Objective: Recently the demands for the effective and safe depigmentative and anti-aging agents of the skin have increased due to the medical, pharmaceutical and cosmetic reasons. The purpose of this study is to investigate the MKG(Methanol Extract of Kaempferia galanga) and their dermal bioactivity properties related to cosmeceuticals such as depigmentation. Methods: We assessed inhibitory effects of MKG on melanin production in B16/F10 melanoma cells, on mushroom tyrosinase activity, effects of MKG on the expression tyrosinase, TRP-1, TRP-2, GSK-$3{\beta}$, CREB, MITF in B16/F10 melanoma cells without cytotoxicity range. Cell viability was measured by MTT assay and tyrosinase activity was assessed using by DOPA staining, western-blot analysis. We measured inhibition of melanin synthesis and tyrosinase activity by down-regulation of melanogenic enzyme expressions in ${\alpha}$-MSH induced melanogenesis B16/F10 melanoma cells. Results: MKG inhibited tyrosinase-activity, total melanin contents and dendrite out-growth. MKG inhibited melanogenesis by down-regulation of tyorsinase, TRP-1, TRP-2, CREB, and MITF in B16/F10 cells. The treatment with MKG at the 12.5, $25{\mu}g/ml$ level significantly inhibited the melanin synthesis induced ${\alpha}$-MSH in B16/F10 melanoma cells compared with untreated control. Conclusion: These results suggest that MKG inhibit melanin biosynthesis which is involved in hyper-pigmentation. So MKG is considered to be used as a whitening components reducing cytotoxicity.
UV irradiation causes skin-aging involving coarse wrinkles, thickening, dyspigmentation, and rough skin surface. This study was carried out to develop health & functional food by using Korean red ginsneg and Fagopyrum esculentum extract mixture (RGFE) for prevention of skin wrinkles. The RGFE-treated group showed the more effective collagenase inhibition rate than the red ginseng (RG)-treated group. To investigate photo protective effects of RGFE on UV-induced damaged skin, SKH hairless male mice were orally administerd RGFE and regional treatment and irradiated with UV for up to 8 weeks. In RGFE-treated group, better skin, and less wrinkle formation were observed compared with UV group. Epidermal thickness of hairless mouse was significantly decreased in RGFE, RG, and Fagopyrum esculentum (FE) groups compared with UV group. These results demonstrate RGFE have photo-protective effects on UV-damaged hairless mouse skin.

The radish skin and radish greens are an edible part of the radish. But they are removed before eating the radish and used as a byproduct or an animal feed material because of their tough and rough texture. Melanin is a pigment that gives colour to our skin. But increased production of melanin can turn into benign or malignant tumours. These days due to global warming, the amount of Ultra violet (UVB) rays has been extensively increased with sunlight. Due to this, a phenomenon called exogenous photo aging is widely observed for all skin colour and types. As a result of this phenomenon, a set of enzymes called matrix metalloproteinases (MMP's) that serves as degradation enzymes for extracellular matrix proteins mainly collagen is increased, causing depletion in collagen and resulting in early wrinkles formation. Therefore in our study we used the murine melanoma cell line B16/F10 to study the melanogenesis inhibition by Heated radish extract (HRE) in vitro and we used HRM-2 hair less mice exposed to artificial UVB for checking the efficacy of Heated radish extract in vivo. Furthermore, we prepared a 3% Heated radish extract (HRE) cream and checked its effects on human skin. Our results have clearly demonstrated that Heated radish extract (HRE) have potently suppressed the tyrosinase activity and melanin production in B16/F10 cells. It had also reduced the expression of components involved in melanin production pathway both transcriptionally and transitionally. In in vivo studies, HRE had potently suppressed the expression of MMP's and reduced the wrinkle formation and inhibited collagen degradation. Moreover, on human skin, ginseng cream increased the resilience, skin moisture and enhanced the skin tone. Therefore in light of these findings, we conclude that HRE is an excellent skin whitening and antiaging product.
