Evaluation of efficacy of a commercial vaccine stress-relieving agent to reduce the adverse effects of foot-and-mouth disease vaccination site in pig (시판 백신접종 스트레스 완화제의 돼지 구제역 백신 접종부위 부작용 발생 저감 효과 평가)

  • In this study, we evaluated the efficacy of a commercial vaccine stress-relieving agent (Anti VS-2 injection) to reduce the adverse effects of foot-and-mouth disease (FMD) vaccination site in 120 pigs on two tested pig farms. The results showed that incidence of pig with abnormal meat, size of abnormal meat and weight of discarded abnormal meat are significantly decreased in Anti VS-2 injection-supplemented FMD vaccinated pig groups (n=60) compared to the non-supplemented FMD vaccinated pig groups (n=60) (P<0.05). And the average economic loss by discard of abnormal meat were estimated 99,382 won or 21,514 won in the non-supplemented FMD vaccinated groups or in the Anti VS-2 injection-supplemented FMD vaccinated groups, respectively, showing that the cost saving effect was 78% (77,869 won). In conclusion, it is expected to reduce the incidence and severity of abnormal meat in the FMD vaccinated pigs and to contribute not only to the reduced economic loss of pig farmers but also to the promotion of food hygiene when the Anti VS-2 injection is mixed with FMD vaccine.

Anti-apoptotic Effect of Steam Exploded Quercus variabilis

  • We hypothesized that the extract from steam exploded Q. variabilis might be cytoprotective for tenofibroblasts cells during oxidative stress. In the present study, the extracts obtained from steam exploded (severity log Ro 4.68) Q. variabilis contained high quantities of phenolics and flavonoids contents. Also, the extracts appear to have, on these tenofibroblasts, a protective effect against oxidative stress. Tenofibroblasts cells incubated with the extracts and stressed with $H_2O_2$ showed an increase in cell viability by MTT assay. The extracts is found to inhibit $H_2O_2$-induced apoptosis in tenofibroblasts cells, as shown by Annexin V/PI double staining analysis. Western blot data showed that in the extracts/$H_2O_2$-treated cells, the extracts inhibited the $H_2O_2$-dependent phosphorylation of ERK and p38. From these results, it is suggested that the extracts showed the protective effect on $H_2O_2$-mediated oxidative stress. The main chemical compounds of the extract was identified as 1,8-cineole by GC-MS analysis. The anti-apoptosis activity is accordingly believed to be attributable to the 1,8-cineole.

Intrathecal administration of naringenin improves motor dysfunction and neuropathic pain following compression spinal cord injury in rats: relevance to its antioxidant and anti-inflammatory activities

  • Background: Spinal cord injury (SCI) is one of the most debilitating disorders throughout the world, causing persistent sensory-motor dysfunction, with no effective treatment. Oxidative stress and inflammatory responses play key roles in the secondary phase of SCI. Naringenin (NAR) is a natural flavonoid with known anti-inflammatory and antioxidative properties. This study aims at evaluating the effects of intrathecal NAR administration on sensory-motor disability after SCI. Methods: Animals underwent a severe compression injury using an aneurysm clip. About 30 minutes after surgery, NAR was injected intrathecally at the doses of 5, 10, and 15 mM in 20 µL volumes. For the assessment of neuropathic pain and locomotor function, acetone drop, hot plate, inclined plane, and Basso, Beattie, Bresnahan tests were carried out weekly till day 28 post-SCI. Effects of NAR on matrix metalloproteinase (MMP)-2 and MMP-9 activity was appraised by gelatin zymography. Also, histopathological analyses and serum levels of glutathione (GSH), catalase and nitrite were measured in different groups. Results: NAR reduced neuropathic pain, improved locomotor function, and also attenuated SCI-induced weight loss weekly till day 28 post-SCI. Zymography analysis showed that NAR suppressed MMP-9 activity, whereas it increased that of MMP-2, indicating its anti-neuroinflammatory effects. Also, intrathecal NAR modified oxidative stress related markers GSH, catalase, and nitrite levels. Besides, the neuroprotective effect of NAR was corroborated through increased survival of sensory and motor neurons after SCI. Conclusions: These results suggest intrathecal NAR as a promising candidate for medical therapeutics for SCI-induced sensory and motor dysfunction.

An Experimental Study on the Anti-Stress Effect by Guibiondamtang (귀비온담탕(歸脾溫膽湯)의 항(抗)스트레스 효과(效果)에 관(關)한 실험적(實驗的) 연구(硏究))

  • This experiment was conducted to study anti-stress effects of Guibiondamtang on several measures of stress, which were number of lever pressing, weight changes, water intake and gastroduodenal ulcer. The following results were observed: 1. There were no significant differences between the control group and the sample group in total number of lever pressing. And no significant differences were observed in the shock duration and the shock length. 2. The median water intake during the stress was 25ml for the sample group and 17ml for the control group and the difference was not statistically significant. 3. The median body weight losess after the psychological stress was given were 28g for the sample group and 35g for the control group and the difference was not statistically significant. 4. The mean frequencies of gastric ulcers induced by the psychological stress were 0.3 for the sample group and 2.3 for the control group and the difference was statistically significant. 5. The mean lengths of gastric ulcers induced by the psychological stress were 0.3mm for the sample group and 2.8mm for the control group and the difference was statistically significant. 6. The mean ratings of gastric ulcers induced by the psychological stress were 0.7 for the sample group and 1.9 for the control group and the difference was stastically significant. Thus it may be concluded that Guibiondamtang has an antistress effect.

