Production of Extracellular Polysaccharide by Perfusion Culture of Angelica gigas Nakai Suspension Cells (배지교환식 고농도 배양에 의한 참당귀 현탁세포 유래 ECP 생산)

  • High-density perfusion cultivation was performed to produce extracellular polysaccharide(ECP) as immunostimulating agents in suspension cell cultures of Angelica gigas Nakai. In batch culture, the maximum cell density was 16.8 gDCW/L at day 6 and 0.9 g/L of ECP was obtained at day 8. When the medium exchange was started at the fifth day after inoculation for the perfusion culture, high concentration of the cells at 23.8 gDCW/L could be achieved with continuous production of ECP. Treatments of ultrasound and Pluronic F-68 were found to be helpful for the secretion of intracellular ECP into the culture medium.

Effects of Pluronic F-68 and Oxygen Vectors on the Cell Growth of Angelica gigas Nakai in Aqueous Two-Phase System (수성이상계에서 Pluronic F-68과 산소전달물질이 참당귀 현탁세포 증식에 미치는 영향)

  • Pluronic F-68 and oxygen vectors were applied to increase the cell growth of Angelica gigas Nakai in aqueous two-phase system (ATPS). ATPS was composed of 3.6% (w/v) polyethylene glycol (PEG) 20,000 and 2.8% (w/v) crude dextran. n-Hexadecane, n-dodecane and FC-40 were used as oxygen vectors to enhance the oxygen transfer in ATPS. With 2$\sim$10 g/L of Pluronic F-68, addition of of n-hexadecane and FC-40 significantly enhanced the oxygen transfer rate as well as the maximum cell mass in a medium with ATPS. However, n-dodecane reduced the cell growth in all treatments. Maximum cell densities were increased up to 27.5% with 10 g/L of Pluronic F-68 and up to 40.2% with 8% (v/v) n-hexadecane compared to those of the controls without Pluronic F-68 and oxygen vectors. It was confirmed that the cell growth could be increased in ATPS using n-hexadecane.

Inhibiting Floral Induction of Angelica gigas NAKAI in the Hilly Altitude Area (중.산간지대에서 참당귀의 화성억제 연구)

  • This study was carried out to investigate the effects of floral inhibition of An-gelica gigas NAKAI in the hilly altitude located in the Northern Gyeongbuk Province from Feb. 1992 to Nov. 1994. The results obtained were as follows : As the cultivated areas are high, rate of bolting was significantly decreased, having high yield, good growth, and medicinal quality. It is considered that the optimal cultivating area was at least above 600m altitude. In the hilly altitude, the more shorten nursery period was, the more decreased rate of bolting was, it results in decreased yield, having no significant differences in contents such as extracts and decursin. In bolting response from temperature treatment of the seedlings, treatment of high temperature was significantly decreased floral induction, but rate of establishment was decreased by decayed root. Bolting rate at different organic resources has more reduced in single fertilization than that of in organic application, but among organic resources, compost of rice straw has the lowest bolting rate. As a result, yield and medicinal qualities at various organic resources were increased in application of organic resources which was no con-siderable tendency among organic resources.

Oral Administration Effects of Herbal Extracts on Atopic Dermatitis in Balb/c Mice Sensitized by Ovalbumin (Ovalbumin으로 유발된 아토피피부염 모델 마우스에 대한 복합한약추출물 경구투여의 효과)

  • Objective: This study was to evaluate whether herbal mixture (HM-A : Houttuynia cordata Thunberg, Rubus coreanus, Rehmannia glutinosa, Prunus yedoensis, HM-B : Houttuynia cordata Thunberg, Rubus coreanus, Rehmannia glutinosa, Angelica gigas nakai) supresses the development of atopic dermatitis in Balb/c mice sensitized by ovalbumin. Methods: Mice were sensitized by intraperitoneal injection of ovalbumin plus aluminum hydroxide hydrate, followed by epicutaneous sensitization for 6 weeks. After induced atopic dermatitis, HM-A and HM-B were orally administrated for two weeks(once a two days) as a 50 mg/kg concentration. After all mice were sacrificed at the end of the experiment, skin and blood were harvested. Results: Oral administration group was reduced the infiltration of eosinophils, mast cells and total T cells on the skin areas as well as blood analysis. Also, cutaneous expression of IL-4,13,17 decreased. Blood IgE level was decreased. Conclusion: These drugs could be potential candidates for the atopic dermatitis.

