구증구포 처리 대두 추출물의 항산화 활성 및 Angiotensin-I Converting Enzyme 저해 효과 (Effect on Angiotensin-I Converting Enzyme Inhibition and Antioxidant Activities of Soybean (Glycine max L.) following Steaming and Drying Nine Times)

  • To increase the functional material content of soybean, a repetitive steaming and drying process was used. We investigated the changes in the total polyphenol content, the antioxidant activity, and the angiotensin-I converting enzyme (ACE) inhibition in soybean following nine rounds of steaming and drying. Soybean was steamed 9 times for 2 h and then dried 9 times from 55℃ to 73℃ for 3 h. The total polyphenol content in the soybean reached a maximum value of 60.47 mg GAE eq./100 g at 73℃ while the total polyphenol content in the raw soybean reached 25.17 mg GAE eq./100 g. In the raw soybean samples, the DPPH radical scavenging activity (5 mg/mL) was 8.04% but it increased by 43.29% after drying 9 times to 73℃. ABTS radical scavenging activity also improved following 9 rounds of steaming and drying. ACE inhibitory activity of the soybean dried 9 times at 73℃ was 58.94% at 10 mg/mL. These results showed that steaming and drying soybean 9 times enhanced the antioxidant activity and the ACE inhibitory activity of soybean. Therefore, more research on the biological and anti-hypertensive activity of soybean using this steaming and drying method is necessary.

맹종죽의 항산화활성을 통한 항고혈압 효능 (Anti-hypertensive Effects of ethanol extract of Phyllostachys Pubescens via Antioxidant Activity)

  • Phyllostachys pubescens (Maengiong-Juk), a kind of the bamboo, was reported to have many beneficial pharmacological actions. in this study, of using 70% ethanol extract of Phyllostachys pubescens we investigated its efficacy on angiotensin converting enzyme (ACE) and antioxidant enzyme activities. In addition, vasorelaxant effect was examined in rat aortic rings. The inhibitory effect of ACE activity by Phyllostachys pubescens extract (PPE) was dose-dependently increased by 61.42% at 10mg/ml. PPE relaxed the pre-contracted rat aortic rings with 10$^{-6}$M phenylephrine, showing about 88% at 4.0mg/ml. Sprague Dawley (SD) rats were given different concentrations of PPE mixed in the drinking water for 10 weeks. PPE did not show any difference with control group in blood pressure, body weight (BW) and food intake. However, it revealed the highest total antioxidative effect at dose of 1.0 g/100 g BW in plasma by TEAC assay. Thiobarbituric acid reactive substance (TBARS) and protein carbonyl levels which are markers of tissue peroxidation, were significantly lowed at the same dosage. Furthermore, hepatic antioxidant enzymes such as total superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), glutathione reductase (GR) and catalase activities were also significantly increased by PPE (1.0 g/100 g BW). In conclusion, we suggest that PPE might have antihypertensive effect through increasing antioxidant activities.

급성 신손상을 동반한 가와사키 쇼크증후군 1예 (Kawasaki Disease Shock Syndrome with Acute Kidney Injury and Hypertension)

  • 가와사키병은 다른 원인이 없는 발열과 함께 피부점막에 특징적인 병변을 보이는, 소아에서는 비교적 흔한 질환으로 일반적으로 급성기에는 혈역학적으로 안정적이다. 저자들은 심한 저혈압과 함께 급성 신손상이 있는 상태에서 내원한 8세 남아가 치료 중에 가와사키병으로 진단된 증례를 경험하였다. 최근에는 가와사키병 초기에 불안정한 혈역학적 상태를 가와사키병 쇼크증후군이라는 개념으로 정의하고 있다. 환아는 면역글로불린 치료 후 임상적으로 회복되었으나 관상동맥 확장 합병증이 확인되어 아스피린을 복용하고 있으며, 가와사키병의 합병증으로서는 비교적 드문 고혈압이 확인되어 베타 차단제를 사용하고 있다. 본 증례를 통해 가와사키병 쇼크증후군의 임상 양상을 알고, 혈관염이라는 가와사키병의 특성을 고려해볼 때 혈압 모니터링의 중요성을 깨닫게 되었다.

