Nutrient Intake, Acid Base Status and Growth Performance of Thalli Lambs Fed Varying Level of Dietary Cation-anion Difference

  • Influence of -110, +110, +220 and +330 mEq/kg of dry matter (DM) dietary cation-anion difference (DCAD) on growth performance of Thalli lambs were examined in a randomized complete block design. Four DCAD diets were randomly allotted to four groups, with ten lambs in each group. A linear increase in nutrient intake was recorded with increasing DCAD level. The digestibilities of nutrients were higher in lambs fed -110 DCAD diet than those fed +110, +220 and +330 DCAD diets. Lambs fed +330 DCAD diet had higher nitrogen balance than those fed -110 and +110 DCAD diets. Blood pH and serum $HCO_3$ increased with increasing DCAD level. Serum chloride was higher in lambs fed -110 DCAD diet, while serum (Na+K)-(Cl+S) increased linearly with increasing DCAD level. Serum calcium increased with decreasing DCAD level while serum magnesium and phosphorus remained unaffected. Lambs fed -110 DCAD diet had higher Ca balance than those fed +110, +220 and +330 DCAD diets. Urine pH increased with increasing DCAD level. Lambs fed +220 and +330 DCAD diets gained more weight than those fed -110 and +110 DCAD diets. In conclusion, increased DCAD level not only increased the dry matter intake but also improved the weight gain of growing Thalli lambs.

The Effects of Dietary Garlic Powder on the Performance, Egg Traits and Blood Serum Cholesterol of Laying Quails

  • This study was conducted to study the effects of dietary garlic powder on laying performance, egg traits and blood serum cholesterol level of quails. A total of three hundred quails (Coturnix coturnix japonica) aged nine weeks were used. They were allocated to 3 dietary treatments. Each treatment comprised 5 replicates of 20 quails. The diets were supplemented with 0, 5 and 10 g/kg garlic powder. The experimental period lasted 21 weeks. The addition of garlic powder did not significantly affect body weight, egg production, feed consumption, feed efficiency, egg shell thickness, egg albumen index, egg yolk index and egg Haugh unit. Adding 5 and 10 g/kg garlic powder to the laying quail diets increased egg weight (p<0.01). Egg yolk cholesterol and blood serum cholesterol concentration were reduced with garlic powder supplementation. The results of this study demonstrated that garlic powder addition had a significant cholesterol-reducing effect in serum and egg yolk without adverse effects on performance and egg traits of laying quails.

IMPACT OF FOURWING SALTBUSH ON FEED AND WATER INTAKE AND ON BLOOD SERUM PROFILE IN SHEEP

  • Sixteen Harnai males were used to evaluate the influence of varying levels of fourwing saltbush hay on feed and water intakes as well as the blood serum mineral status in a completely randomized design. The animals were grouped randomly into four, with four animals in each. The four groups were randomly allotted low, medium, high and very high levels of saltbush hay supplementation in addition to wheat straw. The animals were given fresh water at free of choice. Weekly body weight was recorded for each individual animal. Blood serum was collected for mineral contents. The experiment lasted for eight weeks. The inclusion of saltbush leaves in the diet showed a non-significant effect on the total dry matter intake. There has been a significant increase in the water intake when very high levels of saltbush were included in the ration. Lower levels showed no effect on the water intake. The animals maintained their body weight from week 1 to week 8. No treatment by weeks interactions on the potassium and sodium levels were detected. However higher levels of saltbush increased significantly the potassium and sodium contents in the serum. Calcium contents were significantly(p<0.01) lowered with the inclusion of saltbush leaves in the diet. Whereas Phosphorous contents showed an increasing(p<0.05) trend with the higher levels of saltbush. No clinical or sub-clinical toxicological symptoms were observed in the sheep with the higher mineral contents.

