Transgenic Lettuce Expressing Chalcone Isomerase Gene of Chinese Cabbage Increased Levels of Flavonoids and Polyphenols

  • Flavonoid are large group of the polyphenolic compounds which are distinguished by an aromatic or phenolic ring structure and the phenolic compounds are induced by microbial infection, ultraviolet radiation, temperature and chemical stress. They are known for their antioxidant activity, anti-allergic, anti-inflammatory, anti-microbial and anti-cancer activities. In this study, changes in flavonoid content were investigated using heterologous chalcone isomerase (CHI) expression system. Also, phenolic compounds level was measured to examine the relation between flavonoids and phenols contents. Explants of lettuce (Lactuca sativa L.) were transformed with Agrobacterium tumefaciens LBA 4404 strain containing pFLH-CHI (derived from pPZP2Ha3) vector constructed with CHI gene from Brassica rapa. The putative transgenic plants were confirmed by genomic DNA PCR analysis. Also the transcription levels of the gene were analyzed by semi-quantitative RT-PCR with gene specific primers. The total flavonoid contents were increased at $T_0$ and $T_1$ generations over 1.4 and 4.0 fold, respectively. Total phenol contents also increased at $T_1$ generation. These results indicate that CHI gene plays an important role to regulate the accumulation of flavonoids and its component changes.

Screening of Biological Activities of Grass Clippings from Turfgrass (잔디 예지물의 생리활성 탐색)

  • This study was carried out to investigate the biological activities of grass clippings from turfgrass including Zoysia japonica (TG-1), Zoysia matella (TG-2), Agrostis palustris (TG-3), and Poa pratensis (TG-4). The 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity was relatively higher in TG-1 and -2. Especially, TG-1 exerted the strongest hydroxyl radical scavenging effect, showing 90.40% at the concentration of 100 μg/ml. In addition, TG-1 inhibited the growth of Staphylococcus aureus, Escherichia coli and Helicobactor pylori. TG-1 also showed the highest inhibitory effect of AGS human gastric adenocarcinoma cell growth and nitric oxide production against lipopolysaccharide in RAW 264.7 macrophage cells. In conclusion, among the TG extracts, TG-1 has anti-oxidative, anti-microbial, anti-cancer and anti-inflammatory effect, indicating that TG-1 may be the potential source of functional food.

Fisetin Suppresses Macrophage-Mediated Inflammatory Responses by Blockade of Src and Syk

  • Flavonoids, such as fisetin (3,7,3',4'-tetrahydroxyflavone), are plant secondary metabolites. It has been reported that fisetin is able to perform numerous pharmacological roles including anti-inflammatory, anti-microbial, and anti-cancer activities; however, the exact anti-inflammatory mechanism of fisetin is not understood. In this study, the pharmacological action modes of fisetin in lipopolysaccharide (LPS)-stimulated macrophage-like cells were elucidated by using immunoblotting analysis, kinase assays, and an overexpression strategy. Fisetin diminished the release of nitric oxide (NO) and reduced the mRNA levels of inducible NO synthase (iNOS), tumor necrosis factor (TNF)-${\alpha}$, and cyclooxygenase (COX)-2 in LPS-stimulated RAW264.7 cells without displaying cytotoxicity. This compound also blocked the nuclear translocation of p65/nuclear factor (NF)-${\kappa}B$. In agreement, the upstream phosphorylation events for NF-${\kappa}B$ activation, composed of Src, Syk, and I${\kappa}B{\alpha}$, were also reduced by fisetin. The phospho-Src level, triggered by overexpression of wild-type Src, was also inhibited by fisetin. Therefore, these results strongly suggest that fisetin can be considered a bioactive immunomodulatory compound with anti-inflammatory properties through suppression of Src and Syk activities.

  • Water-insoluble chitosan with molecular weight of 2,000,000, 580,000, 80,000, and 40,000 and more than 90% of degree of deacetylation were prepared to test antibacterial activity of chitosan against a pathogenic bacteria, methicillin resistant Staphylococcus aureus (MRSA). As experimental method, the Shake Flask Method (SFM) and Modified Shake Flask Method (MSFM) were applicated. The anti-microbial activity of chitosan/acetic acid aqueous solution is consistent irrespective of Mw of chitosan. MIC value of SFM measurement was 0.2 ppm, and MIC value of modified SFM measurement was 25 ppm. But MIC value of chitosan/acetic add solution and chitosan treated cotton filter paper was equally 5 ppm. The antibacterial activities of chitosan were different in different test measurements employed. The antibacterial activities of chitosan/acetic acid solution and chitosan treated cotton filter paper were also different. Therefore, it needs to be pointed out that the test measurements of anti-microbial activity have some problems.

