To investigate the anti-diabetic activity of ethanol and aqueous peel and seed extracts obtained from three different grape species (Cambell Early, MBA, and Kyoho), alpha-glucosidase inhibitory activity was examined. All extracts showed anti-diabetic activity, especially aqueous extract exhibited inhibitory effect above 70%. Thus, we used aqueous extract to check the potential hypoglycemic effects in a streptozotocin (STZ)-induced diabetic mice model. The results showed that the blood glucose level of STZ-induced diabetic mice decreased drastically after 3 hr when the aqueous extract of Cambell Early seed was treated to the mice model. The aqueous extract of Kyoho seed lessened blood glucose level by 60%. Together, these data indicate that extracts of grape peel and seed (aqueous or ethanol) may have potential in improving hypoglycemic effects in the diabetic symptoms, suggesting that further investigation on biomarker expressions should be rewarding.
The leaves of Ligularia stenocephala (Compositae) are used as a vegetable food to treat anxiety, weakness and menstrual disorder and are said to be effective against diabetic disease in Korea. This research was aimed to develop this vegetable food as a functional food to treat diabetic disease. Oral administration of MeOH extract exhibited significant hypoglycemic and hypolipidemic effects in streptozotocin (STZ)-induced diabetic rats at 100 and 200 mg/kg dose. This extract inhibited the increase of body weight, water consumption, food intake, blood glucose, lowered atherogenic index, and overcome glucose intolerance in the STZ-treated rat. These results suggest that the leaves of Ligularia stenocephala could be developed as a functional food with anti-diabetic effect.
The extract from Hydnocarpi Semen (HS) has been used to treat leprosy and its anti-inflammatory activity has been reported. However, the effect of HS on the treatment of diabetic or peripheral ulcer is not well known. We therefore examined its wound healing effects on ulcer area in diabetic mice. GC and GC/MS analysis with the total extract of HS show that the main constituents of the extract are chaulmoogric acid, hydnocarpic acid, and gorlic acid. Whereas HS showed wound healing effect in diabetic ulcer, there was no hypoglycemic effect in diabetic mice. The treatment of HS extract significantly decreased the level of total WBC and neutrophils in mice compared to control mice. Cutting ulcer was induced by the round-shaped punch on the backside of diabetic mice and the extract of HS was given orally or topically. The wound area score significantly decreased after treatment of HS at dose of 50 mg/kg. The treatment of HS also induced the activation of macrophages and increased the production of IL-12 and TNF-$\alpha$ in macrophages, indicating that the wound healing by HS extract is associated with the inflammatory effect via the activation of macrophages. Our results suggest that HS extract can be a new therapeutic candidate for treatment of diabetic ulcer.

Many herbal extracts have been reported to have a preventive or therapeutic effect of on diabetes mellitus. Momordica Charantia commonly known as bitter melon or karela has been reported to be a medicinal plant for treating various diseases including cancers and diabetes. The objectives of this study were to investigate anti-diabetic effects of bitter melon (BM) as determined by blood glucose levels, glucose tolerance test (GTT), insulin tolerance test (ITT), insulin and HbA1C activities in serum, serum biochemical and lipid levels, histopathology, immunohistochemistry and AMPK-${\alpha}2$ expression of skeletal muscle in male C57BL/6J db/db mice. There were four experimental groups including vehicle control, BM 10 mg/kg, BM 50 mg/kg, and BM 250 mg/kg. BM at doses of 10, 50, and 250 mg/kg was orally administered to the diabetic mice everyday for 8 weeks. The treatments of BM 10, 50, and 250 mg/kg significantly decreased the blood glucose level in the diabetic mice compared with vehicle control (p < 0.05). The treatments of BM 10 and 50 mg/kg significantly decreased the GTT, ITT and HbA1c levels in the diabetic mice compared with vehicle control (p < 0.05). All BM groups significantly decreased GOT, GPT, BUN, LDL and glucose levels in the diabetic mice compared with the vehicle control mice (p < 0.05). The livers of mice treated with the BM 10, 50, and 250 mg/kg showed a remarkable decrease in the number of lipid droplets compared with the vehicle control. The pancreas of mice treated with the BM 10, 50, and 250 mg/kg showed a remarkable increase in insulin concentration of ${\beta}$-cells compared with the vehicle control. In addition, the treatments of BM 10, 50, and 250 mg/kg actually increased the expression of AMPK-${\alpha}2$ compared with vehicle control. These results suggest that BM has a respectable anti-diabetic effect resulting from inhibition of blood glucose level and lipid level in serum and that consumption of BM may give a benefit for controlling diabetes mellitus in humans.

