PREPARATION OF MULTIFUNCTIONAL LOW MOLECULAR WEIGHT CHITOSAN AND ITS APPLICATION IN COSMETICS.

  • The aim of this study is to elucidate the anti-microbial activity and anti-oxidative activity of water-soluble chitosan with a molecular weight of 5,000-200,000. Water-soluble chitosans have demonstrated a regular anti-microbial activity on the tested strians by the paper disk method. In the MIC (Minimum Inhibitory Concentration) test, CC-01 (MW=5,000) with the lower MW showed the higher MIC value than the higher MW chitosan. The MW of chitosan increase, the MIC decreases. MICs of 4 chitosans(CC-02∼CC-05) against S. aureusTCC 65389, E coli ATCC 8739, p. aeruginosa, ATCC 9027 and C. albicans ,ATCC 10231 were 7.0-39.O$\mu\textrm{m}$, whereas MICs of chitosans against A. niger were over 2.OmM. Formula containing chitosan showed higher anti-microbial activities than the formula made with the chemical preservatives(Methylparaben 0.2% and Imidazolidinyl Urea 0.3%). Among 5 water-soluble chitosans, CC-03(MW=92,163) showed the most potent anti-oxidative activity (IC$\sub$50/ : 0.2mM). In conclusion, the water-soluble low molecular weight chitosan could be served as natural preservatives and antioxidant in cosmetics.

A PROMISING NEW ANTI-WRINKLE INGREDIENT : Pericarpium castaneae extracts

  • Pericarpium castaneae extracts have variously potent activities, such as anti-oxidative activity and free radical scavenging activity. in vivo and in vivo studies both indicate that pericarpium castaneae extracts acts as a flee radical scavenger ($IC_{50}$/: 7.6$\mu\textrm{g}$/ml) stronger than gallic acid($IC_{50}$/: 12.5$\mu\textrm{g}$/ml) and ellagic acid($IC_{50}$/: 15$\mu\textrm{g}$/ml) which could prevent cutaneous UV damages and skin aging. The extracts showed a good effect as a anti-oxidant ($IC_{50}$/: 50$\mu\textrm{g}$/ml). It was shown that the appearance of wrinkle in human skin was reduced by topical application of pericarpium castaneae extracts. And the treatment of human skin with the extracts increased the elasticity and moisture of the skin. We investigated the effect of tile pericarpium castaneae extracts on production of extracellular matrix using cultured A431 fibroblast cells. The results indicated that the extracts had no detectable effect on collagen synthesis, But synthesis of cell adhesion protein was increased by the extracts. The results suggest that increase of cell adhesion protein synthesis by pericarpium castaneae extracts has closely related to reduction of wrinkle in skin.

등수국 잎 추출물 유래 미백 및 항산화 활성 성분 (Whitening and Anti-oxidative Constituents from the Extracts of Hydrangea petiolaris Leaves)

  • 본 연구에서는 등수국 잎 추출물의 미백 및 항산화 활성을 확인하고 유효성분을 분리하여 화학구조를 동정하였다. B16F10 melanoma 세포를 이용한 미백 활성 실험 결과, n-hexane (Hex) 분획물이 세포독성 없는 농도에서 멜라닌 생성 및 세포 내 tyrosinase 효소의 활성을 억제시키고 있음을 확인하였다. 또한 Hex 분획물이 tyrosinase 및 TRP-2 단백질의 발현을 감소시켰다. 반면, ethyl acetate (EtOAc) 분획물은 DPPH 및 ABTS+ 라디칼 소거 활성이 우수하였고, H2O2로 유도된 세포 손상에 대한 세포보호 효과를 나타내었다. Hex 및 EtOAc 분획물의 활성 성분을 규명하기 위해 컬럼 크로마토그래피를 수행하여 8 개의 화합물을 분리하였다; ethyl linoleate (1), ethyl linolenate (2), 1-linoleoyl glycerol (3), 1-linolenoyl glycerol (4), epi-catechin (5), afzelin (6), quercitrin (7), hyperin (8). 분리된 화합물에 대한 항산화 활성 측정 결과, 화합물 5 - 8의 라디칼 소거 활성이 우수함을 확인하였다. 또한 HPLC 분석을 통해 주성분인 quercitrin (7)의 함량을 측정한 결과, 추출물에서 31.3 mg/g, EtOAc 분획물에서 169.8 mg/g이 함유되어 있는 것으로 확인되었다. 이상의 연구 결과를 바탕으로 등수국 잎은 미백 및 항산화 효과를 갖는 천연 화장품 소재로의 개발이 가능할 것이라 사료된다.