Alzheimers disease (AD) is the most prevalent form of neurodegenerations associated with aging in the human population. This disease is characterized by the extracellular deposition of beta-amyloid (A ${\beta}$) peptide in cerebral plaques. The A ${\beta}$ peptide is derived from the ${\beta}$-amyloid precursor protein ( ${\beta}$APP). Photolytic processing of ${\beta}$APP by ${\beta}$-secretase(beta-site APP-cleaving enzyme, BASE) and ${\gamma}$-secretase generates the A ${\beta}$ peptide. Several lines of evidence support that A ${\beta}$-induced neuronal cell death is major mechanisms of development of AD. Accordingly, the ${\beta}$-and ${\gamma}$-secretase have been implicated to be excellent targets for the treatment of AD. We previously found that sesaminol glucosides have improving effect on memory functions through anti-oxidative mechanism. In this study, to elucidate possible other mechanism (inhibition of ${\beta}$-and ${\gamma}$-secretase) of sesaminol glucosides, we examined the improving effect of sesaminol glucosides in the scopolamine (1 mg/kg/mouse)-induced memory dysfunction using water maze test in the mice. Sesaminol glucosides (3.75, 7.5 mg/kg/6ml/day p.o., for 3 weeks) reversed the latency time, distance and velocity by scopolamine in dose dependent manner. Next, ${\beta}$-and ${\gamma}$-secretase activities were determined in different regions of brain. Sesaminol glucosides dose-dependently attenuated scopolamine-induced ${\beta}$-secretase activities in cortex and hippocampous and ${\gamma}$-secretase in cortex. This study therefore suggests that sesaminol glucosides may be a useful agent for prevention of the development or progression of AD, and its inhibitory effect on secretase may play a role in the improving action of sesaminol glucosides on memory function.
사포닌은 동양의 전통적인 약재로 널리 알려진 인삼의 주요한 성분이다. 사포닌의 다양한 생물학적 효능이 밝혀져 있으나, 피부재생과 관련된 효능은 현재까지도 명백하지 않다. 본 연구에서는 세포외 시스템에서 사포닌의 항산화 효과 뿐만 아니라 사람 진피 섬유아세포에서 기질금속단백질 분해효소(MMP)에 대한 효능이 조사되었다. 먼저 MTT assay를 이용한 세포생존력에 대한 사포닌의 효능을 조사한 결과, 10 ${\mu}g$/ml 이하의 농도에서 사포닌은 세포생존력을 증가시켰으나 25 ${\mu}g$/ml 이상의 농도에서는 세포독성을 나타내었다. 항산화에 대한 사포닌 효능을 조사한 결과, 1 ${\mu}g$/ml 이상의 농도에서 사포닌은 환원력 뿐만 $H_2O_2$에 대한 억제 효능을 보여주었다. 특히, DNA 산화에 대한 보호 효과도 나타내었다. 더욱이 gelatin 및 casein zymography 시험결과, 사포닌은 MMP-2를 활성화 시키고 MMP-1의 활성을 증가시키는 것으로 보아, 항노화 및 피부재생 약효제로 잠재적인 가능성이 기대된다.

The effects of Lavandula angustifolia extract fermented with Pediococcus pentosaceus DK1 on UVB-mediated MMP-1 expression and collagen decrease in human skin fibroblasts were determined, and the conversion of its components was also analyzed. Fermentation was performed at varying L. angustifolia extract and MRS medium concentrations, and optimal fermentation conditions were selected. L. angustifolia extracts showed decreased cytotoxicity after fermentation in the fibroblasts. UVB-irradiated fibroblasts treated with fermented L. angustifolia extract showed MMP-1 expression 8.2-14.0% lower than that in UVB-irradiated fibroblasts treated with non-fermented extract. This was observed even at fermented extract concentrations lower than those of non-fermented extracts. Fibroblasts treated with fermented L. angustifolia extract showed 20% less reduction in collagen production upon UVB irradiation than those treated with non-fermented extracts. UVB-irradiated fibroblasts treated with fermented L. angustifolia extracts showed 50% higher inhibition of ROS generation than those treated with non-fermented extract. Luteolin and apigenin glycosides of L. angustifolia were converted during fermentation, and identified using RP-HPLC and LC/ESI-MS. Therefore, the effects of L. angustifolia extract on MMP-1 expression and collagen decrease in UVB-irradiated human skin fibroblasts were increased through fermentation by P. pentosaceus.