Anti-stress effects of ginseng via down-regulation of tyrosine hydroxylase (TH) and dopamine ${\beta}$-hydroxylase (DBH) gene expression in immobilization-stressed rats and PC12 cells

  • Catecholamines are among the first molecules that displayed a kind of response to prolonged or repeated stress. It is well established that long-term stress leads to the induction of catecholamine biosynthetic enzymes such as tyrosine hydroxylase (TH) and dopamine ${\beta}$-hydroxylase (DBH) in adrenal medulla. The aim of the present study was to evaluate the effects of ginseng on TH and DBH mRNA expression. Repeated (2 h daily, 14 days) immobilization stress resulted in a significant increase of TH and DBH mRNA levels in rat adrenal medulla. However, ginseng treatment reversed the stress-induced increase of TH and DBH mRNA expression in the immobilization-stressed rats. Nicotine as a ligand of the nicotinic acetylcholine receptor (nAChR) in adrenal medulla stimulates catecholamine secretion and activates TH and DBH gene expression. Nicotine treatment increased mRNA levels of TH and DBH by 3.3- and 3.1-fold in PC12 cells. The ginseng total saponin exhibited a significant reversal in the nicotine-induced increase of TH and DBH mRNA expression, decreasing the mRNA levels of TH and DBH by 57.2% and 48.9%, respectively in PC12 cells. In conclusion, immobilization stress induced catecholamine biosynthetic enzymes gene expression, while ginseng appeared to restore homeostasis via suppression of TH and DBH gene expression. In part, the regulatory activity in the TH and DBH gene expression of ginseng may account for the anti-stress action produced by ginseng.

Evaluation on Anti-oxidant Activity and Anti-inflammatory Effects for the New Formulation of Gamisoyosan (가미소요산의 새로운 제형에 대한 항산화 활성 및 항염증 효능평가)

  • Objectives : Gamisoyosan (GMS) is a useful prescription for treating insomnia, dysmenorrhea and infertility induced by a stress. Also, GMS has been used traditionally to improve systemic circulation and biological energy production. The purpose of this study was to assess the anti-oxidant activity and anti-inflammatory effects of Gamisoyosan Formulation (Soft extract, GMS-SE). Methods : The biological activities such as anti-oxidant and anti-inflammatory effects were measured through cell line-based in vitro assay. We investigated the anti-oxidant properties of GMS-SE on the 1,1-diphenyl-2-picryhydrazyl (DPPH) radical, contents of total flavonoid and polyphenol. GMS-SE compared to butyl hydroxy anizole (BHA). Furthermore, based on this result the anti-inflammatory effects of GMS-SE have verified by mechanism from LPS- treated Raw264.7 macrophages. Results : The anti-oxidant activities of GMS-SE increased markedly, in a dose-dependent manner. The GMS-SE showed significant scavenging activity (GMS-SE $500{\mu}g/m{\ell}$ : $32.77{\pm}1.65%$, GMS-SE $1000{\mu}g/m{\ell}$ : $45.06{\pm}1.04%$ and BHA $100{\mu}g/m{\ell}$ : $39.25{\pm}2.41%$ for DPPH assay). and, The total phenolic compound and flavonoids contents of GMS-SE were $73.93{\pm}6.87{\mu}g/mg$ and $698.75{\pm}6.78{\mu}g/mg$. GMS-SE which is LPS has diminished in the LPS-induced release of inflammatory mediators (NO, iNOS, COX2 and PGE2) and pro-inflammatory cytokines (TNF-${\alpha}$, IL-6 and IL-$1{\beta}$) from the RAW264.7 macrophages. Moreover, GMS-SE inhibited the activation of phosphorylation of p38 and ERK MAPKs by induced LPS. Conclusion : The present results indicate that GMS-SE has an anti-oxidant and anti-inflammatory properties, therefore may be beneficial in diseases which related to oxidative stress-mediated inflammatory disorders.

