammatory mediators such as NO, TNF-$\alpha$, and $PGE_2$ by lipopolysaccharide (LPS)-stimulated mouse macrophage RAW 264.7 cells was measured. Results: The results showed that extraction solvents (except n-hexane) for S. baicalensis showed significant inhibitory effects on NO, TNF-$\alpha$ and $PGE_2$ production. Especially, methanol was the solvent with the greatest activity against NO and $PGE_2$ production. However, there was no difference between the extracts for inhibitory activity of TNF-$\alpha$. Conclusion: The present study suggests that methanol is a superior extraction solvent than water, ethanol, or n-hexane for maintaining the anti-inflammatory effects of S. baicalensis.
