Oxidative stress has been reported to be one of causes of neuritis. This study examined antioxidative activities of methanol extracts of six amphibian species known to be medicinal animals (Rana catesbeiana, R. coreana, R. rugosa, R. dybowskii, R. nigromaculata, and Hyla japonica) and investigated their effects of inhibiting nitric oxide (NO) production and cytotoxicity on the murine macrophage RAW264.7 cells. As inflammation is closely associated with reactive oxygen species, assays on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, xanthine oxidase inhibitory activity, superoxide anion radical scavenging activity and NO scavenging activity of the extracts of the six species were performed to investigate their antioxidative activity. The results obtained were as follows; All extracts showed antioxidative activity, and the activity of R. dybowskii was the highest in comparison among those. Anti-inflammatory effects of the extracts were also examined, the five extracts except that of R. rugosa did not show cytotoxicity for RAW264.7 cells at the maximal concentration ($1,000{\mu}g\;mL^{-1}$). Selectivity index, meaning NO scavenging activity compared to cytotoxicity, showed the highest level in the extract of R. dybowskii. These results will be very useful basic data for future studies on prevention and treatment of human diseases to understand the biological roles of amphibian extracts throughout the antioxidative or anti-inflammatory pathways.
Effect of cadmium on the early amphibian development was analyzed through FETAX (Frog Embryo Teratogenesis Assay: Xenopus). Embryos manifested concentration-dependent mortality and realformations; shortage of anterior-posterior axis gut realformation, ocular anomalies, bent notochord, misshapen dorsal fin, and derreal blisters. The treatment with 1.5ppm cadmium solution caused 100% mortality and concentration of lppm did not kill the embryos that caused 100% anomaly. The teratogenic index (TI = LC50 /EC50) was 2.8 indicating that $CdCl_2$ is teratogenic for Xenopus laevis. Embryos that were pulsetreated with at early to late blastula stage (St. 3-9) and mid to late blastula stage (St. 6-10) showed relatively strong resistance to cadmium, but the embryos treated at gastrula stage (St. 10-13) showed high mortality. And the embryos treated at tailbud stage (after St. 25) showed highest mortality of any other early stages. Effects of temperature were studied through pulse- treatment during gastrula stage at $20^{\circ}C$ and $30^{\circ}C$. The embryos treated with 7.5ppm at $30^{\circ}C$ and 15ppm at $20^{\circ}C$ caused 100% mortality respectively, indicating that higher temperature had more severe toxic effect. One of the most peculiar effect of cadmium at gastrulation was distortion of the tail. The probable cause of toxic effect of Cd was discussed.
For the study on the sex ratio in natural populations of the different localities, the amphibian species were collected as the materials at the seven localities in Korea, and were obtained the following results. The sex ratios of each amphibian species suggest that each has it's own specific sex rations the following list. Rana nigromaculata H. : 109-39, Rana rugosa SCH. : 67.44, Rana temporaria coreana O. : 158.45, Hyla arborea japonica G. : 137.70. In Rana nigromaculata H. the average sex ratio was 109.39 as the above list, and its was reached statistically significant level according to the different localiies. The sex ratio was generally higher in the eastern area than that in the western area in this country. The sex ratio of the adult frogs of Rana nigromaculata H. was 135.18 , and it has a statistically significant level by each locality but the sex ratio of the younger frogs of the same species was 100.97 which was a little lower than that in the adult, and it was not laid at a significant level according to the localities. There is no doubt that the difference of sex ratios according to the localities was mainly due to the different ratios of the adult frogs which were effected on the constitution of sex by environmental factors in each locality.
To assess the genetic diversity and population structure of Korean K. borealis, allozyme analysis was performed. The average genetic variability of Korean K. borealis populations was %P=13.2, Ho=0.048, and He=0.045. This value was the lowest in comparison with other Korean amphibian species studied. Also, the value was much lower than that of a reference population from Chinese K. borealis (%P=50, Ho=0.125, He=0.172). Wright's F-statistics showed that Korean K. borealis has distinctly low level of gene flow among regional populations (F$_{ST}$=0.339, Nm=0.487) in comparison with other Korean amphibian species studied. However, the average level of genetic divergence among Korean K. borealis populations was moderate (Nei's D=0.020). Therefore, it appeared that low levels of genetic diversity (He=0.045) and gene flow (Nm=0.487) among regional populations ave probably due to the results of decreasing population size and patchy distribution of this species in Korea.