두충차 추출물의 Angiotensin Converting Enzyme 저해효과 (Inhibitory Effects of Eucommia ulmoides Extract on Angiotensin Converting Enzyme)

  • 두충의 기능성 식품으로서 이용 가능성을 평가하기 위하여, 두충의 항고혈압 활성 물질을 분리 및 확인하였고, 각 부위별(잎, 껍질, 줄기) 추출물의 관련물질의 함량 및 ACE 저해활성을 조사하였다. 두충에서 분리한 항고혈압성 물질로 추정되는 8A는 순도 95.64%의 pinoresinol-4,4'-di-O-${\beta}$-D-glucoside(PDG)로서 자연건조 및 볶은 껍질에서 가장 높았으며, 각각 135.13 mg%와 163.67 mg%였다. 또한 두충 추출물의 ACE 저해활성은 10 mg/mL 농도로 처리하였을 때 볶은 잎, 자연건조 껍질 및 볶은 껍질에서 각각 77.56%, 75.73%, 75.73%로 높은 활성을 나타내었으며, 줄기는 상당히 낮게 나타났다. 두충에서 분리한 PDG를 1 mg/mL 처리할 때, ACE 저해활성은 91.87%로서 $Enalapril^{(R)}$의 97.06%보다는 조금 낮았지만, $Captopril^{(R)}$의 90.32% 보다는 약간 높은 항고혈압 활성을 나타내었다.

Effect of Hydroalcoholic Extract of Ribes khorasanicum on Acute Hypertension Induced by L-NAME in Rat

  • Objectives: The aim of this study was to evaluate the effect of Ribes khorasanicum (R. khorasanicum); a plant growing in north Khorasan of Iran; on cardiovascular and stress oxidative in acute hypertension induced by N-nitro-l-arginine methyl ester (L-NAME), anitric oxide synthase inhibitor. Methods: Rats were divided into Control, L-NAME (10 mg/kg), Sodium Nitroprusside (SNP) (50 mg/kg) + L-NAME and three treated groups with R. khorasanicum (4, 12 and 24 mg/kg) groups + L-NAME. L-NAME and SNP were injected intravenously and extract intraperitoneal. In R. khorasanicum groups, L-NAME was injected 30 min after injection of the extract. Systolic blood pressure (SBP), mean arterial pressure (MAP) and heart rate (HR) were recorded continuously using power lab software. At the end of study oxidative stress parameters including of total thiol content (SH), malondialdehyde (MDA), superoxide dismutase (SOD) and catalase (CAT) in heart and aorta of all groups were also measured. Results: In groups 4 and 24 mg/kg extract +L-NAME, there was a non-significant decrease in SBP and MAP compared to L-NAME group but dose 12 mg/kg significantly attenuate the effect of L-NAME(P < 0.05). In L-NAME group the heart and aorta tissues antioxidant enzymes levels decreased, while in treated rats these enzymes significantly increased. Conclusion: The extract of R. khorasanicum in dose 12 mg/kg show anti-hypertensive effect that is mediated by an effect on NO system or antioxidant parameters.

Anti-hypertensive Effects of Artichoke Supplementation in Adults: A Systematic Review and Dose-response Meta-analysis of Randomized Controlled Trials

