Effect of a Freshwater Bivalve (Unio douglasiae) and a Submerged Plant (Potamogeton crispus) on the Growth Inhibition of a Cyanobacterium Oscillatoria sp. (담수패류(Unio douglasiae)와 침수식물(Potamogeton crispus)의 유해 남조 Oscillatoria sp. 성장억제 효과)

  • This study examined the inhibition effects of a freshwater bivalve (Unio douglasiae) and a submerged plant (Potamogeton crispus) on the cyanobacterial bloom (Oscillatoria sp.). The experiment were conducted in aquarium $(50cm{\times}65cm{\times}120cm)$ with lake sediments in the bottom of the aquarium in 10 cm thick. Before the experiments, artificial cyanobacterial bloom was induced with the addition of lake sediment and CB medium. Total 12 transparent acrylic cylinders (${\Phi}19cm$, height 40 cm) were placed in the aquarium, and within which bivalves and plants were placed in various conditions such as the control (C), plant addition (P:5 stems), mussel addition (U:2 individuals), and both mussel and plant addition (PU: the same quantity as used in each treatment). The experiment was conducted in triplicate during 7 days. pH, dissolved oxygen (DO), electric conductivity (EC), salinity, cyanobacterial cell density, chlorophyll-${\alpha}$ concentration, and mussel filtering rate were monitored daily. At the end of the experiment, total phosphorus (TP), total nitrogen (TN), and plant height and weight were measured. Overall, a large degree of cyanobacterial growth inhibition appeared in both P and U treatments, and the effect was highest in the U treatment, followed by P and PU. The combined treatment of both U and P did not show any synergic effects compared to the effect in separated treatment. In all enclosures of the treatments chlorophyll-${alpha}$ (Chl-${alpha}$) concentration decreased until 36 hours after the additions of the plants and mussels. In contrast, Chl-${alpha}$ concentrations increased in PU enclosures after 36 hours. The same trend was shown in the cell density of Oscillatoria. pH and DO gradually decreased until 120 and 144 hours, respectively, in the P and PU enclosures. TP concentration increased in the mussel enclosures (U and PU), while TN concentration largely decreased in the plant enclosures (P and PU). Our results suggest that applied bivalve (Unio) and submerged plant (Potamogeton) seemed to have a potential effect on the growth inhibition of cyanobacteria, but their combined application may have an antagonistic effect to diminish the degree of the inhibition.

Effect of Mixed Application of Cinosulfuron with Dymron on Rice (Cinosulfuron과 Dymron의 혼합처리(混合處理)가 수도(水稻)(Oryza sativa L.)의 초기생육(初期生育)에 미치는 영향(影響))

  • To investigate the safening effect of dymron against cinosulfuron, the responses of 104 rice cultivars and lines to single and mixture treatments of cinosulfuron [3-(4.6-dimethoxy-1,3,5-triazin-2-yl)-1-[2-(2-methoxyethoxy)-phenylsulfonyl]-urea] and dymron [1-(${\alpha}$, ${\alpha}$-dimethylbenzyl)-3-p-tolyl urea] were evaluated. The responses of 104 rice cultivars and lines to cinosulfuron were differed. Among the rice cultivars, growth of Calrose, Dasukei, Hwajinhyeo, and Milyang 105 was significantly inhibited by cinosulfuron but there was no inhibition in Daeseongbyeo, Iri 371, Jangseongbyeo, Taebaegbyeo and IR 74. The japonica types were generally inhibited more than the indica and indica${\times}$japonica types. When Kwangmyungbyeo was treated with the mixtures of cinosulfuron 48g and 96g ai/ha with dymron 450g ai/ha, the inhibition of plant height was significantly reduced regardless of dymron concentrations. With increase in cinosulfuron concentration, the safening effect of dymron against cinosulfuron was more evident in plant height than fresh weight. On the other hand, dymron did not have any safening effects of cinosulfuron on Echinochloa crus-galli at any concentrations. The mixtures of dymron and cinosulfuron produced antagonistic effects on the plant height regardless of rice cultivars and treatment concentrations. At cinosulfuron 24g ai/ha, the effects of the mixtures of cinosulfuron and brassinolide on the plant height of Kwangmyungbyeo and IR 74 were similar irrespective of brassinolide concentrations but the inhibition of Kwangmyungbyeo was greater than that of IR 74 as the concentrations of cinosulfuron increased. The effects of the mixtures of cinosulfuron at concentration of 96g ai/ha with various ABA levels on the plant height of both Kwangmyungbyeo and IR 74 showed synergistic effect regardless of ABA concentration. However, the response of fresh weight of both cultivars treated with cinosulfuron at 96g ai/ha with ABA synergisism was observed in Kwangyungbyeo while IR 74 showed antagonism.

