5-aminoimidazole-4-carboxamide Riboside Induces Apoptosis Through AMP-activated Protein Kinase-independent and NADPH Oxidase-dependent Pathways

  • It is debatable whether AMP-activated protein kinase (AMPK) activation is involved in anti-apoptotic or pro-apoptotic signaling. AICAR treatment increases AMPK-${\alpha}1$ phosphorylation, decreases intracellular reactive oxygen species (ROS) levels, and significantly increases Annexin V-positive cells, DNA laddering, and caspase activity in human myeloid cell. AMPK activation is therefore implicated in apoptosis. However, AMPK-${\alpha}1$-knockdown THP-1 cells are more sensitive to apoptosis than control THP-1 cells are, suggesting that the apoptosis is AMPK-independent. Low doses of AICAR induce cell proliferation, whereas high doses of AICAR suppress cell proliferation. Moreover, these effects are significantly correlated with the downregulation of intracellular ROS, strongly suggesting that AICAR-induced apoptosis is critically associated with the inhibition of NADPH oxidase by AICAR. Collectively, our results demonstrate that in AICAR-induced apoptosis, intracellular ROS levels are far more relevant than AMPK activation.

Structure-Activity Relationships of Gagaminine and Its Derivatives on the Inhibition of Hepatic Aldehyde Oxidase Activity and Lipid Peroxidation

  • In order to determine the structure-activity relationships for antioxidative effects of gagaminine, a steroidal alkaloid isolated from the roots of Cynanchum wilfordi (Asclepiadaceae), two derivatives identified as sarcostin and penupogenin were prepared from gagaminine by hydrolysis and reduction. These compounds were evaluated for the inhibitory effects on the aidehyde oxidase activity and on lipid perbxidation in vitro. Furthermore, their effects were compared with those of gagaminine and the related compounds, cinnamic acid and nicotinic acid. The results of this study prove that the cinnamoyl group in the structure of gagaminine is critical in inhibition of the aldehyde oxidase activity while the nicotinoyl group may be necessary for anti-lipid peroxidation of the compound.

Cilostazol Attenuates 4-hydroxynonenal-enhanced CD36 Expression on Murine Macrophages via Inhibition of NADPH Oxidase-derived Reactive Oxygen Species Production

  • Although anti-atherogenic effects of cilostazol have been suggested, its effects on the expression of SR in macrophages are unclear. This study investigated the role of cilostazol on CD36 expression of murine macrophages enhanced by HNE, a byproduct of lipid peroxidation. The stimulation of macrophages with HNE led to an increased expression of CD36, which was significantly attenuated by NAC, an antioxidant. Moreover, the increased production of ROS by HNE was completely abolished by NADPH oxidase inhibitors, DPI and apocynin, as well as by the 5-LO inhibitor, MK886, but not by inhibitors for other oxidases. This suggested that NADPH-oxidase and 5-LO were major sources of ROS induced by HNE. In addition, HNE-enhanced expression of CD36 was reduced by these inhibitors, which indicated a role for NADPH oxidase and 5-LO on CD36 expression. In our present study, cilostazol was a significant inhibitor of ROS production, as well as CD36 expression induced by HNE. An increase in NADPH oxidase activity by HNE was significantly attenuated by cilostazol, however cilostazol had no effect on HNE-enhanced 5-LO activity. Together, these results suggest that cilostazol attenuates HNE-enhanced CD36 expression on murine macrophages thorough inhibition of NADPH oxidase-derived ROS generation.

St. John's wort(Hypericum perforatum L.)의 생리활성 효과 (Biological activity of St. John's wort (Hypericum perforatum L.))

  • St. John's wort를 성인병 예방을 위한 기능성 식품소재로 이용하기 위하여 추출물의 생리활성효과를 조사하였다. 추출물의 phenol 함량은 열수추출물이 $237.4{\pm}13.2\;{\mu}g/ml$였으며, 알콜추출물은 $246.0{\pm}10.5\;{\mu}g/ml$로 알콜추출물의 phenol 함량이 다소 높게 나타났다. 추출물의 항산화효과는 DPPH가 열수추출물과 알콜추출물이 각각 95.02%, 95.24%로 높게 나타났고, antioxidant protection factor는 알콜추출군이 PF 1.78정도로 지용성물질에 대한 항산화력도 높은 것으로 확인되었으나 열수추출물의 경우 지용성 물질에 대한 항산화력이 거의 없었다. Helicobacter pylori에 대한 추출물의 항균활성은 열수추출물의 경우 clear zone이 나타나지 않았으며 알콜추출물의 경우 $100\;{\mu}g/ml$$150\;{\mu}g/ml$의 농도에서 각각 11 mm와 13 mm의 clear zone이 관찰되어 저해활성이 높은 것으로 판단되었다. Angiotensin converting enzyme 저해효과는 열수추출군에서만 19.15%의 angiotensin converting enzyme에 대한 저해력을 나타내어 고혈압의 예방 효과는 비교적 낮았다. Xanthin oxidase에 대한 억제효과는 열수추출물이 84.79%, 알콜추출물이 100%의 완벽한 저해효과를 나타내어 xanthin oxidase에 대한 높은 저해를 관찰할 수 있었다. 이러한 결과로 St. John's wort는 Helicobacter pylori에 대한 항균제, 항산화제 및 anti-gout 소재로의 개발이 기대되었다.

