The Neuroprotective Effects of Angelicae gigantis Radix on Focal Cerebral Ischemia in the Rat (백서의 국부 뇌경색에 대한 당귀의 신경보호 효과)

  • Current therapy for acute ischemic stroke is highly focused on neuroprotective agents, and many herbal medicines have been challenged for experimental models. The aim of this study is to investigate whether Angelicae gigantis Radix can protect nerve cells against ischemic neural damage of middle cerebral artery occlusion (MCAO) in rats' brains. Rats were treated with Angelicae gigantis Radix immediately after 2 hours of MCAO for 7 days. On the 7th day, the brains of the rats were sliced through the hippocampus and dyedby c-Fos immunohistochemistry stain and cresyl violet stain for microscopic examination. The number of viable neurons and c-Fos immunoreactive cells in CA1 regions was counted. MCAO caused significant decrease in density of neurons and c-Fos immunoreactive cells compared to those of sham-operated rats. Administration of Angelicae gigantis Radix significantly elevated MCAO-induced decrease in density of neurons and c-Fos immunoreactive cells. These results suggest that the neuroprotective effect of Angelicae gigantis Radix against focal cerebral ischemia is related to c-Fos gene expression. Thus, these findings indicate that Angelicae gigantis Radix can be used for treatment and prevention of cerebral ischemia.

Applications of Prescriptions Including Angelicae Gigantis Radix and Cnidii Rhizoma in Dongeuibogam (동의보감(東醫寶鑑) 중(中) 당귀(當歸)와 천궁(川芎)이 등분(等分) 배오(配伍)된 방제(方劑)의 활용(活用)에 대한 고찰(考察))

  • This report describes 214 prescriptions including Angelicae Gigantis Radix and Cnidii Rhizoma in Dongeuibogam. Angelicae Gigantis Radix and Cnidii Rhizoma have been used separately or concurrently in Oriental Medicine for a long time as a treatment for various diseases. The following conclusions are reached through investigations on the applications of prescriptions including Angelicae Gigantis Radix and Cnidii Rhizoma in Dongeuibogam. 1. 34 times(15.9%) prescriptions are recorded in women chapter, 24 times(11.2%) in wind chapter, 13 times(6.1%) in children chapter and 12 times(5.6%) in intumescence chapter, which are arranged in order of frequency. 2. Treating blood, stroke and fetal movement are ranked top, regulating blood, headache, cataract, carbunculosis, pregnancy, abortion, underdevelopment, etc, among 137 symptoms in prescriptions including Angelicae Gigantis Radix and Cnidii Rhizoma. 3. The dosages of Angelicae Gigantis Radix and Cnidii Rhizoma are ranged from 2.5 pun~6 nyang. 1 jeon is recorded 54 times(25.2%), 7 pun is 29 times(13.6%), 5 pun is 24 times(11.2%), 1 nyang is 21 times(9.8%), same amount is 11 times(5.1%), 5 pun is 10 times(4.7%), 8 pun is 9 times(4.2%) used among 187 prescriptions including Angelicae Gigantis Radix and Cnidii Rhizoma. 4. The dosages of Angelicae Gigantis Radix and Cnidii Rhizoma are ranged from 2.5 pun~6 nyang. 2.5 pun~1.5 jeon, 2 jeon~6 nyang, same amount which are recorded by percent(decoction groups: the other groups) are 69.0%(98.0:2.0%), 25.5%(18.5:81.5%), 5.1%(18.2:81.8%) respectively. The less using dosage of Angelicae Gigantis Radix and Cnidii Rhizoma has a more percentage of decoctions groups, The more using dosage of Angelicae Gigantis Radix and Cnidii Rhizoma has a more percentage of the other groups except decoctions. The less using prescriptions have an effects of controling menstrual pain, enhancing blood and chi, etc, as a whole. The more using prescriptions as a form of compound powder have an effects in epilepsy, chronic diseases, etc. 5. The 214 prescriptions including Angelicae Gigantis Radix and Cnidii Rhizoma in Dongeuibogam are mainly composed of Goongguitang, Samooltang, Sagoonjatang and Gobangpoongtang, etc.

