진피로부터 정제한 Syringin의 항산화 및 항균 활성 (The Anti-oxidant and Anti-microbial Activities of Purified Syringin from Cortex Fraxini)

  • 본 연구에서는 물푸레나무 껍질인 진피로부터 Syringin을 효과적으로 분리, 정제하고 항산화 및 항균 활성을 평가하여 기능성 소재로써의 가능성을 탐구하고자 하였다. Syringin은 DPPH radical 소거활성 평가에서 50 ㎍/ml의 적은 농도로도 BHT보다 높은 활성을 나타내었으며 ABTS radical 소거활성 평가에서는 모든 농도에서 BHT의 활성과 비슷한 정도의 활성을 나타냈다. PF 측정에서는 Syringin의 농도가 증가함에 따라 활성이 다소 높아지는 듯했으나 증가폭이 크지 않았고 모든 농도에서 1.2 PF 이상의 활성이 나타나 항산화력이 있다고 판단하였다. TBARs 활성 측정에서는 모든 농도에서 BHT의 활성보다 낮지만 농도 의존적으로 항산화력이 증가하는 것을 확인할 수 있었다. 그람양성, 그람음성, 진균에 대한 생육 억제 활성 평가에서는 L. monocytogenes KCTC 13064, S. aureus KCTC 1916, E. coli KCTC 2571, H. pylori HPKCTC B0150의 네 균주에 대한 생육억제환의 크기가 Syringin의 농도에 의존적으로 증가하였고 C. albicans ATCC 10231에 대해서는 생육억제환이 관찰되지 않았다. 위의 결과를 종합한 결과 Syringin의 수용성 물질의 항산화 활성은 지용성 물질의 항산화 활성보다 우수하고 진균을 제외한 그람양성 및 그람음성 균주에 대한 성장을 억제함으로써 항균 활성을 나타낸다고 판단하였다. 본 연구를 토대로 Syringin의 항산화 및 항세균 기작에 대한 연구와 다른 생리활성 작용, 인체 적용성에 대한 연구를 추가로 진행한다면 천연 유래의 안전한 기능성 소재로 활용 가능할 것으로 사료된다.

Inhibition Effect Against Elastase, Collagenase, Hyaluronidase and Anti-oxidant Activity of Thinning Green Ball Apple

  • The thinning Green ball apple was extracted using water and ethanol and a phenolic concentration of thinning Green ball apple was $50-200{\mu}g/mL$. The water and ethanol extracts of thinning Green ball apple showed 94.69% and 92.24% 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity and 100.30% and 99.16% 2,2'-Azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging activity at phenolic concentration of $200{\mu}g/mL$, respectively. The water and ethanol extracts of thinning Green ball apple showed antioxidant protection factor of 1.76 antioxidant protection factor and 1.76 antioxidant protection factor, respectively. The water and ethanol extracts showed 101.46% and 99.64% anti-oxidative effect on thiobarbituric acid reactive substances at phenolic concentration of $200{\mu}g/mL$. Hence, the water and ethanol extracts of thinning Green ball apple can be considered a potential anti-oxidant. The water and ethanol extracts showed 33.28% and 32.14% hyaluronidase inhibition, respectively, at phenolic concentration of $150{\mu}g/mL$. The water and ethanol extracts showed 47.33% and 40.92% elastase inhibition and 46.19% and 65.58% collagenase inhibition at phenolic concentration of $200{\mu}g/mL$, respectively. About these experiments, thinning Green ball apple was found to exhibit anti-oxidation activity as well as hyaluronidase, elastase and collagenase inhibitory activities. Therefore, thinning Green ball apple can be considered a potential source for functional food.

효소 처리에 의한 흑미 호분 추출물의 산화방지와 항염증 활성 증진 (Improvement of anti-oxidant and anti-inflammatory activities of aleurone layer extracts of black rice (Oryza sativa L.) by enzyme treatment)

