  • A simple and efficient synthesis of various $\alpha$-aminophosphonates (3a-l) by the reaction of substituted aromatic/heterocyclic aldehydes, 2-amino-6-methoxy-benzothiazole and dibutyl/diphenyl phosphites under microwave irradiation without catalyst was accomplished. The phosphonates were characterized by elemental analysis, IR, $^1H$, $^{13}C$- and $^{31}PNMR$ spectra. They showed promising antimicrobial, anti-oxidant activities depending on the nature of bioactive groups at the $\alpha$-carbon.

  • The anti-oxidant activities of the various fractions from the herbs of Artemisia apiacea were investigated. The n- hexane and n-butanol fractions were found to cause significant free radical scavenging effects on DPPH. their scavenging potencies as indicated in $IC_{50}$ values. being 230.1 and 183.7 g/ml. respectively. The n-butanol fraction exhibited a significant decrease in serum transaminase activities elevated by hepatic damage induced by $CCl_{4}$-intoxication in rats. All fractions tested exhibited a lipid peroxidation causing a significant decrease in MDA production in TBA-reactant assay. (omitted)

천초근 에탄올 추출물의 항산화 효능 및 대장암 세포 억제 효과 (Anti-colorectal Cancer and Anti-oxidant Activities of Rubiae radix Ethanol Extract in vitro)

  • 본 연구는 꼭두서니의 뿌리인 천초근의 물 추출물과 에탄올 추출물을 이용하여 대장암 세포에 대한 암세포 성장 억제 및 사멸효과가 있는지 알아보고자 수행하였다. ERA(천초근 에탄올 추출물)은 폴리페놀($45.77{\pm}2.03mg/g$)과 플라보노이드($22.82{\pm}1.33mg/g$)를 함유하고 있었으며, $H_2O_2$에 의해 증가된 ROS(reactive oxygen species)를 억제하는 효과를 나타냈지만 WRA(천초근 물 추출물)은 효과가 없었다. 또한 ERA는 $500{\mu}g/m{\ell}$의 농도로 대장암 세포주(HCT-116)에 처리했을 때 세포사멸을 유도할 뿐만 아니라 caspase-3 단백질 활성화, DNA fragmentation 및 apoptotic cell death를 일으키는 것으로 확인되었다. 이는 ERA가 HCT-116 세포주에 대해 apoptosis(세포자멸사)를 통해 항암효과를 나타내는 것으로 생각되지만 다른 연구결과들과 비교하였을 때 농도 대비 효능이 미미하다. 따라서 천초근 에탄올 추출물에 대장암 세포의 성장을 억제하는 유효성분을 분석하여 그 효능을 탐색하는 추가실험이 필요할 것으로 생각된다.

모링가:두충 2:1 (g:g) 복합 조성물의 in vitro 항산화 및 항염 효능 연구 (In vitro Anti-oxidant and Anti-inflammatory Effects of Moringa Folium and Eucommiae Cortex 2:1 (g/g) Mixed Formula)

  • The purpose of study is to evaluate in vitro anti-oxidant and anti-inflammatory effects of Moringa Folium and Eucommiae Cortex 2:1 (g/g) mixtures (MEMix). HaCaT and human normal dermal fibroblast were treated with 0.01-1 mg/mL of MEMix to monitor cytotoxicity. Radical scavenging activities of MEMix were examined by DPPH assay. To explore anti-inflammatory effect, Raw 264.7 cells were pretreated with MEMix for 1h and subsequently exposed to LPS for 18h. NO release and cytotoxicity of Raw 264.7 cells were measured by adding Griess and MTT reagents, respectively. TNF-α, IL-1β, IL-6, and PGE2 productions were examined by ELISA. Immunoblot analysis was conducted to examine COX-2 expression in MEMix pretreated Raw 264.7 cells. Up to 1 mg/mL concentration, treatment of MEMix for 24 h did not affect normal dermal fibroblast viability and significantly reduced cell viability of HaCaT cells with no concentration dependency. MEMix increased DPPH radical scavenging activity with concentration dependency. Radical scavenging activities by 1 mg/mL of MEMix was comparable with 30 µM of trolox. Pretreatment of MEMix did not change the reduction of Raw 264.7 cell viability. Exposure of LPS in Raw 264.7 cells significantly increased NO, TNF-α, IL-1β, IL-6, and PGE2 productions, and MEMix pretreatment attenuated these productions by LPS concentration dependently. However, pretreatment with MEMix did not change COX-2 expression by LPS in Raw 264.7 cells. MEMix showed in vitro anti-oxidant and anti-inflammatory activities. MEMix would be useful candidate agent against inflammation.

