This paper presents the development of a tacho generator that is applicable to a DC motor for anti-aircraft weapon systems. In general, devices such as tacho generators and resolvers are used as feedback devices for controlling DC motors. A tacho generator with a wide operating temperature range was developed, which has robust characteristics against shock loads and vibrations according to the operational characteristics of anti-aircraft weapon systems. The target specifications were set based on the requirements of the tacho generator currently in operation. A rotor coupled to the shaft of the motor and a stator coupled to the housing of the motor were then designed and manufactured. The inductance was 31.0 mH, the terminal resistance was 147.7 ohms, and the rotational measurement factor was satisfactory under both normal operation and operating conditions after the maximum speed for the standard of 9.500 ± 0.475 V/krpm. In addition, the environmental suitability of the applied equipment was confirmed through the rate of change in unit temperature, and it was found that the temperature characteristics were all within 0.03 %/℃.
The aim of this work was to investigate the cellular responses and proteomic analysis of Bacillus cereus MH-2 exposed to EGCG. Strain MH-2 was isolated from commercial Ssamjang and has the hemolytic activity. Survival of the MH-2 strain with time in the presence of different concentrations of EGCG under sublethal conditions was monitored. The amount of alginate from MH-2 strain decreased depending on the increasing concentrations of EGCG and increased depending on the exposure time at any particular EGCG concentration. Analysis of SDS-PAGE and Western blot using anti-DnaK and anti-GroEL revealed that two stress shock proteins, 70 kDa DnaK and 60 kDa GroEL were found to decrease in proportion to the EGCG concentration in exponentially growing cultures. Scanning electron microscopic analysis demonstrated the presence of protrusions and fused rod forms on the cells treated with EGCG. 2-DE of soluble protein fractions from MH-2 cultures showed 20 protein spots changed by EGCG exposure. These proteins involved in enterotoxins (hemolysin BL lytic component L1 and hemolysin BL-binding protein), chaperons (DnaK and GroEL), cell defense (peptidase M4 family proteins), and various biosynthesis and energy metabolism were identified by peptide mass fingerprinting using MALDI-TOF. These results provide clues for understanding the mechanism of EGCG-induced stress and cytotoxicity on B. cereus MH-2.
The aim of this work was to investigate the synergically bactericidal effects and cellular responses of tea polyphenols (TPP) and imipenem on imipenem-resistant Pseudomonas aeruginosa. Imipenem-resistant Ps. aeruginosa was isolated from patient in hospital. The bactericidal effects of TPP and imipenem were evaluated on the basis of its minimum inhibitory concentrations (MIC). The combined use of TPP and imipenem resulted in 16-fold and 8-fold reductions in the MICs of imipenem for the imipenem-susceptible and imipenem-resistant Ps. aeruginosa, respectively. The bactericidal effects of the imipenem and TPP against the Ps. aeruginosa was evaluated using the time-kill assay. The synergetic effects of the combinations of TPP and imipenem against Ps. aeruginosa were confirmed. Western blot using anti-DnaK and anti-GroEL monoclonal antibodies was performed to investigate the expression of stress shock proteins (SSPs) in imipenem-susceptible and imipenem-resistant strains exposed to TPP. The amount of SSPs were induced as the exposure time increased and decreased. The molecular weights of DnaK and GroEL were 70 kDa and 60 kDa, respectively. SDS-PAGE with silver staining revealed that the amount of lipopolysaccharides (LPS) increased or decreased in the strain treated to different concentrations and exposing periods of TPP. Scanning electron microscopic analysis demonstrated the presence of umblicated and wrinkled surfaces for cells treated with TPP or imipenem.

