This study was evaluated the antiatopic activity of silkworm feces extracts in DNCBinduced NC/Nga atopy mice. It was found that each level of IgE and histamine in blood was significantly decreased in the silkworm feces extracts treatment groups, compared with the DNCB-induced atopy control group. When the silkworm feces extracts was applied to the atopy mice, it could be observed that its skin recovered to normal condition with the skin surface being clean and smooth without any tissue. The results suggest that the application of silkworm feces extracts effect on atopic model through a inhibition of histamine emissions with reducing the levels of blood IgE in NC/Nga atopy mice.
Objectives : The aim of this study is to investigate the effect of natural carbonated hot spring water (NCHW) on DNCB-induced NC/Nga mice, an atopic dermatitis (AD) model. Methods :At first, NC/Nga mice were prepared and induced to have atopic lesion on their back skin by dinitrochlorobenzene (DNCB). They swam for 20 minutes everyday in tanks where the one is filled with $28^{\circ}C$ NCHW, the other is with $25^{\circ}C$ carbonated hot spring water and another is with artificial carbonated hot spring water (ACHW). After 3 weeks, We assessed the skin clinical score and macroscopic appearance, total IgE, IgG1 levels, WBC differential counting, IFN-${\gamma}$, IL-4, IL-13, TNF-${\alpha}$ production levels and histological changes. Results : There are meaningful results of improving atopic lesion-state by relieving the count of total IgE, IgG1 levels, WBC differential counting, IFN-${\gamma}$, IL-4, IL-13, TNF-${\alpha}$ production levels and recovering skin clinical score in the group with $28^{\circ}C$ NCHW in comparison with the other groups. Conclusions : The NCHW may have potential as an effective treatment for atopic dermatitis.
Objectives : The aim of this study is to investigate anti-atopic dermatitis effect using ato-tang.Methods : Ato-tang was external treatment to NC/Nga mice for 4 weeks, where atopic dermatitis was induced by DNCB at 1% and 0.4% for 3 weeks. Atopic dermatitis index score was measured using eye observation and picture evaluation. The histopathological change of dorsal skin was observed by hematoxylin and eosin (H&E) staining. Cytokines including IL-4, IL-5 and IL-13 were measured by Luminex or reverse transcriptase polymerase chain reaction (RT-PCR) analysis and immunoglobulin E (IgE) was measured by ELISA reader.Results : The dorsal skin of Ato-tang group showed decrease in erythema, pruritus, dry skin, edema, excoriation, erosion and lichenification level through naked eye observations. Immunoglobulin cell infiltration and the thickness of epidermis were significantly decreased in the dorsal skin compared to control. Production of Th2 cytokines (IL-4, IL-5, IL-13) and IgE level in serum were all significantly decreased, in comparison with control. In addition, mRNA expression level of Th2 cytokines (IL-4, IL-5, IL-13) in spleen was decreased, in comparison with control.Conclusion : The results indicated that external treatment of ato was improved skin barrier function in the symptoms of atopic dermatitis disease. Also, atopic dermatitis factors where cytokine as well as immunoglobulin E in serum and mRNA expression were decreased, respectively, in comparison with control. Therefore, we suggest that ato could be effectively used as a external therapeutic drug based on atopic dermatitis factors.

