Re-evaluation of the Optimum Dietary Vitamin C Requirement in Juvenile Eel, Anguilla japonica by Using L-ascorbyl-2-monophosphate

  • This study was conducted to re-evaluate the dietary vitamin C requirement in juvenile eel, Anguilla japonica by using L-ascorbyl-2-monophosphate (AMP) as the vitamin C source. Five semi-purified experimental diets were formulated to contain 0 ($AMP_0$), 30 ($AMP_{24}$), 60 ($AMP_{52}$), 120 ($AMP_{108}$) and 1,200 ($AMP_{1137}$) mg AMP $kg^{-1}$ diet on a dry matter basis. Casein and defatted fish meal were used as the main protein sources in the semi-purified experimental diets. After a 4-week conditioning period, fish initially averaging $15{\pm}0.3$ g (mean${\pm}$SD) were randomly distributed to each aquarium as triplicate groups of 20 fish each. One of five experimental diets was fed on a DM basis to fish in three randomly selected aquaria, at a rate of 3% of total body weight, twice a day. At the end of the feeding trial, weight gain (WG) and specific growth rate (SGR) for fish fed $AMP_{52}$ and $AMP_{108}$ were significantly higher than those recorded for fish fed the control diet (p<0.05). Similarly, feed efficiency (FE) and protein efficiency ratio (PER) for fish fed $AMP_{52}$ were significantly higher than those for fish fed the control diet (p<0.05). Broken-line regression analysis on the basis of WG, SGR, FE and PER showed dietary vitamin C requirements of juvenile eel to be 41.1, 41.2, 43.9 and 43.1 (mg $kg^{-1}$ diet), respectively. These results indicated that the dietary vitamin C requirement could range from 41.1 to 43.9 mg $kg^{-1}$ diet in juvenile eel when L-ascorbyl-2-monophosphate was used as the dietary source of vitamin C.

Influence of Quaternary Benzophenantridine and Protopine Alkaloids on Growth Performance, Dietary Energy, Carcass Traits, Visceral Mass, and Rumen Health in Finishing Ewes under Conditions of Severe Temperature-humidity Index

  • Twenty $Pelibuey{\times}Katahdin$ ewes ($35{\pm}2.3kg$) were used to determine the effects of the consumption of standardized plant extract containing a mixture of quaternary benzophenanthridine alkaloids and protopine alkaloids (QBA+PA) on growth performance, dietary energetics, visceral mass, and ruminal epithelial health in heat-stressed ewes fed with a high-energy corn-based diet. The basal diet (13.9% crude protein and 2.09 Mcal of net energy [NE] of maintenance/kg of dry matter) contained 49.7% starch and 15.3% neutral detergent fiber. Source of QBA+PA was Sangrovit RS (SANG) which contains 3 g of quaternary benzophenathridine and protopine alkaloids per kg of product. Treatments consisted of a daily consumption of 0 or 0.5 g SANG/ewe. Ewes were grouped by weight and assigned to 10 pens (5 pens/treatment), with two ewes per pen. The experimental period lasted 70 days. The mean temperature humidity index during the course of this experiment was $81.7{\pm}1.0$ (severe heat stress). There were no treatment effects on water intake. Dry matter intake was not affected (p = 0.70) by treatments, but the group fed SANG had a numerically (11.2%) higher gain in comparison to the control group, SANG improved gain efficiency (8.3%, p = 0.04), dietary NE (5.2%, p<0.01) and the observed-to-expected NE (5.9%, p<0.01). Supplemental SANG did not affect ($p{\geq}0.12$) carcass characteristics, chemical composition of shoulder, and organ weights (g/kg empty body weight) of stomach complex, intestines, and heart/lung. Supplemental SANG decreased liver weight (10.3%, p = 0.02) and increased visceral fat (16.9%, p = 0.02). Rumen epithelium of ewes fed SANG had lower scores for cellular dropsical degeneration (2.08 vs 2.34, p = 0.02), parakeratosis (1.30 vs 1.82, p = 0.03) and neutrophil infiltration (2.08 vs 2.86, p = 0.05) than controls. It is concluded that SANG supplementation helped ameliorate the negative effects of severe heat on growth performance of feedlot ewes fed high-energy corn-based diets. Improvement in energetic efficiency may have been mediated, in part, by anti-inflammatory effects of supplemental SANG and corresponding enhancement of nutrient uptake.

