The present study describes the ameliorating effect of $Ca^{++}\;on\;Cd^{++}$ toxicity on the germination, early growth of mungbean seedlings, nitrogen assimilation enzyme. s-nitrate reductase (NR), nitrite reductase (NIR), anti-oxidant enzymes (POD, CAT and SOD) and on the accumulation of hydrogen peroxide and sulphydryls. $Cd^{++}$ inhibited seed germination and root and shoot length of seedlings. While NR activity was down- regulated, the activities of NIR, POD and SOD were up- regulated with $Cd^{++}$ treatment. $Cd^{++}$ treatment also increased the accumulation of sulphydryls and peroxides, which is reflective of increased thiol rich proteins and oxidative stress. $Ca^{++}$ reversed the toxic effects of $Cd^{++}$ on germination and on early growth of seedlings as well as on the enzyme activities, which were in turn differentially inhibited with a combined treatment with calcium specific chelator EGTA. The results indicate that the external application of $Ca^{++}$ may increase the tolerance capacity of plants to environmental pollutants by both up and down regulating metabolic activities. In order to find the antioxidative compounds, fractionation of the MeOH extract of the leaves of Crataegus pinnatifida guided by DPPH scavenging test furnished seven phenolic compounds, $quercetin-3-O-{\alpha}-L-rhamnopyranosyl(1{\rightarrow}6)-{\beta}-D-glucopyranoside$ (1), myricetin-3-O-rhamnose (2), $quercetin-3-O-{\alpha}-L-rhamnopyranosyl-(1{\rightarrow}6)-{\beta}-D-galctopyranoside$ (3), $quercetin-3-O-{\beta}-D-galactopyranoside$ (4), quercetin (5), $apigenin-8-C-{\beta}-L-rhamnopyranosyl-(1{\rightarrow}6)-{\beta}-D-glucopyranoside$ (2'-O-rhamnosylvitexin) (6) and (-)-epicatechin (7). All of isolated compounds showed the significant antioxidative effect on DPPH free radical scavenging test and TBARS assay.

Objectives : This study was to investigate the anti-oxidative and anti-inflammatory effect of Lonicera japonica water extracts (LE) on Ulcerative Colitis Induced by DSS (Dextran Sulfate Sodium) in Mice. Methods : Colitis was induced by DSS in Balb/c mice. The sample group was divided into three. The mice in control group were not inflammation-induced. The pathological group was composed of untreated colitis elicited mice. The experimental group was administered Lonicera japonica water extracts (LE) after colitis elicitation. The effects on ulcerative colitis were evaluated the anti-oxidant effect, inhibition of COX-2 mRNA expression, the morphological change of colonic mucosa, decrease effect of HSP 70 and COX-2 in mucosa. Results : The SOD ability of LE was dose-dependently increased and the LPS-induced COX-2 mRNA expression of LE was dose-dependently decreased. LE showed the protective effects on DSS-induced experimental colitis. LE inhibited shortening of colon length, the hemorrhagic erosion in colonic mucosa. LE also showed the decrease effect for HSP70 and COX-2 in mucosa. Conclusions : The current results demonstrate the clinical utility of LE in traditional medicine and indicate the possible treatments for ulcerative colitis from natural products. Further investigations for exact mechanisms will be needed.