  • Despite controversies, no earlier study has systematically summarized findings from earlier studies on the effect of artichoke supplementation on blood pressure. Therefore, current systematic review and meta-analysis was done on the effect of artichoke supplementation on systolic blood pressure (SBP) and diastolic blood pressure (DBP) in adults. Five databases were searched from inception to January 2022 using relevant keywords. All randomized clinical trials investigating the impact of oral artichoke supplementation on any of the blood pressure parameters including SBP or/and DBP were included. Out of 1,507 citations, 7 trials that enrolled 472 subjects were included. Artichoke supplementation resulted in significant reduction in SBP (weighted mean difference [WMD], -2.01 mmHg; 95% confidence interval [CI], -3.78, -0.24; p = 0.026) and DBP (WMD, -1.45 mmHg; 95% CI, -2.81, -0.08; p = 0.038). Greater effects on SBP were detected in trials using ≤ 500 mg artichoke, lasted > 8 weeks, participants aged < 50 years' old and sample size ≤ 70. There was also a similar impact of artichoke on DBP. However, significant non-linear associations were found between artichoke supplementation dosage and study duration with both SBP (for dosage: pnon-linearity = 0.002, for duration: pnon-linearity = 0.016) and DBP (for dosage: pnon-linearity = 0.005, for duration: pnon-linearity = 0.003). We found a significant reduction in both SBP and DBP following artichoke supplementation in adults. It could be proposed as a hypotensive supplement in hypertension management.

소아복막투석의 현황: 다기관 공동연구 결과보고 (Current Status of Children on Peritoneal Dialysis in Korea : A Cross-Sectional Multicenter Study)

  • 목적 : 복막투석요법은 말기 신부전 환아에서 신이식 다음으로 추천되는 신대체 요법으로 현재 말기 신부전 환아들의 장기 생존율 향상으로 복막투석요법 환아 수가 점차 증가하고 있다. 현재 한국 내 복막투석요법 소아에 대한 현황을 파악하고자 이번 연구를 시행하였다. 방법 : 2008년 5월 국내 4개 대학병원을 대상으로 전자우편 설문조사를 시행하였고 현재 3개월 이상 복막투석 중인 103명의 소아를 대상으로 자료를 분석하였다. 결과 : 대상 환아 103명의 남녀비는 1.6:1로 남아가 많았고, 평균 연령은 $11.5{\pm}4.9$세(0-19세), 투석시작 전 말기 신부전의 원인 중 원발성 사구체 질환이 34%로 가장 많았다. 복막투석의 형태는 지속적 외래 복막투석법이 42.7%로 가장 많았고, 주간 전체 Kt/V는 $2.1{\pm}0.7$ (0.3-4.1), 복막평형검사상 저 투과성이 36.8%, 저평균 투과성이 31.6%으로 가장 많은 비율을 차지했다. 체중 표준편차점수(Z-score)는 $-1.00{\pm}1.20$(-4.54-+2.50), 신장 표준편차점수(Z-score)는 $-1.55{\pm}1.65$(-9.42-+1.87)였고, 성장호르몬은 24.3%에서 투여 중이었다. 38.8%의 환아에서 이완기 혹은 수축기 혈압 95 백분위수 이상의 고혈압을 보였고, 64.0%의 환아가 항고혈압제 투여 중이었다. 평균 혈청 혈색소는 $10.5{\pm}1.4$ g/dL, 평균 혈청 칼슘은 $9.7{\pm}0.7$ mg/dL, 평균 혈청 인은 $5.4{\pm}1.4$ mg/dL, 평균 부갑상선 호르몬은 $324.2{\pm}342.8$ pg/mL였다. 결론 : 국내 복막투석요법 소아에서 원발성 사구체 질환이 소아 말기 신부전의 가장 흔한 원인이고, 지속적 외래 복막투석법이 가장 흔히 사용되는 복막투석 형태이며, 복막평형검사상 저 투과성과 저평균 투과성이 가장 많았다. 대부분의 환아가 적절한 신장과 체중을 유지하고 있었으나 저신장 환아의 경우 성장 호르몬 치료가 제대로 이루어지지 않고 있었고 여전히 많은 환아에서 빈혈과 고혈압이 관찰되었다. 혈청 칼슘과 인은 비교적 정상범위를 유지하고 있으나 많은 환아가 이차성 부갑상선 기능 항진증을 보였다.

