우리의 이전의 연구에서 Citrus auranifoli 오일은 Malassezia furfur와 Malassezia pachydermatis에 대해 항곰팡이 활성을 나타낸다는 것을 보여주었다. 본 연구에서 Citrus auranifoli 오일의 주요 성분인 리모넨(limonene)의 M. furfur와 M. pachydermatis에 대한 저해효과를 디크스 확산 방법(disk diffusion method)에 의해 측정하였고, Beas-2B 세포에 대한 세포독성을 cytopathic effect reduction 방법에 의해 측정하였다. Limonene의 최소곰팡이저해농도(minimum fungicidal concentration)는 양성대조구인 itraconazole보다 더 낮았고, 100 ${\mu}g$/mL의 농도에서 Beas-2B cells에 대한 세포독성을 나타내지 않았다. 그러나 itraconazole은 같은 농도에서 약한 세포독성을 나타내었다. 그러므로, 우리의 결과에서 limonene는 비세포독성과 함께 M. furfur and M. pachydermatis의 성장저해를 하는데 효과적이라는 것을 보여준다.
비듬균(Malassezia furfur) 는 두피 질환을 일으키는 중요한 요소로써 오늘날 이러한 균들을 치료하는 치료제는 거의 없다. 몇몇의 전의 연구에서 오일들의 다양한 생물학적 활성이 보고되어졌지만 그것들의 항비듬균 활성은 거의 연구되지 않았고, 특히 Malassezia furfur에 대한 저해 활성은 보고되어 있지 않다. 따라서, 이 연구에서는 Malassezia furfur에 대한 식물 오일들의 저해활성을 평가하고, 이러한 오일 중에서 가장 활성이 좋은 오일을 가스 크로마토그래피(Gas chromatography-mass spectrometry)에 의해 성분을 분석하였다. 1차적으로 108개 오일의 스크리닝 과정에서 17개 오일[Allium sativum L., Carum carvi L., Coriandrum sativum L., Myroxylon balsamum var. pereirae Royle, Nepeta cataria L., Origanum vulgare L., Mentha pulegium L., Satureja hortensis L., Thymus vulgaris L. Thymus vulgaris L., Cinnamomum zeylanicum Blume, Pimenta dioica(L.) Merr., Eugenia caryophyllata Thumb.(Clove bud), Eugenia caryophyllata Thumb.(Clove leaf), Cymbopogon nardus L., Cymbopogon martinii Stapf., Citrus aurantifolia Swing. and Pimenta racemosa(Mill.) J.W.Moore] 이 2 mg/mL 의 농도에서 대조구인 이크라코나졸(itraconazole)과 비슷한 활성을 나타내었다. 이러한 17개 오일 중에서 Citrus auranifolia 오일이 가장 활성이 좋았으며, 처리농도 (1.5 mg/mL, 1.0 mg/mL, 0.5 mg/mL와 0.1 mg/mL) 에 따라 활성이 감소하는 경향을 보였다. 또한 Citrus auranifoli 오일의 가스크로마토그래피(GC-MS) 성분분석 결과 limonene, ${\gamma}$-terpinene and terpinolene의 함유율이 높은 것으로 나타났다. 그러므로 Citrus auranifolia 오일은 곰팡이 유래 질병들을 치료하기 위한 활성성분을 함유하고 있는 것으로 보인다.

