퀴노아를 첨가한 청국장의 발효기간에 따른 품질 변화 (Changes in Quality Characteristics of Cheonggukjang added with Quinoa during Fermentation Period)

  • In this study, the quality characteristics of cheonggukjang with addition of different quinoa were investigated. We evaluated the quality and sensory characteristics of cheonggukjang, including the pH, amino nitrogen, slime contents, color value, and total aerobic bacteria. Moreover, The anti-oxidant activities were measured as total polyphenol content, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, and superoxide dismutase (SOD) activity. The pH and amino nitrogen content were noted to significantly increase during the fermentation period in all the samples. The slime content of cheonggukjang added with quinoa increased with increased fermentation time, but cheonggukjang with addition of 20% of quinoa was decreased. The L value and b value decreased significantly with increased fermentation time, but the a value increased significantly. The microbial tests of cheonggukjang with addition of quinoa showed that the aerobic micro-organisms count was 7.45~9.10 Log cfu/g. Total polyphenol contents increased in all groups during the fermentation period, and activity increased with an increased percentage of added quinoa. The DPPH radical scavenging activity and SOD-like activity of cheonggukjang (with addition of quinoa) were also significantly higher than those of the control. The sensory quality of quinoa 10% cheonggukjang was stronger in flavor, and taste, and demonstrated a higher level of overall acceptability, when compared to the other groups.

큰잎모자반 뿌리 및 줄기 추출물의 항산화 효과 (Antioxidant Effect of Sargassum coreanum Root and Stem Extracts)

  • The present study was to investigate the antioxidant activity in ethanol and water extracts of root and stem of Sargassum coreanum. Antioxidant activities were evaluated by total polyphenol contents, DPPH radical scavenging activity, chelating effect, reducing power, and rancimat method. Total polyphenol contents of ethanol and water extracts were 32.79 mg/g and 15.55 mg/g, respectively. Ethanol extract showed higher DPPH radical scavenging activity than water extract and similar activity to BHT. Reducing power of extracts was increased in a concentration-dependent manner and ethanol extract had more reducing power than water extract. Ethanol and water extracts have little chelating effect at all concentrations. Antioxidant index (AI) of ethanol extract measured by Rancimat was higher than that of water extract, but their AI was lower than that of BHT. These results indicate that ethanol extract of S. coreanum root and stem has more potent antioxidant activity than water extract through DPPH radical scavenging and reducing power, and could potentially be used as a good source of natural antioxidants.

  • There was an increasing interest that herbal medicine and natural material extracts were proved processes of antioxidant, cosmeceutical activity and the other effects. The aim of this study was to assess the antioxidant of extraction of four kinds from Prunus armeniaca L., Reynoutria elliptica, Curcuma aromatica, Lithospermum erythrorhizon. RE (Reynoutria elliptica) and CA (Curcuma aromatica) have good electron donating ability. The water and ethanol extract of RE at a 100 ppm concentration showed over 70%, the water extract at 500 ppm concentration showed 83% and the ethanol extract at 100 ppm concentration showed 86% of CA. Xanthine oxidase inhibition activity of the water extract of LE (Lithospermum erythrorhizon) at a 1,000 ppm concentration showed over 44%, on the other hand, RE showed in all lowest effect and there was no inhibition activity of a couple more extracts. In the measurement of nitrite scavenging activity, all extracts showed highly scavenging activity. Especially the water and ethanol extract of RE showed over 99% at 500 ppm, also LE showed over 40% at 10 ppm concentration.

Caffeic Acid의 항산화 활성 및 Amyloid beta와 LPS에 의한 C6 Glial 세포의 산화적 스트레스 보호 효과 (Antioxidant Activity and Protective Effect of Caffeic Acid against Oxidative Stress Induced by Amyloid Beta and LPS in C6 Glial Cells)