Efficacy of Cr (III) Supplementation on Growth, Carcass Composition, Blood Metabolites, and Endocrine Parameters in Finishing Pigs

  • The study was conducted to evaluate the effects of trivalent chromium from different sources on growth, carcass composition, and serum parameters in finishing pigs. Ninety-six crossbred pigs with an initial average body weight of 65.57${\pm}$1.05 kg were blocked by body weight and randomly assigned to four treatments with three replicates. Pigs were offered one of four diets including a control diet or the control diet supplemented with 200 ${\mu}g/kg$ chromium from either chromium chloride ($CrCl_{3}$), chromium picolinate (CrPic) or chromium nanocomposite (CrNano) for 40 days. After completion of the feeding trial, eight pigs from each treatment were selected to collect blood samples, and slaughtered to measure carcass composition. The results showed that supplemental chromium had no significant effect on growth performance, while CrNano increased carcass lean proportion and loin Longissimus muscle area (p<0.05), and decreased carcass fat proportion and 10th rib backfat depth (p<0.05). CrPic supplementation also resulted in lower fat proportion and larger Longissimus muscle area (p<0.05). The addition of Cr from CrNano or CrPic decreased serum glucose (p<0.05) and increased concentrations of total protein and free fat acid in serum (p<0.05). Serum urea nitrogen, triglyceride and cholesterol were decreased (p<0.05), and serum high density lipoprotein and lipase activity were increased (p<0.05) with the supplementation of CrNano. Serum insulin was decreased (p<0.05) by supplemental Cr from CrNano or CrPic, and serum insulin-like growth factor I was increased significantly in the CrNano treated group. These results suggest that chromium nanocomposite has higher efficacy on carcass composition in pigs compared to the traditional chromium sources.

Effects of dietary supplementation with Pediococcus acidilactici ZPA017 on reproductive performance, fecal microbial flora and serum indices in sows during late gestation and lactation

  • Objective: This study was conducted to determine the effects of dietary supplementation with Pediococcus acidilactici (P. acidilactici) ZPA017 as a probiotic on reproductive performance, fecal microbial flora and serum indices in sows during late gestation and lactation. Methods: A total of 94 sows (Large White×Yorkshire, average 4.50 parities) were randomly allotted to two dietary treatments: control diet and the diet supplemented with P. acidilactici ZPA017 (2.40×109 colony-forming unit/kg of diets). The study started at d 90 of gestation and conducted until d 28 of lactation. Results: Compared to sows fed the control diet, supplementation of P. acidilactici ZPA017 increased the number of weaning piglets, weaning weight of litter and piglets, survival rate of piglets at weaning (p<0.05), and decreased diarrhea rate of piglets in lactation (p<0.05). Dietary P. acidilactici ZPA017 increased fecal Lactobacillus populations (p = 0.030) and reduced fecal Escherichia coli and Staphylococcus aureus populations (p<0.05) of sows at weaning. Moreover, the supplementation of P. acidilactici ZPA017 increased serum concentrations of immunoglobulin G, immunoglobulin A and total protein (p<0.05), while decreased serum haptoglobin concentration and alanine aminotransferase activity (p<0.05) of sows at weaning. Conclusion: Administration of P. acidilactici ZPA017 in diets during late gestation and lactation had positive effects on the reproductive performance, intestinal microflora balance and immunity of sows.

Effects of prolonged photoperiod on growth performance, serum lipids and meat quality of Jinjiang cattle in winter