  • The aim of this study was to investigate the effects of dietary supplementation of pelleted-Italian Ryegrass (IRG) as a source of fiber on reproduction performance in pregnant sows. A total of 24 pregnant sows were randomly assigned to four dietary treatments, which was given a corn-soybean diet with 0%, 10%, 20%, and 30% pelleted-IRG from 105 days prepartum to 7 days postpartum. During experimental period, the sows fed the IRG supplemented diet showed the lower feed intake than the sows fed the control diet (p<0.05). The changes of body weight in sows from initial to pre- and/or post-partum was significantly smaller in sows fed the IRG supplemented diet than control group. It is thought that the lower weight gain in IRG supplemented groups is caused by low feed intake. Although there was no significant difference, sows fed the IRG supplemented diet tended to increase the litter size and birth weight in piglets compared with sows fed the control diet. This result suggests that the dietary supplementation of IRG has the positive effects to improve the reproductive performance in sows. But, the excessive feeding of IRG to sows might cause to retard the days of return to estrus, and decrease the contents of solid, milk protein, and milk fat in colostrum. Thus, the addition of about 10% IRG is desirable to increase the reproductive performance. Meanwhile, the feeding energy diet is better effective than feeding the fiber diets to improve overall productivity in sows after postpartum.

Oligosaccharides are a key factor in prediction of amino acid digestibility in soybean meal of different origins when fed to growing pigs

  • Objective: The objective of this experiment was to determine apparent ileal digestibility (AID) and standardized ileal digestibility (SID) of crude protein (CP) and amino acid (AA) in 15 sources of soybean meal (SBM) produced from soybeans from different countries and subsequently to establish equations for predicting the AID and SID in SBM based on their chemical composition. Methods: Eighteen barrows ($57.9{\pm}6.1kg$) fitted with a simple T-cannula were allotted into three $6{\times}6$ Latin square designs. Each period comprised a 6-d adaption period followed by a 2-d collection of ileal digesta. The 15 test diets included SBM as a sole source of AA in the diet. Another nitrogen-free diet was used to measure basal endogenous losses of CP and AA. Chromic oxide (0.3%) was used as an inert marker in each diet. Results: The AID of lysine in SBM from China and USA tended to be greater than in SBM from Brazil (p<0.10). The SID of valine and proline in SBM from China was greater than in SBM from Brazil (p<0.05). The SID of lysine, threonine, cysteine and glycine in SBM from China tended to be greater than in SBM from Brazil (p<0.10). From a stepwise regression analysis, a series of AID and SID prediction equations were generated. The best fit equations for lysine in SBM were: AID lysine = 1.16 sucrose-1.81 raffinose+82.10 ($R^2=0.69$, p<0.01) and SID lysine = 1.14 sucrose-1.93 raffinose-0.99 ether extract (EE)+85.26 ($R^2=0.77$, p<0.01). Conclusion: It was concluded that under the conditions of this experiment, the oligosaccharides (such as sucrose and raffinose) can be used to predict the AID and SID of AA in SBM with reasonable accuracy.

Effects of Cultivars and Seeding Dates on Chemical Composition and Energy Content of Switchgrass (Panicumvirgatum L.) in Republic of Korea

  • The objective of this study was to determine the best performing switchgrass (Panicumvirgatum L.) cultivar with three different seeding dates as a bioenergy source in Republic of Korea. Split-plot in time with three replications was performed and three switchgrass cultivars, Carthage (CT), Cave-in-Rock (CIR), and Forestburg (FB) were used in this experiment from 2009 to 2012. Plots were seeded on April 23, May 4, and May 13, 2009 and were harvested once in November each year. No fertilizer was applied to the field for the first year; however, in second and third years (June 2010 and May 2011, respectively), N, $P_2O_5$ and K2O fertilizers were applied in 67,45 and 90 kg $ha^{-1}$, respectively. Soil pH (5.9) and climate condition including temperature ($10.4{\sim}17.5^{\circ}C$) and precipitation (89.4~109.8 mm $month^{-1}$) were suitable for switchgrass cultivation. Total dry matter yields were higher in CT and CIR compared to FB and were 16.9, 15.9, and 4.5 ton $ha^{-1}$, for CT, CIR, and FB, respectively (p<0.0001). The samples were analyzed for dry matter (DM), crude protein (CP), neutral detergent fiber (NDF), acid detergent fiber (ADF), crude fiber (CF), ether extract (EE), and ash. No significant differences in energy content (p = 0.96) and chemical composition among cultivars (p>0.05) were found. Seeding dates did not affect DM yield (ton $ha^{-1}$), chemical composition and energy content significantly (p>0.05). Significant difference was observed for heights among CT, CIR, and FB (177.59, 169.98, and 94.89 cm, respectively, p = 0.0002). In conclusion, based on soil characteristics and climate condition in Korea compared to other countries, switchgrass can be cultivated successfully. Considering dry matter yield and energy content of these three cultivars of switchgrass CT and CIR adapted better to climate in Middle Eastern of Republic of Korea than Forestburg for bioenergy purpose.