반응표면분석법을 이용한 천마의 최적증자조건 설정 (Optimal Steaming Condition of Gastrodia elate Blume (Chunma) using Response Surface Methodology (RSM))

  • 천마를 채취한 그대로 이용하는 데에는 생체보존에 따른 불편, 독성 그리고 자극성 등의 부작용으로 간단한 전처리 과정이 필요하다. 본 연구에서는 천마를 이용한 가공제품의 개발을 위한 전처리 과정 중 증자 조건별로 천마의 항산화적 특성을 반응 표면 분석법으로 모니터링하여 최적 증자 조건을 설정하였다. 총페놀성 화합물의 함량은 증자온도 $94.43^{\circ}C$, 증자시간 42.38min에서 최적이었고 전자 공여능은 증자온도 $101.67^{\circ}C$, 증자시간 41.09min에서 최적조건이었다. 항산화능 및 아질산염 소거능은 $100^{\circ}C$, 40min의 동일조건에서 최적으로 나타났다. 위 항산화적 특성의 증자조건을 종합하여 superimposed contour map으로 최적조건을 예측한 결과, 증자온도 96{\sim}107{\circ}C$, 증자시간 $28{\sim}46min$으로 나타났다. 예측된 임의의 증자조건 $100^{\circ}C$, 40min으로 실제 항산화 관련값을 측정한 결과 예측값의 $95.9%{\sim}115.6%$으로 유효성이 높게 나타났다.

청화탕(淸華湯)의 항산화 및 항염증 효능 (Effect of CHT in anti-oxidative and anti-inflammatory related factors)

  • To investigate the clinical aspects of CHT in atopic dermatitis (AD) treatments, the effect of CHT in anti-oxidative and anti-inflammatory cytokines were tested. 100% or higher cell viability was observed in all tested groups from 25 to 200 ${\mu}g/m{\ell}$ using Raw 264.7 cells. CHT showed dose-dependent DPPH scavenging activity, with more than 90% scavenging activities at 800 ${\mu}g/m{\ell}$ concentrations. CHT showed dose-dependent suppression activity of ROS production, especially at 200 ${\mu}g/m{\ell}$ of 37.5%. CHT decreased NO production activity, with significant decrease of 33.2% at 200 ${\mu}g/m{\ell}$. IL-6, MCP-1, TNF-${\alpha}$ production rate were decreased by approximately 25% when Raw 264.7 cells were treated with LPS and with CHT of 200 ${\mu}g/m{\ell}$. Also, IL-$1{\beta}$ production rate was decreased by 25% at 100 ${\mu}g/m{\ell}$. The results above indicate that CHT significantly reduces the effect of oxidative and inflammatory cytokines. The use of CHT in dermatitis can be widely suggested.

Anthocyanins Inhibits Oxidative Injury in Human Retinal Pigment Epithelial ARPE-19 Cells via Activating Heme Oxygenase-1

