당귀속 2종 식물의 수용추출액과 휘발성물질이 종자발아와 유묘생장에 미치는 영향 (The Effects of Aqueous Extract and Volatile Substances of Two Angelica Plants on Seed Germination and Seedling Growth)

  • 참당귀와 일당귀의 수용추출액과 휘발성물질이 식물의 종자발아와 유묘생장에 미치는 영향을 알아보기 위하여 실험실 실험을 실시하였다. 참당귀의 수용추출액에 의한 참당귀의 종자발아율은 농도가 증가함에 따라 증가하였고 일당귀는 추출액의 농도증가에 따라 감소하였다. 일당귀의 수용추출액에 의해서는 참당귀와 일당귀의 종자발아가 억제되었다. 참당귀의 수용추출액에 의한 참당귀와 일당귀의 유묘생장은 10%농도에서는 대조구보다 높았으나 50%와 100% 농도에서는 농도증가에 따라 비례적으로 감소하였고, 일당귀의 추출액에 의한 참당귀와 일당귀의 유묘생장은 수용추출액의 농도가 증가함에 따라 뚜렷한 억제효과를 보였다. 참당귀와 일당귀의 휘발성물질에 의해서 상추의 종자발아는 전혀 영향을 받지 않았다. 참당귀와 일당귀의 휘발성물질로 처리한 경우, 대조구에 비하여 실험구의 상추의 유근신장이 억제되었지만 농도에 따른 변화는 보이지 않았다.

  • Angelicae gigantis radix is the root of the perennial plant, which belongs to the family Umbelliferae. However, this herbal drug is represented quite different chemical components according to its different genus name, though other herbal drugs (i.e. Leonuri Herba, Xanthii Fructus and so on) show similar constituents on the same name. The root of Angelica gigas containing the coumarin compounds is commonly used in Korea, while Angelica sinensis and Angelica acutiloba including phthalide compounds are used in China and Japan, respectively as Angelicae gigantis radix. In this paper, a nearinfrared spectroscopic method was developed to determine genus name of Angelica spp., especially A. gigas and A. sinensis which are commonly misused in herbal markets. X-ray fluorescence spectrometry and electronic nose have been also applied as nondestructive methods to discriminate A. gigas from A. sinensis according to their specific properties.

당귀 추출물 정맥 주사가 Middle Cerebral Artery Occlusion 모델 흰쥐에서 Gliosis 억제에 미치는 영향 (The Effect of Intravenous Injection of the Water Extract of Angelica gigas Nakai on Gliosis in the Middle Cerebral Artery Occlusion Rats)

  • Objectives : Gliosis becomes physical and mechanical barrier to axonal regeneration. Reactive gliosis induced by middle cerebral artery occlusion is involved with up-regulation of CD81 and GFAP (Glial fibrillary acidic protein). The current study is to examine the effect of the Angelica gigas Nakai(intravenous injection. 100 mg/kg twice in a day) on CD81 and GFAP of the rat in the brain after middle cerebral artery occlusion. Methods : Cerebral infarction was induced by middle cerebral artery occlusion. And after intravenous injection of water extract of Angelica gigas Nakai, the size of cerebral infarction was measured. Examination of optical microscope were also used to detect the expression of CD81 and GFAP in the brain of the rat. Results : The following results were obtained : We found that size of cerebral infarcion induced by MCAO (Middle Cerebral Artery Occlusion) in rats were decreased after intravenous injection of Angelica gigas Nakai. We injected the extract of Angelica gigas Nakai to the MCAO in rats, and the optical microscope study showed that Angelica gigas Nakai had effect on protecting the cells of hippocampus. We found that GFAP, CD81 and ERK of the brain in rats with cerebral infarction after MCAO were meaningfully decreased after intravenous injecting Angelica gigas Nakai. We found that c-Fos expression of the brain in rats with cerebral infarction after MCAO were significantly increased after intravenous injecting Angelica gigas Nakai. Conclusions : These results indicate that Angelica gigas Nakai could suppress the reactive gliosis, which disturbs the astrocyte regeneration in the brain of the rat with cerebral infarction after MCAO by controlling the expression of CD81 and GFAP. And the effect may be modulated by the up-regulation of c-Fos and ERK.

A Flavone Glycoside from Angelica gigas Roots

  • A flavone glycoside was isolated from the roots of Angelica gigas (Umbelliferae) and identified as diosmin $[diosmetin-7-O-{\alpha}-{_L}-rhamnopyranosyl \;(1{\rightarrow}6)-{\beta}-{_D}-glucopyranoside]$ by spectroscopic methods. This is the first report of a flavone gylcoside from Angelica species.

