Microstructural and Melting Characteristics of Imitation Cheese Analog (모방치즈의 조직과 융점특성 연구)

  • The imitation processed cheese (IPC), formulated with delactosed non fat dry milk (DENFDM) only, showed the smallest melting area. Calcium caseinate sample showed the largest spread. Satistically protein source as a major ingredient for the IPC yielded significantly different melting areas. In a similar fashion, initial melting temperature was markedly and significantly influenced by protein source. In effect of addition of DENFDM on microstructure of IPC analog revealed that as the fraction of DENFDM was decreased, the network was much more uniform and the fat globules were also better dispersed compared to DENFDM cheese analog. Therefore the results of this study help predict that melting and microstructural characteristics are largely but not solely dependent on the protein source. The DENFDM has a potential beneficial effect as a partial replacement of caseinate in the formation of IPC to characteristic close to processed cheese.

  • This study describes the effects of 1,25-dihydroxyvitamin D3[1,25(OH)2D3, calcitriol] analog, 1,25(OH)2-16ene-23yne-D3 on proliferatin and differentiatin of the human histiocytic lymphoma cell line U937. This paper also describes the effects of 1,25(OH)2-16ene-23yne-D3 on ${\gamma}$-interferon(IFN-${\gamma}$) synthesis by phytohemagglutinin-activated peripheral blood lymphocytes(PBLs). In the present investigation, 1,25(OH2)-16ene-23yne-D3 was compared to the natural metablite of vitamin D3, 1,25(OH)2D3. 1,25(OH)2-16ene-23yne-D3 was more potent than 1,25(OH)2D3 for inhibition of proliferation and induction of differentiation of U937 cells, Its effects on inhibition of proliferation was about 30-fold more potent than 1,25(OH)2D3. On induction of differentiation as measured by nonspecific esterase (NSE) activity and morphologic change, this analog morphologically and functionally differentiated U937 cells to monocyte-macrophage phenotype showing a decrease of N/C ration in Giemsa staining and the increase of adherence ability of surface. After 3 days in culture, a more significant supression of IFN-${\gamma}$ synthesis analog on supression of IFN-${\gamma}$ synthesis was a dose-dependent manner, with peak activity at 10-7M. The strong direct effects of 1,25(OH)2-16ene-23yne-D3 on cell proliferation and cell differentiation, make this compound an interesting candidate for clinical studies for several types of malignancies, and the effects on supression of IFN-${\gamma}$ synthesis provide the further evidence for a role of 1,25(OH)2-16ene-23yne-D3 in immunoregulation.

Evaluation of Rheological and Sensory Characteristics of Plant-Based Meat Analog with Comparison to Beef and Pork

  • This study explored the physicochemical, textural, and sensorial properties of a meat analog (MA) as compared to beef and pork meats. Results illustrate that MA patties had lower moisture, fat, and protein content, as well as higher ash and crude fiber than beef and pork. Likewise, MA patties had a higher pH, lightness (L*), and redness (a*) than either beef or pork. Pork meat exhibited the highest released water (RW) and cooking loss (CL) values, followed closely by MA with beef displaying the lowest values. Regardless of patty type, the post-cooking diameter patties were reduced significantly (p<0.05). However, the Warner-Bratzler shear force (WBSF), hardness, chewiness, and gumminess of beef were significantly higher than that of either pork or MA. The visible appearance of MA patties had more porous and loose structures before and after cooking. Consequently, based on sensory parameters, MA patties demonstrated the higher values for appearance and firmness, followed by beef and pork respectively, although the difference was not statistically significant. Therefore, the current study demonstrated that some physicochemical, textural, and sensory characteristics of beef and pork exhibited the most similarity to MA.