Isolation and Identification of a Streptomyces sp. that Produces Antibiotics Against Multidrug - Resistant Acinetobacter baumannii (다제내성 Acinetobacter baumannii의 생장을 억제하는 항생물질을 생산하는 방선균의 분리.동정 및 항균효과)

  • I isolated the actinomycete strain KH223 from soil samples collected from the Kye Ryong mountain area. This strain is antagonistic to the multidrug-resistant Acinetobacter baumannii. KH223 was confirmed as belonging to the genus Streptomyces based on the scanning electronmicroscopy(SEM) observations of the diaminopimelicacid(DAP) type and morphological and physiological characteristics. Comparison of the 16S rDNA nucleotide sequences revealed that KH223 has a relationship with Streptomyces galbus. Production of antibiotics by KH223 was most favorable when cultured on a glucose, polypeptone, and yeast extract(PY) medium for 6 days at 27$^{\circ}C$. The supernatant was found to exhibit an antimicrobial effect on various kinds of bacteria and fungi. Particularly, butanol and ethylacetate extracts of KH223 and cyclo(trp-trp) exhibited significant activity against A. baumannii at concentration ranges of 0.8-12.5 ${\mu}g$/mL, 5.0-25 ${\mu}g$/mL and 12.5${\rightarrow}$100 ${\mu}g$/mL, respectively. Moreover, in contrast to cyclo(trp-trp) had shown to activity against Micrococcus luteus JCM 1464 at the concentration of 12.5 ${\mu}g$/mL, the butanol extract of KH223 showed significant activity against Bacillus subtilis IAM 1069 and Micrococcus luteus JCM 1464 at the concentration of 0.4 and 0.8 ${\mu}g$/mL, respectively. These results suggest that KH223 may have a great potential in the production of new antibiotics to combat multidrug-resistant pathogens and further studies may be warranted for the same.

Effect of a Common Medium on the Growth of Nitrogen Fixer Rhizobium and Phosphate Solubilizer Bacillus megaterium (질소고정균(Rhizobium)과 인산가용화균(Bacillus megaterium)의 동시배양을 위한 배양조건 탐색)

  • Mass culturing of two beneficial organisms used as biofertilizers for crops would reduce the risks in production and minimize the capital involved and this demands appropriate media that supports both organism and also selection of organisms that are not antagonistic to each other. A study was initiated to culture a nitrogen fixer (Rhizobium) and phosphate solubilizer (Bacillus megaterium) in a single medium and to study their growth patterns and shelf life in carrier. The growth of Rhizobium and Bacillus megaterium was assessed in different media and a slight modification in the traditional yeast extract mannitol media promoted the growth of both the organisms. The growth of the individual organisms in the modified medium was assessed by estimating the population at regular intervals and compared to their original medium. Maximum population of Rhizobium and phosphobacteria was at 60 hr when the phosphiobacteria inoculation of later was after 48 hr of Rhizobium inoculation. The shelf life of the individual inoculants in the inoculant containing both the organism in a sterile carrier base revealed no significant differences compared to individual organisms inoculated in a sterilized carrier. The population of both organisms in carrier based mixed inoculant remained at $10^8$ cells till 90 days.

Diversity of Root-Associated Paenibacillus spp. in Winter Crops from the Southern Part of Korea

  • The genus Paenibacillus is a new group of bacilli separated from the genus Bacillus, and most of species have been isolated from soil. In the present study, we collected 450 spore-forming bacilli from the roots of winter crops, such as barley, wheat, onion, green onion, and Chinese cabbage, which were cultivated in the southern part of Korea. Among these 450 isolates, 104 Paenibacillus-like isolates were selected, based on their colony shape, odor, color, and endospore morphology, and 41 isolates were then finally identified as Paenibacillus spp. by 16S rDNA sequencing. Among the 41 Paenibacillus isolates, 23 were classified as P. polymyxa, a type species of the genus Paenibacillus, based on comparison of the 16S rDNA sequences with those of 32 type strains of the genus Paenibacillus from the GenBank database. Thirty-five isolates among the 41 Paenibacillus isolates exhibited antagonistic activity towards plant fungal and bacterial pathogens, whereas 24 isolates had a significant growth-enhancing effect on cucumber seedlings, when applied to the seeds. An assessment of the root-colonization capacity under gnotobiotic conditions revealed that all 41 isolates were able to colonize cucumber roots without any significant difference. Twenty-one of the Paenibacillus isolates were shown to contain the nifH gene, which is an indicator of $N_{2}$ fixation. However, the other 20 isolates, including the reference strain E681, did not incorporate the nifH gene. To investigate the diversity of the isolates, a BOX-PCR was performed, and the resulting electrophoresis patterns allowed the 41 Paenibacillus isolates to be divided into three groups (Groups A, B, and C). One group included Paenibacillus strains isolated mainly from barley or wheat, whereas the other two groups contained strains isolated from diverse plant samples. Accordingly, the present results showed that the Paenibacillus isolates collected from the rhizosphere of winter crops were diverse in their biological and genetic characteristics, and they are good candidates for further application studies.

