Effect of Gleditsia sinensis Lam. Extract on Physico-Chemical Properties of Emulsion-Type Pork Sausages

  • This study was performed to investigate the effect of Gleditsia sinensis Lam. extract on the physicochemical properties of emulsion-type pork sausages during storage at $10^{\circ}C$ for 4 wk. Treatments were as follows: (C, control; T1, sodium ascorbate 0.05%; T2, Gleditsia sinensis Lam. 0.05%; T3, Gleditsia sinensis Lam. 0.1%; T4, Gleditsia sinensis Lam. 0.2%; T5, Gleditsia sinensis Lam. 0.1% + sodium ascorbate 0.05%). The values of pH, moisture content, lightness, redness, and sensory attributes were all significantly decreased, while the yellowness, chroma, hue angle, and texture properties were increased during storage with increase of the Gleditsia sinensis Lam. extract added. In addition, the antioxidant activity and antimicrobial activity in the sausages displayed significant increases (p<0.05). Therefore, although it was concluded that the addition of Gleditsia sinensis Lam. extract is not effective for improvement of the physical properties compared to chemical additives in sausages, it could be applied to meat products as a natural preservatives.

The Effect of Microbial Extracts on the Cell Activation and Inhibition Associated with Atopic Dermatitis

  • Atopic dermatitis (AD) is an inflammatory, relapsing, chronic skin disease and lesions in AD are frequently colonized with Staphylococcus aureus (S. aureus). Activation of T cells and IgE production by staphylococcal enterotoxins B (SEB) plays a crucial role in the pathogenesis of AD. Enterococcus faecalis (E. faecalis) is a nonpathogenic bacterium and produces the probiotic products that have been shown to have inhibitory effects on inflammatory responses. In present study, we carried out to assess the anti-inflammatory role of lyzed E. faecalis against the damaging effects of SEB on AD related immune responses. Furthermore, we attempted to determine whether the co-cultured lyzed E. faecalis can influence the colonization of S. aureus. As a result, we identified the effect of E. faecalis lysate as a potent therapeutic agent for atopic dermatitis (AD). E. faecalis lysate reduces the productions of total IgE and cytokines of AD-related immune cells in response to SEB stimulation. The proliferation of S. aureus was also inhibited by E. faecalis lysate. In conclusions, E. faecalis lysate may improve the skin-defense system disturbed by atopic condition, and may prevent subsequent secondary infection of S. aureus and development of AD.

Studies for Component Analysis and Antioxidant Effect, Antimicrobial Activity in Acanthopanax senticosus HARMS (가시오갈피 (Acanthopanax senticosus) 열매의 성분 분석과 항산화 효과 및 항균활성)

  • In the present studγ, we investigated the component analγsis, measurements of antioxidant activities and anti-microbial activities from fruit of Acanthopanax senticosus HARMS in order to detect the biological activities and develop novel functional resources. Different solvents extraction ratios were different according to different solvent extraction of Acanthopanax senticosus HARMS. Composition analysis we highly composed as 72.33%. The minerals of different organs were highly composed of potasium as 5951.3 mg/100g. The monosaccharldes are composed of the arabinose, xylose, mannose, galactose, glucose. Antioxidamt activity was measured in water extracts: $57.3\;{mu}/ml$, ethanol extracts $630.1\;{\mu}g/ml$, methanol extracts: $248.5\;{\mu}g/ml$, 71% ethanol extracts: $198.97\;{\mu}g/ml$, 75% methanol exracts: $96.77\;{\mu}g/ml$, chloroform extracts. $1194.83\;{\mu}g/ml$ at $IC_{50}$ value. The antimicrobial activities were observed in extracts from Acanthopanax senticosus HARMS against am negative bacteria and gram positive bacteria.

Effect of Reducing Agent, Sodium Hydrosulfite on the Natural Indigo Dyeing of Silk Fabric (환원제 Sodium Hydrosulfite를 이용한 천연쪽의 견 염색효과)

  • Effect of reducing agent sodium hydrosulfite on the natural indigo on the silk fabrics were examined to improve traditional dyeing method. K/S values of dyed fabrics was monitored with various dyeing temperature time, amount of dye and reducing agent. The dyeability of silk fabrics was improved by using reducing agent, sodium hydrosulfite; K/S value (7.20) was higher then that (1.09) of traditional method at the frist dyeing. Natural indigo dye extracted from dyed silk fabrics are composed of isomer, indigo (67.3%) and indirubin (32.5%). However, silk fabrics showed excellent anti-microbial activity regardless of the dyeing methods.

