The High Production of Multimeric Angiotensin-converting-enzyme-inhibitor in E. coli

  • Multimeric angiotensin-converting-enzyme-inhibitor (ACE}) containing a trypsin cleavable linker peptide between ACEI was constructed. We made synthetic DNA coding for the ACEI peptide with asymmetric and complementary cohesive ends of linker nucleotides. A tandemly repeated DNA cassette for the expression of concatameric short peptide multimers was constructed by ligating the basic units. The resultant multimeric peptide expressed as soluble and trypsin treated peptide was shown at the same retention time with chemically synthetic ACEI by HPLC. The present results showed that the technique developed for the production of the ACEI multimers with trypsin cleavable linker peptides can be generally applicable to the production of short peptide.

  • The fruits of Eleutherococcus sessiliflorus (Rupr. & Maxim.) S. Y. Hu (Araliaceae), as edible fruits, were traditionally used for ingredients of wine or tea in Eastern Asia. In addition to, the fruits of E. sessiliflorus were known for having antioxidant and anti-inflammatory effects. Recently, we investigated that the ethanolic extracts of E. sessiliflorus fruits (DHP1501) have effects on hypertension via vasorelaxation and decrease of blood pressure. In the present study, we investigated that the gene and protein expression levels of endothelial nitric oxide synthase (eNOS) was increased by treatment of DHP1501 in HUVECs. Moreover, we confirmed the angiotensin converting enzyme inhibitory activity of DHP1501 through in vitro tasks. Therefore, DHP1501 could be a candidate of functional food for alleviating hypertension.

Effects of ${\alpha}-,\;{\beta}-Adrenergic$, and Calcium Channel Blockers on Renin- Angiotensin System in Perfused Rat Heart

  • ${\alpha},\;{\beta}-Adrenergics$, and calcium channels were known to be related to inducing cardiac hypertrophy. Recently, it was reported that the cardiac renin-angiotensin system (RAS) was an important factor in ventricular hypertrophy. The present study was aimed to investigate the effects of ${\alpha},\;{\beta}-adrenergic$, and calcium channel blockers that might be involved in the regulation of cardiac RAS. The reverse transcription-polymerase chain reaction (RT-PCR) was used to detect the expression of renin gene in the perfused rat heart. Changes in angiotensin converting enzyme (ACE) activity and cyclic AMP (cAMP) content which were thought to play a role in inducing cardiac hypertrophy were measured in the perfused rat heart. The expression of renin gene was not only increased by isoproterenol with metoprolol-pretreatment but also increased by vasopressin treatment in the presence of calcium channel blocker, nifedipine or verapamil. Either prazosin alone or norepinephrine with prazosin-pretreatment significantly increased the ACE activity. However, isoproterenol with metoprolol-pretreatment significantly decreased the ACE activity. On the other hand, the ACE activity was not changed by vasopressin, nifedipine, or verapamil treatments. The content of cAMP was significantly increased by either isoproterenol or vasopressin treatment. According to these results, renin gene expression was associated with ${\beta}2$ - adrenoceptor and calcium channel. ACE activity was associated with ${\alpha}-\;and{\beta}2$ - adrenoceptor. In conclusion, ${\beta}2$ - adrenoceptor was important in cardiac renin gene expression and ACE activity and ${\alpha},\;{\beta}$ -adrenergic, and calcium channel blockers might be involved in the regulation of cardiac RAS in a complicated way.

국산 홍합과 뉴질랜드 초록입 홍합 열수 추출물의 알코올분해효소 활성에 미치는 영향 및 DPPH 라디칼 소거능과 Angiotensin Converting Enzyme 저해 활성 (Effects of Hot Water Extracts of Domestic Blue Mussel and New Zealand Green Lipped Mussel on Alcohol Metabolizing Enzymatic, DPPH Radical Scavenging, and Angiotensin Converting Enzyme Inhibitory Activities)