  • Anthocyanins belong to phenolic pigments and are known to have various pharmacological activities. This study aimed to investigate whether anthocyanins could inhibit hydrogen peroxide (H2O2)-induced oxidative damage in human retinal pigment epithelial ARPE-19 cells. Our results indicated that anthocyanins suppressed H2O2-induced genotoxicity, while inhibiting reactive oxygen species (ROS) production and preserving diminished glutathione. Anthocyanins also suppressed H2O2-induced apoptosis by reversing the Bcl-2/Bax ratio and inhibiting caspase-3 activation. Additionally, anthocyanins attenuated the release of cytochrome c into the cytosol, which was achieved by interfering with mitochondrial membrane disruption. Moreover, anthocyanins increased the expression of heme oxygenase-1 (HO-1) as well as its activity, which was correlated with the phosphorylation and nuclear translocation of nuclear factor-erythroid-2 related factor 2 (Nrf2). However, the cytoprotective and anti-apoptotic effects of anthocyanins were significantly attenuated by the HO-1 inhibitor, demonstrating that anthocyanins promoted Nrf2-induced HO-1 activity to prevent ARPE-19 cells from oxidative stress. Therefore, our findings suggest that anthocyanins, as Nrf2 activators, have potent ROS scavenging activity and may have the potential to protect ocular injury caused by oxidative stress.

Schisandrae Fructus ethanol extract attenuates particulate matter 2.5-induced inflammatory and oxidative responses by blocking the activation of the ROS-dependent NF-κB signaling pathway

  • BACKGROUND/OBJECTIVES: Schisandrae Fructus, the fruit of Schisandra chinensis Baill., has traditionally been used as a medicinal herb for the treatment of various diseases, and has proven its various pharmacological effects, including anti-inflammatory and antioxidant activities. In this study, we investigated the inhibitory effect of Schisandrae Fructus ethanol extract (SF) on inflammatory and oxidative stress in particulate matter 2.5 (PM2.5)-treated RAW 264.7 macrophages. MATERIALS/METHODS: To investigate the anti-inflammatory and antioxidant effects of SF in PM2.5-stimulated RAW 264.7 cells, the levels of pro-inflammatory mediator such as nitric oxide (NO) and prostaglandin E2 (PGE2), cytokines including interleukin (IL)-6 and IL-1β, and reactive oxygen species (ROS) were measured. To elucidate the mechanism underlying the effect of SF, the expression of genes involved in the generation of inflammatory factors was also investigated. We further evaluated the anti-inflammatory and antioxidant efficacy of SF against PM2.5 in the zebrafish model. RESULTS: The results indicated that SF treatment significantly inhibited the PM2.5-induced release of NO and PGE2, which was associated with decreased inducible NO synthase and cyclooxygenase-2 expression. SF also attenuated the PM2.5-induced expression of IL-6 and IL-1β, reducing their extracellular secretion. Moreover, SF suppressed the PM2.5-mediated translocation of nuclear factor-kappa B (NF-κB) from the cytosol into nuclei and the degradation of inhibitor IκB-α, indicating that SF exhibited anti-inflammatory effects by inhibiting the NF-κB signaling pathway. In addition, SF abolished PM2.5-induced generation of ROS, similar to the pretreatment of a ROS scavenger, but not by an inhibitor of NF-κB activity. Furthermore, SF showed strong protective effects against NO and ROS production in PM2.5-treated zebrafish larvae. CONCLUSIONS: Our findings suggest that SF exerts anti-inflammatory and antioxidant effects against PM2.5 through ROS-dependent down-regulating the NF-κB signaling pathway, and that SF can be a potential functional substance to prevent PM2.5-mediated inflammatory and oxidative damage.

The effect of Panax notoginseng saponins on oxidative stress induced by PCV2 infection in immune cells: in vitro and in vivo studies

