  • Ion-selective electrode responsive to the thiocynate ion prepared by using the quaternary ammonium salts as a active material and PVC as a membrane matrix. The effect of chemical structure and composition of active material, and the membrane thickness on the linear response. the detection limit, and Nernstian slope of the electrode studied. Under the above optimum conditions of membrane, the effect of pH and the selectivity coefficients to various interfering anions were compared and investigated. It was concluded that the functions of thiocynate ion-selective electrode(ISE) were closely related to the chemical structure of the quaternary ammonium salts. The linear response, and the detection limit of the electrode potential increased with the increase of the carbon chain length of the alkyl group in the quaternary ammonium salts in the ascending order of Aliquat 336T, TOAT, TDAT, and TDDAT. The optimum membrane thickness was 0.3mm. The electrode characteristics was better with the decrease of the concentration of active material, and the best concentration was 3 weight percent. The membrane potential was independent of the pH variation in the region from pH 2 to 12. The order of the selectivity coefficients is as follows:Cl $O_4$$^{[-10]}$$I^{[-10]}$ >N $O_3$$^{[-10]}$ >B $r^{[-10]}$$F^{[-10]}$ >C $l^{[-10]}$ >O $A_{c}$ $^{[-10]}$ 〓S $O_4$$^{2-}$.

The PVC Membrane Electrode for Measuring Hazardous Anion in Waste Water Process (폐수처리 공정중 유해음이온을 측정하기 위한 PVC 막 전극)

  • The perchlorate, thiocyanate, and nitrate ion-selective PVC membrance electrode for measuring hazardous anion in waste water were developed by incorporating the quaternary ammonium salts as active material. Ion-selective characteristics in waste water were studied by the useful pH range, the selective coefficients to various interfering anions, and the stability of electrode potential. DBP was best as a plasticizer. The effect of the membrane thickness on the electrode characteristics was improved with decreasing the membrane thickness, but below the optimum membrane thickness the electrode exhibited an inverse trend. The electrode potential of perchlorate, thiocyanate, and nitrate electrode with TDDA, as active material, was stable within the pH range 4-11, 3-12, and 4-10 repectively. And the long-term potential stability of these electrodes were 3.0, 3.5, and 3.5 months respectively. The order of the selectivity coefficients was as shown below ; $ClO_4{^-}$ > $SCN^-$ > $I^-$ > $NO_3{^-}$ > $Br^-$ > $CN^-$ > $F^-$ > $Cl^-$ > $Ac^-$ > $H_2PO_4{^-}$, $SO_4{^-}$.

Spectrophotometric Determination of Submicrogram Amounts of Orthophosphate Suitable for Water Analysis (水質分析을 目的한 極微量燐酸鹽의 高感度比色定量法)

  • The critical examination of the spectrophotometric method for determining microgram quantities of phosphate by the n-butyl acetate extraction as molybdophosphoric acid and subsequent development of the molybdenum blue has been made. In this procedure from 2 to 8 ${\mu}g$. of phosphate-phosphorus can be determined under optimum conditions. The final concentration of ammonium molybdate and the final acidity of perchloric acid for the formation of heteropoly acid are suitable to be ranges of 0.5 to 1.1% and 0. 5 to 1. 1 N respectively, and subsequently extracted with 10 ml. of n-butyl acetate. The extract is developed to molybdenum blue with 5.0 ml. of 1. 3% stannous chloride in 1N hydrochloric acid. The color is stable for at least one hour in the use of perchloric acid for the condensation. In order to determination of submicrogram amounts of phosphate, the sensitivity of the molybdenum blue method is hardly sufficient, a sensitive and stable molybdenum(V)-thiocyanate complex method has been investigated. By the procedure less than 1.2 ${\mu}g$. of phosphate-phosphorus can be determined with an accuracy of less than 5% the relative error. The molybdenum(Ⅵ) extracted by the above procedure is reduced to molybdenum(V) in the extract directly with a solution of 4 to 10% of stannous chloride, 0.5 to 1.5 mM of copper, and 0.1 to 0.9 N of perchloric acid as final concentration in 4.3 to 6.3 N of hydrochloric acid or 9.0 to 13.0 N of sulfuric acid by heating for one minute in boiling water, after cooling, the molybdenum(V)-thiocyanate complex color is developed by adding 6.0 M ammonium thiocyanate solution making the final concentration to be in a range of 0.4 to 0.9 M. This procedure the very sensitive, reliable, and stable can be applied to determining submicrogram amounts of phosphate in natural waters with a precision of 1.6 ${\times}\;10^{-2}$ the standard deviation as absorbance.