The Effect of Radix Angelicae gigantis aqua-acupuncture on the hypothyroidism induced by thiourea in rats (당귀약침(當歸藥鍼)이 흰쥐의 갑상선기능(甲狀腺機能)에 미치는 영향(影響))

  • In order to study the effect of aqua-acupuncture with Radix Angelicae gigantis extract solution by acuncture point. Radix Angelicae gigantis aqua-acupuncture was carried on every day for 3 days and 5 days 7 days respectively on the corresponding loci of Chondol(CV22) and non-acupuncture points (loci at the root of tail), after inducing hypothyroidism by thiourea administration and thereafter the contents of serum triiodothyronine$(T_3)$, thyroxine$(T_4)$, triiodothyronine uptake, free $T_3$, free $T_4$, total protein, albumin, creatinine, creatine, BUN, cholesterol, triglyceride, and total lipid were measured in hypothyroidism rats. The following results have been obtained : 1. $T_3$ was increased with statistical significance by Radix Angelicae gigantis aquq-acupuncture for 3 days, 5 days and 7 days Chondol(CV22) and non-acupuncture points as compared with the control group. 2. $T_3$ uptake was increased with statistical significance by Radix Angelicae gigantis aquq-acupuncture for 3 days, 5 days and 7 days to Chondol(CV22) as compared with the control group. 3. Free $T_4$ was increased with statistical significance by Radix angelicae gigantis aqua-acupuncture for 3 days, 5 days and 7 days to Chondol(CV22), and non-acupuncture points respectively as compared with the control group. 4. Albumin was decreased with statistical significance by Radix Angelicae gigantis aqua-acupuncture for 3 days, and 5 days and 7 days to Chondol(CV22), and non-acupuncture points respectively as compared with the control group. 5. Creatine was decreased with statistical significance by Radix Angelicae gigantis aqua-acupuncture for 3 days, and 5 days and 7 days to Chondol(CV22), and non-acupuncture points respectively as compared with the control group. 6. Cholesterol was decreased with statistical significance by Radix Angelicae gigantis aqua-acupuncture for 3 days, and 5 days and 7 days to Chondol(CV22) and non-acupuncture points respectively as compared with the control group. 7. Triglyceride was decreased with statistical significance by Radix Angelicae gigantis aqua-acupuncture for 3 days, and 5 days and 7 days to Chondol(CV22) and non-acupuncture points respectively as compared with the control group. 8. Total lipid was decreased with statistical significance by Radix Angelicae gigantis aqua-acupuncture for 3 days, and 5 days and 7 days to Chondol(CV22) and non-acupuncture points respectively as compared with the control group.

Effects of Hot Water Soluble Extract from Angelicae Gigantis Radix on the Lipid Metabolism and Antioxident Defense System in rats fed Hypercholesterol Diet (당귀 열수추출물이 고콜레스테롤 식이를 공급한 흰쥐의 지질대사와 황사화계에 미치는 영향)

  • To investigate the effects of hot water soluble extract from Angelicae Gigantis Radix on lipid metabolism and antioxidant defense system in rats fed hypercholesterol diet, Sprague-Dawley male rats were fed either hypercholesterol diet with water or hot water soluble extract from Angelicae Gigantis Radix. Hypercholesterol diet contained 18 % beeftallow and 5 % cholesterol. After 4 weeks of experimental periods serum and liver obtained. The level of serum lipid and the activities of GOT and GPT was measured. The level of lipid and TBARS and the activity of GSH-Px was measured also. The results obtained were: 1) Food intake in the experimental group supplied hot water soluble extract from Angelicae Gigantis Radix was low significantly. 2) In the experimental group supplied hot water soluble extract from Angelicae Gigantis Radix of serum total lipid and total cholesterol was significantly low. 3) Serum GOT activity was significantly low in the experimental group supplied hotwater soluble extract from Angelicae Gigantis Radix. There was no difference of serum GPT activity between control and experimental group supplied hot water soluble extract from angelical Gigantis Radix. 4) There was no difference of lipid composition, TBARS level and GSH-px activity in liver between control and experimental group. The result of this study indicates that the hot water soluble extract from Angelicae Gigantis Radix was effective in decreasing the food intake of the experimental animals in the hypercholesterol diet and also effective in decreasing the levels of serum total lipid, total cholesterol and GOT activity. However it was not effective to change the level of lipid, TBARS and GSH-Px activity of liver.