울릉도 자생식물인 섬괴불나무(Lonicera morrowii A.Gray) 잎 추출물의 생리활성 (Physiological activities of leaf extract of Lonicera morrowii A.Gray, a plant native to Ulleungdo)

  • 본 연구에서는 섬괴불나무 잎으로부터 phenol성 화합물의 생리활성 탐색을 통하여 기능성 소재로 활용 가능성을 살펴보았다. 인체에 무해한 용매인 열수와 ethanol로 추출 했을 때 열수는 3.53 mg/g, 40% ethanol은 2.82 mg/g의 TPC를 나타내었으며, 추출물을 이용하여 항산화 활성, xanthine oxidase (XOase), angiotensin converting enzyme (ACE), α-glucosidase 억제효과 및 항균 활성을 검정하였다. 그 결과 섬괴불나무 잎의 열수와 ethanol 추출물 50-200 ㎍/mL TPC에서 농도의존적으로 DPPH 유리라디칼 소거활성 및 PF가 증가하는 경향을 보였고, 50 ㎍/mL의 저농도에서도 모두 매우 높은 활성을 나타내었다. XOase 저해효과에서는 200 ㎍/mL TPC에서 열수와 ethanol 추출물이 각각 76.35, 99.83%의 높은 저해효과 나타냈으며, ACE 저해효과 또한 200 ㎍/mL TPC에서 열수와 ethanol 추출물 각각 79.06, 87.14%의 저해효과를 확인하였다. α-Glucosidase 저해효과를 측정한 결과 200 ㎍/mL TPC에서 열수와 ethanol 추출물 각각 80.45, 63.58%의 저해효과를 보였다. 생육저해환으로 항균활성을 측정 결과 열수 추출물 200 ㎍/100 μL TPC에서 H. pylori, P. acne균에 대해 각각 11.5, 18.5 mm의 clear zone을 형성 하였고, ethanol 추출물 200 ㎍/100 μL 농도에서는 P. acne균에 대해 10 mm의 clear zone을 형성하였다. 따라서 본 연구를 통하여 섬괴불나무 잎 추출물의 항산화 활성, xanthine oxidase (XOase), angiotensin converting enzyme (ACE), α-glucosidase 억제효과 및 항균 활성이 있다고 판단되었다.

경옥고(瓊玉膏)의 성분 분석 및 추출물별 항산화(抗酸化) 효능 비교 (An Analysis of the Gyungokgo's Ingredients and a Comparison Study on Anti-oxidation Effects According to the Kinds of Extract)

  • Objectives : To estimate the value of the Gyungokgo as therapeutic agent preventing against aging with an analysis of the ingredients and the bio-activating effects by enzymologic methods. Methods : A quantitative analysis of general ingredients' of the Gyungokgo's extract was done first. The effects on electronic donating ability, SOD-like activity, nitric oxide inhibition, xanthine oxidase inhibition, whitening effect have been investigated in the physiological activity measurement of function experiment. Results : The contained hydrolyzed amino acid is Valine, Aspartic acid, Arginine, Isoleucine and the contained free amino acid is Arginine, Phenylalanine, Valine, Glycine. The derivative of free amino acid is Phosphoserine, Carnocine, ${\gammer}$-Aminoisobutyric acid. And the Gyungokgo contains 14 species of minerals, K>Na>Ca>Mg>Fe>Al>Mn. Then, to assure of the Gyungokgo's anti-oxidation, these following subjects -polyphenol, electronic donating ability, SOD-like activity, nitric oxide inhibition, xanthine oxidase inhibition, tyrosinase inhibation- are analyzed and show high activity especially the most in chloroform extracts, (every ingredients written by the order of high amount) Conclusions : The Gyungokgo contains many materials functioning as anti-oxidation, neurotransmitter, anti-fatigue and immune agent.