Background: Ranavirus is an emerging infectious disease which has been linked to mass mortality events in various amphibian species. In this study, we document the first mass mortality event of an adult population of Dybowski's brown frogs (Rana dybowskii), in 2017, within a mountain valley in South Korea. Results: We confirmed the presence of ranavirus from all collected frogs (n = 22) via PCR and obtained the 500 bp major capsid protein (MCP) sequence from 13 individuals. The identified MCP sequence highly resembled Frog virus 3 (FV3) and was the same haplotype of a previously identified viral sequence collected from Huanren brown frog (R. huanrenensis) tadpoles in South Korea. Human habitat alteration, by recent erosion control works, may be partially responsible for this mass mortality event. Conclusion: We document the first mass mortality event in a wild Korean population of R. dybowskii. We also suggest, to determine if ranavirus infection is a threat to amphibians, government officials and researchers should develop continuous, country-wide, ranavirus monitoring programs of Korean amphibian populations.
세계 양서류 감소의 주요 요인으로 감염성 질병이 활발하게 연구되고 있으며, 야외 병원균 전파의 가능한 근원지로서 양서류 증양식장들이 언급되고 있으나, 국내에서 관련된 연구수행은 매우 미진한 상태이다. 본 연구에서는 인제, 괴산, 공주에 위치한 북방산개구리 증양식장의 내 외부에서 채집된 성체와 유생의 피부와 내부 장기들에서 확인되는 세균과 곰팡이 콜로니 수의 지역 간, 증양식장 내 외부 간 차이 여부를 파악하고, 나아가 유생으로부터 발견되는 세균의 종류를 16s rDNA 서열 비교를 통해 규명하고자 하였다. 연구결과, 성체의 경우 괴산에서 채집된 성체의 피부와 소화관으로부터 표집된 세균 콜로니 수와 피부와 간으로부터 표집된 곰팡이 콜로니 수가 인제에서 확인된 콜로니 수보다 의미 있게 많았으나 증양식장 내 외 간에는 두 지역 모두에서 어느 부위에서도 차이가 없었다. 유생의 경우 공주에서 채집된 유생의 내부로부터 표집된 곰팡이 콜로니 수가 인제에서 확인된 콜로니수보다 많은 경향을 보였다. 증양식장 내 외부의 비교 결과, 피부에서의 박테리아 수는 인제 증양식장 내부, 개체 내부에서의 박테리아 수는 공주 증양식장 외부에서 더 많은 콜로니 수가 확인되었다. 성체의 건강지수가 좋을수록 피부의 박테리아 콜로니 수와 피부와 심장의 곰팡이 콜로니 수가 적었으나, 유생에서는 관련성이 없었다. 유생으로부터 검출된 세균을 분류한 결과, 4개 문 내, 15개 속에 속하는 세균이 검출되었으나 증양식장 간 및 증양식장 내 외부 간 차이는 분명하지 않았다. 우리의 결과는 증양식장의 지역과 증양식장 내 외부 간의 서로 다른 조건들이 양서류에 있는 세균과 곰팡이의 군집크기 차이를 유발하며, 이것이 증양식 북방산개구리의 건강상태에 영향을 미칠 가능성이 있음을 보여준다. 더불어, 우리의 결과는 성공적인 양서류 증식장 운영과 증식장으로부터 질병의 가능한 야외전파를 막기 위하여, 증식장 내 효율적인 질병관리의 필요성이 높음을 시사한다.