Background: Malassezia, a lipophilic yeast, is a causative agent for dandruff and seborrheic dermatitis. Many biological agents have been studied for anti-Malassezia effect but further studies are needed for their clinical application. Objective: The study was conducted to evaluate the inhibitory effect of different natural essential oils and a fruit extract on Malassezia species in an in vitro study and a clinical trial. Methods: The antifungal effects of natural essential oils and a fruit extract on Malassezia species (M. furfur and M. sympodialis) were evaluated by measuring the minimum inhibitory concentration (MIC) and minimum fungicidal concentration (MFC) and using the disc diffusion method. Natural essential oils of citron seed, lavender, and rosemary and citrus junos fruit extract were used for the in vitro study. The clinical trial was conducted with a shampoo containing four ingredients. A total of 22 subjects used the shampoo every day for 4 weeks and were evaluated using clinical photography, trichoscopy, and sebumeter at baseline, 2 weeks, and 4 weeks after treatment. Results: Antifungal activity of agents was relatively lower in lavender and rosemary essential oils at MIC and MFC. Disc diffusion method revealed same results. In the clinical trial, the amount of sebum decreased statistically significantly and erythema, dandruff, and lesion extent also improved. Conclusion: The natural essential oils and fruit extract are effective for suppressing Malassezia activity, therefore these might be used as an alternative for treatment of dandruff and seborrheic dermatitis.
Herbal mixture water extract of (Phellodendron amurense, Scuellaria baiklensis, Spphora flavescens, Lithospermum erythrorhizon, Mellaphis chinesis, Alumite, Zanthoxylum schinifolium, Glycyrrhiza uralensis), which exhibit several beneficial effects including acne and skin diseases, was tested for anti-microbial activity and anti-inflammation effects. The herbal mixture extract showed antimicrobial activity against Stapylococcus epidermis, Propionibacterium acne, and Malassezia furfur. The growth of Stapylococcus epidermis, Propionibacterium acne, and Malassezia furfur was inhibited allergy and LPS induced cyclooxygenase-2(COX-2)gene expression in RAW24.7macrophage. The results indicated th ear swelling and histamine release induced by compound 48/80 were dose-dependently reduced, ranging 11-38% and 11-56%, respectively. Furthermore the extract inhibited the expression of LPS-induced COX-2 proteins and mRNAs without an appreciable cytotoxic effects on RAW264.7 cells. The cytotoxicity of the extract using M7T assay showed the cytotoxicity of 7 and 18% against L929 cell line. Based on these results, it is concluded that the herbal mixture water extract can be applied to the acne and skin diseases therapy.
Effect of the plant essential oils on the growth of Malassezia pachydermatis was evaluated and the essential oils of Ocimum basilicum L., Melaleuca alternifolia (Maid. & Bet.) Cheel, and Rosa damascene Mill. were the most active against M. pachydermatis and their activity were high than that of itraconazole at 2 mg/mL. The major constituents of the three oils by GC-MS analysis were linalool (21.83%) and estragole (74.29%) for O. basilicum, a-terpinolene (17.96%) and terpinen-4-01 (45.54%) for M. alternifolia, and a-citronellol (59.98%) and geraniol (27.58%) for R. damascene. Results showed that these selected three oils could be effective toward controlling M. pachydermatis opportunistic infections.
The Malassezia fungi are responsible for various human skin disorders including dandruff and seborrheic dermatitis. Of the Malassezia fungi, Malassezia globosa (M. globosa) is one of the most common in human scalp. The completed genome sequence of M. globosa contains four putative cytochrome P450 genes. To determine the roles of Malassezia P450 enzymes in the biosynthesis of ergosterol, we isolated MGL3996 gene from M. globosa chromosomal DNA by PCR. The MGL3996 gene encodes an enzyme of 616 amino acids, which shows strong similarity with known CYP52s of other species. MGL3996 gene was cloned and expressed in Pichia pastoris (P. pastoris) heterologous yeast expression system. Using the yeast microsomes expressing MGL3996 protein, a typical P450 CO-difference spectrum was shown with absorption maximum at 448 nm. SDS-PAGE analysis revealed a protein band of apparent molecular weight 69 kDa and Western blot with anti-histidine tag antibody showed that MGL3996 was successfully expressed in P. pastoris. Cloning and expression of a new P450 gene is an important step to study the P450 monooxygenase system of M. globosa and to understand the role of P450 enzymes in pathophysiology of dandruff.
