혼합물 실험계획법에 의한 머위 및 부원료의 혼합비율 최적화 (Optimization Mixture Ratio of Petasites japonicus, Luffa cylindrica and Houttuynia cordata to Develop a Functional Drink by Mixture Design)

  • 호흡기계 질환에 효과적이라고 알려져 있는 식물성분들을 이용하여 기능성 음료를 개발하고자, 혼합물 실험계획법을 통해 혼합 최적 비율을 찾기 위한 실험을 수행하였다. 즉, 머위 30~70%, 수세미 10~30% 및 어성초 10~30%의 비율로 혼합하여 중앙점 반복을 포함하여 총 12개의 실험점을 설계하였고, 이 설계를 바탕으로 혼합물은 121℃에서 45분간 중탕 추출을 하여 anti-allergy activity 및 anti-microbial activity를 측정하였다. anti-allergy activity 효과는 Response surface와 trace plot을 보면 머위의 함량이 많을수록 좋은 효과를 나타내었고 수세미는 다소약한 영향을 미친다는 것을 알 수 있었다. 다중회귀분석 결과 상관계수 R2=82.10%이며 유의적인 회귀식으로 설명이 가능한 것으로 확인되었다. 따라서 최적비율은 머위 0.75, 수세미 0.14 그리고 어성초 0.11의 비율로 결정되었다. anti-microbial activity는 혼합비율에 따른 각 추출물들이 그람양성균인 Staphylococcus aureus (KCCM 40881), Staphylococcus epidermidis (KCCM 35494)에 대해서는 유효하나 그람음성균인 Escherichia coli (KCCM 11234), Pseudomonas aeruginosa (KCCM 11328)에 대해서는 미미하거나 효과가 나타나지 않았다.

  • 말오줌나무 잎 추출물의 생리활성 및 항균활성을 측정한 결과, 말오줌나무 잎 추출물의 생리활성은 말오줌나무 잎 추출물에 함유되어있는 phenolic성분들에 의해 효과가 발현되는 것으로 판단되었다. 말오줌나무 잎으로부터 생리활성에 관여하는 phenolic 화합물을 물과 에탄올로 추출하였을 때 각각 11.60±0.18과 12.39±0.07 mg/g의 함량을 나타내었다. 말오줌나무 잎의 ACE 저해 활성은 50-200 ㎍/mL의 phenolic을 함유한 물과 에탄올 추출물에서 각각 71.94-92.08와 48.42-78.33%의 저해 활성이 측정되었으며, 농도의존적인 양상을 나타내었다. 말오줌나무 잎 추출물의 XOase에 대한 추출물의 저해 활성은 50-200 ㎍/mL의 phenolic을 함유한 물과 에탄올 추출물에서 각각 42.11-100와 35.61-100%의 매우 우수한 저해효과를 나타내었다. 말오줌나무 잎 추출물에 대한 HAase 저해활성은 200 ㎍/mL의 phenolic을 함유한 농도에서 물과 에탄올 추출물이 각각 25.35±0.41와 43.38±2.26%의 HAase 저해활성을 나타내었다. 말오줌나무 잎 추출물의 항균활성은 충치균인 S. mutans와 식중독균인 S. aureus 및 여드름균인 P. acnes에 대한 우수한 항균 효과를 나타내었다. 따라서 말오줌나무는 고혈압억제, 통풍억제 및 염증억제 작용 등의 생리활성 및 S. mutans와 S. aureus 및 P. acnes에 대한 항균활성이 우수한 것으로 확인되어 말오줌나무를 활용한 기능성 식품 소재로서 산업화에 활용할 수 있을 것으로 생각되었다.

Synthesis and In-vitro Activity of Some New Class of Thiazolidinone and Their Arylidene Derivatives

  • In an attempt to find a new class of anti microbial agents, a series of thiazolidinone and their 5-arylidene derivatives containing 4-(4-methyl benzamido)-benzoyl moiety were synthesized via the reaction of benzocaine with appropriate chemical reagents. These compounds were screened for their antibacterial activity against Gram-positive bacteria (Bacillus subtilis and Bacillus thuringiensis), Gram-negative bacteria (Escherichia coli, Pseudomonas aeruginosa) and antifungal activity against Botrytis fabae, Fusarium oxysporan and Candida albicans. On the other hand the synthesized compounds were also screened for their anti tubercular activity. IR, $^1H$ NMR, $^{13}C$ NMR and MS spectral analyses established the structures of the newly synthesized compounds. The results revealed that some of these compounds have shown promising antimicrobial and anti tubercular activity in comparison with standard drugs.

