Comparative study on bone regeneration between silk mat incorporated 4-hexylresorcinol and collagen membrane

  • The purpose of this study was (1) to demonstrate the anti-microbial properties of 4-hexylresorcinol (4HR) loaded silk mat and (2) comparison of bone formation between 4HR incorporated silk mat and collagen membrane. Anti-microbial properties of 4HR incorporated silk mat was done after sterilization of silk mat (autoclaving and ethylene oxide gas). For the evaluation of bone formation, bilateral bony defects (size: 8 mm) were prepared in the parietal bone of the rabbits (n=10). 4HR incorporated silk mat (size: $10{\times}10mm$) was applied on the right defect. For the comparative purpose, the same size of commercial collagen membrane was applied on the left defect. The anti-microbial properties of 4HR incorporated silk mat were maintained after sterilization process. When compared bone mineral density and bone volume, there was no statistically significant difference between groups at 4 weeks and 8 weeks after operation (p>0.05). In conclusion, 4HR incorporated silk mat could be autoclaved without concern of anti-microbial properties loss. In addition, 4HR incorporated silk mat showed similar bone regeneration to collagen membrane. Therefore, 4HR incorporated silk mat might be considered for the application of open membrane technique.

Anti-oxidant and Anti-microbial Activities of Herb-combined Remedies used in Traditional Korean Medicine for Treating Breast Cancer (유암 처방에 사용되어온 한약재 복합 처방전의 항산화 및 항균활성에 관한 연구)

  • Sipyukmiryuki-eum (SYMYKE), Danjacheongpi-tang (DJCPT), Jipae-san Ⅰ (JPS Ⅰ), Jipae-san Ⅱ (JPS Ⅱ), and Chungganhaeul-tang (CGHUT) are representative herb-combined remedies used in traditional Korean medicine for treating breast cancer patients, as mentioned in "Dongeuibogam." In this study, we investigated the total phenolic contents (TPCs) and the anti-oxidant and anti-microbial activities of hot water and 70% ethanol extracts of these herbal prescriptions. Among the five herb-combined remedies, the extraction yields of the hot water extracts and 70% ethanol extracts were the highest in JPS Ⅱ (34.30%) and DJCPT (30.50%), respectively. The TPCs of the hot water extracts from the herb medicines were rich in the order of JPS Ⅰ < CGHUT < JPS Ⅱ < SYMYKE < DJCPT. In addition, the 70% ethanol extracts from the herb medicines were rich in the order of JPS Ⅱ < JPS Ⅰ < CGHUT < SYMYKE < DJCPT. Among them, DJCPT and SYMYKE displayed a strong anti-oxidant capability, which was determined using ferric-reducing anti-oxidant power and scavenging of 2,2'-diphenyl-1-picrylhydrazyl and 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonate) cationic radical activity assays. In addition, anti-microbial activities against Staphylococcus aureus and Escherichia coli were stronger in the 70% ethanol extracts than in the hot water extracts. Together, these findings reveal a positive relationship between TPCs and their anti-oxidant activities.

  • This study was conducted to estimate if the level of several chemical ingredients including alkanolamines or ethanolamines (EA) examined in the specific synthetic metalworking fluid (MWF) ‘A’ can cause anti-microbial activity and health effect. Three water-based MWF products (‘A’, ‘B’, and ‘C’) were studied every week for two months. Chemical ingredients such as formaldehyde, boron, EA, and copper were examined. In the sump where MWF ‘A’ was used, not only the total level of EA, monoethanolamine(MEA), diethanolamine(DEA) and triethanolamine(TEA), but also boron level were significantly higher than those of the other MWFs. ANOVA statistical tests indicated that levels of pH, alkalinity, boron, MEA, DEA and TEA in MWF ‘A’ were significantly higher than those in other MWF types. Correlation tests also found that levels of pH, alkalinity, boron, MEA, DEA and TEA in MWF ‘A’ are significantly correlated. We suggested the assumptions that excessive concentrations of EA, and borate at a high pH level, may cause anti-microbial resistance synergically. To demonstrate this assumption, additional study is needed to examine the relationship between the levels of microbes and excessive concentrations of EA, and borate at a high pH level.

  • The most extensively used synthetic antioxidants are butylated hydroxyanisole (BHA), butylated hydroxy toluene (BHT) and tert-butylhydroquinone (TBHQ). However BHT and BRA have been suspented of being responsible for liver damage andcar cinogenesis. Therefore, the importance of the search and exploitation of natural antioxidant, especially of plant origin, has greatly increased in recent years. Plant contain a wide variety of chemicals that have potent biological effects. As a result, there has been a growing interest in the use of herbs as a source of therapeutic drugs. The aim of this study was to assess the antioxidant and cosmeceutical of natural materials. The antioxidant and cosmeceutical activity of natural materials were investigated by hydroxyl radical scavenging, superoxide dismutase (SOD) -like, xanthine oxidase inhibition, tyrosinase inhibition, anti-microbial and astringent.

