Effects of Zinc on Oral Bacteria and Volatile Sulfur Compound (VSC) in Oral Cavity (구강내 세균과 휘발성 황화합물에 대한 아연의 영향)

  • Zinc compounds have been used in various fields - cosmetics, medicine, and dentistry -because of its effective functions to human tissues or organs. Especially, it is well known that zinc has many biologic effects in oral cavity. Zinc ion can affect various oral microorganisms, resulting in reduction of oral bacteria, dental plaque, and dental caries. Also, zinc ion has an ability to reduce amounts of oral anaerobic bacteria and oral VSC and can reduce oral malodor. The author summarized the characteristics and toxicity of zinc, several forms of zinc compounds applied in human tissues, and reviewed biologic effects of Zinc in oral cavity (anti-bacterial effects, anti-plaque effects, anti-caries effects, and anti-VSC effects of zinc). Because of many advantages of zinc in oral cavity, it can be concluded that application of zinc compound to various oral diseases will be extended and activated, and promising.

Synthesis and In-vitro Activity of Some New Class of Thiazolidinone and Their Arylidene Derivatives

  • In an attempt to find a new class of anti microbial agents, a series of thiazolidinone and their 5-arylidene derivatives containing 4-(4-methyl benzamido)-benzoyl moiety were synthesized via the reaction of benzocaine with appropriate chemical reagents. These compounds were screened for their antibacterial activity against Gram-positive bacteria (Bacillus subtilis and Bacillus thuringiensis), Gram-negative bacteria (Escherichia coli, Pseudomonas aeruginosa) and antifungal activity against Botrytis fabae, Fusarium oxysporan and Candida albicans. On the other hand the synthesized compounds were also screened for their anti tubercular activity. IR, $^1H$ NMR, $^{13}C$ NMR and MS spectral analyses established the structures of the newly synthesized compounds. The results revealed that some of these compounds have shown promising antimicrobial and anti tubercular activity in comparison with standard drugs.

Probiotic Property and Anti-Obesity Effect of Lactiplantibacillus plantarum KC3

  • Lactic acid bacteria are representative probiotics that have beneficial effects on humans. Nineteen strains among the 167 single strains from kimchi was selected and their physiological features were investigated. The selection of a strain was based on strong enzyme (lipase, α-amylase, and α-glucosidase) inhibitory activities and anti-obesity effects in the adipocytes. For the final selection, the strain Lactiplantibacillus plantarum KC3 was tested for its potential as a starter. To assess its functionality, a freeze-dried culture of L. plantarum KC3 was administered to a diet-induced obese mouse model receiving a high-fat diet. The animal group administered with L. plantarum KC3 showed significant body weight loss during the 12-week feeding period compared to the high-fat control group. This study investigated the physiological characteristics of selected strain and evaluated its potential as an anti-obesity probiotic in mice.

In vitro Anti-bacterial and Anti-scuticociliate Activities of Extract and Bromophenols of the Marine Red Alga Polysiphonia morrowii with Structure-activity Relationships (홍조류 모로우붉은실(Polysiphonia morrowii)의 추출물과 이로부터 분리된 브로모페놀계 화합물의 in vitro 항균·항스쿠티카충 활성 및 구조-활성 상관성)

  • Scuticociliates are regarded as serious pathogens in marine aquaculture worldwide. In Korea, they cause mass-mortalities in fish such as the commercially important olive flounder Paralichthys olivaceus. In particular, mixed infections of scuticociliates with pathogenic bacteria have been commonly reported. During efforts to identify natural marine-algae derived products that possess anti-bacterial and anti-scuticociliate properties, we found that an 80% methanolic extract of the red alga Polysiphonia morrowii Harvey exhibits both anti-scuticociliate activity against Miamiensis avidus, which is a major causative agent of scuticociliatosis, and anti-bacterial activities against fish pathogenic bacteria. Activity-guided fractionation and isolation of the 80% methanolic extract of P. morrowii yielded three bromophenols, which were identified as 3-bromo-4,5-dihydroxybenzyl methyl ether (1), 3-bromo-4,5-dihydroxybenzaldehyde (2) and urceolatol (3) based on spectroscopic analyses. 3-bromo-4,5-dihydroxybenzyl methyl ether (1) showed the highest anti-bacterial and anti-scuticociliate activities, with a minimal inhibitory concentration (MIC) of $62.5{\mu}g/mL$ (against Vibrio anguillarum) and minimal lethal concentration (MLC) of 62.5 ppm (in seawater). Investigations of the anti-bacterial and anti-scuticociliate activities of seventeen bromophenol derivatives, including the three isolated natural bromophenols, showed that the existence of an electron donating group or atom with a non-covalent electron pair at $C_4$ of the 2-bromophenol structure may be important in anti-scuticociliate activity. These findings suggest that the extract and bromophenol derivatives of P. morrowii may provide useful alternatives in aquaculture anti-scuticociliate therapies.

Screening and Characterization of Lactic Acid Bacteria Strains with Anti-inflammatory Activities through in vitro and Caenorhabditis elegans Model Testing

  • The present study was conducted to screen candidate probiotic strains for anti-inflammatory activity. Initially, a nitric oxide (NO) assay was used to test selected candidate probiotic strains for anti-inflammatory activity in cultures of the murine macrophage cell line, RAW 264.7. Then, the in vitro probiotic properties of the strains, including bile tolerance, acid resistance, and growth in skim milk media, were investigated. We also performed an in vitro hydrophobicity test and an intestinal adhesion assay using Caenorhabditis elegans as a surrogate in vivo model. From our screening, we obtained 4 probiotic candidate lactic acid bacteria (LAB) strains based on their anti-inflammatory activity in lipopolysaccharide (LPS)-stimulated RAW 264.7 cell cultures and the results of the in vitro and in vivo probiotic property assessments. Molecular characterization using 16S rDNA sequencing analysis identified the 4 LAB strains as Lactobacillus plantarum. The selected L. plantarum strains (CAU1054, CAU1055, CAU1064, and CAU1106) were found to possess desirable in vitro and in vivo probiotic properties, and these strains are good candidates for further investigations in animal models and human clinical studies to elucidate the mechanisms underlying their anti-inflammatory activities.