Effect of High Phytase Inclusion Rates on Performance of Broilers Fed Diets Not Severely Limited in Available Phosphorus

  • Phytate is not only an unavailable source of phosphorus (P) for broilers but it also acts as an anti-nutrient, reducing protein and mineral absorption, increasing endogenous losses and reducing broiler performance. The objective of this study was to investigate the anti-nutritional effects of phytate by including high levels of phytase in diets not severely limited in available P. A total of 768 male Arbor Acres broilers were distributed in six treatments of eight replicate pens of 16 birds each consisting of a positive control diet (PC), positive control with 500 FTU/kg phytase, negative control (NC) diet with lower available P and calcium (Ca) levels and the same NC diet with 500, 1,000 or 1,500 FTU/kg phytase. Body weight gain (BWG), feed intake (FI), feed conversion ratio (FCR) and mortality were determined at 21 and 35 d of age while foot ash was determined in four birds per pen at 21 d of age. FI, FCR and foot ash where not affected by the lower mineral diets at 21 d of age nor by the enzyme inclusion but broilers fed lower Ca and available P diets had lower BWG. At 35 d of age no difference was observed between broilers fed the positive or NC diets but broilers fed 500, 1,000 and 1,500 FTU/kg on top of the NC diet had better FCR than broilers fed the positive control diet. When compared to birds fed a diet adequate in P, birds fed the same diet included with 500, 1,000 and 1,500 FTU/kg of phytase in marginally deficient available P and Ca diets had an improvement of performance. These results support the concept that hydrolysing phytate and reducing the anti-nutritional effects of phytate improves bird performance on marginally deficient diets that were not covering the P requirement of birds.

Effects of Monensin Administation on Mammary Function in Late Lactating Crossbred Holstein Cattle

  • An experiment was carried out to study the effect of monensin administration on mammary functions in crossbred Holstein cattle. Fourteen non-pregnant late lactating crossbred Holstein cattle, approximately 270 days postpartum, were selected for the experiment. They were divided into two groups of 7 animals each. Seven animals in the treated group were given sodium monensin orally in a slow-release capsule. Animals in both control and treated groups were fed the similar diet to maintain milk production and body score at 2.5. Rice straw was fed as a source of dietary fiber throughout the experimental period. After monensin administration, a significant increase in the molar percent of ruminal propionate (p<0.05) and a significant decrease in the molar percent of ruminal acetate (p<0.05) were apparent in comparison to the pretreated period. The ratio of acetate to propionate concentration decreased significantly after monensin administration (p<0.05), while it was maintained at the similar level throughout the period of experiment in the control group. Monensin did not affect the molar percent of ruminal butyrate and valerate. The concentration of milk allantoin between the control group and monensin treated group was not different. An excretion rate of allantoin in milk decreased in animals treated with monensin (p<0.05). Mammary blood flow did not show significant difference between control and monensin treated groups. The plasma glucose concentration, arteriovenous concentration difference and mammary gland uptake of glucose remained constant in both groups. Milk yield of the later stage of lactation in the control group declined during lactation advance while a tendency to increase in the milk yield was apparent after 21 days monensin administration. Milk compositions for concentration of lactose, fat and protein in both control group and monensin treated group did not change throughout the experimental periods. From these results, it can be concluded that the action of monensin could affect the ruminal fermentation pattern. Monensin could not increase milk yield in the late lactating period.