향미 개선을 위한 침지 발아 로부스타 커피의 다중 기능성 평가 (Multifunctional evaluation of soaking-germinated Robusta coffee for flavor improvement)

  • 본 연구는 커피의 향미개선에 효과가 있다고 알려진 침지 발아를 로부스타 커피에 적용하여 그 유용성을 검증하였다. 베트남 로부스타 커피 생두를 정제수와 함초 추출물에 침지 발아시켜 정수발아(WGC)와 함초발아(SGC) 원두를 제조하고 에스프레소 커피로 추출한 다음 그 기능성을 평가하였다. 400 ㎍/mL 농도에서 Folin-Ciocalteu법으로 분석한 총 폴리페놀 함량은 WGC (20.17%)>NGC (19.45%)>SGC (16.71%) 순으로 WGC의 총 폴리페놀 함량이 가장 높았다. 비발아 커피 뿐만 아니라 발아커피는 25~100 ug/ml 모든 농도에서 DPPH 제거능으로 평가된 실험에서 우수한 항산화 활성을 나타내었다. XOD 저해 효과 또한 400 ㎍/mL 농도에서 WGC 42%, NGC 39%, SGC 32%로 관찰되었다. 비발아 및 발아커피는 멜라닌 생합성의 주요 단계에 관여하는 효소인 tyrosinase 활성을 억제하였으며, α-MSH로 유도된 B16F10 세포에서 멜라닌 생성을 억제하였다. 이 결과는 대조약물 arbutin과 유사한 결과를 나타내었으며, NGC, SGC, WGC는 모두 미백효능이 있는 것으로 확인되었다. Oil red O 염색을 통해 NGC, SGC, WGC의 지방분화 억제효과를 측정한 결과 분화된 대조군 보다 로부스타 커피를 처리한 시료들의 지방구 양이 유의적으로 감소된 것을 확인할 수 있었다. 이상의 연구결과 로부스타 커피의 향미를 개선하기 위해 물 또는 함초 추출물을 사용하여 침지 발아된 커피는 항산화, 미백, 항비만 등 다양한 기능성을 나타내는 것을 시사한다.

발효 옻 추출물의 생리활성 및 단회 경구 투여 독성시험 (Biological activities and single oral dose toxicity in rat of fermented Rhus verniciflua extract)

  • In this study, we investigated the biological activities such as anti-tumor, anti-oxidant and antiinflammatory activities as well as single oral dose toxicity of fermented Rhus verniciflua extract (FRVE). In order to examine anti-tumor activity of FRVE, the sarcoma 180 cells were treated with FRVE at various concentrations (0.03, 0.3, 3 and 30 mg/mL) in microtetrazolium (MTT) assay. In MTT assay, all the cells treated with FRVE at various concentrations have shown a significant difference compared with control (p < 0.05). In xanthine oxidase inhibition assay to examine the antioxidant activity, the xanthine oxidase inhibition rate of FRVE at 1.5 mg/mL and 15 mg/mL was $85{\pm}15.01%$ and $99{\pm}16.02%$, respectively. Nitric oxide production in RAW 264.7 cells showed that FRVE showed a significant anti-inflammation effect at 3 mg/mL (p < 0.05). In single oral dose toxicity study, no differences were observed between control and treated groups in clinical signs, body weight gains, feed and water consumptions. The results indicated that lethal dose 50 ($LD_{50}$) of FRVE was found to be higher than 5,000 mg/kg in this experiment. From the above results, we may suggest that FRVE might have useful as a material for functional food and/or animal pharmaceutics.

피부의 노화와 주름 억제, 미백효과를 가지는 전통 한약재의 탐색 (Research of Traditional Herbal Medicines for Anti-aging, Inhibition Effect of Wrinkle and Whitening Effect in the Skin)

  • The aims of this study are to search herbal medicines that have anti-aging, anti-wrinkle, anti-oxidant and whitening effects. This is preliminary study as a finding for excellent and specific natural agents which inhibit the skin aging and whitening formation. In this study, the articles and the documents, which have been published in domestically or internationally, have been searched and analyzed. Articles were gathered by taking advantage of Society of Cosmetic Scientists of Korea's documents, Pub med database, Korean Medical Database and Korean Studies Information Service System. Among the many species of herbal medicines, we selected the herbal medicines showing superoxide scavenging activities, and 1.1-diphenyl-2-picrylhydrazyl(DPPH) scavenging effects. The herbal medicines were showed anti-wrinkle effects on Metrixmetallo proteinase (MMP-1) inhibition activity and elastase inhibition activity. Also we found herbal medicines that have highly effect of tyrosinase inhibition, L- DOPA inhibition and melanin synthesis inhibition.