Antilisterial Activity of Bacteriocin Produced by Enterococcus faecium MJ5-14 (Enterococcus faecium MJ5-14가 생산한 박테리오신의 항리스테리아 활성)

  • Enterococcus faecium MJ5-14 isolated from Meju produced a bacteriocin, which was antagonistic towards Listeria monocytogenes. Bacteriocin activity reached a maximum (640 BU/mL) after incubation for 12 hr, the early stationary phase, then dropped after the late stationary phase. Bacterocin of E. faecium MJ5-14 was extremely active against a wide range of Listeria species, including L. monocytogenes with sensitives up to about 640 BU/mL. In case of mixed culture with 105 CFU/mL L. monocytogenes and 105 CFU/mL E. faecium MJ5-14, the inhibitory effect against L. monocytogenes at $37^{\circ}C$ was higher than at $25^{\circ}C$. The mode of action was identified as bactericidal, because the addition of 100 BU/mL this bacteriocin to cell suspensions of L. monocytogenes KCTC 3569, led to a marked decrease in the number of viable cells. Further, when held in contact with bacteriocin of E. faecium MJ15-14 for 12 hr, L. monocytogenes KCTC 3569 displayed the disruption of the cells and an important efflux of the intracellular material.

Changes in the Endothelin-1-induced Contraction of Aorta in Streptozotocin-induced Diabetic Rats

  • Vascular diseases are significant complications of diabetes mellitus (DM), and the endothelial cells may play a pivotal role in the development of vascular disease in DM. Endothelin-1 (ET-1) released from endothelium is a potent vasoconstrictor peptide and circulating level of ET-1 is increased in a variety of disease states. The purpose of this study was to determine the changes of responsiveness to ET-1 in DM, and we experimented on the changes in the ET-1-induced contraction, levels of nitrite and lipid peroxidation, and ET-1 immunoreactivity in aorta from streptozotocin-induced DM rats. DM was induced by single injection of streptozotocin (55 mg/kg, i.p.). The immunoreactive ET-1 levels in endothelial layer of thoracic aorta were much higher in DM rats than control rats. Nitrite in tissue homogenate was decreased and plasma nitrite was increased in DM rats. Malondialdehyde (MDA) was significantly increased in DM rats and cGMP was not significantly different between control and DM rats. ET-1 produced concentration- dependent contractile responses that are significantly attenuated in DM rats compared to controls. In the presence of selective $ET_A$ receptor antagonist BQ610, the maximum contraction was decreased and the concentration ratios for BQ610 yielded $pA_2$ values of 7.3 (slope, 0.65) in control rats, whereas BQ610 had no antagonistic effect on ET-1-induced contraction in DM rats. However, pretreatment with BQ788, an $ET_B$ receptor antagonist, maximum response was decreased and the dose-response curves for ET-1 were shifted to the right in both groups and $pA_2$ values were 7.9 and 7.7 (slope, 1.05 in control and DM rats), respectively. IRL 1620 and sarafotoxin S6c, $ET_B$ agonists, induced relaxation in control rats but not in DM rats. These results indicate that endothelial cell dysfunction and enhanced immunoreactivity of ET-1 have been found in DM rat and ET-1-induced contraction was attenuated in DM rat. These attenuated responses might be at least in part caused by the alteration of $ET_A$ receptor properties (e.g. desensitization), and partly related with an alteration in intracellular mechanism for contraction to ET-1.

Comparative Study of Labor Disputes in the Period of Restructuring: the Cases of Hyundai Motor and Power Generation Companies (구조조정기 노사분쟁의 사례비교연구: 현대자동차와 발전회사의 분규를 중심으로)

  • This paper analyzes the two cases of labor disputes (Hyundai Motor in 1998 and Power Generation Companies in 2002) in the period of restructuring, by applying the behavioral theory of labor negotiations as a comparative framework. The paper compares th backgrounds of the labor disputes, core issues, bargaining processes, and evolutionary patterns and consequences of the labor disputes at the two cases. The common features, found in the two dispute cases, are strong mistrust and exclusive bargaining attitude between labor unions and management, little feasibility of contract zone in bargaining proposals by the two parties, heteronomous dispute resolution by the intervention of the government, and the lack of learning effect gained from the experience of labor disputes. This comparative case study identifies that the confrontational labor-management relations at the firm level is re-produced by a regressive process of the following circulation: labor-management distrust $\rightarrow$ interest conflict in bargaining demand $\rightarrow$ exclusive bargaining attitude $\rightarrow$ the experience of antagonistic dispute $\rightarrow$ deepened distrust. In conclusion, four parties-labor unions, management, the government, and public press - are required to make much effort to replace the vicious circle of labor-management confrontation by a virtueous cycle of labor-management cooperation.