The Effect of Antibacteria and Antioxidantion Activities from Needle Leaf Tree (침엽수 추출물의 항균.항산화 효과)

  • We studied anti-microbial and anti-oxidant activities of bio-active components in stem of Juniperus chinensis L. and leaf of Abies koreana Wilson. Those plants of needle leaf tree family were reported to contain anti-cancer compounds. The anti-bacterial activity was tested by Broth dilution method against Escherichia coli and Staphylococcus aureus. As results, Juniperus chinensis L. and Abies koreana Wilson extracts showed 17.0% and 8.5% higher anti-bacterial activity than methyl paraben, respectively. The free radical scavenging activity of Juniperus chinensis L. and Abies koreana Wilson extracts showed 45 % and 44 % at 5,000 ppm. We measured polyphenol (catechin equivalent) and flavonoids quantity. The Juniperus chinensis L. extract contained 312 mg/g of polyphenol and 105 mg/g of flavonoids. The Abies koreana Wilson extract contained 280 mg/g of polyphenol and 103.8 mg/g of flavonoids. The cytotoxicity of extracts was measured by neutral red assay. Extracts did not affect the viability of CCK-986sk cells up to a concentration of 1,250 ppm. In conclusion, these data suggest that extracts of needle leaf trees would be usefull as antiseptic agents and anti-oxidants for cosmetic products.

Development of Microbial Inoculant Using By-product of Oriental Herbal Medicine (한약재박을 이용한 미생물제제의 개발)

  • The development of microbial inoculant was conducted using a by-product of oriental herbal medicine. The constituent of the by-product, which was high in organic matter, was 11.3% of crude protein, 5.1% of crude lipid, 49.7% of NDF (neutral detergent fiber), and 33.8% of ADF (acid detergent fiber). Microorganisms isolated from the by-product of oriental herbal medicine were 35 species. Among them, 6 bacterial species, 4 fungal species, 2 actnomycetes species, and 1 yeast species were effective in the utilization of the by-products. The 13 strains screened were tested for the plant growth-promoting effect in soybean seedling. BL-333 strain was found to increase the soybean yield by about 23% as compared with control. The strain BL-333 was identified as Paenibacillus marcerans. P. marcerans BL-333 showed high anti-fungal activities against virulent fungi, especially Fusarium sp. and Collectotrichum sp. Yields of plants which were inoculated with microbial inoculant prepared with P. marcerans BL-333 and by-product of oriental herbal medicine were found to be higher than control by $3{\sim}24%$. The yield was especially promoted in lettuce, radish, chinese cabbage and cucumber plants.

Anti-bacterial Effects of Aqueous Extract Purified from the Immature Cone of Red Pine (Pinus densiflora) (미성숙 솔방울 열수추출물의 항균성)

  • Red pine (Pinus densiflora) is widely used traditional medicine, pharmacological and nutritional values from which the phytochemical compounds are derived. The present study was aimed to examine the antibacterial effects in the absence and presence of a immature red pine cone extract against 13 microorganisms. The components in the aqueous extract from immature red pine cone were identified by GC-MS. About 1.4% of total polyphenolic compound was measured in aqueous extract collected from immature red pine cone. Also, the high concentration of ${\beta}$-phellenandrene, ${\alpha}$-pinene, limonene, bornyl acetate and aldehyde was detected in total ion chromatograms. Of total 13 microorganisms, 4 microorganisms including Pseudomonas aeruginosa, Vibrio cholera, Listeria monocytogenes, Klebsiella pneumonia were effectively killed by aqueous extract of immature red pine cone. The highest anti-bacterial effect was detected in P. aeruginosa, followed by V. cholera, L. monocytogenes and K. pneumonia. In case of P. aeruginosa, the largest diameter of inhibition zone was maintained to 1/2 solution treated cells and slightly decreased at 1/4 and 1/8 solution treated cells. Also, in test used V. cholera and L. monocytogenes, the inhibition zone was strongly formed in only 1 and 1/2 solution treated cells, while K. pneumonia showed the very small diameter of inhibition zone in all concentrations. Therefore, these results suggested that the aqueous extracts of immature red pine cone should be considered as a new and potentially important anti-bacterial substrate to effectively prevent the microbial infection and penetration.