  • This study was investigated the radical scavenging effect and the protective activity of caffeic acid (CA) against oxidative stress. CA showed strong 1,1-diphenyl-2-picrylhydrazyl radical (DPPH) and hydroxyl radical ( OH) scavenging activity, showing 42.00% and 87.22% at 5 μM concentration of DPPH and ·OH scavenging activity, respectively. Furthermore, we studied protective activity of CA from amyloid beta (A${\beta}$25-35) and lipopolysaccharide (LPS) induced neuronal cell damage and neuronal inflammation using C6 glial cells. The treatment of A${\beta}$25-35 to C6 glial cell showed declines in cell viability and high generation levels of reactive oxygen species (ROS). However, the treatment of CA increased cell viability. The treatment of 5 ${{\mu}M}$ CA led to the elevation of cell viability from 59.28% to 81.22%. In addition, the production of ROS decreased cellular levels of ROS by the treatment of CA. The treatment of LPS to C6 glial cells increased significant elevation of nitric oxide (NO) production, while CA decreased NO production significantly. The production of NO increased by the treatment of LPS to 131.08%, while CA at the concentration of 1 ${{\mu}M}$ declined the NO production to 104.86%. The present study indicated thatCA attenuated A${\beta}$25-35-induced neuronal oxidative stress and inflammation by LPS, suggesting as a promising agent for the neurodegenerative diseases.

Extremely Low Frequency Magnetic Field is an Environmental Stress Factor by Exerting Oxidative Stress

  • The previous study reported the biological effect of magnetic field exerted by acting on endocrine and anti-oxidant system. The present study aims to study whether ELF-MF (extremely low frequency magnetic field) affects the physiological endocrine systems such as thyroid and whether ELF-MF affects the defense system against oxidative stress when it alters the function of thyroid. Finally, we correlate the effects of MF on oxidative stress, and adrenal and thyroid with an environmental stress factor. We exposed sham or MF to rats for 5 or 25 days. After the exposure, we determined pain sensitivity, level of TSH, $T_3$ and free $T_4$ in plasma. We also assayed in whole brain, lipid peroxidation, the activity of enzymatic anti-oxidant defense including superoxide dismutase(SOD) and glutathione peroxidase (GPx), and non enzymatic defense such as reduced or oxidized glutathione contents. MF induced the hypersensitivity to thermal stimuli with the reduction of latency. $T_3$ and $T_4$ levels were also increased by the exposure of MF. In addition, we observed the rise of MDA level in rat brain by MF although the MF did not change superoxide dismutase and glutathione peroxidase activity. The effect of MF on both reduced and oxidized glutathione results in decrease in reduced or oxidized glutathione in whole brain. In every experiment, there was no significant difference in MF influence between short term (5 days) and long term (25 days) exposure. Taken together, MF exposure affects the thyroid hormonal control in brain. The elevated thyroid hormone acts on brain, leading to hyper-utilization of oxygen. This phenomenon may be correlated with oxidative stress resulting from MF exposure. In conclusion, we suggest that MF exposure may be an environmental stress by exerting oxidative stress.

아마인 및 아마인유의 유지자원으로서의 특성 (Characterization of flaxseed and flaxseed oil as edible oil resources)

  • 황색 아마인과 갈색 아마인의 일반성분 분석과 아미노산 조성, 지방산 조성, 토코페롤 함량, 총 폴리페놀 함량 및 전자 공여능을 측정하여 각 유지들의 특성을 비교하였다. 일반성분의 경우 수분과 탄수화물은 37% 정도를 차지하고 있으며 조지방은 35%내외로 대두의 20%보다는 높은 함량을 나타내었다. 아마노산 조성은 황색과 갈색 아마인의 차이는 없었으며 주요 구성 아미노산은 glutamine, glycine, arginine 등이었고, 지방산 조성의 경우는 두 아마인의 지방산 조성에 차이가 있으며 황색과 갈색 아마인의 linolenic acid와 oleic acid의 함량은 각각 56.60, 31.38 과 18.24, 39.16으로 나타나 갈색 아마인이 황색 아마인보단 산화 안정성이 우수할 것으로 생각된다. ${\gamma}$-토코페롤의 경우는 황색 아마인이 20.59 mg/100 g으로 갈색 아마인보단 높았지만 들깨의 30.10 mg/100 g에는 미치지 못했다. 총 폴리페놀은 황색 및 갈색 아마인이 각각 10.78, 29.88 mg/100 g으로 갈색 아마인이 높은 함량을 나타냈으며, 전자 공여능 또한 2,500 mg%의 농도에서 갈색 아마인이 77.81%으로 가장 좋은 활성을 나타냈다. 따라서, 황색 아마인과 비교하여 갈색 아마인이 유지의 산화안정성 및 저장성이 우수할 것으로 생각되며, 기존 참기름 및 들기름과 비교하여서도 영양학적인 측면과 산업적인 가치가 높다고 할 수 있다.