Influence of Calcium Supply on the Growth, Calcium and Oxalate Contents, Mineral Nutrients and Ca-oxalate Crystal Formation of Cucumber (오이생육, 칼슘, 옥살산 및 무기성분 함량 및 칼슘-옥살산염 형성에 대한 칼슘처리 효과)

  • Although the roles of calcium in plant are widely known, little is known about on an antagonistic effect of macro elements, oxalate biosynthesis and main shape of crystal in cucumber plant organs. Seeds of cucumber (Cucumis sativus cv. Ijoeunbackdadagi) were germinated in perlite tray supplied with distilled-deionized water. Seedlings were transplanted into aerated containers with a half strength of Ross nutrient solution. Ca levels treated in media were as follows; No-Ca, $Ca(NO_3)_2$ 0.25, 1.25 and 2.5 mmol $L^{-1}$, and $Ca(NO_3)_2$ 2.5 mmol $L^{-1}$ + $CaCl_210$, 25 and 50 mmol $L^{-1}$. Ca-deficient and -excessive conditions severely reduced cucumber growth, as compared to the control, and adversely affected an accumulation of macro elements (N, P, K, and Mg). Calcium favorably induced oxalate (acid-soluble) synthesis in leaves and roots of cucumber plant, but not in stem. Acid-soluble oxalate contents in leaves proportionally increased with Ca supply levels (0.91, P<0.001), however, this pattern was not observed in stem and roots. Ca-oxalate crystal formation and compositional analysis were examined using SEM-EDS technique in cucumber leaves. The main type of crystal revealed a prismatic crystal and main components were Ca, Na and Cl.

Evaluation of Bio-Control Efficacy of Trichoderma Strains against Alternaria alternata Causing Leaf Blight of Ashwagandha [Withania somnifera (L.) Dunal]

  • Ashwagandha is an important ancient medicinal crops, being affected with many diseases, among which leaf blight caused by Alternaria alternata has become the constraint resulting in huge yield losses. Continuous usage of chemical methods leads to environment, soil and water pollution. Whereas biological control of diseases is long lasting, inexpensive, eco-friendly and harmless to target organisms. In this context, it is aimed to evaluate five Trichoderma strains viz. Trichoderma virens IMI-392430, T. pseudokoningii IMI-392431, T. harzianum IMI-392432, T. harzianum IMI-392433 and T. harzianum IMI-392434 as bio-control efficacy against A. alternata and growth promoting effect in Ashwagandha. All the Trichoderma strains had varied antagonistic effects against the pathogen. In dual culture technique, the strain T. harzianum IMI-392433 showed maximum percentage inhibition of mycelial growth (54.89%) followed by T. harzianum IMI-392432 (53.83%), T. harzianum IMI-392434 (48.94%) and T. virens IMI-392430, (43.62%) against the pathogen, while the least inhibition percentage was observed with the T. pseudokoningii IMI-392431 (36.60%). The culture filtrate of the Trichoderma strain, T. harzianum IMI-392433 recorded highest inhibition on the mycelial growth (39.05%) and spore germination (80.77%) of pathogen and the lowest was recorded in T. pseudokoningii IMI-392431 (20.45 and 50%). Moreover, seeds treated with spore suspension of the strain T. harzianum IMI-392433 reduced the percentages of disease severity index significantly. The strain T. harzianum IMI-392433 also significantly increased seed germination %, seedling vigor and growth of Ashwagandha. The correlation matrix showed that root yield per plant of Ashwagandha had significant and positive correlation with plant height (r=0.726⁎⁎), number of leaf (r=0.514⁎⁎), number of primary branch (r=0.820⁎⁎), number of secondary branch (r=0.829⁎⁎), fresh plant weight (r=0.887⁎⁎), plant dry weight (r=0.613⁎⁎), root length (r=0.824⁎⁎), root diameter (r=0.786⁎⁎), root dry weight (r=0.739⁎⁎) and fresh root weight (r=0.731⁎⁎). The significant and negative correlation (r=-0.336⁎⁎) was observed with the root yield and percentages of disease severity index. The study recognized that the T. harzianum IMI-392433 strain performed well in inhibiting the mycelial growth and reduced the percentages of disease severity index of pathogen as well as increased the plant growth in Ashwagandha.