Optimization of Physical Conditions for Caviar Analog Preparation Using Calcium-alginate Gel Capsules

  • High prices, overfishing, and contamination have limited the availability of natural caviar as a food product. We attempted to apply encapsulation by calcium-alginate gel membranes to caviar analog preparation in an effort to produce a high-quality replacement for natural caviar. Physical conditions of stirring speed $(X_1,\;rpm)$ and gelation time $(X_2,\;min)$ as the independent variables for gelation were optimized by response surface methodology. Sphericity $(Y_1,\;%)$, diameter $(Y_2,\;mm)$, membrane thickness $(Y_3,\;mm)$, rupture strength $(Y_4,\;g)$, and rupturing deformation $(Y_5,\;mm)$ were used as the dependent variables to compare characteristics of the capsules for caviar analogs with natural caviar. The values of the independent variables as evaluated by multiple response optimization were $X_1=-0.1271 (278 rpm) and $X_2=0.4436$ (12.2 min), respectively. Predicted values of the four dependent variables were $Y_1=97.7%,\;Y_2=2.97mm,\;Y_4=1,465g,\;and\;Y_5=1.15mm$. Membrane thickness $(Y_3)$ was eliminated from the dependent variables for multiple response optimization because it could not be measured with an image analyzer. The experimental values prepared under the optimal conditions for verification nearly coincided with the predicted values and satisfied the conditions of natural caviar.

Effects on Quality Characteristics of Extruded Meat Analog by Addition of Tuna Sawdust (참치 톱밥의 첨가가 압출성형 인조육의 품질 특성에 미치는 영향)

  • In this study, tuna sawdust was added to extruded meat analog in order to develop a meat analog with high quality. Addition of tuna sawdust has merit for utilizing a byproduct from poultry processing. Physicochemical characteristics were examined through the extrusion cooking process. The basic mixture of sample mixed with 65% deffated soy flour 25% isolated soy protein, and 10% corn starch was setup as the raw material. Three kinds of samples were made in total by addition of 15% and 30% tuna sawdust to this mixture. The extrusion process had a screw speed of 250 rpm, die temperature of $140^{\circ}C$, and moisture content of 50%. As addition of tuna sawdust increased, breaking strength and density decreased, specific length increased, and integrity and water holding capacity decreased. Likewise, nitrogen solubility index and protein digestibility decreased as addition of tuna sawdust increased. DPPH radical scavenging activity increased as addition of tuna sawdust addition, whereas it decreased as storage period increased to 30 or 60 days. The value of rancidity decreased as addition of tuna sawdust increased. However, 60 days later, radical scavenging activity increased more or less, and a significant difference was detected 150 days later. In conclusion, addition of tuna sawdust increased soft texture, and nutrition of the basic mixture sample. The process promoting functionality such as improvement of antioxidant function was confirmed through this study.

Effects of Heat, Salt and Hydrocolloid Treatments on Flying Fish Cypselurus agoo Roe Analogs Prepared Using Calcium Alginate Hydrogels

  • The present study was conducted to elucidate effects of heat, salt and hydrocolloid treatments on flying fish Cypselurus agoo roe analogs prepared using calcium alginate gel. The changes in size, sphericity and rupture strength of the analogs as affected by treatments of heat, sodium chloride and hydrocolloids were investigated. The size (mm), sphericity (%), and rupture strength (kPa) of the analogs were $2.2{\pm}0.1$, $98.2{\pm}0.2$, and $74.7{\pm}1.7$, respectively. When the analogs were heated at $95^{\circ}C$ in water, the size was slightly decreased. The rupture strength by curing with 2% sodium chloride was slightly increased. Sphericity didn't show significant differences by sodium chloride and heat treatment. The rupture strength of the analogs was slightly decreased by heat treatment, whereas remarkably decreased by curing with sodium chloride. In order to prevent a remarkable decrease in rupture strength of the analogs by curing with sodium chloride, the analogs were treated with hydrocolloids such as xanthan gum, gum guar, glucomannan, pectin and gelatin. The hydrocolloids treated analogs showed an increment in size and no significant changes in sphericity. On the other hand, the rupture strength of the hydrocolloids treated analogs exhibited remarkable increase than that of untreated ones.

