항고혈압성 안지오텐신 전환효소 저해제를 생산하는 새로운 효모의 선별 및 저해물질 최적 생산조건 (Screening New Antihypertensive Angiotensin I-Converting Enzyme Inhibitor -Producing Yeast and Optimization of Production Condition)

  • 항고혈압 효능이 우수하며 부작용이 없는 새로운 고혈압 예방 소재를 개발하고자 한국 식품연구원에서 분리, 동정한 48종의 효모들의 PD broth 배양 농축물들을 대상으로 angiotensin 전환효소(ACE) 저해활성을 조사하여 활성이 우수한 효모들을 선정한 다음 ACE 저해물질의 생산 최적 조건을 검토하였다. ACE 저해활성은 Saccharomyces cerevisiae Y183-3 균주의 PD broth 배양 농축물이 71.8%의 가장 높은 ACE 저해활성을 보여 최종 우수 균주로 선발하였다. 그리고 이 Y183-3 효모를 $30^{\circ}C$에서 36시간 PD broth에 배양했을 때 ACE 저해물질이 가장 많이 생산되었다.

담수어 열수추출물 및 효소가수분해물의 Angiotensin I 전환효소 저해작용 (Angiotensin I Converting Enzyme Inhibitory Activity of Hot-Water Extract and Enzymatic Hydrolysate of Fresh Water Fish)

  • 담수산 어류의 고도이용과 이들 추출물의 기능적 특성 해명을 위하여 전통적인 건강보양식품으로 애용되어 온 담수산 어류를 대상으로 열수추출물과 효소 가수분해물을 제조하고 이들의 ACE 저해작용 및 gel 여과에 의한 ACE 저해획분을 분리하고 그 특성을 알아 보았다. 열수추출물과 효소 가수분해물의 ACE 저해활성은 담수어류 중 잉어가 가장 높았으며, 효소가 수분해물이 열수추출물에 비하여 강한 경향을 보였다. Ethanol가용성 획분이 침전획분에 비하여 강한 ACE 저해능을 나타내었으며, 잉어 효소 가수분해물의 70% ethanol 가용성 획분이 가장 높은 ACE 저해율을 나타내었다. ACE 저해효과를 지니는 활성획분의 분자량은 열수추출물에서는 약 1,400이었으며 효소 가수분해물에서는 1,400 보다 다소 큰 것으로 나타났고, 시료 중 잉어 효소 가수분해물이 가장 높은 ACE 저해활성을 나타내었다. 활성회분의 아미노산 조성은 열수추출물에 있어서는 glycine, alanine, leucine, proline등의 합량이 많았으며 효소 가수분해물에 있어서는 aspartic acid, glutamic acid, glycine, alanine, valine, leucine, proline 등의 함량이 많았다. 활성획분의 $IC_{50}$/은 효소가수분해물이 열수추출물에 비하여 낮은 경향이었다.

제주도에 자생하는 양치식물의 생리활성 검색 (Screening of Biological Activities in Fern Plants Native to Jeju Island)

  • 제주도에 자생하는 양치식물 25종을 대상으로 항산화활성, ACE 저해활성, APN 저해활성 및 $\alpha$-amylase 저해활성을 검색하였다. 항산화활성은 쇠고비와 일색고사리에서 비교적 높아서 항산화 음료로 애용되고 있는 차나무 잎보다도 더 높았다. ACE 저해활성과 APN 저해활성은 전체적으로 낮았으나, 쇠고비에서 각각 26.7%와 28.2%으로 비교적 높은 저해활성을 나타내었다. $\alpha$-Amylase 저해활성은 10종이 50% 이상의 높은 활성으로 보였으며, 그 중에서 쇠고비와 홍지네고사리의 저해활성이 특히 높았다. 이상의 결과로부터 쇠고비는 항산화활성뿐만 아니라 ACE 저해활성, APN 저해활성과 $\alpha$-amylase 저해활성 모두 다른 식물에 비해 높아 생리활성 물질의 분리 및 동정 등 식품소재 및 식물생약으로 이용하기 위한 후속 연구가 기대된다.