Antimicrobial Activity of Resveratrol Oligomers and Flavonoids from the Stems of Vitis coignetiae Pulliat and the Seeds of Perilla frutescens (L.) Britton (머루 줄기와 자소자로부터 분리한 Resveratrol 올리고머와 Flavonoid의 항균효과)

  • We studied the antimicrobial activities of five compounds isolated from the stems of Vitis coignetiae Pulliat and the seeds of Perilla frutescens (L.) Britton. Based on spectroscopic evidence, compounds 1 to 5 were characterized as resveratrol, $\varepsilon$-viniferin, ampelopsin E, apigenin, and luteolin, respectively. The antimicrobial activities against Gram-positive (Staphylococcus aureus) and -negative (Pseudomonas aeruginosa) bacteria and a fungus (Candida albicans) were investigated using the disc diffusion and broth dilution methods. C. albicans was not inhibited by the five compounds. Compounds 2 and 5 had significant anti-microbial activity against S. aureus, and the 50% inhibitory concentration ($IC_{50}$) of compound 2 against S. aureus was 7.2 ${\mu}M$. Compounds 4 and 5 significantly inhibited P. aeruginosa and the minimum inhibitory concentration (MIC) of compounds 2 and 5 was 0.07 and 2.0 ${\mu}M$, respectively. Compounds 2, 4, and 5 had strong anti-microbial activity against S. aureus and P. aeruginosa.

Anti-microbial and Anti-thrombosis Activities of Lees of Sweet Potato Soju (고구마 소주 주박의 항균 및 항혈전 활성)

  • Sweet potato soju (SPS), a form of traditional distilled alcoholic liquor in Korea, is manufactured by the distillation of fermented broth under normal pressure, thus providing it for a uniquely smooth taste infused with the flavor of sweet potato. After distillation, the lees of SPS is produced as by-product and discarded. In this study, the ethanol and hot water extracts of lees of SPS, and their subsequent organic solvent fractions using hexane, ethylacetate (EA), butanol, and water residue were prepared in an effort at the efficient re-use of the lees of SPS. The ethanol extraction yield was 1.36-fold higher than that of the hot water extraction, and the EA fraction revealed the highest total polyphenol content among the solvent fractions. The various extracts and solvent fractions did not demonstrate hemolytic activity at up to 0.5 mg/ml concentrations against human red blood cells. In the bioactivity assay, only the EA fraction displayed a broad spectrum of anti-microbial activity against different pathogenic and food spoilage bacteria, and demonstrated significant anti-coagulation activity by inhibitions of thrombin, prothrombin and blood coagulation factors. Furthermore, only the EA fraction from the hot water extract of the lees of SPS showed anti-platelet aggregation activity, which is comparable to aspirin (a commercially available drug). Our results suggest that the EA fraction of the hot water extract prepared from the lees of SPS has a high potential as a novel resource for anti-microbial and anti-thrombosis agents.

Physical and Chemical Composition, Anti-Microbial Activity and Cytotoxicity of Kiwifruit Skin (식품표시 관련 교육경험에 따른 중학생들의 식품표시에 대한 인식과 활용실태)

  • This study was conducted to examine the chemical composition of kiwifruit skin, and to est its anti-microbial activities and cytotoxicities, thus, exploring ways for the economic utilization of kiwifruit skin. Four varieties of kiwifruit were examined: Daeheung, Bidan, Haegeum No.1 and Hayward. Vitamin C content in the fruit skins of Bidan, Daeheung, Haegeum No.1 and Hayward were 72.44, 67.22, 62.51 and 61.44mg/100g, respectively. Total amino acids content in the fruit skins of Bidan, Haegeum No.1, Hayward and Daeheung ere 808.31, 706.02, 629.07 and 464.83mg/100g dry weight, respectively. K and Ca content ere $17.20-45.70{\mu}g/mL$ and $4.58-10.15{\mu}g/mL$. While, other inorganic matter contents were below $4.89{\mu}g/mL$. Anti-microbial activity of kiwifruit skin extracts, in terms of the diameter of inhibition zone when tested against five gram positive and three gram negative microbial trains (even in the concentration of 2,000mg/L), was less than 14.1mm. The hyperplasia inhibition of lung cancer cells by methanol extracts from Bidan and Haegeum No.1 using concentrations of 800mg/L were 27.7% and 14.5%, however, those from Daeheung and Hayward were below 3% Consequently, it will be useful to know that kiwifruit skin can be added to processed goods which demand for higher concentrations of vitamin C, amino acids, K and Ca.

