[Purpose] While the anti-obesity effects of exercise and capsiate are well-observed individually, the effect of exercise with capsiate intake has not been systematically explored yet. Therefore, the purpose of this study is to investigate whether the anti-obesity effects of exercise training can be further enhanced by capsiate intake. [Methods] 8-week-old male mice were divided into 3 groups (n = 8 per group): sedentary group (SED; nontrained), exercise-trained group (EXE) and exercise-trained group with 10 mg/kg of capsiate intake (EXE+CAP). All mice were offered high-fat diet and water ad libitum. The mild-intensity treadmill training was conducted 5 times a week for 8 weeks. After 8 weeks, metabolism during exercise and abdominal fat weight were measured. [Results] Body weight and the rate of total abdominal fat were significantly less in EXE+CAP than in SED but not between EXE and SED. The average of respiratory exchange rate during exercise was significantly much lower in EXE+SED (p = 0.003) compared to the difference between EXE and SED (p = 0.025). Likewise, the fat oxidation during exercise was significantly much higher in EXE+SED (p = 0.016) compared to the difference between EXE and SED (p = 0.045). Then, the carbohydrate oxidation during exercise was significantly much lower in EXE+SED (p = 0.003) compared to the difference between EXE and SED (p = 0.028). [Conclusion] In conclusion, the anti-obesity functions of exercise training can be further enhanced by capsiate intake by increasing fat oxidation during exercise. Therefore, we suggest that capsiate could be a candidate supplement which can additively ameliorate obesity when combined with exercise.

The objective of this study was to investigate the anti-obesity effects of adding Momordica charantia (MC) and Chrysanthemum zawadskii var. latilobum (CZ) extracts to drinking water on obesity-induced mice. A total of 84 eight-week-old C57BL/6N male mice with an initial body weight (BW) of 28.11 ± 1.39 g were used in this study. All treatments were fed a high-fat diet for d 28. Mice were randomly divided into seven drinking treatments (six replicate each treatment) based on the initial BW. Treatments are as follows: control (CON), normal tap water, MC 1, CON with 1% MC aqueous extract, MC 2, CON with 2% MC aqueous extract, CZ 1, CON with 1% CZ aqueous extract, CZ 2, CON with CZ aqueous extract (2%), MCZ 1, CON with 1% MC aqueous extract and 1% CZ aqueous extract, MCZ 2, CON with 2% MC aqueous extract and 2% CZ aqueous extract. During the entire period, the CZ 1, MCZ 1, and MCZ 2 significantly decreased (p < 0.05) gain to feed than CON. The CON significantly higher (p < 0.05) water intake than other treatments on d 0 to 14. The MCZ 1 significantly decreased (p < 0.05) relative (ratio of absolute organ weight to BW) organ weights, including retroperitoneal white adipose tissue (RWAT) weight and inguinal white adipose tissue (IWAT) weight, compared to CON. In conclusion, our study suggests that there was no significant difference in the anti-obesity effects between MC and CZ, and MCZ 1 has synergistic effects by regulating adipose tissue.
Juglans mandshurica Maxim. walnut (JMW) is well-known for the treatment of dermatosis, cancer, gastritis, diarrhea, and leukorrhea in Korea. However, the molecular mechanism underlying its antiobesity activity remains unknown. In the current study, we aimed to determine whether JMW can influence adipogenesis in 3T3-L1 preadipocytes and high-fat diet rats and determine the antioxidant activity. The 20% ethanol extract of JMW (JMWE) had a total polyphenol content of 133.33 ± 2.60 mg GAE/g. Considering the antioxidant capacity, the ABTS and DPPH values of 200 ㎍/ml of JMWE were 95.69 ± 0.94 and 79.38 ± 1.55%, respectively. To assess the anti-obesity activity of JMWE, we analyzed the cell viability, fat accumulation, and adipogenesis-related factors, including CCAAT-enhancer-binding protein alpha (C/EBPα), sterol regulatory element-binding protein-1c (SREBP1c), peroxisome proliferator-activated receptor-gamma (PPARγ), fatty acid synthase (FAS), and acetyl-CoA carboxylase (ACC). We found that total lipid accumulation and triglyceride levels were reduced, and the fat accumulation rate decreased in a dose-dependent manner. Furthermore, JMWE suppressed adipogenesis-related factors C/EBPα, PPARγ, and SREBP1c, as well as FAS and ACC, both related to lipogenesis. Moreover, animal experiments revealed that JMWE could be employed to prevent and treat obesity-related diseases. Hence, JMWE could be developed as a healthy functional food and further explored as an anti-obesity drug.