분리대두단백질 가수분해물의 Angiotensin-I Converting Enzyme 저해효과 (Effect of Angiotensin-I Converting Enzyme Inhibitory from Hydrolysate of Soybean Protein Isolate)

  • 분리대두단백질의 가수분해물을 제조 시, 최적 가수분해 조건을 설정하기 위하여 총 7종의 단백질 가수분해 효소를 이용하여 효소선정, 효소농도, 가수분해시간 및 분리대두단백질분말의 첨가량에 따른 단백질의 함량 정도를 나타내는 280 nm의 흡광도 측정과 수율을 대신한 brix 측정, 가수분해물의 ACE 저해활성 등을 조사하였다. 분리대두단백질 분말에 증류수를 5%의 분산농도로 하여 분해효소 Alcalase 2.4 L을 1%(v/w) 첨가하여 $65^{\circ}C$에서 4시간 동안 가열하여 가수분해하였다. 가수분해를 마친 후 효소의 반응정지를 위하여 $80^{\circ}C$에서 5분간 열처리한 후, $4^{\circ}C$로 냉각하여 원심분리(7,000 rpm, 40 min) 하였다. 원심분리 한 상등액을 취하여 여과한 후, $60^{\circ}C$에서 농축하였다. 농축액을 냉동시켜, 진공 동결건조 하여 분리대두단백질 가수분해물을 제조하였다. 분리대두단백질 가수분해물의 수율은 38.32%를 나타냈으며, ACE 저해활성($IC_{50}$)은 $79.94 {\mu}g/mL$를 나타내었다.

Production of Antihypertensive Angiotensin I-Converting Enzyme Inhibitor-Enriched Edible Yeast Using Gugija (Lycium chinesis Mill)

  • To produce bioactive compound enriched yeast using medicinal Gugiga (Lycium chinensis Mill), several edible Saccharomyces species were cultured in Gugija extracts added yeast extract, peptone and dextrose medium (GE - YEPD medium) at $30^{\circ}C$ for 24 hr, and their growth were determined. Growth of Saccharomyces cerevisiae K-7 and Sacchromyces cerevisiae ACTC 7904 were better than those of the other yeasts. Two yeasts were selected and then determined their some physiological functionalities after cultivated the yeasts in the GE - YEPD medium and compared those grown on YEPD medium. Antihypertensive angiotensin I-converting enzyme (ACE) inhibitory activity of S. cerevisiae K-7 grown on GE - YEPD medium was about 20% higher than that grown on YEPD medium. Superoxide dismutase-like activity of S. cerevisiae ACTC 7904 was also about 12% more high. However, the other physiological functionalities were almost same or lower. Optimal addition concentration of Gugija extract was 10%, and maximally growth and ACE inhibitory activity of S. cerevisiae K-7 were shown when the strain was cultured in 10% Gugija extracts containing YEPD medium at $30^{\circ}C$ for 12 hr.

구증구포 처리 대두 추출물의 항산화 활성 및 Angiotensin-I Converting Enzyme 저해 효과 (Effect on Angiotensin-I Converting Enzyme Inhibition and Antioxidant Activities of Soybean (Glycine max L.) following Steaming and Drying Nine Times)

  • To increase the functional material content of soybean, a repetitive steaming and drying process was used. We investigated the changes in the total polyphenol content, the antioxidant activity, and the angiotensin-I converting enzyme (ACE) inhibition in soybean following nine rounds of steaming and drying. Soybean was steamed 9 times for 2 h and then dried 9 times from 55℃ to 73℃ for 3 h. The total polyphenol content in the soybean reached a maximum value of 60.47 mg GAE eq./100 g at 73℃ while the total polyphenol content in the raw soybean reached 25.17 mg GAE eq./100 g. In the raw soybean samples, the DPPH radical scavenging activity (5 mg/mL) was 8.04% but it increased by 43.29% after drying 9 times to 73℃. ABTS radical scavenging activity also improved following 9 rounds of steaming and drying. ACE inhibitory activity of the soybean dried 9 times at 73℃ was 58.94% at 10 mg/mL. These results showed that steaming and drying soybean 9 times enhanced the antioxidant activity and the ACE inhibitory activity of soybean. Therefore, more research on the biological and anti-hypertensive activity of soybean using this steaming and drying method is necessary.

Development of effective heparin extraction method from pig by-products and analysis of their bioavailability

  • This study was conducted to develop an effective heparin extraction method by using low-cost and highly effective enzymes from six pig by-products (liver, lung, heart, stomach, small intestine, and large intestine), and analyze their bioavailability. Low-cost and highly effective enzymes (alkaline-AK and papain) and a common enzyme (trypsin) were used for the heparin extraction. The angiotensin I- converting enzyme (ACE) inhibitory activity and the antimicrobial activity of extracted heparin were analyzed to verify their bioavailability. The average amount of heparin extracted per kilogram of pig by-products was 439 mg from the liver, 127 mg from the lung, 398 mg from the heart, 261 mg from the stomach, 197 mg from the small intestine, and 239 mg from the large intestine. Various enzymes were used to extract heparin, and the amount of extracted heparin was similar. Based on 1 g of pig by-product, the enzymes trypsin, papain, and alkaline-AK could extract 1,718 mg, 1,697 mg, and 1,905 mg of heparin, respectively. Heparin extracted from pig by-products showed antihypertensive activity and antimicrobial activity against Staphylococcus aureus at low populations. These results indicated that heparin can be obtained from pig by-products at a low cost.