Nodakenin and Decursin Contents of Fermented Angelicae Gigantis Radix by 4 Species Strain (4종의 발효균주가 당귀의 nodakenin과 decursin의 함량에 미치는 변화)

  • Objectives : The purpose of this study was to investigate the nodakenin and decursin contents in each fermented Angelicae gigantis Radix by 4 species of ferment strains. Methods : The strains of fermented Angelicae gigantis Radix were Lactobacillus plantarum(SE1), L. acidophilus(AC), Bacillus subtilis(B2) and B. licheniformis(BL2). The fermentative changes of nodakenin and decursin were analyzed using HPLC. Results : All of 4 species strains reduced nodakenin and decursin concentration in Angelicae gigantis Radix by fermentation. But fermentability were difference between strains. The nodakenin concentration of AC was lower than B2. The decursin concentrations of SE1 and AC were lower than B2 and BL2. Conclusions : 4 species of fermental strain were difference decomposable rate with nodakenin and decursin in fermented Angelicae gigantis Radix.

A Study on the Standardization of Ligustici Rhizoma and Angelicae Tenuissimae Radix including the Comparison for the Biological Activity on the Active Ingredients (Ligustici Rhizoma(고본(藁本))와 Angelica tenuissimae Radix(한국고본(韓國藁本)뿌리)의 규격화(規格化) 및 유효성분(有效成分)의 진통효과(鎭痛效果) 비교(比較)에 관한 연구(硏究))

  • Major separation for the active ingredients and characterization of chemical properties in conjunction with screening test on animal were performed in order to analyze and standardize Ligustici Rhizoma or Angelicae Tenuissimae Radix as an important oriental herbal medicine for antiphlogistic or an important oriental herbal medicine for antiphlogistic or an anodyne. Furthermore the structure, composition and contents of ingredients for essential oil in Angelicae Tenuissimae Radix(Suckpo, Korea) were determined by means of Ge/MS followed by screening test on Z-ligustilide(82%) known as major ingredient as well as butylidenephthalide collected by HPLC with normal phase semiprep-column. The total active ingredient in Ligustici Rhizoma from China or Angelicae Tenuissimae harvested at Choonyang(Kyungnam, Korea), Jungsun(Kangwon, Korea), Suckpo(Kyungnam Korea), Youngchun(Kyungnam, Korea) have been determined showing higher abundant for three times on the product in Korea compared to that in China. In addition, the major component in Ahgelicae Tebyussunae Radux extract was found to be Z-ligustilide(70-80%) which is very different from that in Ligustici Rhizoma senkyunolide(39%) as major species. For screening test of Ligustici Rhizoma or Angelicae Tenuissimae Radix extracts toward the target animal, the efficiency has been shown the similarity on both extracts. Taking into account the level of ingredient, the total efficiency may be three times higher on Angelicae Tenuissimae Radix in Korea compared to Ligustici Rhizoma in China. As a result of present study, it is preferable to distinguish between Ligustici Rhizoma and Angelicae Tenuissimae Radix for better usage of oriental herbal medicine because of very different composition and abundant in spite of their similar screening effect.

Compositions of Astragali Radix and Angelicae Radix by DPPH Radical Scavenging Activity (당귀(當歸)와 황기(黃芪)의 배합 변화가 DPPH 자유기 소거에 미치는 영향 연구)

  • Objectives : This study was conducted to compare antioxidant activity of ratio-related Angelicae Radix and Astragali Radix compositions by DPPH radical scavenging activity. Methods : The species of Angelicae Radix was compared as Angelica gigas, A. sinensis and A. acutiloba. Then various compositions of Angelicae Radix with Astragali Radix were prepared with different ratio. Diphenyl-picryl-hydrazyl (DPPH) radical scavenging activity was measured after 10, 20 and 30 minutes. Each extracts were tested by 1, 5, 10, 50, 100, 500 and 1,000 ${\mu}g/ml$ concentrations. Results : The results showed that Angelica gigas, A. sinensis and A. acutiloba scavenged DPPH radical with 66.8, 61.7 and 56.7% in 1,000 ${\mu}g/ml$ concentration, respectively. According to this result, Angelicae Radix and Astragali Hadix compositions with 100:0, 10:1, 5:1, 3:1, 1:1, 1:3, 1:5, 1:10 and 0:100 ratio were scavenged DPPH radical with 56.3, 49.9, 61.2, 55.7, 54.7, 55.0, 52.8 ,md 50.2% in 1,000 ${\mu}g/ml$ concentration, respectively. The extract of Angelicae Radix and Astragali Radix compositions with 100:0, 10:1, 5:1, 3:1, 1:1, 1:3, 1:5, 1:10 and 0:100 ratio scavenged DPPH radical with the $IC_{50}$ being 85,21, 1016.26, 656.51, 785.67 and 956.74 ${\mu}g/ml$ respectively. Conclusion : In conclusion, the extract of Angelicae Radix and Astragali Radix compositions with 1:1 ratio showed dose-dependent DPPH radical scavenging activity.