This study was conducted to investigate the anti-hyperlipidemic and anti-oxidative activities of Ikgihwaetang-gamibang (IK). We observed the effect of IK on lipid factor, inflammation factor, body weight and organ weight, and liver after administering IK to hyperlipidemia mice triggered by high fat diet. Additionally, we used Raw 264.7 cells to investigate the changes in anti-oxidative activity and the effects of anti-inflammation. Administration of IK resulted in reduction of total cholesterol, LDL cholesterol, TG, and glucose in blood serum while a clear increase in HDL cholesterol. The production of IL-1β and IL-6, and ROS and NO also decreased. The results showed that IK has anti-hyperlipidemic and anti-oxidative effects, and suggested its potential use in prevention and treatment of hyperlipidemia and its associated diseases.
Objectives: The mushroom Ganoderma lucidum has been widely used as a traditional herbal medicine for many years. Although several studies have focused on the anti-oxidative activity of this mushroom, the molecular mechanisms underlying its activity have not yet been clearly established. The present study investigated the cytoprotective effect of ethanol extract of Ganoderma lucidum (EGL) against oxidative stress (hydrogen peroxide, $H_2O_2$) and elucidated the underlying mechanisms in a C2C12 myoblast cell line. Methods: Oxidative stress markers were determined by using the comet assay to measure reactive oxygen species (ROS) generation and deoxyribonucleic acid (DNA) damage. Cell viability and Western blotting analyses were employed to evaluate the cellular response to EGL and $H_2O_2$ in C2C12 cells. Transfection with nuclear factor erythroid 2-related factor 2 (Nrf2)-specific small interfering ribonucleic acid (siRNA) was conducted to understand the relationship between Nrf2 expression and $H_2O_2$-induced growth inhibition. Results: The results showed that EGL effectively inhibited $H_2O_2$-induced growth and the generation of ROS. EGL markedly suppressed $H_2O_2$-induced comet-like DNA formation and phosphorylation of histone H2AX at serine 139 ($p-{\gamma}H2AX$), a widely used marker of DNA damage, suggesting that EGL prevented $H_2O_2$-induced DNA damage. Furthermore, the EGL treatment effectively induced the expression of Nrf2, as well as heme oxygenase-1 (HO-1), with parallel phosphorylation and nuclear translocation of Nrf2 in the C2C12 myoblasts. However, zinc protoporphyrin IX, a HO-1 inhibitor, significantly abolished the protective effects of EGL against $H_2O_2$-induced accumulation of ROS and reduced cell growth. Notably, transient transfection with Nrf2-specific siRNA attenuated the cytoprotective effects and HO-1 induction by EGL, indicating that EGL induced the expression of HO-1 in an Nrf2-dependent manner. Conclusion: Collectively, these results demonstrate that EGL augments the cellular anti-oxidant defense capacity through activation of Nrf2/HO-1, thereby protecting C2C12 myoblasts from $H_2O_2$-induced oxidative cytotoxicity.

The crude ethanol extracts and their solvent-partitioned fractions derived from the leaf and twig of Viburnum dilatatum Thunb. were investigated for their 1,1-diphenyl-2-picrylhydrazyl (DPPH) scavenging efficacy. The results showed that the butanol-soluble fraction ($SC_{50}\;=\;110.30\;{\mu}g/mL$) exhibited higher anti-oxidant activity than the crude ethanol extract ($SC_{50}\;=\;117.03\;{\mu}g/mL$) in the DPPH assay model. Then, the effects of the same extract samples on the production of nitric oxide were examined in LPS-stimulated RAW264.7 cells. Although the hexane and methylene chloride-soluble fraction showed a weak anti-oxidant activity, they exhibited potent inhibitory activity of NO production above 50 % at a concentration of $10\;{\mu}g/mL$. The hexane-soluble fraction also showed the inhibitory effect on mRNA expression of pro-inflammatory mediators such an TNF-$\alpha$, IL-$1{\beta}$, IL-6, iNOS and COX-2 in LPS-stimulated RAW264.7. These results suggest that the solvent extracts of Viburnum dilatatum Thunb. could be used as an anti-irritation ingredient.
The herbal extract (YMT_02) is a modified herbal extracts from Yukmijihwangtang (YMJ) to promote memory-enhancing. The YMJ extracts has been widely used as an anti-aging herbal medicine for hundred years in Asian countries. The purpose of this study is to; 1) quantitatively evaluate the memory-enhancing effect of YMT_02 by hehavior task, 2) identify candidate genes responsible for enhancing memory by cDNA microarray and 3) assess the anti-oxidant effect of YMT_02 on PC12 cell. Memory retention abilities are addressed by passive avoidance task with Sprague-Dawley (SD) male rat. Before the training session, the rats are subdivided into four groups and administrated with YMT_02, Ginkgo biloba, Soya lecithin and normal saline for 10 days. The retention test was performed. 24 hours after the training session. The retention time of the YMT_02 group was significantly (p<0.05) delayed $({\sim}100%)$, whereas Ginkgo biloba and Soya lecithin treatment delayed 20% and 10% respectively. The hippocampi of YMT_02 and control group were dissected and mRNA was further purified. After synthesizing cDNA using oligo-dT primer, the cDNA were applied and mRNA was further purified. After synthesizing cDNA using oligo-dT primer, the cDNA were applied to Incyte rat GEMTM 2 cDNA microarray. The microarray results show that prealbumin(transthyretin), phosphotidy lethanolamine N-methyltransferase, and PEP-19 are expressed abundantly in the YMT_02 treated group. Especially, PEP-19 is a neuron-specific protein, which inhibits apoptotic processes in neuronal cell. On the other hand, transcripts of RAB15, glutamate receptor subunit 2 and CDK 108 are abundant in control group. Besides, neuronal genes involved in neuronal death or neurodegeneration such as neuronal-pentraxin and spectrin are abundantly expressed in control group. Additionally, the YMT_02 shows an anti oxidative effect in the PC12 cell. The list of differentially expressed genes may implicate further insight on the action and mechanism behind the memory-enhancing effect of herbal extracts YMT_02, for example, anti-apoptotic, anti-oxidative, and neuroprotective effects.

