전호(Anthriscus sylvestris) 뿌리 정유의 항균 및 항산화 작용 (Antibacterial and Antioxidant Activities of the Essential Oil from the Roots of Anthriscus sylvestris)

  • To develop a new effective and safe natural antibiotics and antioxidant the essential oil was extracted from the roots of Anthriscus sylvestris by steam distillation. Its composition was analyzed by GC-MS. The activities of the essential oil fraction and its main components were evaluated against antibiotic-susceptible and -resistant strains of some food-born bacteria. In addition the synergism was examined with this oil combined with antibiotic by checkerboard titer test. The antioxidant activities were determined by in 1,1-diphenyl-2-picryl-hydrazil (DPPH) free radical scavenging activity test and reducing power assay. The essential oil fraction of A. sylvestris revealed significant inhibiting activities against antibiotic-susceptible and -resistant species of Vibrio and Shigella with MICs ranged from 1.00~4.00 mg/ml. It showed synergistic or additive effects when it was combined with amphicillin or trimethoprim/sulfamethoxazole (1 : 9). Additionally, the essential oil fraction of A. sylvestris exhibited significant DPPH free radical scavenging activity and the reducing power.

  • In the present paper, morphological characteristic and PCR analysis were carried not to identify the medicinal origins of Peucedanum decursivum and Anthriscus sylvestris. Some samples in Korean. markets were Peucedanum decursivum misused with rhizomes of Anthriscus sylvestris. The main. origin of Peucedanum decursivum producted was quite different from those of Anthriscus sylvestris in the morphological characteristics. And PCR analysis, the three primer has developed into anew tool for identification of Peucedanum decursivum and Anthriscus sylvestris.

Water Extracts of Anthriscus sylvestris Leaf induces Apoptosis in FaDu Human Hypopharynx Squamous Carcinoma Cells

  • Anthriscus sylvestris (L.) Hoffm. is a perennial herb found widely distributed in various regions of Korea, Europe, and New Zealand. The root of A. sylvestris have been extensively used in the treatment for antitussive, antipyretic, cough remedy in Oriental medicine, but the physiologically active function of the leaf of A. sylvestris is as yet unknown. In this study, we investigated the anti-cancer activity and the mechanism of cell death of water extracts of leaf of Anthriscus sylvestris (WELAS), on human FaDu hypopharyngeal squamous carcinoma cells. Our data showed that WELAS treatment inhibited cell viability in a concentration- and time-dependent manner. In addition, the treatment of WELAS markedly induced apoptosis in FaDu cells, as determined by the viability assay, DAPI stain and FACS analysis. WELAS also increased the proteolytic cleavage of procaspase-3, -9 and PARP (poly(ADP-ribose) polymerase). In addition, exposure to WELAS decreased the expression of Bcl-2 (an anti-apoptotic factor), but increased the expression of Bax (a pro-apoptotic factor), suggesting that mitochondria-dependent apoptotic pathways are mediated in WELAS-induced apoptosis. Taken together, these results indicate that water extracts of leaf of A. sylvestris inhibits cell growth and induces apoptosis via the mitochondrial-dependent apoptotic pathway in FaDu human hypopharyngeal squamous carcinoma cells. Therefore, we propose that the water extracts of leaf of A. sylvestris is a novel chemotherapeutic drug, having growth inhibitory properties and induction of apoptosis in human oral cancer cells.

전호 지하부와 지상부 추출물의 주요성분 및 생리활성 비교 (Comparison of Main Compounds and Physiological Activities of Anthriscus sylvestris (L.) Hoffm. Roots and Aerial Parts Extracts)

  • The number of people suffering from diabetes have been increased around the world. In this study, we investigated the antidiabetic and antioxidant effects of Anthriscus sylvestris(L.) Hoffm and its main compounds. It was divided into root(R) and aerial part(AP) for comparative analysis. Total polyphenol, total flavonoid content was higher in AP extract, but nodakenin content was higher in R(1169.13 ± 6.00 mg/g) extract. Antioxidant activity was also higher in AP extract. To compare antidiabetic efficacy, we analyzed the effects of R and AP extracts on ɑ-glucosidase inhibition(AGI), dipeptidyl peptidase-4(DPP-4) and protein tyrosine phosphatase(PTP)1B activity. R and AP extracts showed similar effects on AGI and DPP-4 activity in a concentration dependent manner, and there was no effect on PTP1B activation. Glucose uptake(139.51 ± 3.19%) in 3T3-L1 cells was more effective in the AP extract-treated group than the positive control, rosiglitazone, group. Both R and AP extracts were effective in protecting against pancreatic beta cell damage caused by hyperglycemia. These results suggest that Anthriscus sylvestris(L.) Hoffm. could be used as a candidate for diabetes treatment.