Angiogenic Inhibition Effects of Several Herbs Supplementing Qi and Blood (수종 보기보혈 한약의 혈관신생 억제효과)

  • Two of the essential processes required for metastasis are neoangiogenesis and tumor cell invasion of basement membranes (BM) and extracellular matrix (ECM). Recently, data showed that herbs removing blood stasis has an anti-angiogenic effects. Tonifying vital Qi and eliminating pathogenic factor was a basic modality in Oriental oncology. In this study, we investigated several Qi and Blood tonics for potent angiogenic inhibitors. Methanol extracts of samples inhibited the proliferation of ECV-304 at the concentration of 100 ㎍/㎖. Zizyphi Fructus, Glycyrrhizae Radix, Angelicae Gigantis Radix decreased the gelatinolytic activity of MMP-9 from ECV-3Q4, at the concentration of 100 ㎍/㎖ in gelatin zymography. In in vitro invasion assay, herbs inhibited the invasion activity of ECV-304 by 53% of control (Ginseng Radix), 39% (Zizyphi Fructus), 36% (Angelicae Gigantis Radix), 25% (Glycyrrhizae Radix). Ginseng Radix inhibited the capillary-like tube formation of ECV-304 at the concentration of 160 ㎍/㎖, Angelicae Gigantis Radix and Paeoniae Radix Alba inhibited at the concentration of 320 ㎍/㎖. These results indicated that Ginseng Radix, Glycyrrhizae Radix, and Angelicae Gigantis Radix could be considered as potent angiogenic inhibitiors.

Hygienic Quality and Safety of Gamma Irradiated Angelicae Gigantis Radix and Aloe (감마선조사에 의한 당귀와 알로에의 위생화 및 안전성 평가)

  • Gamma irradiation was applied to Angelicae gigantis radix and Aloe to improve their hygienic quality. The effective dose of irradiation was 7 kGy in Angelicae gigantis radlx and 5 kGy in Aloe for the sterilization of all contaminated microorganisms tested. After 8 months of storage at room temperature, no growth of microorganisms was observed in the irradiated products. The safety of these products were evaluated by Salmonella typhimurium reversion assay and in vivo micronucleus assay using mouse bone marrow cells. They were negative in the bacterial reversion assay with S. typhimurium TA 98, TA100, TA1535 and TA1537. In the in vivo mouse micronucleus assay, they did not show any clastogenic effect at all doses tested. These results indicate that the gamma irradiation of Angelicae gigantis radix at 12 kGy and of Aloe at 10 kGy have no genotoxic effects under these experimental conditions.

Studies on Pest Insects of Crude Drugs (생약 및 한약재의 저장 중 발생하는 곤충류)

  • In order to estimate accurate quality of crude drugs in which pest was found during the storage, damaged 15 items were collected on current markets. We identified insect name by investigation of insect morphology, bug cluster, and morphology of damaged crude drugs. Three kind of pests were identified in the above 15 items as follows: A: Pyralis sp. (Pyralidae) was observed in damaged Platycodi Radix. B: Anthreus verbasci (Anobiidae) was found out in damaged Angelicae gigantis Radix, and Dioscoreae Rhizoma. C: Stegobium pamceum (Anobiidae) was discovered in the 12 crude drugs as follows: Puerariae Radix, Angelicae koreanae Radix, Angehcae tenuissimae Radix, Codonopsis Radix, Ledebouriellae Radix, Ginseng Radix alba, Angelicae dahuricae Radix, Belamcandae Rhizoma, Paeoniae Radix, Cnidii Rhizoma, Alismatis Rhizoma, and Fritillariae Bulbus.
