Anti-inflammatory activity of aqueous methanolic extract of Swietenia mahagoni (L.) Jacq. (Meliaceae) leaves

  • Pharmacological investigations were carried out with aqueous methanolic extract (AME) of Swietenia mahagoni (L.) Jacq. (Meliaceae) leaves. Acute toxicity studies revealed that the $LD_{50}$ dose of AME was 600 mg/kg, i.p. AME was found to possess significant anti-inflammatory activity in acute, sub-chronic and chronic models of inflammation. AME selectively inhibited cyclooxygenase (COX)-2 activity, which is involved in arachidonic acid metabolism and biosynthesis of prostaglandins under inflammatory conditions. Treatment with AME significantly enhanced total peritoneal cell count and the number of macrophages in normal mice, which revealed that AME may also alter the immune response along with its anti-inflammatory effect. The saponins or the alkaloids present in AME may be responsible for the anti-inflammatory activity.

소염진통제 약물에 대한 In vitro 피부자극 시험연구 (In Vitro Skin Irritation Test of Anti-Inflammatory Drugs)

  • In vitro skin iritation of anti-inflammatory drugs was investigated in terms of the cytotoxicity method to human skin fibroblast cells. Five anti-inflammatory drugs (Diclofenac, Naproxen, Meclofenamic acid, Ibuprofen and Fnoprofen) which are commercially available as oral preparations or injections were tested. The cytotoxicity of 5 chemicals was evaluated by using MTT[tetrazolium salt 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide] assay. NRU (neutral red uptake) assay and Alamar Blue assay after fibroblast cells had been exposed to the chemicals for 24 hours or 489 hours. The $IC_{50}$ values of the chemicals showed the comparative strength of cytotoxicity as following order of Meclofenamic acid>Diclofenac>Fenoprofen>Ibuprofen>Naproxen. The values of $IC_{50}$ determined by Alamar Blue assay were lower than those of MTT and NRU assay. These data suggest Alamar Blue assay can be useful method for assessing in vitro skin irritation potential of anti-inflammatory drugs.

  • Most inflammatory disorders are usually treated using anti-inflammatory drugs including non-steroidal anti-inflammatory drugs (NSAID) and steroidal anti-inflammatory drugs (SAID). Prolonged uses of NSAIDs and SAIDs may frequently cause adverse side-effects such as nausea, vomiting, diarrhea, constipation, decreased appetite, kidney and liver failure, ulcers, and prolonged bleeding after an injury or surgery. Thus, it is necessarily required to develop a new anti-inflammatory drug with little side-effects. Dandelion (Taraxacum officinale) possesses the therapeutic abilities to eliminate body heat and toxins and to remove swelling and inflammation. In order to verify the anti-inflammatory activity of dandelion, TPA(12-O-tetra decanoylphorbol-acetate)-induced or croton oil-induced acute edema was developed in the mouse ears, and dandelion extract dissolved in acetone was applied to both sides of inflamed ears. It was found that dandelion could significantly reduce the ear swelling, compared to that of non-treated control. In the case of $20{\mu}{\ell}$ application of $100mg/m{\ell}$ dandelion solution (DA-100), its anti-inflammatory effect was comparable to that of indomethacin, a non - steroidal anti-anflammatory drug. Taken together, it could be concluded that topically applied dandelion extract exhibited its potentials as a new drug candidate with an effective anti-inflammatory activity.

이비인후과 염증질환에 대한 데옥시리보뉴클레아제-브로멜라인정의 효과 조사 (The Effect of Deoxyribonuclease-Bromelain Tablet in Patients with Inflammation of Otorhinolaryngology)

  • The author has tested the clinical efficacy and safety of an anti-inflammatory drug in patients with inflammation of the otnrhinolaryngology. Deoxyribonuclease prepared from bovine pancreas destroyed fibers and reduced the viscosity of the exudates. Bromelain is a group of proteolytic enzymes which are known to have an effect on inflammation, swelling, and pain in inflammatory disorders. The mucolytic and anti-inflammatory properties of deoxyribonuclease associated with bromelain in inflammation of otorhinolaryngology were assessed in 67 patients. A standard dose of 3 tablets of deoxyribonuclease bromelain tablet was given for not less than 7 days. In all tile groups of patients considered, the drug showed a considerable therapeutic effectiveness. At the end of 7-28 days treatment, more than $89.6\%$ of treatment was improved. These findings suggest that it could be permitted to use deoxyribonuclease-bromelain tablet in patients with inflammation of the otorhinolaryngology.