  • 본 연구에서는 흑미 호분을 실험소재로 선택하여, 흑미 호분에 효소(lipase, lecitase, lipopan)를 처리함으로써 산화방지와 항염증활성을 증진하고자 하였다. 항산화 활성을 확인해보고자 DPPH 라디칼 제거능과 ABTS 라디칼 제거능을 실시하였다. 그 결과 효소 처리군이 무 처리군에 비해 산화방지 활성이 증가됨을 확인할 수 있었고, 특히 라이페이스를 처리한 후 추출한 시료에서 산화방지활성 증진이 가장 효과적이었다. 이것은 총 안토사이아노 사이드 함량 측정 결과와 일치한 것을 확인하였다. 이러한 산화방지 활성의 증가에서 라이페이스의 작용이 흑미 호분 겉면의 지방 분해를 도와 산화방지능과 관련된 유효성분들을 효과적으로 추출한 것으로 생각된다. 항염증 활성을 비교하기 위해서는 효소처리한 흑미 호분 추출물(OLAE)과 LPS를 함께 처리하여 RAW 264.7 세포가 생산하는 NO의 양과 염증성 사이토카인의 분비량을 측정해 보았다. 효소 처리를 한 경우, 라이페이스와 lecitase를 연이어 처리한 경우의 시료에서 NO의 생성이 억제되고, 염증성 사이토카인($TNF-{\alpha}$, IL-6)의 분비량이 감소됨을 확인하였다. 항염증 활성의 증진은 두 가지의 효소를 처리하는 과정 중에 일어나는 유효성분의 변화 또는 성분의 전환이 되었기 때문이라 여겨지며, 이를 확인하는 후속연구가 필요하다고 생각된다. 이상의 결과는 부산물로 버려지는 흑미 호분을 효소 처리 과정을 이용하여 만든 흑미 호분층 추출물(OLAE)로 활용하면 유용하고 안정하고 효과적인 산화방지 및 항염증 기능성 식품 소재 개발로 가능함을 제시할 수 있다.

석류 추출물의 항산화 작용 (Anti-oxidant Activities of Punica granatum Extracts)

  • 강한 천연 생리활성물질인 폴리페놀과 탄닌을 다량 함유한 석류를 노화방지제 및 기능성화장품의 화장품 원료로 이용할 수 있는 가능성을 제시하고자, 과즙과 과피 및 종자유로 나누어 분리추출한 후, DPPH radical과 superoxide의 소거작용, 세포내 $H_2O_2$와 hydroperoxide 및 superoxide 생성 억제작용에 미치는 영향에 관하여 실험하여 항산화 작용을 비교 평가하였다. 석류과피 메탄올 추출액은 전반적으로 가장 유의성 있는 항산화 작용을 보였다. 자체적인 항산화 작용뿐만 아니라 세포내에서 free radical이 생성되는 것을 억제하는 작용도 있는 것으로 여겨지며, 석류과즙 농축액은 free radical을 소거하는 항산화 작용이 있으며, 또한 석류 종자유는 강한 hydroperoxide 생성 억제효과를 나타내어, 특히 hydroxy기를 가진 readical의 생성을 억제하는데 효과가 있는 것으로 사료된다. 이상과 같은 결과로 보아, 석류의 과즙보다는 과피의 추출물이 항산화 작용이 더 뛰어난 것을 알 수 있었으므로 피부미용에 관련된 건강보조식품 개발시 과피성분을 함유한 추출물을 원료로 사용하는 것이 효과적이라고 사료되며, 특히 피부 주름 방지제 등과 같은 기능성 화장품을 개발하고자 할 때는 석류 종자유를 화장품 원료로 하여 개발하는 것이 더 효과적이라고 사료된다.

한국산 및 Canada산 High Bush Blueberry 열매의 항산화 활성 비교 (Comparative Anti-oxidant Activity of Korean and Canadian High Bush Blueberry Fructus)

  • Blueberries (Ericaceae) are cultivated worldwide, and are used not only as foodstuff but also for relievement of eyestrain. Bluberry species representatively includes highbush blueberry (V. corymbosum L.), lowbush blueberry (V. angustifolium $A_{IT}$.), rabbiteye blueberry (V. ashei $R_{EADE}$), and bilberry blueberry (V. myrtillus L.). All of these species contain large amounts of phenolics and anthocyanins. In this regard, we isolated six compounds from Korea cultivated blueberry and identified as 3-O-caffeoylquinic acid (1), 5-O-caffeoylquinic acid (2), myricetin-3-O-${\beta}$-D-galactoside (3), quercetin-3-O-rutinoside (4), ethyl-3-O-caffeoylquinic acid ester (5), ethyl-5-O-caffeoyl quinic acid ester (6) by $^1H$-NMR, $^{13}C$-NMR and MS. Anti-oxidative activities of six compounds were verified by anti-oxidant assay such as DPPH, ABTS and Hypoxanthine/xanthine oxidase system. And then, anti-oxidant activities of Korea blueberry and Canadian were compared with each other. These results support that Korean blueberry has also the possibility to be potential supplementary material as healthy food like Canadian blueberry. Therefore, Korean blueberry can be used as a substitute of Canadian blueberry.