당뇨유발 흰쥐에 있어서 산화적 스트레스에 대한 함박잎새버섯의 효과 (Effects of Hanbag Mushroom(Grifola frondosa) on Oxidative Stress in Diabetic Rats)

  • 본 연구는 당뇨병으로 인한 산화적 스트레스에 대한 함박잎새버섯분말의 효과를 밝히기 위하여 SD계 흰쥐를 STZ로 당뇨를 유발하여 간 및 신장 조직에서 조사하였다. 또한 당뇨흰쥐에 함박잎새버섯분말 1-2% 첨가하여 6주간 식이하였다. 산화적 스트레스의 지표물질인 LPO를 비롯하여 유발원 XOD 활성도를 측정하였다. 또한 이에 따른 간조직 손상 확인을 위해 혈청 ALT와 AST 활성도를 측정하였다. 특히 함박잎새버섯분말의 항산화적 효능을 위해 이들 지표물질들과 더불어 항산화체계의 중요 요소인 GSH 농도와 GST 활성도를 당뇨군, 당뇨-잎새버섯분말투여군 그리고 정상군에서 측정하였다. 당뇨군은 정상군과 비교하여 LPO 농도를 비롯하여 XOD 활성도가 유의하게 높았다. 특히 이러한 결과로 추정되는 간 조직 손상이 정상군보다 유의하게 높은 ALT 및 AST 활성도가 혈청에서 확인되었다. 그러나 당뇨-잎새버섯분말투여군에서는 LPO 농도, XOD 활성도를 비롯하여 조직손상의 지표인 ALT 및 AST 활성도가 당뇨군보다 유의하게 감소하였다. 항산화물질인 GSH 농도는 당뇨군 및 당뇨-잎새버섯분말투여군 비교에서 유의한 차이가 없었으나 GST 활성도는 당뇨-잎새버섯분말투여군이 당뇨군보다 유의하게 높았다. 따라서 당뇨유발성 산화적 스트레스에 대한 잎새버섯분말의 효능은 GSH 농도 변화보다 GST 활성도를 증가시키고 또한 산화적 스트레스의 유발원인 XOD 활성도 감소의 유도를 통해 이루어지는 것으로 추정된다. 결론적으로 당뇨는 산화적 스트레스를 증가시키며 조직손상을 유발한다. 그러나 함박잎새버섯분말은 항산화물질 및 효소계의 활성도를 증가시켜 당뇨유발-산화적 스트레스 감소를 유도하여 조직 손상을 감소시키는 것이 확인되었다.

염수초 포제법에 따른 두충의 항산화 및 항염증 활성 변화 비교연구 (Salt-water Processing-dependent Change in Anti-oxidative and Anti-inflammatory Effects of Cortex Eucommiae)

  • Objectives The present study aimed to investigate the change in marker compounds, anti-oxidative and anti-inflammatory effects of salt-water processed Cortex Eucommiae. Methods To evaluate the influence of processing on anti-oxidant effect of Cortex Eucommiae, changes in total phenol, total flavonoid, 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging, 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) free radical scavenging, and ferric reducing antioxidant power (FRAP) between processed and raw Cortex Eucommiae were assessed. In addition, nitrite assay was conducted to determine the influence of processing on anti-inflammatory effect of Cortex Eucommiae. Cell viability was also examined as to elucidate whether processing affects cytotoxicity of Cortex Eucommiae. Finally, high-performance liquid chromatography (HPLC) analysis was conducted to monitor changes in pinoresinol diglucoside amount of processed and raw Cortex Eucommiae. Results Salt-water processed Cortex Eucommiae showed higher total phenol and flavonoid amount, compared to raw Cortex Eucommiae. Furthermore, anti-oxidative activity of processed Cortex Eucommiae was improved as discovered in DPPH, ABTS, and FRAP assays. Anti-inflammatory effect of Cortex Eucommiae was also enhanced following salt-water processing, as evidenced in nitrite assay. HPLC analysis found that the amount of pinoresinol diglucoside, widely known as the marker compound of Cortex Eucommiae, increases through salt-water processing. All experiments were performed with non-toxic concentration of Cortex Eucommiae; processing did not affect the cytotoxicity of Cortex Eucommiae up to the currently adopted concentration. Conclusions The present results support that salt-water processing of Cortex Eucommiae is beneficial in terms of marker compound amount, anti-oxidative, and anti-inflammatory activities. Additional investigations are needed to standardize the processing method of Cortex Eucommiae.