The reason that aromatherapy began to draw attention again as the alternative therapy is because the essential oils that are used for aromatherapy not only allow the human body to maintain homeostasis but also have an effect on curing a mental feeling of fatigue and because these efficacies and effects are proceeding with being proved scientifically. Also, with coming to be high in a voice of concern about severally side effects that the modern medicine and the synthetic chemistry have, the aromatherapy came to be paid attention as the replacement therapy that can substitute for and supplement this. In particular, at the time of requiring a study on a substance to substitute for this after having been announced in Japan for 1998 a risk of Zincpyrithione that is being used as a component of dandruff treatment, it is thought that the aroma oil that was known for having an anti-dandruff effect has a merit of being easy in a user's access, thus it will be effective in curing the symptom of dandruff. Accordingly, this study conducted the following experiment in a bid to examine whether the aroma oil has the anti-microbial effect on the actual malassezia. First of all, it measured the minimum concentration of aroma oil in which the growth of malassezia is inhibited, by carrying out MIC(Minimum Inhibition Concentration) test in terms of anti-microbial activity against Malassezia furfur KCTC 7744 while using oils of Patchouli, Rosemary, Lavender, and Tea tree among aroma oils that are being used for dandruff treatment. As a result of that, Tea tree oil has the most excellent anti-microbial activity, and the measurement result of anti-microbial activity was shown in order of Lavender oil>Rosemary oil>Patchouli oil. Next, it carried out the is actually eased by applying Tea tree oil directly to the scalp of dandruff. In consequence of the experiment, the massage of Tea tree oil indicated the result of relieving the symptoms of dandruff such as scale, itch, pain, inflammation, and sebum, and among these symptoms of dandruff, it was shown to be most effective in easing itch. Especially, this clinical experiment was indicated that the massage of Tea tree oil has the most effect on relieving itch of targets with a dry-skin type who had no experience of dandruff treatment.
본 연구는 정향, 황련 추출물을 혼합하여 Malassezia furfur에 대한 항균 능력을 확인하고 최적의 혼합물을 만들어 항산화 및 항염증 능력을 평가하기 위해 수행되었다. 정향과 황련을 70% 에탄올, 100% 메탄올, 열수로 추출하여 항균 능력을 평가하여 정향은 100% 메탄올 추출물, 황련은 열수 추출물이 가장 항균 능력이 높은 것을 확인하였다. 또한 두 추출물을 혼합하여 비율별로 항균 능력을 평가하였을 때 9:1 비율이 가장 우수한 활성을 보였고 혼합물의 항산화 활성이 뛰어남을 확인하였다. Raw 264.7 세포에서 생존율에 영향을 미치지 않는 1, 10, 50, 100 ㎍/mL에서 LPS를 이용하여 염증 반응을 유도하고 항염증 활성을 확인한 결과 10, 50, 100 ㎍/mL에서 No 생성 저해율과 IL-6 발현 저해율 및 COX2, iNOS 단백질 발현 저해 활성이 뛰어남을 확인하였다. 본 연구를 통해 정향 메탄올 추출물 및 황련 열수 추출물의 혼합물은 항균, 항염증 효능이 뛰어나기에 기능성 화장품의 천연 소재로써 사용이 가능할 것으로 사료된다.