급성염증 동물모델에서 연옥분과 연옥수의 염증억제 효과 (In Vivo Studies on Anti-inflammatory Activity of Nephrite)

  • Most inflammatory disorders are usually treated using anti-inflammatory drugs including non-steroidal anti-inflammatory drugs (NSAID) and steroidal anti-inflammatory drugs (SAID). In a prolonged use, however, they may frequently produce adverse side-effects. Thus, it is necessarily required to develop a new anti-inflammatory drug with little side-effects. Nephrite has been widely used by traditional oriental medicine to cure the various chronic diseases. In order to verify the anti-inflammatory activity of nephrite, the TPA (12-O-tetradecanoylphorbol-acetate) or the croton oil-induced edema was developed in the mouse ears and the nephrite powder suspension or the nephrite water was directly applied to the ear edema. It was found that nephrite could significantly reduce the ear swelling implying its strong potential as an active anti-inflammatory agent when comparing to indomethacin, a non-steroidal anti-inflammatory drug.

  • Objectives : This study was conducted to investigate the research trends of herbal medicine prescriptions and herbal medicines with anti-inflammatory effects. Methods : Among the papers published in oriental medicine from 2015 to 2020(1st half), experimental papers to reveal anti-inflammatory effects were collected, and finally 198 papers were selected. The anti-inflammatory effects of each herb were summarized, and were analyzed by year, journal, experimental subjects and experimental types. Results : 1. Of the 198 papers, there were 186 papers proving anti-inflammatory drug efficacy, 6 papers comparing efficacy between drugs, and 6 papers comparing the efficacy of packaging or storage of drugs. 2. The number of anti-inflammatory drug articles published from 2015 to the first half of 2020 was 35(2015), 35(2016), 38(2017), 44(2018), 34(2019), 12(2020). 3. Papers were published in a total of 18 journals, of which 58(29.3%) were published by the Korea Journal of Herbology. 4. There were 101 papers on single herbs(51.0%) and 78(39.4%) papers on combination drugs. 71 types of single herbs were used in a total of 101 papers, of which Cheongyeol was the most common with 28. 5. Of the 198 papers, 87 in vivo papers and 125 in vitro papers were published, and 14 papers were both conducted. In vitro experiments demonstrated the efficacy of anti-inflammatory drugs in various inflammatory indicators such as NO, PGE2, and in vivo experiments demonstrated the efficacy of anti-inflammatory drugs in various inflammatory diseases such as musculoskeletal, digestive, dermal diseases.

Anti-pyretic and anti-inflammatory activity of chloroform extract of Croton roxburghii in standard animal models

  • The chloroform extract of Croton roxburghii (Family: Euphorbiaceae) was evaluated for its antipyretic effects in Brewer's yeast induced hyperthermia in rats. The anti-inflammatory effect of the Croton roxburghii was also evaluated by using carrageenan, dextran, histamine, serotonin induced rat paw oedema and cotton pellet induced granuloma (chronic) models in rats. The chloroform extract of Croton roxburghii (CECR) exhibited significant anti-pyretic and anti-inflammatory effect at the dose 50, 100 and 200 mg/kg. Maximum inhibition (55.32%) was notedat the dose of 200 mg/kg after 3 h of drug treatment in carrageenan induced paw oedema, whereas the Indomethacin (standard drug) produced 61.33% of inhibition. The extract exhibited significant anti-inflammatory activity in dextran induced paw edema in a dose dependent manner. In the chronic model (cotton pellet induced granuloma) the CECR (200 mg/kg) and Indomethacin (10 mg/kg) showed decreased formation of granuloma tissue by 52.32% and 56.32% respectively. The extract also exhibited a significant antipyretic response in Brewer's yeast induced pyrexia in rats. Thus, the present study revealed that the CECR exhibited significant antipyretic and anti-inflammatory activity in the tested animal models.

  • Loxoprofen sodium is an orally used anti-inflammatory, analgesic and antipyretic agent. For alleviation or inhibition of the pain symptoms regardless of referred or superficial pain, the prompt absorption of a drug and its immediate bioavailability might be generally required for a formulation or development of a new drug. Therefore, in intramuscular administration, its anti-inflammatory, analgesic, and antipyretic effects were evaluated compared with an oral administration in animals. The occurrence of gastric damages which is common in nonsteroidal anti-inflammatory drugs was also observed.

Analgesic and Anti-inflammatory Activities of Momordica dioica Fruit Pulp

  • In the present study Momordica dioica fruit pulp was successively extracted with hexane and methanol. To the concentrated methanolic extract ethyl acetate was added and ethyl acetate soluble portion was separated. Both hexane extract (HE) and ethyl acetate soluble portion (EASP) of methanolic extract was vacuum dried to yield the respective HE and EASP. HE and EASP were evaluated for its analgesic and anti-inflammatory activities in a dose of 50 and 100 mg/kg in mice and rats. Both HE and EASP significant exhibited analgesic and anti-inflammatory activities when compare to standard drug.

  • The leaves of Clerodendron trichotomum Thunberg(CTL) is used in Chinese folk medicine for anti-inflammatory properties. We studied on the anti-inflammatory effects of CTL extracts in rat. mouse and Raw 264.7cell. 1 mg/kg of 30%. 60% methanol fraction of CTL and 1 mg/kg of indomethacin as the standard anti-inflammatory drug were administrated into rats, respectively. Carrageenan was injected subcutaneously to induce hind paw edema in rats. (omitted)