In order to develop a functionally improved kochujang with antiobesity effects, garlic-added kochujang was prepared with freeze-dried garlic powder and followed by fermentation for 60 days at $30^{\circ}C$. Antiobesity effect of the garlic-added kochujang was investigated by measuring the leptin secretions and mRNA expression levels of obesity-related gene such as TNF${\alpha}$, PPAR${\gamma}$, C/EBP${\alpha}$, and SREBP1c, in cultured 3T3-L1 adipocytes. Fermentation of garlic-added kochujang led to decreased levels of leptin secretions and reduced the mRNA expression levels of TNF${\alpha}$, PPAR${\gamma}$, C/EBP${\alpha}$, and SREBP1c in the 3T3-L1 adipocytes. Accordingly, these results suggest that the addition of garlic to kochujang has a potential as a valuable functional food for controlling obesity.
Objectives : Obesity is a metabolic disease resulted from various causes including nutritional disproportion, hormonal imbalance, and hypothalamic nuclei disorder. Samhwang-tang(Sanhuang-tang, SHT) is composed of 5 herbs (Ephedrae Herba, Scutellariae Radix, Typhae pollen, Acori Rhizoma and Ansu Semen) and is frequently used for obese patients. This study investigated anti-obesity effects of SHT on the rats. Methods : Obesity was induced by high fat diet (HFD), monosodium L-glutamate injection (MSG) and ovariectomy (OVX) in the Sprague-Dawley rats. Water extract of SHT was treated for 2 weeks, then changes of body weight gain and epididymal fat cell size were measured. In OVX animals, c-Fos and neuropeptide Y (NPY) expressions in hypothalamic nuclei were observed with immunohistochemistry. Results : SHT treatment demonstrated significant decrease of body weight gain in obesity induced by HFD and OVX, but not in obesity induced by MSG. SHT treatment demonstrated significant decrease of fat cell size in obesity induced by HFD, but not in obesity induced by MSG. On c-Fos and NPY expressions in hypothalamic nuclei, SHT treatment resulted significant decrease of immuno-reactivity of c-Fos and NPY in obesity induced by OVX. Conclusions : These results suggest that SHT has a weight loss effect against obesity induced by nutritional disproportion of diet intake and hormonal imbalance, especially without central nervous system disorder.
Resveratrol (3,4,5-trihydroxy-trans-stilbene), a phytoalexin found in grape skin, grape products, and peanuts as well as red wine, has been reported to have various biological and pharmacological properties. The purpose of this study was to investigate the anti-obesity effect of resveratrol-amplified grape skin extracts on adipocytes. The anti-obesity effects of grape skin extracts were investigated by measuring proliferation and differentiation in 3T3-L1 cells. The effect of grape skin ethanol extracts on cell proliferation was detected by the MTS assay. The morphological changes and degree of adipogenesis of preadipocyte 3T3-L1 cells were measured by Oil Red-O staining assay. Treatment with extracts of resveratrol-amplified grape skin decreased lipid accumulation and glycerol-3-phosphate dehydrogenase activity without affecting 3T3-L1 cell viability. Grape skin extract treatment resulted in significantly attenuated expression of key adipogenic transcription factors, including peroxisome proliferator-activated receptor, CCAAT/enhancer-binding proteins, and their target genes (FAS, aP2, SCD-1, and LPL). These results indicate that resveratrol-amplified grape skin extracts may be useful for preventing obesity by regulating lipid metabolism.