  • 우리나라 남해안 연안에서 주로 양식되는 진주담치(국산 홍합)와 세계적으로 기능성이 잘 알려진 뉴질랜드 초록입 홍합의 열수 추출물을 제조하여 생리활성을 비교하였다. 모든 홍합 추출물들은 알코올대사와 관련한 alcohol dehydrogenase(ADH)와 acetaldehyde dehydrogenase(ALDH)의 활성을 상승시켰으며, 특히 ADH의 활성을 크게 향상시켰다. 국산 홍합의 육질 추출물은 초록입 홍합의 육질 추출물에 비하여 ADH 상승 효과는 비슷하였으며, ALDH 상승 효과는 약간 낮았다. DPPH 라디칼 소거능으로 조사한 항산화능은 국산 홍합의 육질 추출물이 초록입 홍합의 육질 추출물 보다 높았다. 항고혈압과 관련한 ACE 저해능의 경우에는 국산 홍합 육질 추출물이 초록입 홍합의 육질 추출물에 비해 10 mg/mL 농도에서는 낮았으나 20 mg/mL 이상의 농도에서는 유의차(P<0.05)를 보이지 않았다. 이상의 결과는 국내에서 양식되고 있는 홍합도 세계적으로 초록입 홍합에 못지않은 생리활성을 가지고 있음을 시사한다.

헛개나무의 Angiotensin 전환 효소 저해 및 항산화 활성 (Inhibitory Activity on Angiotensin Converting Enzyme and Antioxidant Activity of Hovenia dulcis Thunb. Cortex Extract)

  • 식물 유래 기능성 식품 소재발굴을 위한 기초연구의 일환으로 헛개나무 수피의 항고혈압, 항산화 활성과 총페놀화합물의 함량을 분석하였다. 헛개나무 수피 메탄올 추출물과 물 추출물의 ACE 효소 저해 활성은 $4,000\;{\mu}g\;m{\ell}^{-1}$의 농도에서 각각 81.1% 및 75.8%였으며, 용매별 분획물중에서는 에테르 분획 및 에틸아세테이트 분획이 각각 66.6% 및 75.9%로 높은 효과를 나타내었다. Superoxide 라디칼 소거능은 메탄올 추출물과 물 추출물이 $200\;{\mu}g\;m{\ell}^{-1}$의 농도에서 모두 99.5%이상의 활성을 보여 매우 우수하였으며, 분획물 중에서는 에틸아세테이트 및 에테르 분획이 90.8% 및 85.3%로 높은 효과를 나타내었다. DPPH 라디칼 소거능은 에틸아세테이트 분획 $(RC_{50},\;30.9{\mu}g\;m{\ell}^{-1})$이 실험된 조추출물 및 분획물들 중 가장 우수한 것으로 확인되었고 사람 저밀도지단백질 (LDL)의 산화에 대해 헛개나무 수피 조추출물은 $50\;{\mu}g\;m{\ell}^{-1}$이상의 농도에서 80% 이상의 저해효과를 나타내었다. Linoleic acid의 자동산화에 대한 항산화 효과는 $25\;{\mu}g\;m{\ell}^{-1}$의 농도에서 반응 5일째까지 메탄을 추출물이 ${\alpha}-tocopherol$보다 더 우수한 저해활성을 나타내었으며 총 페놀 함량은 메탄올 및 물 추출물이 7.2% 및 3.6%를, 분획물 중에서는 에틸아세테이트 분획이 60.8%로 조추출물 및 분획물 중에서 가장 높은 함량을 보였다.

Production of Antihypertensive Angiotensin I-Converting Enzyme Inhibitor-Enriched Edible Yeast Using Gugija (Lycium chinesis Mill)

  • To produce bioactive compound enriched yeast using medicinal Gugiga (Lycium chinensis Mill), several edible Saccharomyces species were cultured in Gugija extracts added yeast extract, peptone and dextrose medium (GE - YEPD medium) at $30^{\circ}C$ for 24 hr, and their growth were determined. Growth of Saccharomyces cerevisiae K-7 and Sacchromyces cerevisiae ACTC 7904 were better than those of the other yeasts. Two yeasts were selected and then determined their some physiological functionalities after cultivated the yeasts in the GE - YEPD medium and compared those grown on YEPD medium. Antihypertensive angiotensin I-converting enzyme (ACE) inhibitory activity of S. cerevisiae K-7 grown on GE - YEPD medium was about 20% higher than that grown on YEPD medium. Superoxide dismutase-like activity of S. cerevisiae ACTC 7904 was also about 12% more high. However, the other physiological functionalities were almost same or lower. Optimal addition concentration of Gugija extract was 10%, and maximally growth and ACE inhibitory activity of S. cerevisiae K-7 were shown when the strain was cultured in 10% Gugija extracts containing YEPD medium at $30^{\circ}C$ for 12 hr.