형방지황탕(荊防地黃湯) 전탕액이 노화쥐의 신장, 비장, 방광 세포의 항산화능에 미치는 영향 (Anti-Oxidant Effect of Hyeongbangjiwhang-tang Decoction in Kidney, Bladder and Spleen Cell of SD Rats)

  • 1. Objectives The purpose of this study is to investigate the anti-aging and anti-oxidant effects of Hyeongbangjiwhang-tang decoction(HGD) in SD rats. 2. Methods This experiment was used the tissue of kidney, bladder and spleen cells of 6, 52 and 68 weeks old SD rats. Each age group was again divided into three groups. One group, as normal group, was not-treated cells, another group, as control group, was saline-treated cells, and the last group, as experimental group, was HG-treated cells. After culture for 48 hours, each groups measured the level of SOD, GSH, MDA and NO in the tissue of kidney, bladder and spleen cells. 3. Results and Conclusions The activity of SOD were significantly increased in kidney cell of 68 w-HGD group, bladder cell of 52 w-HGD group and in spleen cell of 52,68 w-HGD group compared with those of the control groups. The level of GSH were significantly increased in bladder cell of 52w-HGD group and in spleen cell of 68w-HGD group compared with those of the control groups. The level of MDA were significantly decreased in bladder cell of 68 w-HGD group and in spleen cell of 68w-HGD group compared with those of the control groups. The level of NO were significantly decreased in kidney cell of 68 w-HGD group, bladder cell of 52, 68w-HGD groups, and in spleen cell of 68w-HGD group compared with those of the control groups. These results suggest that HGD(Hyeongbangjiwhang-tang) has anti-oxidant effects in aged rats.

Anti-oxidative and Anti-inflammatory Activities of Polysaccharide isolated from Korean-Style Soy Sauce

  • Soy sauce is one of the representatives of traditional fermented foods in Korea. However, studies on soy sauce are relatively insufficient in Korea compared to Japan. In this study, antioxidant and anti-inflammatory activities of polysaccharides were measured by polysaccharides isolated from two different soy sauces, Korean and Japanese (KSS-0 and JSS-0). KSS-0 was purified into two fractions using gel chromatography and named them as KSS-I and KSS-II. To investigate the antioxidant activity of the polysaccharides, we measured the polyphenol content and radical scavenging activity using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS). To investigate the anti-inflammatory activity of polysaccharides, we used RAW 264.7 macrophage cells and induced inflammation using lipopolysaccharide (LPS). Then, we measured levels of inflammatory mediators such as nitric oxide (NO) and tumor necrosis factor (TNF)-α. Among the four polysaccharides, KSS-II showed the highest antioxidant activity and had good anti-inflammatory activity; KSS-II decreased inflammatory mediators in a dose-dependent manner. In conclusion, the polysaccharide isolated from Korean soy sauce (KSS-II) showed better anti-oxidant and anti-inflammatory activities than polysaccharides isolated from Japanese soy sauce, and may be useful as substances for functional foods.

얌빈 추출물의 항산화 효능과 멜라닌 생성 억제효과 (Antioxidant Activity and Melanin Inhibitory Effects of Yambean (Pachyrhizus erosus) Extract)