This study set out to identify problems with amphibian habitation by the wetland types and improve their habitation environment in urban forest wetlands, thus creating a habitat for amphibians. Study site include forest swamps in Jatjul Park as well as Yeoji neighborhood Park in Guro-gu, and in Choansan neighborhood Park in Dobong-gu. The forest swamp in Jatjul Park gets its water from Mt. Maebong and it is a former escalated farmland-turned wetland. The swamp area is $2,500m^2$, a forest zone and a landscape planting site are 83.27% and 6.70% each. Target species Seoul pond frogs are inseparable from rice fields because they live in a short radius of and lay eggs in or near paddy fields, and Rana nigromaculata have similarities with Rana plancyi chosenica in choosing their habitats. There was need for paths that would lead to other paths so amphibians would spread to other parts of the forest and for measures to secure open water. Modifying a variety of routes for water, human and animals along with building a buffer to keep the core habitation zones were required. The forest swamp in Yeonji neighborhood Park used to be a water reservoir on the foot of Mt. Gunji. The swamp area is $1,980m^2$, a forest zone and farmland account for 80.61% and 4.88% each. Non-point pollutants from upstream along run into the subject forest marsh, bare ground on the around swamp and steep stone embankments obstructed amphibians. Target species was Bufo gargarizans that live in forests and edges of hills and spawn in deep water. The forest swamp in Choansan neighborhood Park gets its water from Mt. Choan and it is close to its water source that it is a mountain stream forest wetland. The basin and the swamp are $35,240m^2$ and $250m^2$ in size respectively. A forest zone accounts for 90.20%, high stone embankments laid in refurbishing the valley obstruct amphibians and there is water shortage in times of droughts. Target species were Rana coreana, Rana dybowskii and Hynobius leechii that live in mountain valleys, streams and wetlands and lay eggs in forest marshes and rocks in valleys. Looking into the three swamps of amphibian habitation, I came to conclusions that those wetlands were suitable for their amphibians but man-made facilities blocked their corridors leading to other corridors and even killed off target species in some parts of those swamps by destroying those parts. Amphibians live in water, on ground and underground at different stages of life. Hence, we should take this fact into consideration when planning their habitats and design core habitation zones, buffers zone and use zones accordingly. Buffer zones ought to be between core habitation zones and surrounding trees. Aiming at protecting core habitation zones, buffers should be in harmony with habitation zones. Use zones should be minimized in size and not in direct contact with core habitation zones.
스테로이드 합성효소 중 $17\;{\alpha}-hydroxylase/17,20-lyase(P450_{C17})$는 progesterone을 $17\;{\alpha}-hydroxyprogesterone$을 거쳐 androstenedione으로 변환을 담당하는 효소이다. 양서류 난소에서 스테로이드 합성의 분자적 조절과정의 연구에 사용할 목적으로 북방산 개구리(Rana dybowskii) 난소에서 $P450_{C17}$ cDNA를 클로닝 하였다. 북방산 개구리 난포세포의 cDNA library 검색을 통해 분리된 약 2.5kb의 cDNA는 529개의 아미노산을 가진 단일 번역틀을 가지고 있었다. 개구리 $P450_{C17}$의 아미노산 서열은 Xenopus와는 76%, 닭과는 63%, 그리고 사람과는 약 45%의 동일성을 보여 주였고, 동시에 진화적으로 척추동물에서 매우 잘 보존된 아미노산 서열을 가지고 있었다. 노던 분석에서 개구리의 $P450_{C17}$ 전사체는 난소에서만 2.5kb와 3.6kb 크기의 두 종류가 발견되었다. 그리고 개구리 Rana $P450_{C17}$ cDNA는 비스테로이드 합성 세포인 COS-1세포에서 분명한 $17\;{\alpha}-hydroxylase/17,20-lyase$ 활성을 주었다. 따라서 클로닝된 개구리 $P450_{C17}$ 유전자는 양서류의 난소에서 스테로이드 합성의 분자적 기작을 연구하는데 매우 유용할 것으로 사료된다.
상처회복과정은 염증, 재상피화, 그리고 기질의 재형성등을 포함한 포괄적인 생물학적 반응이다. 생물학적 분류체계에서, 회복능력은 매우 다양한데, 일반적으로 하등동물이 고등동물에 비교해 단순하며 뛰어난 것으로 알려져 있다. 따라서, 하등동물들을 이용한 실험모델은 상처회복과 치유의 기작을 연구하는데 자주 이용되고 있다. 양서류를 이용한 피부 상처회복과정 동안의 조직화학적 특성들을 규명하기 위하여 한국산 무당개구리(Bombina orientalis)를 이용하였다. 1일, 10일 그리고 16일의 조직에서 점액물질이 활발하게 분비되는 것을 확인하였으며, PAS (periodic acid shiff)와 Alcian blue (pH 2.5) 반응에 강하게 염색되었다. PTAH(phosphotungstic acid hematoxylin) 염색을 통하여 상처 후 10일 조직에서 상처주변의 진피층에서 콜라겐의 합성이 증가하는 것을 확인하였다. 또한, 세포의 유사분열의 시점을 확인하기 위한 MG-P (methyl green pyronin)염색에서 초기에는 3시간과 6시간에 강한 염색반응을 관찰하였고, 이는 23일과 27일 조직에서도 동일한 패턴으로 이어졌다.
This paper describes a neural network modelling of a vertebrate retina using a discrete-time and discrete-space approach based on neuro-anatomical data, and the computer simulations of the model which approximate the frog/amphibian negro-physiological data. It then compares them and describes how such a model can be beneficially used for confirming the hypothesis of a given neural system and further predict yet unknown experimental data.