Allergies are immediate hypersensitive responses to antigens and interleukin (IL)-4 is involved in the initiation and development of allergic responses. $Rb_1$ has been known to have a variety of biological activities including anti-inflammatory activity, but the effect of $Rb_1$ on allergic responses is not known yet. The present study was undertaken to examine whether $Rb_1$ has an inhibitory effect on allergic response in mouse model. In allergic mouse model, our results showed that topical application of $Rb_1$ on atopic dermatitis (AD)-like skin lesions improved skin condition and inhibited starching behaviors. In addition, $Rb_1$ application not only suppressed mRNA expression of IL-4 and IL-10, but also prevented the nuclear factor of activated T cells 1 transcription. Moreover, $Rb_1$ application suppressed IL-4's secretion. Taken together, these results suggest that $Rb_1$ has a potent inhibitory effect in AD-related T cell cytokine production and may be a candidate for therapeutic agent in allergy.
Objectives : HRHDT has been known as a useful prescription with antibiotic, anti-inflammatory, antioxidative and immunosuppressive activity. To evaluate anti-atopic dermatitis effect of HRHDT, we treated HRHDT in NC/Nga mice model skin induced contact hypersensitivity. Methods : Contact hypersensitivity, a local inflammatory response of skin, was induced by spreading the back skin of NC/Nga mice with 0.4${\sim}$1% DNCB. HRHDT was prepared by dissolving 3% HRHDT in solution and treated 3 weeks on the back skin. Results : HRHDT significantly reduced TEWL and erythema by 0.4${\sim}$1% of DNCB treatment compared to control group. HRHDT also reduced IgE on serum obtained from blood of DNCB-treated mice. Conclusions : These results showed that HRHDT could be used as a pharmaceutical material with anti-atopic dermatitis effect by reducing IgE in contact, hypersensitivity atopic dermatitis NC/Nga mice model by DNCB.
Allergic diseases have rapidly increased in recent years. Mast cells play a critical role in IgE-mediated allergy responses and, therefore, closely associated with rhinitis, asthma, and atopic dermatitis. We studied anti-allergic effect of Penthorum chinense extract (PCE) in vitro and in vivo. PCE inhibited the degranulation of mast cells by antigen stimulation and its effect was dose-dependent and reversible in mast cells. PCE also suppressed the production of inflammatory cytokines such as TNF-${\alpha}$ and IL-4 by antigen in mast cells. Mechanistically, PCE inhibited the activation of Syk/LAT pathway which is a key signaling pathway for the activation of mast cells by antigen. Notably, PCE suppressed IgE-mediated allergic responses by antigen in mice. These results strongly suggest that PCE is a potential candidate for anti-allergic treatment.
In order to investigate the possibility of Kamisamultang(KMSMT) as therapeutic for the treatment of atopic dermatitis(AD), cytotoxicity, anti-oxidant activity, modulatory and suppression activities of KMSMT were tested. 90% or higher cell viability was observed in all tested groups from 25 to 200 ${\mu}g/m{\ell}$ using Raw 264.7 cells. KMSMT showed dose-dependent DPPH scavenging activity, with more than 80% scavenging activities at 400 and 800 ${\mu}g/m{\ell}$ concentrations. KMSMT showed dose-dependent suppression activity of reactive oxygen species(ROS) production, especially at 200 ${\mu}g/m{\ell}$ of 42.6%. KMSMT decreased nitric oxide(NO) production activity dose dependently, expecially at 200 ${\mu}g/m{\ell}$ of 30.9%. IL-$1{\beta}$, IL-6, MCP-1, TNF-${\alpha}$ production rate were decreased by 45.7%, 15.5%, 8.9%, 16.5% respectively when Raw 264.7 cells were treated with LPS and with KMSMT of 200 ${\mu}g/m{\ell}$. However, only IL-$1{\beta}$ and MCP-1 showed significant changes. The results above strongly suggest the modulatory and suppressive effect of KMSMT. The results above indicate that KMSMT significantly reduces the effect of oxidative and inflammatory cytokines. The use of KMSMT in atopic dermatitis can be widely suggested.
The in vivo and in vitro effects of oriental herb extracts of Cassia obtusifolia, Taraxacum platycarpum and Ulmusmacrocarpa on anti-atopic allergic reaction were evaluated in this study. A mixture of these extracts exhibited more potent anti-allergic activities in human mast cells than those from individual extracts. The herbal mixture significantly inhibited the release of compound 48/80-induced $\beta$-hexosaminidase release in the human mast cell line, HMC-1. The mixture also suppressed the production of PMA and A23187-induced inflammatory cytokines in HMC-1 cells. To further investigate the in vivo effects of the herbal mixture, a Dermatophagoides farinae (DF)-induced atopic dermatitis mouse model was utilized. Oral administration of the herbal mixture significantly decreased the ear thickness and swelling in DF treated NC/Nga mice in a dose dependent manner. Furthermore, serum levels of IgE and interleukin-4 (IL-4) were significantly decreased, whereas interferon-gamma (IFN-$\gamma$) levels were increased in the mixture administrated groups when compared to the control. Taken together, our data indicate the possibility of using a mixture of the oriental herb extract to relieve symptoms of atopic dermatitis.

To investigate the clinical aspects of CHT in atopic dermatitis (AD) treatments, the effect of CHT in anti-oxidative and anti-inflammatory cytokines were tested. 100% or higher cell viability was observed in all tested groups from 25 to 200 ${\mu}g/m{\ell}$ using Raw 264.7 cells. CHT showed dose-dependent DPPH scavenging activity, with more than 90% scavenging activities at 800 ${\mu}g/m{\ell}$ concentrations. CHT showed dose-dependent suppression activity of ROS production, especially at 200 ${\mu}g/m{\ell}$ of 37.5%. CHT decreased NO production activity, with significant decrease of 33.2% at 200 ${\mu}g/m{\ell}$. IL-6, MCP-1, TNF-${\alpha}$ production rate were decreased by approximately 25% when Raw 264.7 cells were treated with LPS and with CHT of 200 ${\mu}g/m{\ell}$. Also, IL-$1{\beta}$ production rate was decreased by 25% at 100 ${\mu}g/m{\ell}$. The results above indicate that CHT significantly reduces the effect of oxidative and inflammatory cytokines. The use of CHT in dermatitis can be widely suggested.

The experiment was conducted to validate anti-inflammatory effects of pinitol from bean. It was evaluated for some molecule targets by wound healing assay and RT-PCR. The results of wound healing assay was shown dose-dependent inhibition of cell migration in cancer cells and inhibited RNA expression of ICAM-1, CD 44, MMP-17, MMP-14 and ARF2. Immune suppression activity in a mouse provoked by DNFB observed that inflammatory reaction with pinitol were reduced ear swelling and inflammatory cells infiltration in mouse atopic models. The result confirmed that pinitol have the effect of dose-dependent immune suppression activity.