자연발증적 고혈압 흰쥐에서 무청 건조분말의 항고혈압 효과 (Antihypertensive Properties of Dried Radish Leaves Powder in Spontaneously Hypertensive Rats)

  • The study aim was to investigate the antihypertensive effect after oral supplementation of dried radish leaves powder (DRLP). Angiotensin-converting enzyme (ACE) activity was measured by spectrophotometric assay. The systolic blood pressure (SBP) was measured in spontaneously hypertensive (SHR) and normotensive rats (Wistar) by the tail cuff method after a 4-week diet with DRLP at the level of 2.5% or 5%. The supplementation of DRLP decreased SBP of SHR although the 5% supplementation level did not show any more pronounced effect than the 2.5% level did. The decrease in the SBP observed for both 2.5% and 5% DRLP was accompanied by significant increases of the urinary Na and K excretion. The DRLP supplementation showed a potent ACE-inhibitory activity in pulmonary tissue from both hypertensive and normotensive rats. However, the DRLP supplementation did not affect the SBP in normotensive rats. These results indicated that DRLP exerted an antihypertensive effect in SHR due to the decreased ACE activity and increased urinary Na excretion.

Lack of any Association between Insertion/Deletion (I/D) Polymorphisms in the Angiotensin-converting Enzyme Gene and Digestive System Cancer Risk: a Meta-analysis

  • Objective: To investigate the association between the gene polymorphisms of angiotensin-converting enzyme (ACE) and digestive system cancer risk. Method: A search was performed in Pubmed, Medline, ISI Web of Science and Chinese Biomedical (CBM) databases, covering all studies until Sep 1st, 2013. Statistical analysis was performed by using Revman5.2 and STATA 12.0. Results: A total of 15 case-control studies comprising 2,390 digestive system cancer patients and 9,706 controls were identified. No significant association was found between the I/D polymorphism and digestive cancer risk (OR=0.93, 95%CI = (0.75, 1.16), P=0.53 for DD+DI vs. II). In the subgroup analysis by ethnicity and cancer type, no significant associations were found for the comparison of DD+DI vs. II. Results from other comparative genetic models also indicated a lack of associations between this polymorphism and digestive system cancer risks. Conclusions: This meta-analysis suggested that the ACE D/I polymorphism might not contribute to the risk of digestive system cancer.

The Relationship between Physiological Activity and Cell Number in Dolsan Leaf Mustard Kimchi (Brassica juncea)

  • Changes in antioxidative activity and Angiotensin converting enzyme(ACE) inhibitory activity in juice prepared from Dolsan leaf mustard kimchi (DLMK) ar various fermentation temperatures were investigated. Antioxidative activity of juice from optimally ripened DLMK at 20 and 3$0^{\circ}C$ showed 80 and 83%, respectively. Juice from 10-day fermented DLMK at 3$0^{\circ}C$ showed 62% inhibitory activity against the ACE. In the juice fermented DLMK at 2$0^{\circ}C$~3$0^{\circ}C$, physiological activity was higher than that of the 4~1$0^{\circ}C$. In particular, optimally ripened DLMK at 3$0^{\circ}C$ showed the highest physiological activity. The physiological activity in DLMK juice at the fermentation period increased significantly with an increase in the growth of microbes. Consequently, a maximum physiological activity was shown at the maximum cell number. These results suggest that the microorganisms in DLMK juice would play an important role in the physiological activity.

Isolation of Angiotensin Converting Enzyme Inhibitors from Compositae Plants

  • Thirty plants of the Compositae family were screened for inhibitory activity of angiotensin converting enzyme (ACE). Among them, Chrysanthemum boreale Makino and Ixeris dentate Nakai were selected for further investigation since they had the highest inhibitory activity. Crude water extracts of the flowers of Chrysanthemum boreale Makino and the roots of Ixeris dentate Nakai were prepared by heating at 95$^{\circ}C$ and 6$0^{\circ}C$ for 2 hr, respectively, followed by centrifugation at 8000$\times$ g far 30 min. Crude extracts were then filtered using YM-3 and YM-1 membranes. The ACE inhibitors were isolated using consecutive chromatographic methods including: Sephadex G-15, ion exchange, FPLC, and reverse phase HPLC. The inhibitors were identified to have molecular masses of 204 and 283 daltons, respectively, by mass spectrometry. These results demonstrate that crude extracts of Compositae plants may be useful as functional food ingredients with anti-hypertensive properties.