  • Bioassay-guided chemical investigation of the roots of Anthriscus sylvestris (L.) Hoffm. resulted in the isolation of nine compounds, whose structures were determined by spectroscopic methods. Compound 1 was isolated from this plant for the first time and compounds 3 and 9 were first found from this genus. Different polar fractions of A. sylvestris extract and compounds 1, 6-8 and 9 were evaluated for antitumor activities against HepG2 (human hepatocellular carcinoma), MG-63 (human osteosarcoma cells), B16 (melanoma cells) and HeLa (human cervical carcinoma cells) lines by the MTT method. The petroleum ether fraction of A. sylvestris extract exhibited excellent inhibitory activity with an $IC_{50}$ value of $18.3{\mu}g/ml$. Among the isolates from the petroleum ether fraction, compound 7 showed significant inhibition against the growth of the four tumor cells with $IC_{50}$ values ranging from $12.2-43.3{\mu}g/ml$.

패독산류의 구강편평세포암종 및 악성중피종에 대한 항암 활성 (Anticancer Effect of Paedoksans for Oral Squamous Cell Carcinoma and Malignant Pleural Mesothelioma)

  • In order to search for anticancer agent as therapy of oral squamous cell carcinoma (OSCC) and malignant pleural mesothelioma (MPM) from Korean traditional prescriptions, we selected 58 traditional prescriptions based on a review of the Korean traditional medicine books. Among the selected traditional prescriptions, only water extracts of paedoksan (敗毒散) showed relatively good cytotoxicity at the concentration of $50{\mu}g/ml$. Additionally, we evaluated cytotoxicity for various paedoksans and each herbal ingredient in paedoksans. The root of Anthriscus sylvestris exhibited more cytotoxic effect than any other ingredients in paedoksans. Bioactivity-guided fractionation of the MC layer of Anthriscus sylvestris led to the isolation of deoxypodophyllotoxin (DPT). DPT exhibited dose-dependently significant cytotoxicity against OSCC and MPM cell with nM range. Therefore, DPT from A. sylvestris might be a potential candidate as an effective anticancer therapeutic agent for OSCC and MPM.

  • Chinese crude drug 'JeonHo (前胡)' has been used mainly cough and dyspnea in respiratory infections. To clarify the botanical origin of 'JeonHo' in Korean market, the anatomical characterization of Anthriscus sylvestris, Angelica decursiva and Peucedanum praeruptorum were studied. As a result, it was clarified that 'JeonHo' in Korean market was the root of Peucedanum praeruptorum.

사람 섬유아세포에서 전호(Anthriscus sylvestris Hoffmann)추출물이 콜라겐 생합성에 미치는 영향 (Effects of Anthriscus sylvestris Hoffmann Extract on the Biosynthesis of Collagen in Human Dermal Fibroblasts)

  • 본 연구에서는 전호추출물의 주름개선에 대한 효능을 측정하기 위하여 인간의 섬유아세포에 전호추출물을 처리하여 총콜라겐과 type I procollagen, MMP-1의 생합성에 미치는 영향을 조사하였다. 전호추출물을 인간 섬유아세포에 처리 시 콜라겐의 생합성은 증가시켰지만, MMP-1의 발현에 대해서는 영향을 미치지 못하였다. 이는 전호추출물이 자외선에 의해 영향을 받는 광노화보다는, 노화에 의해 콜라겐 생합성율이 감소되는 내인성노화에 훨씬 유효함을 시사한다. 전호추출물을 처리시 95% EtOH 추출물의 경우 25%까지 총콜라겐의 생합성을 증가시켰고, EtOAc층은 28%, EtOAc의 E6 소분획의 경우 50% 증가시켰으므로, 콜라겐 생합성에 영향을 미치는 유효성분은 EtOAc층에 대부분 함유되어 있을 것으로 사료되고, 이는 확인되지 않은 미지의 효능 성분이 존재하거나, 다양한 활성 성분들의 상호 작용에 의한 것으로 판단된다. 결론적으로 전호추출물은 콜라겐 생합성을 촉진함으로 내인성노화에 의한 주름생성 및 탄력저하를 개선할 수 있는 천연 유용자원으로 이용할 수 있을 것으로 사료된다.

  • Activity-guided fractionation of the roots of Anthriscus sylvestris resulted in the isolated and characterization of five cytotoxic compounds, deoxypodophyllotoxin (1), falcarindiol (2), and angeloyl podophyllotoxin (5) from the hexane soluble fraction and morelensin (3), bursehernin (4) from the choloroform soluble fraction. It is the first report of the occurrence of compound 5 in nature.