  • Objectives : Yam bean (Pachyrhizus erosus) possess various nutrients, it has been widely used as traditional cosmetic material in Indonesia. The aim of this study was to investigate the anti-oxidant activity and the anti-melanogenic effect of Yambean (Pachyrhizus erosus) extract and its fractions. Methods : The anti-oxidant activity of yam bean extract assessed based on total polyphenol, flavonoid contents, DPPH and ABTS radical scavenging assay. To evaluate anti-melanogenic effects and cytotoxicity of Yambean extract and its fractions, B16F10 melanoma cell was used. Results : In results, total polyphenol content of yam bean water extract (YW) and Yambean 70% ethanol extract (YE) were $1.18{\pm}0.03mg/g$ (mg of gallic acid/g of sample), $1.16{\pm}0.01mg/g$. Total flavonoid contents of YW, YE were $3.55{\pm}0.06mg/g$ (mg of naringin/g of sample), $1.78{\pm}0.03mg/g$. Moreover, YE scavenged DPPH and ABTS effectively in $4mg/m{\ell}$ compared to YW. Cytotoxicity of YE and its fractions in B16F10 melanoma cell was measured using MTT assays. It had no cytotoxicity up to $500{\mu}g/m{\ell}$. Melanin accumulation in B16F10 melanoma cell was induced using alpha-melanocyte stimulating hormone (${\alpha}-MSH$) and 3-isobutyl-1-methylxanthine (IBMX). B16F10 melanoma cell treated with $10-500{\mu}g/m{\ell}$ YE and hexane, ethyl acetate, butanol, $H_2O$ fractions for 24h. Non treated B16F10 melanoma cell (Control) markedly increased melanin contents. In contrast, YE ethylacetate fraction effectively suppressed melanin accumulation in a dose-dependent manner. Conclusion : In conclusion, these results suggest that Yambean extract has the potential as a cosmetic material which possess anti-oxidant and anti-melanogenic activities.

Anti-oxidant and anti-inflammatory activities of the various kinds of herbal tea

  • Objectives : Reactive oxygen species (ROS) are involved in a wide spectrum of diseases including chronic inflammation and cancer. In this study, we investigated the antioxidant activities and anti-inflammatory effects of the extracts from the herbal teas such as Lonicera japonica Thunberg (L. japonica), Chrysanthemum morifolium Ramat (C. morifolium), Mentha arvensis L. (M. arvensis), and P.rhizoma. Methods : Anti-oxidant activity was evaluated using DPPH radical scavenging assay and $Fe^{2+}$ chelating assay. And DNA cleavage assay was performed to evaluate an anti-oxidative effect. Anti-inflammatory effect was performed using NO generation assay and western blot in LPS-stimulated RAW264.7 cell line. Results : L. japonica scavenged DPPH radical by 9.8% at 12.5 ${\mu}g/ml$, 24.8% at 25 ${\mu}g/ml$, 34.3% at 50 ${\mu}g/ml$, 61.1% at 100 ${\mu}g/ml$ and 75.8% at 200 ${\mu}g/ml$, respectively. In addition, C. morifolium and M. arvensis removed DPPH radical by 15.6% and 10.4% at 12.5 ${\mu}g/ml$, 34.8% and 22.8% at 25 ${\mu}g/ml$, 66.9% and 43.3% at 50 ${\mu}g/ml$, 87.4% and 69.1% at 100 ${\mu}g/ml$, and 92.1% and 73.2% at 200 ${\mu}g/ml$, respectively. However, P. rhizoma did not affect on DPPH radical scavenging. The $Fe^{2+}$ chelating activity was highest in L. japonica, but lowest in P. rhizoma among the herbal teas. In addition, the extracts from L. japonica, C. morifolium and M. arvensis inhibited oxidative DNA damage via its anti-oxidant activity. In anti-inflammatory effect, the extracts from C. morifolium inhibited NO production. In addition, it suppressed the $NF-{\kappa}B$ signaling pathway in LPS-stimulated RAW 264.7 cells. Conclusions : Together, this study indicates that L. japonica, M. arvensis and C. morifolium possess the protective effect against the oxidative DNA damage. Furthermore, C. morifolium exerts an anti-inflammatory effect.

비타민 나무 Seed 추출물의 항산화 활성 및 항염증에 관한 연구 (A Study on Anti-oxidant Activity and Anti-inflammatory Action of Sea Buckthorn Seed Extract)

  • This study aimed to observe the effects of Sea Buckthorn seed extracts (SEB) on anti-oxidation and antiinflammation. SEB were found to strongly depress ROS production depending on concentration, and 46.5% anti-oxidation effects were found in the top concentration of 100 ${\mu}g/mL$. Histamine and arachidonic acid release were measured to observe the effects of SEB on anti-inflammation. SEB inhibited histamine release and arachidonic acid release on dose dependent manner. Based on the results above, the conclusion is drawn that SEB have the potential to be used as natural materials that are effective for anti-oxidation and antiinflammation.