  • Objective: This study was conducted to investigate the potential effects of prolonged photoperiod on the serum lipids, carcass traits, and meat quality of Jinjiang cattle during winter. Methods: Thirty-four Jinjiang bulls aged between 14 and 16 months were randomly assigned to two groups that were alternatively subjected to either natural daylight +4 h supplemental light (long photoperiod, LP) or natural daylight (natural photoperiod, NP) for 96 days. The potential effects on the levels of serum lipids, carcass traits, meat quality, and genes regulating lipid metabolism in the intramuscular fat (IMF) of the cattle were evaluated. Results: Jinjiang cattle kept under LP showed significant increase in both dry matter intake and backfat thickness. the serum glucose and the plasma leptin levels were significantly reduced, while that of melatonin and insulin were observed to be increased. The crude fat contents of biceps femoris muscle and longissimus dorsi muscle were higher in LP than in NP group. In longissimus dorsi muscle, the proportions of C17:0 and C18:0 were significantly higher but that of the C16:1 was found to be significantly lower in LP group. The relative mRNA expressions in IMF of longissimus dorsi muscle, the lipid synthesis genes (proliferator-activated receptor gamma, fatty acid-binding protein) and the fatty acid synthesis genes (acetyl-coa carboxylase, fatty acid synthetase, 1-acylglycerol-3-phosphate acyltransferase) were significantly up-regulated in LP group (p<0.05); whereas the hormone-sensitive lipase and stearoyl-CoA desaturase 1 were significantly down-regulated in LP than in NP group. Conclusion: Prolonged photoperiod significantly altered the growth performance, hormonal levels, gene expression and fat deposition in Jinjiang cattle. It suggested that the LP improved the fat deposition by regulating the levels of different hormones and genes related to lipid metabolism, thereby improving the fattening of Jinjiang cattle during winter.

Effect of alcohol dehydrogenase 1C (ADH1C) genotype on vitamin A restriction and marbling in Korean native steers

  • Objective: This work was to find the correlation of alcohol dehydrogenase 1C (ADH1C) genotype with vitamin A reduction and carcass traits during the vitamin A restriction period. Methods: In study 1, 60 Korean native steers were fed a diet (890 IU/kg) with 8,000 IU and 0 IU of supplemental premix vitamin A/kg of dry matter (DM) for control and treatment group, respectively. The levels of serum vitamin A were analyzed through high preparative performance liquid chromatography, and the ADH1C genotype was analyzed based on polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP; 78.1% TT type, 21.9% TC type); however, CC type was not found. Then, the interaction between ADH1C and carcass traits on the vitamin A restriction was investigated in study 2. A total of 136 Korean native steers were fed a diet that included 930 IU/kg vitamin A of DM. Results: Serum vitamin A in treatment was reduced to 112.4 IU/dL in steers with TT type of ADH1C, while for steers with TC type the concentration of serum vitamin A was dropped to 79.5 IU/dL (p<0.1) in study 1. This showed that TC type had the potential to lower serum vitamin A concentration during vitamin A restriction compared to TT type. In study 2 we found that eye muscle area, marbling and carcass weight in Korean native steers with TC type were higher than in steers with TT type (p<0.05). Conclusion: The interaction between vitamin A restriction and TC type of ADH1C gene could have the potential of increasing the marbling in Korean native steers. These results indicated that steers with TC type of the ADH1C gene were more sensitive to the change of serum vitamin A than TT types. Furthermore, this finding has the potential to enable a higher marbling score under the condition of vitamin A restriction in Korean native steers.

다양한 혈청 물질의 첨가가 염소 수정란의 체외배양에 미치는 영향 (Influence of various serum supplement on in vitro culture for goat embryos)