Evaluation of Twice Decorticated Sunflower Meal as a Protein Source Compared with Soybean Meal in Pig Diets

  • A series of four experiments was conducted to compare nutritional values of decorticated sunflower meals against soybean meal, in diets for pigs from weaning (Exp. 1 and 2) to finishing (Exp. 3 and 4). All experimental diets were prepared compensating for the energy content by using vegetable oil and the lysine content was matched using synthetic L-Lysine HCl. Twenty-one day old pigs were fed either corn-soybean meal based diet (CSBM) or corn- twice-decorticated sunflower meal based diet (CDSM) for four weeks (Exp. 1). There was no difference in performances between treatment groups. In Exp. 2, corn-non-decorticated sunflower meal based diet (CNSM) was added to the existing two treatments. Twenty-one day old pigs were fed three experimental diets for four weeks. Pigs fed CNSM had a lower weight gain and feed intake than other treatments (p<0.05). There was no difference between pigs fed CSBM and CDSM (Exp. 2). Growth performance of growing pigs was also greater (p<0.05) in pigs fed corn starch-twice- decorticated sunflower meal based diet (CSDSM) than pigs fed corn starch-non-decorticated sunflower meal based diet (CSNSM) during the eight week feeding trial (Exp. 3). There was no difference between pig fed corn starch-soybean meal based diet (CSSBM) and CSDSM (Exp. 3). In Exp. 4, growing pigs were fed three experimental diets (CSBM, CDSM, and barley-twice- decorticated sunflower meal based diet; BDSM) until the slaughter. There was no difference in growth performance of pigs during growing and finishing periods among treatments. However, pigs fed CSBM had a higher carcass dressing percentage (p<0.05) than pigs fed CDSM and BDSM. Pigs fed BDSM diet had a lower fat tissue percentage than other groups (p<0.05). The twice-decorticated sunflower meal can be used as a substitute for soybean meal in pig diets. The performances of piglets and growing-finishing pigs were not affected when soybean meal was replaced by twice-decorticated sunflower meal. This substitution needs the contribution of synthetic lysine and vegetable oil as sources of complementary nutrients to match the nutrient profile.

Isolation, Production, and Characterization of Protease from Bacillus subtilis IB No. 11

  • A potent protein degrading bacterium was isolated from soil samples of different environments. Polyphasic taxonomic studies and phylogenetic 16S rRNA sequence analyses led to identify the isolate IB No. 11 as a strain of Bacillus subtilis. The isolated strain was recognized to produce protease constitutively, and the maximum production (1.64 units/ml) was attained in a shake flask culture when the isolate was grown at $40^{\circ}C$, for 32 h in basal medium supplemented with starch (0.25%) and gelatin (1.25%) as sole carbon and nitrogen source, respectively. The optimum pH and temperature for the protease activity were determined to be pH 7.0 and $50^{\circ}C$, respectively. $Ca^{2+}$ and $Mn^{2+}$ enhanced remarkably the protease activity but neither showed positive effect on the protease's thermal stability. In addition, it was observed that the protease was fairly stable in the pH range of 6.5-8.0 and at temperatures below $50^{\circ}C$, and it could be a good candidate for an animal feed additive. The inhibition profile of the protease by various inhibitors indicated that the enzyme is a member of serine-proteases. A combination of UV irradiation and NTG mutagenesis allowed to develop a protease hyper-producing mutant strain coded as IB No. 11-4. This mutant strain produced approximately 3.23-fold higher protease activity (6.74 units/mg) than the parent strain IB No. 11 when grown at $40^{\circ}C$ for 32h in the production medium. The protease production profile of the selected mutants was also confirmed by the zymography analysis.

Different Levels of N Supply Impacts on Seed Yield by Modulating C and N Metabolism in Brassica Napus

  • Oilseed rape is known to crop having low nitrogen use efficiency (NUE) but requires high levels of N fertilizer. NUE is associated with N remobilization from source to sink organ, consequently affects seed yield. Remobilization of leaf N is also related to transport of C/N metabolites in phloem. However, interaction between seed yield and phloem transport was not fully documented. In response to seed yield, N and C metabolites and their transport into seed from bolting to pod filling stage investigated in two contrasting genotypes (Capitol and Pollen) cultivated under ample (HN) or limiting nitrate (LN) supply. Seed yield was significantly reduced in N limitation and its reduction rate was much lower in Capitol than in Pollen compared to HN treated plants. Amino acid and protein content was higher in Capitol than in Pollen at bolting stage. They gradually decreased during plant development but not significant between two cultivars and/or two treatments. Glucose, fructose and sucrose content were 1.8-,1.6- or 1.25-fold higher in LN condition than in HN condition, respectively. Amino acid and sucrose content in phloem were largely higher in Capitol than in Pollen under LN condition. These results indicate that the higher seed yield might be related to greater transport ability of amino acid and sucrose in phloem under LN condition.