활성산소종으로 유도되는 연골 세포의 apoptosis는 퇴행성 관절염의 발병 기전에 중요한 역할을 한다. Schisandrin 속의 과일에서 발견되는 생체 활성 화합물인 schisandrin A는 여러 가지 약리학적 작용을 하는 것으로 보고되고 있다. 현재까지 schisandrin A의 유도체들의 항산화 효과에 대해서는 여러 연구가 보고되었지만, schisandrin A의 항산화 효능의 분자 기전은 아직 미해결 상태로 남아 있다. 본 연구는 SW1353 인간 연골세포에서 산화적 스트레스($H_2O_2$)에 대한 schisandrin A의 세포 보호 여부를 조사하였다. 본 연구의 결과에 의하면 schisandrin A는 PARP 단백질의 분해와 caspase-3의 활성 차단을 통해 $H_2O_2$에 의해 유도된 성장 억제와 apoptosis를 유의적으로 억제하였다. 이러한 schisandrin A의 anti-apoptotic 효과는 미토콘드리아 기능 손상의 억제와 pro-apoptotic Bax의 발현 증가 및 anti-apoptotic Bcl-2의 발현 감소의 차단과도 관련이 있었다. 또한, schisandrin A는 ROS의 생성과 DNA 손상 마커인 H2AX의 인산화도 효과적으로 저해하였다. 따라서 SW1353 연골세포에서 schisandrin A는 산화적 스트레스에 의한 ROS 생성의 억제를 통하여 apoptosis와 DNA 손상을 보호하였음을 알 수 있었다. 결론적으로 본 연구의 결과는 schisandrin A가 ROS의 과잉 생산으로 인한 산화적 장애에 치료적 잠재력이 있음을 보여준다.
본 연구는 패션 프룻(passion Fruit) 열매의 과육과 씨 추출물의 항염증, 항산화, 항노화 및 피부 미백의 기능성 효과에 관한 것이다. 패션 프룻 열매의 추출물은 피부 첩포에 의한 일차 자극 인체적용시험 결과 피부 자극 지수는 0.00으로 판정되었다. 또한 염증매개물질 LPS (Lipopolysaccharides)를 처리하여 Raw 264.7 대식세포를 활성화시켜 NO를 유발하고 패션 프룻 열매 추출물과 반응시켰을 때 LPS만 처리한 그룹의 NO 발생량 보다 현저히 낮은 NO 발생량을 보임으로써 패션 프룻 열매 추출물의 우수한 항염증 효과를 확인할 수 있었다. 그리고 DPPH (2,2-diphenyl-1-picrylhydrazyl) 시험을 통해 패션 프룻 열매 추출물이 5.11%의 ABTS 라디칼 소거능을 보이면서 항산화 효과가 있음을 확인하였다. 피부의 콜라겐 I 형의 합성은 노화와 다양한 피부 질병의 척도인데 인간의 프로콜라겐 I 알파 1 활성도 분석을 통하여 패션 프룻은 프로콜라겐의 합성을 약간 감소시킴을 확인하였다. 또한 멜라닌 억제 시험을 통하여 패션 프룻 열매 추출물의 미백 효능을 확인하였는데 대조군으로 식약처에서 고시한 미백 기능성 성분인 알부틴(arbutin)을 2% 이상 배합한 시료는 월귤나무에서 얻은 히드로 퀴논을 반응시켜 얻은 유기화합물로 일반적으로 백색 분말의 형태로 존재하며 멜라닌 생성을 촉진하는 티로시나아제를 억제하는 효능 효과가 있어 미백 개선의 기능성 성분으로 사용된다. 그러므로 이러한 자연에서 얻은 패션 프룻 열매 추출물의 미백 기능성 효능 확인은 화장품 및 식품의 기능성 원료의 개발에 기여할 수 있을 것으로 기대된다.
Toll 유사수용체의 TLR4는 세포자연사(apoptosis)와 관련하여 세포의 생존과 증식에 영향을 미치는 것으로 알려져 있다. 본 연구에서는 TLR4의 활성이 부인과 질환 특이적 종양세포의 세포사멸기작에 어떠한 영향을 미치는지 살펴보았다. TLR4의 활성에 의한 세포자연사를 확인하기 위하여 부인암 조직(자궁경부암, 자궁내막암, 난소암)에서 종양세포를 분리하여 초대배양시스템을 구축하였고, lipopolysaccharide (LPS)에 의한 TLR4의 활성유도 과정에서 종양세포의 형태학적 변화를 살펴보았다. 또한, TLR4 매개성 세포사멸 기작을 확인하기 위하여 역전사 중합효소 연쇄반응(RT-PCR)을 통해 유전자 분석을 진행하였다. 연구 결과, 부인암의 초대배양세포에서 세포접촉저지(contact inhibition)현상이 감소되었고, 세포의 배가시간(doubling time)이 단축되어, 종양세포의 성장률 변화를 확인하였다(p<0.05). 자궁근육층(정상조직)의 초대배양세포에서는 민무늬근육 확인 인자인 ITGA5 (an alpha5 integrin marker)의 유전자 발현이 나타났으나, 자궁경부조직의 초대배양세포에서는 발현변화를 확인할 수 없었다. 종양세포의 유전자분석 결과에서 p53과 같은 종양억제인자의 발현이 유의적으로 증가한 반면(p<0.05), 세포사멸 신호기작과 관련하여 TLR4와 Caspase-3의 발현은 감소하였다(Caspase-3, p<0.05). LPS를 처리한 종양세포에서는 LPS 비처리군과 비교 시, TLR4의 발현증가와 함께 Caspase-3의 발현변화가 동반되었다. 이러한 결과들은 TLR4 매개성 apoptosis 유도가 종양세포의 증식억제에 중요한 영향을 미치는 것을 의미하며, TLR4 신호기작을 이용한 종양세포의 새로운 치료적 접근법을 제시할 것으로 기대한다.