  • 본 연구는 염소 수정란의 체외배양용 배지에 FBS, gBS 및 PVA 첨가시 배발달 및 apoptosis 발생률을 분석하여 체외배양시 각 첨가물의 효과를 조사하였다. 체외성숙 후 체외수정을 실시하여 체외배양 배지에 10% FBS, 10% gBS 및 10% PVA를 첨가하고 배발달 효율과 배반포의 품질을 확인하기 위해 TUNEL assay를 통해 apoptosis 발생 비율을 조사한 결과, 난할률 및 배반포 형성률에서 처리군 모두 대조군(무혈청) 보다 유의적으로 높은 결과를 보였다. 특히 gBS와 PVA 처리군에서 각각 31.95, 35.29%로 유의적으로 가장 높은 배반포 형성률을 보였다. 또한 TUNEL assay의 결과에서도 배발달 비교실험에서와 같이 대조군에서 가장 적은 총세포수와 가장 높은 apoptosis 비율을 보였으며, gBS와 PVA 처리군에서 가장 많은 총세포수 및 가장 적은 apoptosis 비율을 보였다. 염소의 체외배양 효율 향상을 위한 본 연구의 결과, 무혈청 배지나 FBS첨가 보다는 gBS나 PVA의 첨가가 배발달 효율뿐만 아니라 배반포 품질에도 긍정적임을 알 수 있었으나, gBS내 확인되지 않는 물질들의 위험성을 감안했을 때 PVA가 보다 안전하고 효율적일 것으로 생각된다.

Biological effects of dietary probiotics on blood characteristics in Hanwoo heifers subjected to lipopolysaccharide (LPS) challenge

  • This study hypothesized that dietary feed additive containing probiotics alter either immune-related serum substances or serum metabolites in Hanwoo heifers. A probiotic treatment was given at 0.5% top-dressing of concentrate diet for 6 months. The change of immunological indicators in the blood was analyzed under LPS (Lipopolysaccharide) challenge. One day before administration of LPS, all heifers were fitted with an indwelling jugular vein catheter for serial blood collections. Both a serum tube and an EDTA-coated tube were collected at 30-min intervals from - 2 to 8 hours relative to the LPS challenge at time 0 ($1{\mu}g/kg$ of BW). Serum was used for analyzing albumin (ALB), glucose (GLU), total protein (TP), triglycerides (TG), phosphorus (IP), and non-esterified fatty acids (NEFA). Plasma was used for analyzing white blood cell (WBC), red blood cell (RBC), platelet (PLT) and inflammation-related factors (NE, LY, MO, EO, BA, Hb, HCT, MCV, MCH, MCHC, RDW, MPV). There were significant differences in ALB, GLU, TG, IP, and NEFA concentration with the passage of hours post challenge (p < 0.05). The level of ALB, GLU, TG, and IP showed significant difference (p < 0.05) between treatments. However, none of the data showed interaction between time and treatments (p > 0.05). The level of WBC, EO, LY, and MO were reduced after LPS challenge (p > 0.05). In conclusion, LPS challenge after dietary supplementation of probiotics changed the levels of both serum metabolites and inflammation-related factors. The increase of GLU and TG indicated a probiotics-positive response under LPS challenge (p < 0.05).

Effects of Serum Addition and Different Culture Media on Growth of Porcine Preantral Follicles In Vitro

  • Current developments in IVF and animal cloning have resulted in increasing demand for large quantities of oocytes and ovarian follicles at specific stages of development. These medical and scientific needs may be met by developing an optimal culture system for preantral follicles. In this study, we investigated the growth of porcine preantral follicle cultures in different media and in the presence and absence of serum. Follicles were manually dissected from ovaries obtained from prepubertal gilts at a local slaughterhouse, and cultured for 3 days in M199 or NCSU23 medium supplemented with porcine FSH, transferrin, L-ascorbic acid and insulin. Follicle diameters were measured on day 1 and 3 of culture. In Experiment 1, the effect of supplementing culture medium with fetal calf serum (FCS) on porcine preantral follicle growth was examined. In the group of cultures supplemented with FCS, follicle diameter after 3 days of culture, survival rate and antrum formation rate in the FCS group were significantly higher than those of the control group. In Experiment 2, the effects of culture medium (M199 and NCSU23) on follicle growth were compared. Follicle diameters were increased in the M199 group, compared with those in NCSU23 (p<0.05), but we observed no significant differences in survival and antrum formation rates between cultures grown in the two media. In conclusion, supplementation of the culture medium with serum enhances preantral follicle growth and antrum formation, and M199 is superior to NUSU23 for porcine preantral follicle culture in vitro.