Supplementation of Essential Oil Extracted from Citrus Peel to Animal Feeds Decreases Microbial Activity and Aflatoxin Contamination without Disrupting In vitro Ruminal Fermentation

  • Long-term storage of feeds or feedstuffs in high temperature and humid conditions can be difficult because of microbial contamination. Essential oil isolated from industrial waste citrus peel could be used as a preservative because it is likely to have anti-bacterial and anti-fungal activity. Our objective was to determine whether different levels (0.028, 0.056 and 0.112 g/kg) of citrus essential oil (CEO) would provide anti-microbial activity and enhance preservation of animal feed without influencing rumen fermentation. At 0.112 g/kg, CEO inhibited growth of Escherichia coli (ATCC 25922) and Salmonela enteritidis (IFO 3313). Growth of E. coli recovered after 24 h of incubation, but S. enteritidis continued to be inhibited for 72 h. Preservation of antibiotic-free diets for swine was assessed by observing anti-aflatoxin activity. Aflatoxin was detected in control feed samples on days 16 (8 ppb) and 21 (8 ppb) and in anti-fungal agent (AA) treated samples on days 16 (2 ppb) and 21 (4 ppb). However, aflatoxin was not detected in feed samples treated with CEO. Treatment with CEO and AA did not influence ruminal pH, dry matter digestibility (DMD) or organic matter digestibility (OMD) over 48 h of incubation in rumen fluid. Acetate and propionate were slightly higher with CEO treatment (p<0.05), but total concentration of volatile fatty acid (VFA) was not significantly affected by treatment. Ammonia-N concentration was slightly higher for the control treatment (p<0.05). This study showed that treating feed with CEO enhances preservation of animal feed without influencing in vitro rumen fermentation.

Identification of Uncharacterized Anti-microbial Peptides Derived from the European Honeybee (꿀벌 Apis mellifera에서 유래 한 특성화 되지 않은 항균성 펩티드의 동정)

  • The European honeybee (Apis mellifera L.) has multiple anti-microbial peptides, but many were unknown and demands for their characterization have increased. This study therefore focused on identifying novel anti-microbial peptides (AMPs) from A. mellifera L. To obtain high-throughput transcriptome data of the honeybee, we implemented next-generation sequencing (NGS), isolating novel AMPs from total RNA, and generated 15,314 peptide sequences, including 44 known, using Illumina HiSeq 2500 technology. The uncharacterized peptides were identified based on specific features of possible AMPs predicted in the sequencing analysis. AMP5, one such uncharacterized peptide, was expressed in the epidermis, body fat, and venom gland of the honeybee. We chemically synthesized this peptide and tested its anti-bacterial activity against Gram-negative Escherichia coli (KACC 10005) and Gram-positive Bacillus thuringiensis (KACC 10168) by anti-microbial assay. AMP5 exhibited anti-bacterial activity against E. coli (MIC50=22.04±0.66 μM) but not against B. thuringiensis. When worker bees were injected with E. coli, AMP5 was up-regulated in the body fat. This study therefore identified AMP5 in adult European honeybees and confirmed its anti-bacterial activity against Gram-negative E. coli.

Effects of Egg Shell Membrane Hydrolysates on Anti-Inflammatory, Anti-Wrinkle, Anti-Microbial Activity and Moisture-Protection

  • This study was conducted to examine the effects of eggshell membrane hydrolysates (ESMH) on the anti-inflammatory, anti-wrinkle, anti-microbial activity, and moisture-protection for cosmetic use. Whole ESMH (before fractionation), and fraction I (>10 kDa), fraction II (3-10 kDa), and fraction III (<3 kDa) of the hydrolysates were assessed in this experiment. As lipopolysaccharide (LPS) and IFN-${\gamma}$ caused the inflammation on Raw264.7 cell, whole ESMH and fraction I showed to be effective in inhibiting the induction of cell inflammation depending on the concentration, and also showed outstanding effect to suppress the skin inflammation. Fraction I inhibited collagenase and elastase activities to a greater extent than the other fractions, while all fractions had antibiotic effects at concentrations of 10 mg/disc and 20 mg/disc. In addition, it showed the moisture protection effects of skin on the holding amount and losing amount of moisture in upper-inner arm of the human body with a relatively low loss rate in skin, which confirmed that the hydrolyzed fractions of ESM helps to form the superior protective layer of moisture. It was concluded that ESMH fractions with different molecular weights, especially the 10 kDa fraction, have anti-lipopolysaccharide, anti-IFN-${\gamma}$-induced inflammation, anti-collagenase and elastase activities, and thus can be used as a cosmetic agent to protect skin.

  • This study is development of UV-cured coating compounds which has more improved anti-stain and anti-biosis, about surface prevention of PVC Tile. We added water-soluble anti-static and anti-microbial agent to the resin. The process has prevented electrostatic and bacterial contagious disease. The result, which added 15wt% of water-soluble anti-static and 1wt% anti-microbial agent and coated with No.12 Bar-coater, coating composition had optimum surface property. It appear electric resistance($10^9{\Omega}/cm^2$), anti-stain (Ink Test, Dust Test), anti-biosis (99.99%), and adhesive power(100%).