본 연구는 수분함유량이 많아 장기간의 보관이 어려운 아이스플랜트의 저장성을 높이기 위하여 발효하였을 경우, triglyceride, cholesterol 합성 및 tyrosinase 활성 등의 변화를 조사하였다. 유기산의 함량은 발효 후에는 lactic acid가 $23.672{\pm}3.74mg/g$으로 가장 많았으며 malic acid, acetic acid citric acid의 순이었다. 발효 전 후의 cyclitol 화합물의 변화를 보면, D-pinitol은 각각 $6.670{\pm}0.187mg/g$과 $5.541{\pm}0.088mg/g$으로 감소하였으나, myo-inositol은 $0.653{\pm}0.022mg/g$과 $3.848{\pm}0.216mg/g$, D-chiro-inositol은 $3.848{\pm}0.216mg/g$과 $4.945{\pm}0.098mg/g$으로 증가하였다. HepG2 세포 내 중성지방의 감소율은 발효 전 후에 각각 $3.04{\pm}1.70{\sim}35.73{\pm}5.06%$와 $5.13{\pm}0.90{\sim}53.32{\pm}1.79%$로서 발효 후 저해효과가 약간 높게 나타났으며, 대체로 극성이 큰 n-butanol과 aqueous 분획에서는 각각 $25.93{\pm}1.47{\sim}35.73{\pm}5.06%$과 $26.26{\pm}2.07{\sim}53.32{\pm}1.79%$로서 발효 후에 비교적 큰 폭으로 중성지방의 축적을 저해하였다. HepG2 세포 내 콜레스테롤의 감소율도 발효 전 후에 각각 $4.02{\pm}1.50{\sim}56.34{\pm}5.55%$와 $6.56{\pm}1.46{\sim}64.52{\pm}5.50%$ 정도로서 발효 후에 콜레스테롤의 합성 저해효과가 약간 크게 나타났다. 특히 처리농도가 $100{\mu}g/ml$ 이상에서 n-hexane, chloroform 및 ethylacetate 분획에서는 발효 전 후에 각각 $8.15{\pm}0.60{\sim}9.16{\pm}0.71%$와 $9.81{\pm}1.26{\sim}12.96{\pm}0.97%$로서 저해효과가 크지 않았으나, n-butanol과 aqueous 분획에서는 발효 전 후에 각각 $28.68{\pm}0.86{\sim}56.34{\pm}5.55%$와 $28.32{\pm}2.87{\sim}64.52{\pm}5.50%$로서 비교적 큰 폭으로 콜레스테롤의 합성을 저해하였다. Tyrosinase 활성억제효과도 발효 후에 약간 증가하는 경향이었다. 기질로서 L-tyrosine을 사용하였을 경우, 극성이 큰 aqueous 분획에서 발효 전 후에 각각 $60.58{\pm}4.035$와 $63.35{\pm}4.35%$로서 저해활성이 가장 높았으며, 양성대조군(arbutin, $100{\mu}g/ml$)의 72%에 달하는 활성이었다. 또한 기질로서 L-DOPA을 사용하였을 경우에도 aqueous 분획에서 발효 전 후에 각각 $56.85{\pm}1.57%$와 $59.38{\pm}1.74%$로서 저해활성이 가장 높았으며 양성대조군(kojic acid, $100{\mu}g/ml$)의 88% 이상의 높은 활성을 가지고 있었다. 전체적으로 발효 아이스플랜트의 중성지방 및 콜레스테롤 합성억제 효과, tyrosinase 활성억제효과는 대체로 발효 전에 비하여 다소 향상되었다. 특히 극성이 큰 용매의 분획추출물에서 높은 활성을 나타내었다. 이러한 결과를 바탕으로 발효 등의 다양한 가공방법이 개발되어 저장성과 활용도가 높아질 수 있는 계기가 되기를 기대한다.

회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