This study was carried out to investigate the optimal mixed extract with Bambusae Caulis in Liquamen in order to strengthen anti-diabetic effects on the hyperglycemia induced by streptozotocin in mice. The original Bambusae Caulis in Liquamen filtered and refined. The effects of Bambusae Caulis in Liquamen and Mixed extracts(Bamboo Juice) with Bambusae Caulis in Liquamen were administered to mice for 4weeks and its anti-diabetic effect examined. Mice used in this experiment were divided into three groups and saline(control), Bamboo Juice mixed with refined Bambusae Caulis in Liquamen(BJ+BCL.D) and distrilled water mixed with refined Bambusae Caulis in Liquamen(DW+BCL.D) were given orally for 28days respectively. And then, experemental groups were observed in terms of blood sugar, creatinine, BUN and GPT. The amount of glucose was significantly decreased in the Bambusae Caulis in Liquamen and Mixed extracts(Bamboo Juice) with Bambusae Caulis in Liquamen-treated groups compared with the control group(P<0.01). The amount of creatinine, BUN and GPT did not show any differences among Control, BJ+BCL.D and DW+BCL.D groups. In conclusion. it was found that Bambusae Caulis in Liquamen and Mixed extracts(Bamboo Juice) with Bambusae Caulis in Liquamen were nontoxic to kidney and liver and also effective on murine hyperglycemia induced with STZ. Mixed extracts(Bamboo Juice) were more effective for decreasing blood glucose than Bambusae Caulis in Liquamen D. BJ+BCL.D can be used as optimal mix material with Bambusae Caulis in Liquamen D for control Diabetes.
Ginseng is a popular medicinal plant highly valued throughout the world. Asian ginseng is one of the most common species of ginseng. It has long occupied a significant position in oriental medicine and has been justified its name as the 'king herb'. As a nutritional supplement, ginseng is an extremely common and popular herbal medicine in the United States and Canada in recent decades. The multiple constituents of ginseng possess equally multifaceted pharmacological actions as demonstrated by numerous studies. Ginseng root and its constituents influenced the central nervous system, endocrine, cardiovascular, gastrointestinal system, sexual, renal organ and immune system, etc. One important action is its anti-hyperglycemic effect. Previous studies on ginseng demonstrate that only the root of ginseng has been used in the treatment of diabetes, while the other parts of ginseng plant were always neglected. Recently, we analyzed the constituents of ginseng berry, leaf and discovered that ginseng berry, leaf extracts and its total ginsenosides have the ability to reduce hyperglycemia and body weight and increase the peripheral glucose utilization in obese or diabetic ob/ob or db/db mice. Our data suggest that all parts of ginseng plant, including root, berry, leaf and stem exhibit potent anti-hyperglycemic and anti-obese effects and may provide an opportunity to develop a novel class of anti-diabetic agents.

The leaves of Phaleria nisidai Kaneh. have been used as a beverage for tonic and immune-enhancing effect in Palau. Recently, extract of the plants showed anti-diabetic effect in clinical trial and possibility could be used as a medicine for diabetes. However, in vitro and in vivo experiments which elucidate cytotoxicity and biological mechanism have not been studied yet. To begin the series of in vitro and in vivo anti-diabetic studies, we evaluated cell viability of pancreatic beta cell lines to figure it out toxicity of the extract. The leaves of Phaleria nisidai were extracted with distilled water and 70% EtOH, and followed by evaporation and lyophilization. And, MIN6 and RIN-m5F cell lines were used for assessment of cell viability. MTT assay was adopted to evaluated a toxicity. Any of extract and concentration used in this study did not show cytotoxicity in the cell lines. Furthermore, morphological change was not observed. These results means Phaleria nisidai might be a medicinal resource when further biological and safety studies would be performed.

Diabetic neuropathy is characterized by the decrease of cell viability in neuron, which is induced by the hyperglycemia. HT22 cell is the neuron cell line originated from hippocampus. Ginsenosides have been reported to retain anti-diabetic effect. However, the preventive effect of ginsenosides in the condition of diabetic neuropathy was not elucidated. Thus, this study was conducted to examine the protective effect of ginsenoside total saponin (GTS), panoxadiol (PD), and panoxatriol (PT) in the high glucose-induced cell death of HT22 cells, an in vitro cellular model for diabetic neuropathy. In present study, high glucose increased lactate dehydrogenase(LDH) activity, the lipid peroxide(LPO) formation and induced the decrease of cell viability. These effects were completely prevented by the treatment of GTS, but partially prevented by the treatment of PD and PT. High glucose also increased the expression of Bax and cleaved form of caspase-3 but decreased that of Bcl-2. These effects of high glucose on Bax, Bcl-2 and cleaved form of caspase-3 were completely prevented by the treatment of GTS, but partially prevented by the treatment of PD and PT in HT22 cells. In conclusion, ginsenosides prevented high glucose-induced cell death of hippocampal neuron through the inhibition of oxidative stress and apoptosis in HT 22 cells.
The present study was carried out to summarize the effect of ginseng in the experimental diabetic rats by meta-analysis related studies. The association measure to test effect of ginseng was the mean difference(MD) between group of rats induced streptozotocin(STZ) and group of rats induced STZ treated with ginseng about the considered effect factors. The level of FI, glucose and TG were significantly reduced(< 0.01), and the level of glycogen was significantly increased by treatment with ginseng (< 0.01) After checking the indication of publication bias for the combined MDs by using the funnel plots, the anti-diabetic effects of ginseng is clearly presented in FI, glucose, TG and glycogen (< 0.05).