Thioacetamide로 유발한 간손상 모델에서 계혈등(鷄血藤)의 간보호 효과 (Protective Effect of Spatholobi Caulis in Thioacetamide induced Acute Liver Injury of Rat)

  • Objectives : This study was undertaken to investigate the hepatoprotective effect of Spatholobi Caulis water extract (SC) to thioacetamide (TAA)-induced acute liver injury (ALI) in rats. Methods : The rats were injected intraperitoneally with TAA (200 mg/kg body weight) and orally administered SC (100 or 200 mg/kg b.w.) daily for 3 days. Liver biomarkers were assessed by serum glutamic oxaloacetic transaminase (GOT), glutamic pyruvic transaminase (GPT), and ammonia levels. Malondialdehyde (MDA) was measured both serum and liver tissue. In addition, nicotinamide adenine dinucleotide phosphate (NADPH) oxidase, anti-oxidant, and inflammation-related proteins were investigated by western blot analysis. Histological examination further confirmed though hematoxylin and eosin stain. Results : The SC treatment reduced liver function markers like GOT and GPT and also remarkably decreased ammonia level. Moreover, the elevated MDA level in TAA-induced group was significantly reduced by SC treatment. NADPH oxidase expression associated with oxidative stress including NOX2, NOX4, and p47phox markedly inhibited by SC administration. SC treatment exerted anti-oxidant effect through the increase of anti-oxidant enzyme including superoxide dismutase (SOD), Catalase, and heme oxygenase-1 (HO-1). The protein expressions of inflammatory cytokines such as tumor necrosis factor-�� (TNF-��), IL-6, and IL-1�� induced by nuclear factor-kappa B (NF-��B) activation were modulated through blocking the phosphorylation of inhibitor of nuclear factor ��B�� (I��B)��. SC treatment also improved histological alterations. Conclusion : These findings suggested that SC administration may be a potential candidate for the prevention or treatment of ALI.

고추냉이 추출액의 항산화 효과 (Anti-oxidant Effect of Wasabia Japonica Extracts)

  • This study was carried out to investigate the anti-oxidant effect of Wasabia japonica(WJ). The anti-oxidant activities of various extracts from WJ were investigated. The water and ethanol extracts of WJ leaf were found to cause significant free radical scavenging effects on DPPH and nitric oxide(NO). Sprague-Dawley male rats were randomly assigned to one normal diet and three high cholesterol diet groups which contained 1%(w/w) cholesterol. After 4weeks cholesterol diet, the groups of high cholesterol diet were classified to control(normal diet only), WJR5(normal diet and 5% WJ root) and WJL5(normal diet and 5% WJ leaf). Liver xanthine oxidase(XO) activity in the rats of high cholesterol diet was decreased up to the levels of normal diet group according to WJ diet. The results indicated that WJ possesses antioxidant effects through free radical scavenging effects on DPPH, NO and the decreasing of XO activity. In view of these results, WJ is expected to be an effective material for the anti-oxidant.

감마선을 조사한 桃仁皮(도인피)의 항산화활성 (Anti-oxidative Activities Gamma Irradiated Shell of Persicae semen)

  • Objectives : This study were carried out to investigate the anti-oxidative activities of $\gamma$-SPE(gamma irradiated shell of Persicae semen extract with ethanol). Methods : Anti-oxidative was measured by DPPH free radical scavenging activities, nitric oxide radical scavenging activities, SOD-like activities, xanthine oxidase radical scavenging activities, nitrite scavenging activities and tyrosinase inhibition activities. Results : Effects of free radial scavenging was increased by irradiation. Tyrosinase inhibition activities was decreased by $\gamma$-SPE. Conclusions : Results suggested that $\gamma$-SPE have a potential as a new anti-oxidative material.

목질진흙버섯 Phellinus linteus HN1009K의 항산화 활성과 항염증효과 (Antioxidant and Anti-inflammatory Effects of Phellinus linteus HN1009K)

  • 최근 농가에서 재배하고 있는 상황버섯 종에 비하여 균사생장이 빠르고 자실체 발생과 생육이 빠른 P. linteus HN1009K (HK1009)가 개발되었다. 본 연구는 HK1009의 균사체와 자실체 에탄올 추출물의 항산화 및 항염증효과를 기존에 재배되고 있는 P. linteus (PL, Korea Sangwhang)과 P. baumii (PB, Jangsu Sanghwang)를 대조구로 하여 비교분석 하였다. HK1009는 균사체와 자실체 추출물 모두 PL과 PB에 비하여 항산화 활성이 높았으며 xanthine oxidase분석에서는 PL, HK1009, PB순으로 항산화 활성이 높게 나타났다. RAW264.7세포를 이용한 상황버섯 균사체 추출물 PB(EX), PL(EX), HK1009(EX)시료의 NO assay 결과는 PB, PL, HK1009 모두 농도 의존적으로 원액 $200{\mu}g/ml$ 에서는 $7{\sim}8{\mu}M$ 정도로 염증 억제 효과가 뛰어났다. RAW 264.7 세포에 대한 MTT assay 결과에서 PB(EX), PL(EX), HK1009(EX) 추출물의 원액처리구 에서도 세포의 생존능이 70% 이상을 유지하여 세포 독성이 없는 것으로 판단되었다.