Schisandrae Fructus ethanol extract attenuates particulate matter 2.5-induced inflammatory and oxidative responses by blocking the activation of the ROS-dependent NF-κB signaling pathway

  • BACKGROUND/OBJECTIVES: Schisandrae Fructus, the fruit of Schisandra chinensis Baill., has traditionally been used as a medicinal herb for the treatment of various diseases, and has proven its various pharmacological effects, including anti-inflammatory and antioxidant activities. In this study, we investigated the inhibitory effect of Schisandrae Fructus ethanol extract (SF) on inflammatory and oxidative stress in particulate matter 2.5 (PM2.5)-treated RAW 264.7 macrophages. MATERIALS/METHODS: To investigate the anti-inflammatory and antioxidant effects of SF in PM2.5-stimulated RAW 264.7 cells, the levels of pro-inflammatory mediator such as nitric oxide (NO) and prostaglandin E2 (PGE2), cytokines including interleukin (IL)-6 and IL-1β, and reactive oxygen species (ROS) were measured. To elucidate the mechanism underlying the effect of SF, the expression of genes involved in the generation of inflammatory factors was also investigated. We further evaluated the anti-inflammatory and antioxidant efficacy of SF against PM2.5 in the zebrafish model. RESULTS: The results indicated that SF treatment significantly inhibited the PM2.5-induced release of NO and PGE2, which was associated with decreased inducible NO synthase and cyclooxygenase-2 expression. SF also attenuated the PM2.5-induced expression of IL-6 and IL-1β, reducing their extracellular secretion. Moreover, SF suppressed the PM2.5-mediated translocation of nuclear factor-kappa B (NF-κB) from the cytosol into nuclei and the degradation of inhibitor IκB-α, indicating that SF exhibited anti-inflammatory effects by inhibiting the NF-κB signaling pathway. In addition, SF abolished PM2.5-induced generation of ROS, similar to the pretreatment of a ROS scavenger, but not by an inhibitor of NF-κB activity. Furthermore, SF showed strong protective effects against NO and ROS production in PM2.5-treated zebrafish larvae. CONCLUSIONS: Our findings suggest that SF exerts anti-inflammatory and antioxidant effects against PM2.5 through ROS-dependent down-regulating the NF-κB signaling pathway, and that SF can be a potential functional substance to prevent PM2.5-mediated inflammatory and oxidative damage.

Antistress effect of Gagamsanzointang on rat chronic stress model (가감산조인탕(加減酸棗仁湯)이 만성 스트레스 모델 쥐에 대한 항스트레스 효과)

  • Objectives : To observe the effect of 4 kinds of Korean medicine and complex extracts on the anti-stress of chronic mild stress Rat model, so as to find out the machanism of its pharmacological effects. Methods : 64 healthy SD Rats were randomly divided into 8 groups. Model Design by chronic stress(CSM), and carried out the forced swimming test. We investigated the body weight, feed and water intake, feed conversion ratio and measured parameters to changes in swimming time, immobility time, 2 and 24 hours of feces, corticosterone hormone and blood glucose level. Results : In this experiments, body weight gain and feed intake were higher in all korean medicinal herb group. Swimming time were increased by Zizyphus jujuba and Gagamsanzointang(SZT) groups and immobility time was significantly lower by Zizyphus jujuba and Fluoxetine(FLX) groups. The result of corticosterone hormone level were significantly lower in Fluoxetine(FLX) and Gagamsanzointang(SZT) but blood glucose was not significantly difference. In 2 hours after fecal frequency result were significantly lower in Zizyphus jujuba group. but SZT group were increased. This result caused by acute stress but 24 hours of feces were restored to normal. Conclusions : 5 kind of Korean Medicinal Herbs extracts has good anti-stress effects, and also to clear the circulatory system and regulate blood glucose and improve bowel function. We suggest that use a single Korean medicine rather than the combination to relieve stress.

  • This study analyzed the mechanical characteristics and evaluated their subsequent performance for two types of seals which reinforced characteristics with lower friction and anti-wear functions among the foremost important features in the automotive engine seals; one with the addition of glass fiber to PTFE(Polytetrafluoro ethylene); the other with the addition of self-lubricant molybdenumin addition to the glass fiber. Based on the configuration design of seal installed to the front part in the automotive engine, this study carried out interpretations on the stress and reaction for those two types of oil seals to compare the maximum stress and contact load generated from the seal steel, rubber and PTFE lip. This study also verified the stress concentration and anti-wear performance through the coefficient of friction, torque and durability test by producing two types of PTFE seals actually.

Experimental Effects of Aucklandiae Radix and Cyperi Rhizoma Extract on Chronic Stress in Rats (목향과 향부자 추출물이 흰쥐의 만성 스트레스에 미치는 실험적 효과)

  • This study aimed to investigate the anti-stress function of Aucklandiae Radix (AR) and Cyperi Rhizoma (CR). The essential oils used in the experiment were extracted from AR and CR using Steam Distillation Extraction and Super critical CO2 extraction. To observe the effects of sample administration, we measured feed intake, leukocytes, red blood cells, hemoglobin, platelets, serum serotonin content, immobility time, climbing time, and swimming time in mice subjected to chronic restraint stress as behavioral changes. The average body weight of all experimental groups increased than the average body weight of the control group. The immobility and climbing times of experimental groups A and B administered with supercritical extraction samples were shorter than those of the other experimental groups and the control group, and the swimming time was longer. The serotonin content in the blood of all experimental groups decreased compared to the normal group, and the serotonin content of the control group was increased. The authors suggest that Korean herbal medicines AR and CR may be utilized as anti-stress flavoring agents based on the above results.