The purpose of this study was to investigate the anti-inflammatory effects of extract from GuBoEum(GBE) on the peritoneal macrophage. To evaluate anti-inflammatory effects of GBE. I measured cytokines (interleukin-6; IL-6, interleukin-12; IL-12, tumor necrosis factor-$\alpha$; TNF-$\alpha$) and nitric oxide (NO) production in lipopolysacchride (LPS)-induced macrophages. Furthermore, I examined molecular mechanism using western blot and also LPS-induced endotoxin shock. Extract from GBE does not have any cytotoxic effect in the peritoneal macrophages. Extract from GBE reduced LPS-induced IL-6, TNF-$\alpha$, IL-12 and NO production in peritoneal macrophages. GBE inhibited the activation of extracelluar signal-regulated kinase (ERK), C-Jun $NH_2$-terminal kinase (JNK) but not of p38, degradation of $I{\kappa}B-{\alpha}$ in the LPS-stimulated peritoneal macrophages. GBE inhibited the production of TNF-$\alpha$, IL-6 and IL-12 in serum after LPS injection. These results suggest that GBE may inhibit the production of TNF-$\alpha$, IL-6, and IL-12 through inhibition of ERK and JNK activation, and that GBE may be beneficial oriental medicine for inflammatory diseases.
Background and Aim: Breast cancer is a major healthcare problem in women. There are many reports about up-regulation of Hsp27 in cancer tissues but less is known about the potential relationship between Hsp27 antibody levels and breast cancer complications. We here investigated concentrations of serum Hsp27 antigen and antibodies in subjects with and without breast cancer and assessed potential associations with two-year disease-free survival, histological grade and number of lymph nodes. Materials and Methods: Specifically, serum Hsp27 antigen and antibody levels from 97 patients with breast cancer, and 65 healthy controls were determined by enzyme-linkedimmunosorbent assays (ELISAs). Results: Serum Hsp27 and antibody levels were significantly (p<0.001) higher in patients with breast cancer compared to the control group, but no relationship were found with two-year disease free survival, histological grade or number of lymph nodes (p> 0.6, 0.2 and 0.9 respectively). Conclusions: Elevated levels of Hsp27 antibody occur with women with breast cancer but do not appear to be associated with the presence of disease clinical complications.
This study was undertaken to evaluate the general pharmacological properties of CJ-11828, an amlodipine adipate, in experimental animals and in vitro system. CJ-11828 had no effects on general behavior, motor coordination, writhing syndromes, pentetrazol-induced chemoshock and electric shock in mice at dose levels of 3,10, anti 30 mg/kg, po. But there were decrease of body temperature, prolongation of sleeping time, and inhibition of intestinal activity in mice treated with CJ-11828 at doses of 10 and 30 mg/kg, po. CJ-11828 decreased the blood pressure in coscuous fog at the dose level of 2mg/kg, po, but it was expected as a result of pharmacological activity of CJ-11828. Any effect on respiratory system was not observed in conscious rat at doses of 3,10, and 30 mg/kg, po. The slight decrease in spontaneous motor activity was observed in mice treated with CJ-11828 at high dose, 30 mg/kg. In vitro experiments, CJ-11828 had no effect on agonists-induced contraction of isolated guinea pig ileum at 0.1, 1, and 10 ${\mu}$M. Based on these results, it was concluded that CJ-11828 had no pharmacological effect ill these studies even up to the 36-fold anticipated clinical dose, 3 mg/kg.
Objective : Banhahubak-tang is indicated for globus hystericus, marked by a subjective sensation as if something stuffed in the throat, chest distress, cough or vomiting, greasy whitish, taut and smooth pulse. In this study, the effects of Banhahubak-Tang extract (BHTe) were tested for anti-stress action. Methods : BHTe was extracted by pure water using electronic extractor and then fed to ICR male mice ($20{\pm}2g$) orally with the dose of 100mg/kg/day for five days. Mice were exposed to sociopsychological stress by restraining and seeing foot shock stressed mice for one hour for five days. Results : BHTe administered group showed a tendency of decreasing of serum corticosterone secretion when compared with control group, and BHTe administration also significantly up-regulated noradrenaline secretions in the dorsal cortex of brain. Lipid peroxidation of the brain tissues of mice were tested by measuring malondialdehyde, but BHTe showed no significant change. The elevated plus-maze test was designed to detect the effect of anxiolytic drugs, and BHTe administered group showed a significant increase of latency time. Conclusions : These results suggest that BHTe can effectively rid the psychological stress and it's related diseases.