이 연구는 개의 귀 속에서 분리한 $Malassezia$$pachydermatis$의 특성과 이들 효모에 대한 essential oil들의 항진균효과를 측정한 결과로서 일반적인 $M.$$pachydermatis$를 분리하기위한 Sabouraud dextrose agar (SDA), olive oil이 첨가된 SDA (SDAO) 배지 및 지방 친화성 효모를 검출하기 위한 Leeming's medium (LM)으로 구성된 세종류의 분리배지가 사용되었다. $Malassezia$ spp는 77.8%의 개로부터 분리되었으며 분리배지에 따라 분리율에 큰 차이를 나타내었으며 지방성분이 포함된 배지에서의 분리율이 일반적으로 사용하는 SDA에서 보다 높았다. 분리된 모든 $Malassezia$ spp는 $M.$$pachydermatis$로 동정되었지만 육안적으로 보이는 집락의 모양과 SDA에서의 증식속도를 고려했을 때 4개의 아종으로 관찰되었다. 배양 72시간 후 집락의 직경이 3 mm 이상인 큰 집락(LC)과 2 mm 정도인 중간 집락(MC) 및 배양 5일 후에도 1 mm 이상으로 크지지 않는 작은 집락(SC), 그리고 SDA에서 증식하지 않는 지방의존 집락(Lipo)으로 분류되었다. SDA, SDAO 및 LM 배지에서 각 type의 효모가 분리된 정도를 보면, LC type 효모는 각각 4, 11, 11개 시료에서, MC type 효모는 각각 2, 3, 1개 시료에서, SC type 효모는 각각 1, 2, 1개시료에서 분리되었으며 Lipo type은 SDAO과 LM 배지 중 3 곳에서 분리되었다. $M.$$pachydermatis$에 대한 essential oil의 항진균효과 측정은 디스크 확산법과 well 확산법에 의해 이루어졌으며, 대부분의 essential oil은 0.5에서 1.0%의 농도에서 $M.$$pachydermatis$ 의 증식을 억제하였다. MIC90과 MIC50은 essential oil의 성분에 따라 다양하였으나, Thyme oil이 가장 억제 효과가 좋았으며 marjoram과 tea tree oil은 비교적 낮은 억제 능력을 나타내었다.
솔잎은 동의보감과 향약생약대사전에 간, 위, 심, 피부 등 질환 치료에 효과가 있다고 하여 예로부터 솔잎을 노화방지와 피부질환 치료에 활용하여 왔다. Malassezia furfur (M. furfur)는 친유성 곰팡이로 인간 피부에 존재하는 흔한 균이다. 이 균은 대게 피부의 각질층에서 주로 발견되며, 어우러기, 지루성 피부염, 비듬, 모낭염, 건성 등의 각종 피부질환의 주요요인이다. 본 연구는 사람의 대부분의 염증실험에서 사용되는 Raw 264.7 세포를 이용하여 솔잎의 항염증 활성을 확인하였다. 솔잎을 에탄올, 물, 메탄올로 추출하여 항산화 효과를 확인한 결과, 메탄올 추출물에서 항산화능이 높게 나타났으며, 이에 따라 헥산, 에틸아세테이트, 부탄올, 물 층으로 나누었다. 이 4층을 이용하여 노화의 원인이 되는 자유라디칼의 소거능을 확인한 결과, 부탄올과 에틸아세테이트 층에서 강한 항산화 활성이 나타났으며, M. furfur에 대한 항균활성을 확인한 결과 부탄올, 에틸아세테이트층과 헥산층에서 항균활성이 나타났다. 또한 티로시나아제 저해에 따른 미백활성은 4층 모두 대조군인 비타민C와 유사하게 나타났다. 세포실험을 위해 먼저 대식세포인 Raw 264.7에 솔잎 분획물을 처리하여 세포독성을 확인 한 결과 Hexane층에서 농도별로 높은 독성이 나타났으며 나머지 층에서는 독성이 나타나지 않음을 확인하였다. 이후 Malassezia furfur로 염증반응이 유도된 Raw 264.7에 에틸아세테이트, 부탄올, 물층을 처리하여 염증에 관련된 유전자들의 발현량을 Real-Time PCR을 통하여 확인하였다. 이때 사용된 유전자는 IL-1β, TNF-α로 에틸아세테이트층이 염증과 깊은 연관이 있는 IL-1β와 TNF-α의 발현량을 낮추는 것으로 확인되었다. 모든 실험결과들을 종합해 보았을 때, 솔잎은 천연자원으로써 피부 건강과 관련된 건강식품, 건강보조제, 화장품 등 폭 넓은 범위로 사용될 수 있을 것으로 사료된다.