효소처리 및 가압추출 공정을 이용한 율피 추출물의 항산화, 피부 미백 및 주름개선 효과 (The anti-oxidant, whitening and anti-wrinkle effects of Castanea crenata inner shell extracts processed by enzyme treatment and pressurized extraction)

  • 본 연구에서는 율피 추출물을 이용하여 화장품 소재로서의 기능성을 조사하고자 효소종류별 및 가압추출 시간별에 따른 항산화 활성과 피부 미백 및 주름개선 효과를 실시하였다. 효소 종류별 및 가압추출 시간에 따른 율피 추출물의 추출수율은 12.42-29.80%로, C30m 추출물이 29.80%로 가장 높게 나타났다. 이화학적 특성으로 총 폴리페놀, 총 플라보노이드 및 총 탄닌 함량은 모두 C30m 추출물($1,000{\mu}g/mL$)에서 각각 22.69 g/100 g, 12.13 g/100 g 및 21.66 g/100 g으로 높은 함량을 나타내었으며, 총 당 함량은 H120m 추출물($1,000{\mu}g/mL$)에서 61.07 g/100 g으로 높은 함량을 나타내었다. 항산화 활성으로 DPPH radical 소거활성, ABTS radical 소거활성, superoxide radical 소거활성 및 FRAP 활성 모두 C30m 추출물($1,000{\mu}g/mL$)에서 각각 89.20%, 81.96%, 67.63% 및 $1,324.79{\mu}M$로 높은 항산화 활성을 나타내었으며, 모든 효소 처리구간에서 가압추출 시간이 증가함에 따라 항산화 활성이 감소하였다. 피부 미백 및 주름개선 효과를 확인하기 위해 tyrosinase 및 elastase 저해활성을 측정한 결과 C30m 추출물($1,000{\mu}g/mL$)에서 각각 61.32% 및 61.06%로 높은 저해활성을 나타내었으며, HE 추출물에서 가장 낮은 저해활성을 나타내었다. 이는 효소처리 및 가압추출에 의해 율피의 생리활성물질이 많이 용출된 것으로 판단되며, 율피의 새로운 추출공정 확립을 통해 항산화, 피부 미백 및 주름개선에 효과가 있는 항노화 기능성 소재로 화장품 제조용 원료 로 적합함을 확인하였다.

폴리감마글루탐산(PGA) 함유량이 증가된 청국장이 조골세포 분화에 미치는 영향 (Effects of Poly-Gamma Glutamate Contents Cheonggukjang on Osteoblast Differentiation)

  • Polyglutamic acid(PGA) 함유량이 증대된 청국장이 조골세포에 미치는 영향을 세포 수준에서 관찰하고자 하였다. 조골세포에 미치는 영향을 관찰하기 위하여 세포 생존율, 염기성 인산분해효소(alkaline phosphatase, ALP) 활성, 골 석회화 형성능, 조골세포 분화 관련 유전자 발현능을 측정하였다. 세포 독성이 없는 농도에서 염기성 인산분해효소 활성을 측정한 결과 농도에 의존적으로 ALP 활성이 증가하였으며, 일반 청국장 A보다 PGA 함유량이 증가한 청국장 B가 더 높은 ALP 활성 증가 효과를 나타내었다. 청국장의 골 석회화 형성능을 측정한 결과 역시 청국장 A보다 B가 더 높은 석회화 형성능을 나타내었다. 또한 분화 관련 유전자인 OCN, OPN, Col1, ALP 역시 청국장 B가 A보다 유전자 발현이 더 높게 나타났다. 이러한 실험 결과를 바탕으로 PGA가 증대된 청국장이 기존 청국장보다 조골세포의 활성에 효과적일 것으로 생각한다.