This study was conducted to investigate the cholesterol and Triglyceride (TG) lowering and anti-obesity effects of water extract of vegetable formula (onion 25%, carrot 20%, Cabbage 20%, Sweet Pumpkins 20%, Chinese plum 5%, turmeric 5%, morus leaves 5%, SLS) in mice fed high fat-diet. ICR mice were divided into 3 groups; a normal diet group (ND), a high-fat diet group (HFD), a high-fat diet and SLS with 300 mg/kg treated group (HFD+SLS). Body fat gain was increased by high-fat diet and HFD+SLS group showed a definite weight loss. The total cholesterol level in the HFD+SLS group was 328.4 mg/dl which was 27.4% lower than that in the HFD group and the cholesterol-lowering effect of SLS was confirmed. The HFD + SLS group showed 118.1 mg/dl and the triglyceride level in the serum was decreased by 88.7% compared to the HFD group and SLS significantly decreased blood triglyceride levels. RT-PCR showed that the expression of PPAR-${\gamma}$ and the target gene SCD-1 was inhibited in a dose-dependent manner in the SLS-treated group. These results suggest that the SLS water extract may have a cholesterol and triglyceride-lowering effect and inhibit the expression of PPAR-${\gamma}$ and SCD-1 to have an anti-obesity effect.

Microbial bioconversion using lactic acid bacteria (LAB) provides several human health benefits. Although whey and whey-derived bioactive compounds can contribute to an improvement in human health, the potential anti-obesity effect of whey bioconversion by LAB has not been well studied. This study aimed to investigate whether bioconversion of whey by Pediococcus pentosaceus KI31 and Lactobacillus sakei KI36 (KI31-W and KI36-W, respectively) inhibits 3T3-L1 preadipocyte differentiation. Both KI31-W and KI36-W reduced intracellular lipid accumulation significantly, without decreasing 3T3-L1 preadipocyte proliferation. In addition, obesity-related transcription factor (peroxisome proliferator-activated receptor γ) and genes (adipocyte fatty acid-binding protein and lipoprotein lipase) were down-regulated significantly in 3T3-L1 cells in the presence of KI31-W and KI36-W. Collectively, these results suggest that bioconversion of whey by LAB exhibits anti-adipogenic activity and may be applied as a therapeutic agent for obesity.

Obesity increases the risk of mortality and morbidity because it results in hypertension, heart disease, and type 2 diabetes. Therefore, there is an urgent need for pharmacotherapeutic drugs to treat obesity. We performed a screening assay using natural products with anti-adipogenic properties in 3T3-L1 cells and determined that tschimganidine, a terpenoid from the Umbelliferae family, inhibited adipogenesis. To evaluate the anti-obesity effects of tschimganidine in vivo. Mice were fed either a normal chow diet (NFD) or a high-fat chow diet (HFD) with or without tschimganidine for 12 weeks. Treatment with tschimganidine decreased lipid accumulation and adipogenesis, accompanied by reduced expression of adipogenesis and lipid accumulation-related factors. Tschimganidine significantly increased the phosphorylation of AMP-activated protein kinase (AMPK) and decreased that of AKT. Depletion of AMPK relieved the reduction in lipid accumulation resulting from tschimganidine treatment. Moreover, tschimganidine administration drastically reduced the weight and size of both gonadal white adipose tissue (WAT) and blood glucose levels in high-fat diet-induced obese mice. We suggest that tschimganidine is a potent anti-obesity agent, which impedes adipogenesis and improves glucose homeostasis. Tschimganidine can then be evaluated for clinical application as a therapeutic agent.
Objectives: The purpose of this study is to review clinical studies related to the effect of Traditional Chinese Medicine (TCM) on moderate to severe obese patients with hypertension. Methods: Clinical studies from the China National Knowledge Infrastructure (CNKI) were searched by specific keywords and criteria. Total of 7 randomized controlled trials were selected and analyzed. Results: Various evaluation methods were used to see the effect of TCM compared to conventional western medicine. Most of the studies used herbal medicine combined with conventional anti-hypertensive western medicine. The effective rate of anti-hypertension was higher when TCM was added to conventional anti-hypertensive medicine. Traditional medicine treatment showed significant effect on lowering blood pressure and body mass index. Conclusions: Based on the results of the clinical studies from China, TCM can be a valuable option for moderate to severe obese patients with hypertension. Applying Traditional medicine has a significant effect on lowering blood pressure and weight loss. Combining herbal medicine can be worthy of clinical promotion and application for moderate and severe obese patients with hypertension.