Metabolism of Poncirin by Intestinal Bacteria (장내 세균에 의한 폰시린의 대사)

  • Poncirin which is one of the flavanone rhamnoglucosides showed anti-inflammatory activity as the major component of fruit of Poncirus trifoliata. Poncirin did not show antiinflammatory effect when it was intraperitoneally administered, but it was very effective when orally administered. Poncirin was not metabolized by blood and liver enzymes but by intestinal bacteria of human and rats. Among the human intestinal bacteria, Streptococcus Y-25 converted poncirin to ponciretin through the poncirenin and Staphylococcus Y-88 converted to ponciretin directly.

Anti-bacterial effects of the Na2O-TiO2-P2O5 glass-ceramics added Ag+ ion (은이온을 함유한 Na2O-TiO2-P2O5 glass-ceramic의 항균 특성)

  • Glass-ceramics composed of $5Na_2O-36CaO-10TiO_2-xP_2O_5$ could be obtained with $P_2O_5$ content as following procedure: 1) leaching out $Ca_3(PO_4)_2$ crystals between $Ca_3(PO_4)_2$ and $NaTi_2(PO_4)_3$ selectively in 1 N HCl solution for 2 days, and 2) exchanging $Na^+$ ion to $Ag^+$ ion in Ag($NO_3$) solution for 1 day. Scanning electron microscope (SEM) and X-rat diffractometer (XRD) were measured to verify the proper synthesis of glass-ceramics. And anti-bacteria test was accomplished using Staphylococus aureus bacteria. In the results of anti-bacteria test, the bacteria were died perfectly after 3 hours.

Comparing Medical Efficacy of Socheongyong-tang with Lactic Acid Bacteria Fermented Socheongyong-tang (소청룡탕과 유산균 발효 소청룡탕의 약리효능의 비교)

  • To compare the medical efficacy of original Socheongyong-tang with fermented Socheongyong-tang, we've studied the two medicines according to the search for optimal bacteria and optimal conditions, component analysis, assessment of medical efficacy and toxicity, and have the result below. The results were obtained as follows: Considering bacterial growth, $CO_2$ gas emission and pH, we examined that using 3 kinds of bacteria(S. cerevisiae KCTC 7913, L. casei KCTC 3109, L. brevis KCTC 3102) is desirable. There is no main difference in optimum conditions between incubator and shaking incubator. And it is considered that ideal fermentation time is 2 days after vaccination. As the result of componential analysis of before and after fermentation, there's a noticeable decrease of total sugar and protein. But there's no alterations in total phenolics compounds and in total flavonoid compounds that influence on medical effect. The result was interpreted that it can promote the assimilation of herbal decoction after fermentation. As the result of medical efficacy assessment, we can check out that there is more anti-oxidating effects in fermented Socheongyong-tang, whereas anti-inflammatory effects and obesity-preventing effects were favorable in original Socheongyong-tang. And there is no main difference of whitening and COX-2 removing effects between before and after the fermentation. As a result of assessing weight change, hepatotoxicity and nephrotoxicity, we can not notice any unusual difference between before and after the fermentation. According to the results above, it is considered that we checked out the optimal bacteria and optimal conditions, advantages and disadvantages of the medical efficacy of original Socheongyong-tang and fermented Socheongyong-tang. And we suggest that there will have to be a following in-depth and systematic research on this subject in the future.

Anti-tumor and immuno-stimulating activity of fungal polysaccharides

  • Low molecular weight molecules and high molecular weight substances were found to have anti-tumor and immuno-modulating activity. Previously polysaccharides have been received much attention because of adhesives, food additives or animal foods (Whistler et al., 1976). In effort of developing new anti-tumor substances with low toxicity, numerous polysaccharides from yeast, algae, bacteria, higher plants and especially fungi have been investigated for anti-tumor and immuno-modulating activities. Thus the high molecular weight molecule was reported to have anti-tumor activity through host mediated immunity. In this brief article, attention will be paid to polysaccharides which is especially fungal origin.

Antioxidant and Whitening Effects of the Fermentation of Barley Seeds (Hordeum vulgare L.) Using Lactic Acid Bacteria (유산균을 이용한 보리의 발효를 통한 항산화 및 미백 효과)

  • Barley (Hordeum vulgare L.), of the Poaceae/Gramineae family, is a common grain in the surrounding area. It has been used in Ancient Egyptian medicine and it has been used worldwide for many years as food and as an ingredient in beer. Barley has been reported to have anti-inflammatory, anti -carcinogenic and anti-diabetic effects. So far, a lot of research has been done on barley but the effects of fermented barley seeds with lactic acid bacteria have not been studied largely. In this study, we investigated the effects of ethanol-extracted barley seeds after their fermentation with lactic acid bacteria. The biological activities of fermented barley seeds with lactic acid bacteria and non-fermented barley seeds were analyzed for total polyphenol content, total flavonoid content, DPPH radical scavenging, superoxide dismutase-like activity and tyrosinase inhibition. These results showed that fermented barley seeds with lactic acid bacteria have more advanced anti-oxidant and whitening properties than non-fermented barley seeds. Hence, we suggest that fermenting barley seeds with lactic acid bacteria can be an impressive material in the food and cosmetic industries.