황연해독탕(黃連解毒湯)과 온청음(溫淸飮)이 고혈압(高血壓) 및 고지혈증(高脂血症)에 미치는 영향(影響) (A Experimental Study on the Effect of Hwangryeonheadock-Tang and Onchung-Eum on Hyperlipidemia & Hypertension)

  • The aim of the present study was to investigate the effect of Hwangryeonheadock-Tang and Onchung-Eum on essential hypertension and hyperlipidemia. Rats were orally administered for 30days with Hwangryeonheadock-Tang and Onchung-Eum and the blood was withdrawn at 10, 20 and 30days after an oral administration. The heart rate, tail blood pressure, plasma renin activity, plasma level of aldosterone. catecholamine, sodium and angiotensin II were measured after an oral administration of Hwangryeonheadock-Tang and Onchung-Eum in spontaneously hypertensive rat(SHR). In addition, serum levels of total cholesterol, triglyceride, HDL-cholesterol. LDL-cholesterol and total lipid were measured cholesterol-fed rats. The results were summarized as follows ; 1. A significant decrease of tail blood pressure was shown at 10, 20 and 30days after Hwangryeonheadock-Tang and Onchung-Eum treatment in SHRs. compared with saline. 2. Heart rate was significantly decreased at 30days in SHRs after Hwangryeonheadock-Tang treatment and at 20, 30days after Onchung-Eum treatment in SHRs. compared with the effects of saline group. 3, A significant decrease of plasma aldosterone level was elicited at 10, 20days after Hwangryeonheadock-Tang treatment in SHRs, compared with the effects of saline group, 4. Plasma renin activity was significantly decreased at 10days after Onchung-Eum treatment compared with the effects of saline group in SHRs. 5. Plasma norepinephrine level was significantly decreased at 20 and 30clays after Onchung-Eum treatment in SHRs, compared with the effects of saline group. 6. A significant decrease of plasma epinephrine level was induced at 30days after Hwangryeonheadock-Tang treatment and at 10, 20 and 30days after Onchung-Eum treatment, compared with the effects of saline group in SHRs. 7. Plasma sodium level was. significantly decreased at 20days after Hwangryeonheadock-Tang and Onchung-Eum treatment, compared with the effects of saline group in SHRs. 8. Plasma angiotensin II level was significantly decreased at 30days after Onchung-Eum treatment, compared with the effects of saline group in SHRs. 9. A significant decrease of body weight was observed at 20 and 30days after Hwangryeonheadock-Tang treatment and at 10, 20 and 30days after Onchung-Eum treatment. compared with the effects of saline group in hyperlipidemia rats. 10. Hwangryeonheadock-Tang and Onchung-Eum showed a significantly decreasing effect at 30days on serum total cholesterol level in hyperlipidemia rats, compared with the saline treatment. 11. Hwangryeonheadock-Tang and Onchung-Eum saw 20 and 30days respectively on serum triglyceride level in the saline treatment. 12. Hwangryeonheadock-Tang and Onchung-Eum decreased on serum HDL-cholesterol level significantly, compared with the saline treatment in hyperlipidemia rats. 13. A significant decrease of serum LDL-cholesterol was observed at 10 and 30days after Hwangryeonheadock-Tang treatment and at 30days after Onchung-Eum treatment, compared with the effects of saline group in hyperlipidemia rats. 14. Hwangryeonheadock-Tang had a significantly decreasing effect at 10, 20 and 30days on serum total lipid level, compared with the saline treatment in hyperlipidemia rats. 15. Hwangryeonheadock-Tang elicited a significantly decreasing effect on weight of kidneys, spleen and testes respectively and Onchung-Eum induced on weight of liver and spleen respectively in hyperlipidemia rats, compared with saline treatment. These Findings suggest a possible anti-hypertensive and anti-hyperlipidemic effect of Hwangryeonheadock-Tang and Onchung-Eum.