  • Background: Panax notoginseng saponins (PNS) are bioactive substances extracted from P. notoginseng that are widely used to treat cardiovascular and cerebrovascular diseases and interstitial diseases. PNS have the functions of scavenging free radicals, anti-inflammation, improving blood supply for tissue and so on. Objectives: The aim of this study was to investigate the effects of PNS on the oxidative stress of immune cells induced by porcine circovirus 2 (PCV2) infection in vitro and in vivo. Methods: Using an oxidative stress model of PCV2 infection in a porcine lung cell line (3D4/2 cells) and mice, the levels of nitric oxide (NO), reactive oxygen species (ROS), total glutathione (T-GSH), reduced glutathione (GSH), and oxidized glutathione (GSSG) and the activities of xanthine oxidase (XOD), myeloperoxidase (MPO) and inducible nitric oxide synthetase (iNOS) were determined to evaluate the regulatory effects of PNS on oxidative stress. Results: PNS treatment significantly reduced the levels of NO and ROS, the content of GSSG and the activities of XOD, MPO, and iNOS (p < 0.05), while significantly increasing GSH and the ratio of GSH/GSSG in infected 3D4/2 cells (p < 0.05).Similarly, in the in vivo study, PNS treatment significantly decreased the level of ROS in spleen lymphocytes of infected mice (p < 0.05), increased the levels of GSH and T-GSH (p < 0.05), significantly decreased the GSSG level (p < 0.05), and decreased the activities of XOD, MPO, and iNOS. Conclusions: PNS could regulate the oxidative stress of immune cells induced by PCV2 infection in vitro and in vivo.

황기(黃芪)추출물의 항산화 효능과 에멀션 점탄성도에 미치는 영향에 관한 연구 (Study on Effects of Anti-oxidant and Viscoelastic on Emulsion by the Extract of Astragalus membranaceus)

  • Objectives : The Astragalus membranaceus has been used to diuretic, tonic, anti-viral, anti-oxidant activities in oriental herb medicine, and in each experiment, proved in this effects. Root of A. membranaceus includes according to flavonoid, saponin, polysaccharide, thus it has been studied for anti-oxidant, anti-inflammatory, anti-cancer activities. However, previous researches have been performed with being limited mainly to food industry. The present study was conducted anti-oxidative activity of A. membranaceus extracts. In addition, The objective of the present study was to determine the possibility for the application of emulsions in the development of A. membranaceus for cosmetics. Methods : In this study, aimed SOD like activity of A. membranaceus extract, effects of A. membranaceus extract on viscoelastic properties of emulsion was measured using rheometer. Results : In the results, 1, 3-butylene glycol extract of A. membranaceus efficiently reduce the active oxygen, SOD like activity of more than 40% decrement depend on concentration. And A. membranaceus extract increase the modulus of elasticity of emulsion, therefore A. membranaceus extract have not influence on stability loss. As such, the A. membranaceus extracts will remain stable when applied to emulsions or skin products. Conclusions : In consequence of this study, A. membranaceus extract can be effectively used in cosmetic emulsions when the relation between natural product extracts and formulation of cosmetics is elucidated. It is highly recommended that rheology be applied to determine the optimal extract concentration for cosmetic formulations.

홍화(紅花), 천궁(川芎), 황기 약침액(藥鍼液)의 뇌교세포주 보호 효과 (Protective effects of Pharmacopuncture Solutions made by Carthmi Flos, Cnidii Rhizoma and Astragali Radix on C6 glioma cells)

  • Objective : This study was carried out to investigate protective effects of Pharmacopuncture Solutions (PSs) made by Carthmi Flos (CF), Cnidii Rhizoma (CR) and Astragali Radix (AR) on C6 glioma cells Methods : We investigated the effects of PSs on proliferation rates and types of C6 cells, and also investigated the effects on LDH release. In addition, protective effects of PSs on oxidative stress induced by hydrogen peroxide and SOD-like activities were also investigated. Results : PSs made by CF, CR and AR did not show cytotoxicity in various concentrations. CF-PS and AR-PS elevated levels of proliferation rates significantly. Treatment with CF-PS lowered level of LDH release in C6 cells. In addition, CF-PS and CR-PS showed protective effects on cell death induced by hydrogen peroxide respectively. Finally, CF-PS group showed high level of SOD-like activity compared to that in CR-PS group. Conclusion : These results suggest that CF-PS can accelerate proliferation of neuroglial cells, and has protective action against oxidative stress, which was involved in anti-oxidative effects such as SODlike activities. In addition, CR has protective effects against oxidative stress, and AR can accelerate proliferation of neuroglial cells.