Enhancement of the Cosmeceutical Activity by Nano-encapsulation of Thiamine Di-lauryl Sulfate (TDS) with antimicrobial efficacy (항균 효능이 있는 비타민 B1 유도체(Thiamine Dilauryl Sulfate:TDS)의 나노입자화를 통한 기능성 향장 활성 증진)

  • This study was to improve cosmetical activity of thiamine di-lauryl sulfate (TDS) by encapsulation of nanoparticle with lecithin. Results showed that most of the nanoparticles containing the TDS were well formed in round shape with below 150 ~ 200 nm diameter as well as they were fairly stable in various pH ranges by measuring zeta potentials. The nanoparticles of TDS resulted in 85% cell viability of human normal fibroblast cells (CCD-986sk) when added at the highest concentration (1.0 mg/mL). The nanoparticles of Acer mono sap showed highest free radical scavengering effect as 88.1% in adding sample (1.0 mg/mL), compared to TDS solution of non-encapsulation (81.6%). The nanoparticles of TDS reduced the expression of MMP-1 on UV-irradiated CCD-986sk cells down to as 41.4%. The TDS solution and nanoparticles showed significant anti-microbial activities agaionst the salmonella typhimurium and listeria monocytogenes at 5 and 6 days as compared with control. Anti-microbial activities of TDS nanoparticles were similar to positive control. These results indicated that TDS nanoparticles may be a source for functional cosmetic agents capable of improving cosmetical activity such as antioxidant, whitening, and anti-wrinkling effects and can be further developed as natural preservative in cosmetics.

Study of Anti-microbe Activity of Essential oil(Unshiu oil) purified from Citrus Unshiu S.Marcov (감귤에서 분리한 정유 성분의 항균활성 연구)

  • Volatile odor substance originating from drying and making dry peel of tangerine from the fruit skin were collected through modern equipment. The anti-microbial activity of the substance against various skin-residing bacteria including acne, dandruff, athelete's foot, and gingivitis inducing microorganisms were tested. Anti-microbial activity was observed in purified oil, where 87 to 92% was D-limonene. Against P. acnes, 103$cfu/m{\ell}$ of P. acnes were suppressed at 0.1% Unshiu oil, and the MIC was measured to be 0.3%. Against P. ovale, a dandruff inducing bacteria, 104$cfu/m{\ell}$were suppressed at 0.1% Unshiu oil, and the MIC was measured to be 0.1%. Against T. rubrum and T. Mentagrophytes, both of which are athelete's foot inducing microorganisms, 83% of T. Mentagrophytes and 99.9% of T. rubrum were suppressed at 0.1% Unshiu oil, and the MIC were 0.3% and 0.05% respectively. Against S. aureus, a skin infection inducing bacteria, 103$cfu/m{\ell}$ of the bacteria were suppressed at 0.1% Unshiu Oil. Against B. subtilis, a non-pathogenic sporulating bacteria, 104$cfu/m{\ell}$ of the bacteria were suppressed at 0.1% Unshiu Oil. Against C. albicans, found in mucous membranes, 104$cfu/m{\ell}$ of the bacteria were suppressed at 0.1% Unshiu Oil. Against Aspergillus niger, an otomycosis inducing microorganism, 99.9% were suppressed at 0.1% Unshiu Oil. The results above indicate that low concentration of purified oil extracted from tangerine had strong antimicrobial activities against bacteria and fungi residing on the skin, and that it may be developed into skin disorder treating products in the future.

Antimicrobial Activities Against Oral Microbes and Growth-inhibitory Effect on Oral Tumor Cell by Extract of Paeonia lactiflora (작약 추출물의 구강병원균에 대한 항균성 및 구강암 세포 증식 억제효과)

  • Paeonia lactiflora was stepwise extracted with hexane, chloroform, ethyl acetate, butanol and water. Anti-microbial activity of each extract was investigated. Methanol extract of P. lactiflora revealed anti-microbial activity against S. mutans, C. albicans, and S. aureus. Also, hexane fraction revealed anti-bacterial activity against S. mutans and ethyl acetate fraction acted as potent anti-microbial agent on C. albicans and S. aureus. The relative growth ratio(RGR) of hexane fraction of P. lactiflora against S. mutans were determined as 77.8% in concentration of 0.125 mg/ml, 98.46% in 0.25 mg/ml and 100% in 0.5 mg/ml. The ethyl acetate fraction of P. lactiflora revealed RGR against C. albicans as 52.5% in concentration of 0.125 mg/ml, 60.83% in 0.25 mg/ml and 78.33% in 0.5 mg/ml. It indicate that increasing concentration increase RGR. The measured minimal inhibitory concentration(MIC) of hexane fraction on S. mutans KCTC 5316 strain was 0.5 mg/ml and MIC of ethyl acetate fraction on C. albicans KCTC 7270 was 2.0 mg/ml. The experiment of inhibition to growth of KB roll(oral squamous cell carcinoma) result 61.9% in butanol, 76.7% in hexane extract of P. lactiflora. The hexane extract exhibit potent inhibition effect to the growth of KB cell. These results suggest that the hexane extract of Paeonia lactiflora has antimicrobial activity against S. mutans and has preventive effect to dental caries in addition to potent inhibition to KB cell growth.