Quality Characteristics and Cardiovascular Activities of Korean Traditional Wines and Liquors

  • The goal of this study was to screen and characterize the physiological functions of Korean traditional wines (TW) and liquors (TL). Forty-two TW and TL were collected and evaluated for quality and cardiovascular activities. Ethanol content ranged from $9.0%{\sim}41%$, and pH ranged from $3.0{\sim}7.8$, and they also contained 0.01% to 0.67% of total acid. Samples contained a maximum of 2.0% of crude protein and $0.1%{\sim}14.0%$ of reducing sugar. Commercial CM-wine showed the highest antihypertensive angiotensin I-converting enzyme (ACE) inhibitory activity, 85.9%. The greatest fibrinolytic activity and platelet aggregation inhibitory activity were also found in commercial CM-wine (31.8U) and commercial SS2-wine (38.6 %), respectively. Commercial SHBI-liquor showed the highest HMG-CoA reductase inhibitory activity, 78%. The ACE inhibitor from commercial CM-wine was a peptide compound and also showed an antihypertensive effect in spontaneous hypertensive rats at a dosage of 1.5 mg/kg.

Comparison of Functional Properties of Blood Plasma Collected from Black Goat and Hanwoo Cattle

  • Slaughterhouse blood is a by-product of animal slaughter that can be a good source of animal protein. This research purposed to examine the functional qualities of the blood plasma from Hanwoo cattle, black goat, and their hydrolysates. Part of the plasma was hydrolyzed with proteolytic enzymes (Bacillus protease, papain, thermolysin, elastase, and α-chymotrypsin) to yield bioactive peptides under optimum conditions. The levels of hydrolysates were evaluated by 15% sodium dodecyl sulfate polyacrylamide gel electrophoresis. The antioxidant, metal-chelating, and angiotensin I-converting enzyme (ACE) inhibitory properties of intact blood plasma and selected hydrolysates were investigated. Accordingly, two plasma hydrolysates by protease (pH 6.5/55℃/3 h) and thermolysin (pH 7.5/37℃/3-6 h) were selected for analysis of their functional properties. In the oil model system, only goat blood plasma had lower levels of thiobarbituric acid reactive substances than the control. The diphenyl picrylhydrazyl radical scavenging activity was higher in cattle and goat plasma than in proteolytic hydrolysates. Ironchelating activities increased after proteolytic degradation except for protease-treated cattle blood. Copper-chelating activity was excellent in all test samples except for the original bovine plasma. As for ACE inhibition, only non-hydrolyzed goat plasma and its hydrolysates by thermolysin showed ACE inhibitory activity (9.86±5.03% and 21.77±3.74%). In conclusion, goat plasma without hydrolyzation and its hydrolysates can be a good source of bioactive compounds with functional characteristics, whereas cattle plasma has a relatively low value. Further studies on the molecular structure of these compounds are needed with more suitable enzyme combinations.

수수 입국과 두류 첨가가 전통주의 품질과 엔지오텐신전환효소 저해활성에 미치는 영향 (Effect of Indian Millet Koji and Legumes on the Quality and Angiotensin I-Converting Enzyme Inhibitory Activity of Korean Traditional Rice Wine)

  • 고혈압 예방 효과를 가진 전통주를 개발하기 위하여 우선 엔지오텐신전환효소(ACE)저해 활성이 있다고 알려진 몇 가지 곡류를 이용하여 입국을 만들어 알콜 발효특성을 조사하였다. 또한 선정된 입국과 각종 두류분말을 덧밥에 첨가하여 전통주를 제조한 후 품질특성과 ACE저해 활성 및 기호도 등을 조사하였다. 수수 입국을 이용하여 전통주를 제조하였을 때 에탄올 생성량은 13.8%로 다른 입국으로 만든 술 보다 약간 낮았으나 ACE저해 활성은 43.3%로 제일 높았다. 또한 녹두가루를 덧밥의 50% 첨가하고 민들레꽃을 1% 첨가한 후 수수 입국과 주모로 $30^{\circ}C$에서 10일간 발효 시켰을때 에탄올 생성량과 항고혈압 활성을 나타내는 ACE저해 활성이 약 69%로 크게 증가하였고 맛과 향도 우수하였다.