Improved Axonal Regeneration Responses in the Injured Sciatic Nerve of Rats by Danggui Treatment (당귀가 rat의 손상된 좌골신경 재생에 미치는 영향)

  • Objective: This study was performed to examine Danggui (DG, Angelica gigas Nakai)'s potential activity for promoting axonal regeneration in the injured peripheral nerve. Methods: Using the sciatic nerve in the rats, DG extract 5 ${\mu}l$(10 mg/ml in 0.5% saline) was dripped into the injury site of the nerve. Results: DG treatment facilitated axonal elongation responses in the distal portion to the injury site. GAP-43 protein levels were upregulated by DG treatment in the injured nerve and also in the DRG, suggesting the induction of GAP-43 expression at gene expression level after nerve injury. Phospho-Erk1/2 protein levels were upregulated in the injured nerve area and also in the DRG, suggesting retrograde transport of phospho-Erk1/2 protein from the injury area to the cell body. Cdc2 protein levels were slightly upregulated by DG treatment. DG treatment increased the number of non-neuronal cells in the distal portion to the injury site. Conclusions: The present data suggest that DG is effective for enhanced axonal regrowth after sciatic nerve injury.

Absorption, Distribution, Metabolism, and Excretion of Decursin and Decursinol Angelate from Angelica gigas Nakai

  • The pharmacokinetics of decursin and decursinol angelate (D/DA) were investigated in male SD rats following oral and intravenous administration. D/DA and metabolites obtained from in vitro samples were evaluated by LC/MS. The levels of D/DA and metabolized decursinol in the blood following oral and intravenous administrations declined according to first-order kinetics, with $T_{1/2}$ values of 56.67, 58.01, and 57.22 h, respectively, being observed after administration of a dose of 2 mg/kg body weight. The large intestine was the major site of disposition following oral administration. These data indicate that D/DA is rapidly absorbed from the gastrointestinal tract. In in vitro experiment utilizing liver microsomal protein, the major metabolic reaction of D/DA occurred to change decursinol. The cumulative biliary, urinary, and fecal excretions of D/DA in bile duct-cannulated rats was $36.10{\pm}2.9%$, $25.35{\pm}3.8%$, and $34.20{\pm}3.2%$, respectively, at 72 h after administration. These results indicate that the absorption of D/DA is almost complete, and that its metabolites are primarily excreted into feces through the bile. These results indicate that D/DA is subject to enterohepatic circulation.

Effect of Pluronic F-68 and Oxygen Vectors on Cell Growth of Angelica gigas Nakai in Fed-batch Culture

  • It has been commonly known that cell growth is inhibited by the lack of dissolved oxygen and mass transfer inhibition of nutrients at stationary phase in fed-batch culture. In this study, Pluronic F-68 and oxygen vectors were added in Angelica gigas Nakai suspension culture in order to enhance cell growth in fed-batch culture. It was observed that the addition of 6%(w/v) Pluronic F-68 promoted cell growth up to 6.1% compared to control and that the use of 4%(v/v) n-hexadecane markedly enhanced cell growth up to 11.4%.

  • This study was carried out to investigate the relation between bolting rate and root yield of Angelica gigas for 3 years from 1992 to 1994. Plant height at early growth stage and bolting rate showed linear regression as y=-8.502+0.5449x. Root yield was highest at the field with 11% bolting. Root yield of Angelica gigas in relation to seedling size was highest from medium and small seedlings with 10 to 15% bolting and from large seedlings with below 10% bolting.