Quercetin is a natural flavonoid phytochemical that is extracted from various plants. Having an advantages due to its varied biological properties, such as anti-inflammatory, anti-viral, anti-oxidant, and anti-cancer effects, quercetin is used to treat many diseases. Recently, it has been reported that autophagy inhibition may play a key role in anti-cancer therapy. Therefore, in this study, we investigated the molecular mechanisms and anti-cancer effects of quercetin in human osteosarcoma cells via autophagy inhibition. We ascertained that quercetin inhibited cell proliferation and induced cell death, these process is demonstrated that apoptosis via the mitochondrial pathway and the caspase cascade. Quercetin also induced autophagy which was inhibited by 3-MA, autophagy inhibitor and the blockade of autophagy promoted the quercetin-induced apoptosis, confirming that autophagy is a pro-survival process. Thus, these findings demonstrate that quercetin is an effective anti-cancer agent, and the combination of quercetin and an autophagy inhibitor should enhance the effect of anti-cancer therapy.
In recent year, We are concerned about anti-aging, disease-protection, long-life, many methods are used in solving this problem. And Those are related with antioxidative ability. Recently, We heard that Cucurbita moschata Duch. has anti-hypertensive effect and good for health. So I let made a experiment for this result of the anti-oxidant effect of Cucurbita moschata Duch. used for 3 methods, those are DPPH radical scavenging activity, Nitric oxide(NO) radical scavenging activity and Bovine serum albumin(BSA). The results of this study were as follows: We measured level of DPPH radical scavenging activity. And we obtained results that the ability of DPPH radical's elimination was increased when concentration of Cucurbita moschata Duch. was to $20\;mg/m{\ell}$. We measured level of Nitric oxide(NO) radical scavenging activity. And we founded that the ability of NO radical's elimination was increased when concentration of Cucurbita moschata Duch. was to $10\;mg/m{\ell}$. When we inspected antioxidation with Bovine serum albumin(BSA). And we obtained that antioxidative ability was increased after$2.5\;mg/m{\ell}$ of Cucurbita moschata Duch.. So I guess that Cucurbita moschata Duch. has effect of antioxidative ability. Hereafter we need differential experimental methods for evidence of antioxidative effect on Cucurbita moschata Duch.

The frequency of obesity has risen dramatically in recent years but only few safe and effective drugs are currently available. In addition, obesity can induce type 2 diabetes (T2DM), hyperlipidemia and fatty liver disease. Recently, protective effect of purslane extract (PE) on obesity has been reported, but little is known about the role and mechanism of PE in obesity. This study aimed to evaluate the effect of PE on obesity and diabetes in obese mice. In addition, the effect of PE was compared with anti-obesity and diabetes drugs. High-fat diet (HFD)-induced obese mice were treated for 8 weeks with drugs as follows: PE, orlistat, metformin, voglibose or pioglitazone. While PE mixed with normal diet did not have any effects on BW in non-obese mice, PE mixed with HFD significantly reduced BW gain, insulin resistance, and glucose intolerance, without affecting food intake and appetite in obese mice. The effect was comparable to the effects of anti-obesity and diabetes drugs. Furthermore, PE significantly increased the activity of hepatocellular anti-oxidant enzymes, leading to protection of liver from oxidative stress in obese mice. These results suggest that PE treatment may be a useful tool for preventing obesity and complication of obesity.