Isolation of a Tryptophan-Overproducing Strain Generated by EMS Mutagenesis of Candida rugosa

  • To isolate a mutant strain which overproduces tryptophan, mutants of Candida rugosa were screened after EMS (ethyl methane sulfonate) mutagenesis. Fluorotryptophan, a tryptophan analogue, was used for selection of a tryptophan-overproducing mutant after mutagenesis. Among 50 mutants, several candidates were selected based on intracellular tryptophan content. Amino acid analysis results showed that C3 was the best strain because it had the highest amount of tryptophan among the mutants.

  • 1, 25(OH)2-23ene-D3 is a novel vitamine D3 analog which has a double bond between C-23 and C-24. We describe the effects of this analog on cell differentiation and cell proliferation in vitro using the human histiocytic lymphoma cell line U937, and on calcium metabolism in rats in vivo. In the present investigation 1, 25(OH)2-23ene-D3 was compared to the natural metabolite of vitamin D3, 1$\alpha$, 25-dihydroxycholecalciferol[1, 25(OH)2-23ene-D3 was more potent than 1, 25(OH)2-23ene-D3 for inhibition of proliferation and induction of differentiation of U937 cells. Especially, its effect on induction of differentiation, as measured by superoxide production and nonspecific esterase(NSE) activity, was about 20-fold more potent that 1, 25(OH)2-23ene-D3. This analog morphologically and functionally differentiated U937 cells to monocyte-macrophage phenotype showing a decrease of N/C ratio in Giemsa staining and the increase of adherence ability to surface. Intraperitoneal administration of 1, 25(OH)2-23ene-D3 to rats showed that the compound had at least 50 times less activity than 1, 25(OH)2-23ene-D3 in causing hypercalcemia and hypercalciuria. The strong direct effects of 1, 25(OH)2-23ene-D3 on cell proliferation and cell differentiation, coupled with its decreased activity of calcium metabolism make this compound an interesting candidate for clinical studies including patients with leukemia, as well as several skin disorders, such as psoriasis.

Enhanced Enzyme Activities of Inclusion Bodies of Recombinant ${\beta}$-Galactosidase via the Addition of Inducer Analog after L-Arabinose Induction in the araBAD Promoter System of Escherichia coli

  • We observed that an inclusion body (IB) of recombinant ${\beta}$-galactosidase that was produced by the araBAD promoter system in Escherichia coli (E. coil) showed enzyme activity. In order to improve its activity, the lowering of the transcription rate of the ${\beta}$-galactosidase structural gene was attempted through competition between an inducer (L-arabinose) and an inducer analog (D-fucose). In the deep-well microtiter plate culture and lab-scale fermentor culture, it was demonstrated that the addition of D-fucose caused an improvement in specific ${\beta}$-galactosidase production, although ${\beta}$-galactosidase was produced as an IB. In particular, the addition of D-fucose after induction led to an increase in the specific activity of ${\beta}$-galactosidase IB. Finally, we confirmed that the addition of D-fucose after induction caused changes in the structure of ${\beta}$-galactosidase IB, with higher enzyme activity. Based on these results, we expect that an improved enzyme IB will be used as a biocatalyst of the enzyme bioprocess, because an enzyme IB can be purified easily and has physical durability.

Production optimization of flying fish roe analogs using calcium alginate hydrogel beads

  • Due to decreased supplies of marine resources and byproducts, new processing technologies for the development of analogs for natural fishery products are becoming increasingly important in the fishing industry. In the present study, we investigated the optimal processing conditions for flying fish roe analogs based on alginate hydrogels. Optimized processing of these analogs was performed by response surface methodology. The optimal processing conditions for the flying fish roe analogs (based on sphericity) were at a sodium alginate concentration of 2.41 %, calcium chloride solution curing time of 40.65 min, calcium chloride concentration of 1.51 %, and a reactor stir speed of $254{\times}g$. When the experiment was performed under these optimized conditions, the size (mm), sphericity (%), and rupture strength (kPa) of the analogs were $2.2{\pm}0.12$, $98.2{\pm}0.2$, and $762{\pm}24.68$, respectively, indicating physical properties similar to their natural counterparts.