Ulmi radicis cortex is a herb medicine which has been used for the treatment of such allergic disease as urticaria, allergic rhinitis and athma. To assess the contribution of an aqueous extract of Ulmi radicis cortex(URC) in systemic anaphylaxis, we used compound 48/80 as a fatal anaphylaxis inducer in rats. URC inhibited anaphylactic shock 100% with a dose of 1.0 mg/g body weight (BW) 1 hr before injection of compound 48/80. URC significantly inhibited serum histamine levels induced by compound 48/80. URC (1.0 mg/g BW) also inhibited to 79.1% passive cutaneous anaphylaxis activated by anti-dinitrophenyl (DNP) IgE. URC dose-dependently inhibited the histamine release from the rat peritoneal mast cells (RPMC) by compound 48/80. Moreover, URC had a significant inhibitory effect on anti-DNP IgE-induced histamine release or tumor necrosis $factor-{\alpha}$ production from RPMC. The level of cAMP in RPMC, when URC was added, significantly increased compared with that of normal control. These results indicate that URC may possess strong antianaphylactic effect.
Safety pharmacological properties of CJ-11555, an anti-cirrhotic agent, were investigated in experimental animals and in vitro test system. CJ-11555 had no effects on normal body temperature in rats, motor coordination, chemoshock induced by pentetrazol, electric shock induced by electric shocker and writhing syndromes in mice at dose levels of 100, 300 and 1,000 mg/kg. CJ-11555 inhibited intestinal activity and prolonged hexobarbital-induced sleeping time in mice at the dose level of 1,000 mg/kg. CJ-11555 affected on general activity and behaviour tests in SD rats, such as lacrimation, ptosis, piloerection, decreased body tone, abnormal dispersion within the cage, diarrhoea, red colored faeces, slight hypothermia and decreased grooming, at the dose level of 1,000 mg/kg in rats. CJ-11555 was effected on cardiovascular and respiratory system in anesthetized beagle dogs, such as tachycardia, increase of mean blood pressure and decrease of PR interval, decrease of respiratory rate and minute volume, at dose levels of 10 and 30 mg/kg. However, these effects were also observed in vehicle treated anesthetized beagle dogs. In in vitro experiments, CJ-11555 inhibited agonists (histamine, acetyl-choline or $BaCl_2$) induced contraction of isolated guinea-pig at the concentration of 30$\times$$10^6$ M. CJ-11555 was weekly inhibited hERG channel current at concentrations of 10 and 30$\times$$10^6$ M, and $IC_{50}$ was estimated to be higher than 30${\times}$$10^6$M. Based on these results, it was concluded that CJ-11555 affected on cardiovascular and respiratory system, general activity and behaviour and hexobarbital-induced sleeping time at the dose level of 1,000 mg/kg and contraction of the smooth muscle and hERG channel current at the concentration of 30$\times$$10^6$ M.
The overproduction of nitric oxide (NO) by inducible nitric oxide synthase (iNOS) is known to be responsible for vasodilation and hypotension observed in septic shock and inflammation. Inhibitors of iNOS, thus, may be useful candidates for the treatment of inflammatory diseases accompanied by overproduction of NO. In the course of screening oriental anti-inflammatory herbs for the inhibitory activity of NO synthesis, a crude methanolic extract of Curcuma zedoaria exhibited significant activity. The activity-guided fractionation and repetitive chromatographic procedures with the EtOAc soluble fraction allowed us to isolate three active compounds. They were identified as 1,7-bis (4-hydroxyphenyl)-1,4,6-heptatrien-3-one (1), procurcumenol (2) and epiprocurcumenol (3) by spectral data analyses. Their concentrations for the 50% inhibition of NO production $(IC_{50})$ in lipopolysaccharide (LPS)-activated macrophages were 8, 75, 77 ${\mu}M$, respectively. Compound 1 showed the most potent inhibitory activity for NO production in LPS-activated macrophages, while the epimeric isomers, compound 2 and 3 showed weak and similar potency. Inhibition of NO synthesis by compound 1 was very weak when activated macrophages were treated with 1 after iNOS induction. In the immunoblot analysis, compound 1 suppressed the expression of iNOS in a dose-dependent manner. In summary, 1,7-bis (4-hydroxyphenyl)-1,4,6-heptatrien-3-one from Curcuma zedoaria inhibited NO production in LPS-activated macrophages through suppression of iNOS expression. These results imply that the traditional use of C. zedoaria rhizome as anti-inflammatory drug may be explained at least in part, by inhibition of NO production.