참당귀 추출분말이 Monosodium Iodoacetate로 유발된 흰쥐의 골관절염에 대한 효과 (Effect of Angelica gigas extract powder on progress of osteoarthritis induced by monosodium iodoacetate in rats)

  • 참당귀 추출물의 골관절염에 대한 억제 또는 치료효과를 알고자 monosodium iodoacetate로 유발한 흰쥐의 골관절염 모델을 사용하여 30마리 6주령 SD흰쥐를 정상군(normal), 비처리군(untreated) 및 참당귀 처리군(treated)으로 각각 10마리씩 나누어 실험하였다. 실험기간 동안 실험 개시일부터 4일 간격으로 체중 변화를 측정하였다. 3주간 참당귀 추출물 처리 후 관절연골 내의 glycosaminoglycan (GAG)의 함량 및 proteoglycan (PG) 함량을 측정 비교하였다. 그 결과, 참당귀 추출물은 MIA로 증가한 GAG를 완화시켰으며, PG의 함량을 회복시켰다. 또한, 활액에서의 TNF-a함량은 미약하게 감소하였다. 한편, 경골 관절연골을 분리하여 Safranin-O 염색으로 관절연골 상태를 확인한 결과, MIA 처리로 유발된 관절연골 소실을 억제하였다. 이 참당귀 추출물을 HPLC로 분석한 결과, 주요 성분인 decursin 및 decursinol angelate의 총 함량은 $10.5{\pm}0.2%$이었다.

Detection of Anticancer Activity from the Root of Angelica gigas In Vitro

  • Anticancer activity of a fraction of the ethanol extract from the root of Korean angelica (Angelica gigas Nakai) was recognized in human cancer cell lines HeLa $S_3$, K-562, and Hep $G_2$. The extract blocked the phorbol ester-inducing megakaryocytic differentiation of K-562 cells, which indicated the modification of protein kinase C (PKC) activity. In vitro assay showed the activation of PKC by the extract. An effective fraction of the Angelica gigas extract, of which $R_f$ value was 0.64 in a thin layer chromatography, was a different component from those of European angelicas. The $ED_50$ value of the fraction was 8, 9, and $16\;\mu\textrm{m}/ml$ against HeLa $S_3\;Hep\;G_2$, and K-562 cells, respectively, while the fraction showed higher $ED_50$ values against normal cell lines.

Multiplex Polymerase Chain Reaction을 이용한 당귀 종 판별 (Development of Multiplex Polymerase Chain Reaction Assay for Identification of Angelica Species)

  • Background: Angelica gigas, A. sinensis, and A. acutiloba are commercially important in the herbal medicine market, and among them, A. gigas has the highest economic value and price. However, their similar morphological traits are often used for fraud. Despite their importance in herbal medicine, recognition of the differences between Angelica species is currently inadequate. Methods and Results: A multiplex polymerase chain reaction (PCR) method was developed for direct detection and identification of A. gigas, A. sinensis, and A. acutiloba. The gene for the distinction of species was targeted at ITS in the nucleus and trnC-petN gene in chloroplasts. The optimized multiplex PCR in the present study utilized each Angelica species-specific primer pairs. Each primer pair yielded products of 229 base pairs (bp) for A. gigas, 53 bp for A. sinensis, 170 bp for A. acutiloba. Additionally non-specific PCR products were not detected in similar species by species-specific primers. Conclusions: In the present study, a multiplex-PCR assay, successfully assessed the authenticity of Angelica species (A. gigas, A. sinensis, and A. acutiloba). and whole genome amplification (WGA) was performed after DNA extraction to identify, the species in the product. The detection method of raw materials developed in the present study could be applied to herbal medicine and health functional food management.

참당귀에서 분리된 내생균의 다양성과 병원균 억제 효과 (Species Diversity and Antifungal Activity of Endophytic Fungi Isolated from Angelica gigas Nakai)

  • 다양한 지역에서 채취한 참당귀(Angelica gigas Nakai)의 잎, 줄기, 뿌리에서 내생균을 분리하였다. 내생균 균주는 형태적특징과 ITS (internal transcribed spacer) 지역의 염기서열을 이용하여 동정하였다. 총 35종의 내생균이 동정되었다. 내생균의 다양성은 참당귀의 조직에 따라 다르게 나타났다. 분리된 내생균은 참당귀 점무늬병균 Phoma sp. Y11 균주와 대치배양하여 항균활성을 측정하였다. 총 14종의내생균이균주 Y11에대해억제효과를나타내었다. 본 연구는 참당귀에서 분리한 내생균이 참당귀의 점무늬병에 대한 생물학적 방제제로 사용될 수 있다는 것을 제시하고 있다.

한국산 당귀의 생약학적 연구 (Pharmacognostical Studies on the Dang Gui from Korea)

  • Chinese crude drug 'Dang Gui' has been used mainly as a tonifying the blood, promoting blood circulation, relieving pain and moistening the bowels. To clarify the botanical origin of 'Dang Gui' from Korea, the anatomical characteristics of Angelica gigas, Angelica sinensis and Angelica acutiloba were studied. As a result, it was clarified that 'Dang Gui' from Korea was the root of Angelica gigas.

  • Systematic fractionation of Angelica gigas roots led to the isolation of linear coumarins such as decursinol angelate, decursin and nodakenin. These compounds were tested for their effects on rat lens aldose reductase. Nodakenin was demonstrated to exhibit significant inhibition of rat lens aldose reductase activity with $IC_{50}$ value of $7.33\Mu\textrm{M}$.