영지버섯 다당체의 항미생물작용 및 항암작용에 관한 연구 (Studies on Anti-Mcirobial and Anti-Cancer Functions of Polysaccharide Extracted from Ganoderma lucidum)

  • This study was carried out to elucidate the immunomodulating activity of protein bound polysac charide extracted from Ganoderma lucidum(PSG). Macrophage drived reactive radicals were known as an effector for antimicrobial and anticancer functions. The promising immune response molecule, reactive oxygen intermediates(ROIs), was determined in TIB 71 cells with PSG at various experimental conditions. Treatment with 0.5mg PSG significantly increased the production of ROIs, superoxide anion as well as hydrogen peroxide, from TIB 71 cells(p<0.001). Under the same concentration, con siderable results were obtained from 24 hour cultivation with 106 cells at 5% CO2 incubator. The cells were trigged with PMA(5.3 M) after primed BCG(100 M) or IFN (100U) alone could not induce the production of ROIs, but it had a significant potentiating effect on ROIs secretion when the cells were treated with PSG.

어성초 추출물이 에멀젼에 미치는 영향에 관한 연구 (Study on the Effects of Houttuynia Cordata Extracts on Emulsions)

  • Objectives : Since cosmetics have been one of the help of life, unlike medicine, natural products have been used for cosmetics, generally giving the image of safety and relief compared to synthetic products. Among them, Houttuynia cordata has been known as a useful herbal medicine with antibiotic, anti-inflammatory and anti-oxidative activities. In this study, we were to use Houttuynia cordata extract on formulation of cosmetic emulsions. Methods : The effects of Houttuynia cordata extract on emulsion stability and viscoelastic properties of emulsion were measured using turbiscan and rheometer. And we assessed the anti-oxidative and antibiotic activities of Houttuynia cordata extract. Results : 1. The results of this investigation for emulsion stability showed that the stability emulsion containing Houttuynia cordata extract was decreased depending on concentration of Houttuynia cordata extract. 2. Super oxide dismutase activity was strongly dependents on concentration of Houttuynia cordata extract. 3. Houttuynia cordata extract showed good anti-microbial activity against Staphylococcus aureus. Conclusions : Houttuynia cordata extract can be effectively used in cosmetic emulsions when the relation between natural product extracts and formulation of cosmetics is elucidated.

  • BMB-CF has been constituted of purified fractions of Scutellaria baicalensis, which has medicinal effect such as anti-microbial. anti-inflammation. fever remedy. anti-oxidation. and anti-aging effect etc.. It has been used in traditional medicine formula from long time ago in the east Asia. It is constituted of the active flavone ingredients such as baicalin. baicalein. DTF(Di-methyl Tetra -hydroxy Flavone), wogonin. wogonoside. $\beta$- Sitosterol. etc.. General purified fractions of Scutellaria baicalensis has the high portion of the baicalin which has the problem of narrow antimicrobial spectrum and compatibility against cosmetic formula. Now. we has been develop the new purificaton process of Scutellaria baicalensis that has the high rate of DTF content, which is improved in antimicrobial activity and cosmetics compatibility. So. we have assure that it is the potent preservative against various cosmetic formula.

Carnosol induces the osteogenic differentiation of bone marrow-derived mesenchymal stem cells via activating BMP-signaling pathway

  • Carnosol is a phenolic diterpene phytochemical found in rosemary and sage with reported anti-microbial, anti-oxidant, anti-inflammatory, and anti-carcinogenic activities. This study aimed to investigate the effect of carnosol on the lineage commitment of mouse bone marrow-derived mesenchymal stem cells (mBMSCs) into osteoblasts and adipocytes. Interestingly, carnosol stimulated the early commitment of mBMSCs into osteoblasts in dose-dependent manner as demonstrated by increased levels of alkaline phosphatase activity and Alizarin red staining for matrix mineralization. On the other hand, carnosol significantly suppressed adipogenesis of mBMSCs and downregulated both early and late markers of adipogenesis. Carnosol showed to induce osteogenesis in a mechanism mediated by activating BMP signaling pathway and subsequently upregulating the expression of BMPs downstream osteogenic target genes. In this context, treatment of mBMSCs with LDN-193189, BMPR1 selective inhibitor showed to abolish the stimulatory effect of carnosol on BMP2-induced osteogenesis. In conclusion, our data identified carnosol as a novel osteoanabolic phytochemical that can promote the differentiation of mBMSCs into osteoblasts versus adipocytes by activating BMP-signaling.