희렴의 Nitric Oxide 유리를 통한 평활근세포에서의 Apoptosis유도 (Production of Nitric Oxide by Siegesbeckia Glabrescens is Associated with Apoptosis of Vascular Smooth Muscle Cell)

  • Apoptosis is the ability of cells to self-destruct by the activation of an intrinsic cellular suicide program when the cells are no longer needed or when they are seriously damaged. Morphologically, apoptosis is characterized by the appearance of membrane blebbing, cell shrinkage, chromatin condensation, DNA cleavage, and the fragmentation of the cell membrane-bound apoptotic bodies. Siegesbeckia glabrescens Makino (Siegesbeckiae Herba, SG) has been widely used as treatments for arthritis, and fever, as well as detoxification properties. The present studies were undertaken to evaluate if SG has an anti-apoptotic property. Cell viability was measured by XTT and tryphan blue stain. Morphological characteristic of human aortic smooth muscle cells(HASMC) were visualized with a phase-contrast microscope. SG significantly reduced HASMC, but not human umbilical vein endothelial cell(HUVEC), viability in a dose-dependent manner. Confluent untreated cells at 24hrs showed normal morphology, flat with a uniform polygonal shape. SG-treated cells (0.5㎎/㎖) at 24hrs showed apoptotic morphology. Cells became irregular with elongated lamellipodia, and exhibited condensed chromatin in nuclei with occasional endoucleation. There was an increase in the number of apoptotic cells rounding-up and being detached from the substrate. TUNEL staining of SG-treated cells showed dark-brown stains in nuclei and cytosol. Caspases are central components of the machinery responsible for apoptosis and are generally divided into two categories; the initiator caspases, which include caspases-2,-8,-9, and -10, and the effector caspases, which include caspases-3,-6, and -7. SG decreased anti-caspase-3 protein expression, which means activation of caspases-3 activity. It has been reported that there is a link between NO formation and apoptosis. NO production was accelerated by SG treatment in HASMC. L-NNA, NOS inhibitor, inhibited SG-induced apoptosis. These results, therefore, indicated that both caspases-3 and NO production are involved in apoptosis in smooth muscle cells. According to these results, SG may have a potential effect in the treatment of hypertensive atherosclerosis.

Cinnamaldehyde Attenuates Cataractogenesis via Restoration of Hypertension and Oxidative Stress in Fructose-Fed Hypertensive rats

  • Objectives: Several studies have revealed that systemic hypertension is strongly associated with cataractogenesis. However, the pathophysiology and treatment is often unclear. In this study, we evaluated the anti-cataractogenic effect of cinnamaldehyde (CA), a natural organic compound, in rats with fructose-induced hypertension. Methods: The rats were divided into six groups. For six weeks, the normal group received a suspension of 0.5% carboxy methyl cellulose (10 mL/kg/day, p.o.) while five other groups received a 10% (w/v) fructose solution in their drinking water to induce hypertension. By the end of the third week hypertension had been induced in all the animals receiving fructose. From the beginning of the fourth week to the end of the sixth week, one of those five groups (control) continued to receive only 10% (w/v) fructose solution, one group (standard) received ramipril (1 mg/kg/day, p.o.) plus 10% (w/v) fructose solution, and three groups (experimental) received CA at doses of 20, 30, and 40 mg/kg/day p.o., plus 10% (w/v) fructose solution. Blood pressure was measured weekly using a non-invasive blood pressure apparatus. After six weeks, the animals were sacrificed, and the anti-cataractogenic effects on the eye lenses were evaluated. Results: Administration of fructose elevated both the systolic and the diastolic blood pressures, which were significantly reduced by CA at all dose levels. In the control group, a significant increase in the malonaldehyde (MDA) level and decreases in the total protein, $Ca^{2+}$adenosine triphosphate (ATP)ase activity, glutathione peroxidase, catalase, superoxide dismutase and glutathione levels, as compared to the normal group, were observed. Administration of CA at all doses significantly restored the enzymatic, non-enzymatic, antioxidants, total protein, and $Ca^{2+}$ATPase levels, but decreased the MDA level, as compared to the control group. Conclusion: The present study revealed that CA modulated the antioxidant parameters of the serum and lens homogenates in hypertension-induced cataractogenic animals.