탁리산 용매 분획물의 in vitro 항산화, 주름 억제 및 미백 효과 (In vitro anti-oxidant, anti-wrinkle and whitening effect of solvent fraction from takli-san)

  • 본 연구에서는 탁리산 열수추출물을 용매 분획하여 항산화, 주름억제, 미백효과에 대한 활성을 검증하여 화장품 소재로서 이용가능성을 연구하였다. 용매 분획물로 DPPH radical scavenge, ABTS radical scavenge, elastase, tyrosinase 저해활성을 확인하였고 에틸아세테이트 분획물(TW-EA)이 가장 높은 저해활성을 나타내었다. 세포내에서의 주름억제, 미백효과를 확인하기 위해서 western blot을 실시하였고 비교 검증을 위해 사용한 positive contol보다 TW-EA의 억제효과가 뛰어난 것을 확인하였다. 이러한 결과로 탁리산의 화장품소재로서 적용 가능성을 검증하였다.

구절초 꽃 추출물의 항산화, 항염증 및 멜라닌 생성 억제 효과에 관한 연구 (A Study on Anti-oxidative, Anti-inflammatory, and Melanin Inhibitory Effects of Chrysanthemum Sibiricum Extract)

  • 본 연구는 구절초 꽃 추출물이 기능성 화장품 소재로서의 개발 가능성을 관찰하기 위하여 항산화 활성과 피부세포에 대한 세포독성, 항염증, 멜라닌 생합성 억제능에 미치는 영향을 관찰하였다. 본 연구 결과 구절초 꽃 추출물에서 폴리페놀과 플라보노이드의 높은 함량과 뛰어난 DPPH radical 소거작용을 확인하였고, Raw 264.7세포와 B16F10세포에 대해 유의한 세포독성을 나타나지 않았으며, Raw 264.7세포에서 LPS에 의해 유도된 NO 생성을 유의하게 억제함으로써 항염증 효과를 확인하였다. 구절초 꽃 추출물이 B16F10세포에 ${\alpha}$-MSH로 멜라닌 생성을 유도한 후 멜라닌 생합성 억제능을 측정한 결과 멜라닌의 생성 증가를 농도 의존적으로 억제되는 것을 확인하였다. 이와 같은 결과로 보아 구절초 꽃 추출물은 세포에 대한 독성이 낮고, 높은 항산화 활성과 항염증, 미백효과를 가진 기능성 화장품소재로써의 활용 가능성이 있을 것으로 사료된다.

지역별 두중(杜仲)의 지표성분 및 항산화 활성 비교 (Comparison of Anti-oxidant Activity and Marker Compounds in Eucommiae Cortex Samples from Regional Groups)

  • Objectives : It is necessary to manage herbal medicines based on effectiveness by comparing the efficacy of herbal medicines by region. In this study, we compared anti-oxidative activity and marker compounds of Eucommiae Cortex by regional groups. Methods : Eucommiae Cortex grown and harvested in Gangjin, Sancheong, Yeongwol, Jangsu, and Jecheon were used. Eucommiae Cortex was extracted in distilled water at $100^{\circ}C$ for 3 hours, and filtered. filtered. Extract samples were freeze-dried at $-80^{\circ}C$. Comparison of anti-oxidant activity in Eucommiae Cortex samples from regional groups was measured in 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging, 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) free radical scavenging, and ferric reducing antioxidant power (FRAP) between regional groups of Eucommiae Cortex. In addition, high-performance liquid chromatography (HPLC) analysis was conducted to compare pinoresinol diglucoside concentration by regional groups. Results : HPLC analysis found that pinoresinol diglucoside concentration, widely known as the marker compound of Eucommiae Cortex, was the highest in Gangwon Yeongwol. There was a significant difference in anti-oxidative activity of Eucommiae Cortex between regional groups as discovered in DPPH, ABTS and FRAP assays. DPPH free radical scavenging was the highest in Jeonbuk Jangsu. ABTS free radical scavenging was the highest in Jeonbuk Jangsu and Chungbuck Jecheon. FRAP was the highest in Jeonbuk Jangsu. Conclusions : Although pinoresinol diglucoside concentration was high, anti-oxidative activity was not proportionately high. Pinoresinol diglucoside concentration was the highest in Gangwon Yeongwol. Anti-oxidative activity was the highest in Jeonbuk Jangsu.