Phytochemical Compounds from the Ethanolic Extract of Gymnema sylvestre, Senna auriculata and Cissus quadrangularis through GC-MS Analysis

  • Plants are a traditional source of many chemicals used as biochemical, flavors, food, color, and pharmaceuticals in various countries, especially India. Most herbal medicines and their derivatives are often made from crude extracts containing a complex mixture of various phytochemical chemical components (secondary metabolites of the plants). This study aimed to identify bioactive compounds from the different parts of the plant from the ethanolic extract of Gymnema sylvestre, Senna auriculata, and Cissus quadrangularis (leaves, flower, stem) by gas chromatography-mass spectroscopy (GC-MS). The gas chromatography - mass spectrometry analysis revealed the presence of various compounds like 3,4-dimethylcyclohexanol, hexanoic acid, D-mannose, and N-decanoic acid. Hence, the Gymnema sylvestre, Senna auriculata, and Cissus quadrangularis may have chemopreventive, anti-cancer, anti-microbial activity, antioxidant, anti-diabetic activity, anti-inflammatory, and antifungal due to the presence of secondary metabolites in the ethanolic extract. These phytochemicals are supported for traditional use in a variety of diseases.

큰실말(Cladosiphon novae-caledoniae Kylin) 부산물의 화장품소재 특성 (Cosmetic Effects of Dietary Fiber from Mozuku, Cladosiphon novae-caledoniae Kylin)

  • 큰실말(Cladosiphon novae-caledoniae kylin, C. novae-caledoniae kylin) 부산물을 이용하여 화장품 소재로써 가능성을 조사하고자, 항산화 효과(DPPH 자유 라디칼 소거 활성), 항균(anti-microbial activity), 미백(tyrosinase inhibition assay), 주름개선(elastase inhibition assay) 및 B16F10 mouse melanoma cell (MTT assay)을 이용한 세포 생존률을 측정하였다. 큰실말 추출물 원물(MC)과 불용성(MI)은 Staphyloccus aureus (S. aureus)와 항생제내성균주인 MRSA에 대하여 활성을 나타내었지만 항곰팡이 활성은 나타내지 않았다. 큰실말 추출물 수용성(MS)은 tyrosine에 대한 tyrosinase 저해효과가 49%로 표준물질인 arbutin 보다 미백효과가 우수하였지만 주름개선효과는 비교적 낮은 활성을 나타내었다. DPPH 라디칼 소거능력은 $500{\mu}g/mL$에서 89%로 나타내었다. 따라서 본 연구인 큰실말 추출물은 화장품 미백소재로써 좋은 후보군임을 확인할 수 있었다.

Function of Blending Essential Oil in the Development of Anti-Dandruff Products

  • Purpose: In this paper, we show our blending ratio of 10 types of Essential Oils that survives beneficial bacteria and kills harmful bacteria in the scalp, and we investigate the possibility of application of our blending ratio to the development of anti-dandruff products and the possibility of being used as a raw material for clinical beauty and customized cosmetics. Methods: The scalp microorganisms used in our study were M. furfur, S. epidermidis, E. coli, and P. nitroreducens. There are a total of 10 Essential Oils such as True Lavender, Lime, Roman chamomile, Rosemary camphor, Cedarwood, Geranium, Clove, Tea tree, Palmalosa, and Peppermint. The antibacterial test of the blended Essential Oil was carried out according to the test method of the standardized evaluation methodology of "Food and Food Additives Code". Since M. furfur is related to the growth of sebum in the scalp, in this study we used the fnLNB and the fnLNA with 20 ㎖ of whole fat cow milk added. Results: The blending ratio of EO, which inhibits dandruff-causing bacteria such as M. furfur, S. epidermidis, E. coli, and does not inhibit P. nitroreducens showing dominant growth in a healthy scalp, was B8(Clove 0.2%, Roman chamomile 0.5%, Tea tree 0.3%), B9(Geranium 0.1%, Palmarosa 0.1%, Roman chamomile 0.5%, Tea tree 0.3%), B10(Clove 0.1%, Geranium 0.1%, Palmarosa 0.1%, Roman chamomile 0.5%, Tea tree 0.2%). Conclusion: It is thought that the blending ratio of BEO obtained as a result of this study can provide a basis for use as an alternative to antibiotics in developing anti-dandruff drugs and emerge as a new alternative to solve scalp microbial imbalance. In order for EO to be used as a useful raw material for anti-dandruff preparation, researches on 1) Standardization (the effects of products differ according to the types, regions, climate, extraction methods, etc.), 2) Antimicrobial effects, 3) Safety, etc., must be established.