Antioxidative Activities of Mixture of Schisandrae Fructus (SF) and Perilae Folium (PF) using Natural Color fixation technic (천연 색소 고정화 기술을 활용한 오미자와 자소엽 추출물 함유 음료의 항산화 활성 연구)

  • Objectives : This study focused on Schisandrae Fructus (SF) and Perilae Folium (PF), traditional medicine herbs and health functional food in Korea, Japan and China. We investigated various pharmacological activities that include a potential source of free radical scavenging, anti-viral, anti-microbial, anti-allergy and anti-inflammatory activities. Methods : We conducted an investigation of total contents of phenolic and flavonoid compounds in these single herbal extraction with/without combined to mixture. We also measured antioxidant activities such as DPPH free radical scavenging, SOD-like scavenging, nitrite scavenging and hydroxyl radical scavenging activities, xanthine oxidase inhibition, linoleic acid peroxidation inhibition, and reducing power. Results : As the results, contents of total phenoilc compounds and flavonoids were higher in those of PF than those of SF. Those of SF+PF mixture showed the synergy effects compared with those of SF and PF single extractions. Activities of DPPH free radical and SOD-like scavenging in 1 mg/mL concentration increased in dose dependent manners. That of SF increased compared with that of PF. That of SF and PF mixture also increased compared with that of BHA as a positive control. The other antioxident activities also showed similar to patten of activity of DPPH free radical scavenging. When combined to SF and PF extractions, there was showed synergic effect compared with those of BHA, excepted activities of xanthine oxidase inhibition and reducing power. Taken together, SF and PF have high phenolic and flavonoid compounds content furthermore, antioxidant activities in SF and PF mixture showed more synergy effect compared with those of BHA. Conclusions : Therefore, these findings suggest that SF and PF mixture may offer functional materials potential for development of functional beverage. But further studies are needed for the identification of the active compounds.

Lactococcus lactis Culture Methods for the Enhanced Depression of Inducers in Atopic Diseases (아토피유발인자 억제효과를 증대하는 Lactococcus lactis의 배양방법)

  • We conducted a screening and checked the cultivation methods of lactic acid bacteria, which have anti-atopic dermatitis functions, by determining the lactic acid bacteria's immune enhancement by FACS, and antimicrobial activity against Staphylococcus aureus. The increase of Tcell CD4+/CD25+/foxp3+ was bigger in Lactobacillus plantarum than Lactococcus lactis subsp. lactis (Lc. lactis) and the antimicrobacterial activity against S. aureus was the opposite. The antimicrobial activity of Lb. plantarum culture with medium containing Lc. lactis culture broth was not enhanced, but the antimicrobial activity of Lc. lactis cultured in a medium containing Lb. plantarum culture broth was enhanced. As the optimal method caltivation of Lc. lactis in a medium containing 10% of heat-killed Lb. plantarum culture broth was chosen. By this method, the antibacterial activity of the pure Lc. lactis culture increased sharply at the end of the log phase, while a restraint effect on the growth of S. aureus increased 1.29 times.

Effects of Bovine α-Lactalbumin Added with Oleic Acid and Microbial Transglutaminase on Cancer Cell Apoptosis (올레산과 미생물 유래 트랜스글루타미나제를 첨가한 우유 알파-락트알부민의 암세포사멸 효과)

  • In order to investigate the biological effects of conformational changes in the folding state of bovine ${\alpha}$-lactalbumin (${\alpha}$-La), the protein was prepared and classified as apo form, microbial transglutaminase (MTGase) added form, or bovine ${\alpha}$-La made lethal to tumor cell (BAMLET) form. Apo ${\alpha}$-La form showed weaker cancer cell inhibitory activity (apoptosis) than native ${\alpha}$-La, which suggests that the metal ion-like $Ca^{2+}$ had a positive effect, whereas BAMLET form showed strong cancer cell apoptotic activity. The BAMLET form seemed to be a molten globule structure that increased hydrophobicity. MTGase added to apo ${\alpha}$-La polymer showed similar anti-cancer activity as native ${\alpha}$-La, and it was well hydrolyzed by digestive enzymes. NMR results showed that BAMLET interacted with oleic acid and produced a complex.