  • One of the most frequently used method for measurement of the degree of sizing (viz., hydrophobicity) is the Stockigt test. However, the Stockigt test was influenced by various factors such as dropping height, dropping amount, dropping speed and viewing angle. The resultant data of the sizing degree on the same specimen also varied according to different testers. Thus, the Stockigt test should be modified to be regarded as a highly reliable and reproducible standard method. For modifying the Stockigt test, it was required to quantify red coloration by reaction between 1% ferric chloride and 2% ammonium thiocyante during Stockigt testing. The cameras capturing the serial images during the red coloration process were the CMOS (Complementary Metal Oxide Semiconductor)-type and CCD (Charge Coupled Device)-type cameras. For measurement based on KS M 7025, the CCD-type camera must be used due to its high resolution, and on the other hand, for measurement based on Tappi Useful Method 429, the CMOS-type camera may be used owing to its low resolution. It was needed to covert the RGB values of a droplet image into HSV(Hue, Saturation, and Value) values because the human eyes are much closer to HSV than RGB. Among HSV values, the Hue value was accepted as the most reliable index consistent with the red coloration process by excluding the surrounding conditions such as light, tester's movement etc.

Avidity of serum immunoglobulin G antibodies to Porphyromonas gingivalis in the elderly with chronic periodontitis

  • The present study aimed at evaluating serum immunoglobulin G (IgG) avidity to Porphyromonas gingivalis in elderly patients with mild and severe chronic periodontitis. The avidity of antibodies against P. gingivalis present in the sera of 18 patients with mild chronic periodontitis and 18 patients with severe chronic periodontitis was evaluated using an ammonium thiocyanate-dissociated enzyme-linked immunosorbent assay (ELISA). The results showed that the mean absorbance value in serum IgG antibody titers was significantly higher in the severe chronic periodontitis group than in the mild chronic periodontitis group ($198{\pm}35ELISA$ unit [EU] vs. $142{\pm}32EU$, p < 0.01). However, there was no significant difference between the two groups in antibody avidity ($65{\pm}57EU$ vs. $54{\pm}27EU$). These findings suggest that humoral immune responses to P. gingivalis between mild and severe chronic periodontitis in elderly patients are characterized by the differences in the quantity rather than the quality of the antibodies.

  • Pyungwi-san(PWS) have been using as a basic prescription of digestive disorder in Korean traditional medicine. This study was performed to examine the in vitro antioxidant activity of the extract using different antioxidant tests including by 1,1-diphenyl-2-picryl-hydrazil (DPPH) radical scavenging, superoxide anion radical scavenging, metal chelating hydrogen peroxide scavenging, lipid peroxydation protective effect and scavenging effect of nitric oxide and peroxynitrite. Herbal-acupuncture solution of PWS(PWS-HS) exhibited a concentration-dependent inhibition of DPPH radical adduct formation and it showed dose-dependent free radical scavenging activity onto superoxide anions. In addition, the result of metal chelating hydrogen peroxide scavenging and ammonium thiocyanate experiments showed that PWS-HS was an active scavenger of hydroxyl radicals. Furthermore, it was also found to be effective in scavenging nitric oxide and peroxynitrite, well-known cytotoxic species that can oxidize several cellular components such as proteins, lipids and DNA.

  • The method of amino acid sequence determination from the C-terminal amino acid is proposed and mass spectrometric identification of thiohydantoins described previously. In this paper was discussed the fragmentation of thiohydantoin-ring by deutero substitution and model tripeptide have been degraded through three stages each, with interpretable results. The conditions employed in this method are mild enough for biological materials. The main features of the method are the following. 1. Thiohydantoins were formed in a non-aqueous medium a mixture of acetic anhydride, acetic acid and ammonium thiocyanate. 2. Mass sepectra of thiohydantoins derived from 20 amino acids were obtained with a mass spectrometer, JEOL model JMS-06H. 3. Cleavage of peptidyl thiohydantoin was made with an acidic from of a cation-exchange resin. (Amberlite IR-120) 4. Separation of the cleaved thiohydantoin and the parent peptide less one amino acid moiety was made by chromatography on a Sephadex G-10 column. 5. The peptide fraction was concentrated by freezedrying. 6. Thiohydantoin derivative of carboxyl terminal amino acid residue was introduced with a direct inlet probe in methanol solution.

Quantitative Determination of Lipid Hydroperoxide in Human Blood Serum by Ferrothiocyanate Method (Ferrothiocyanate법에 의한 혈청 Lipid Hydroperoxide정량)

  • In order to develope the method of quantitative determination of lipid hydroperoxide in human blood serum, we tried the ferrothiocyanate method to total lipids extracted by Bligh-Dyer method and obtained the results as follows. 1. The maximum absorbance showed at the concentration of Mohr's solution, 0.127M at pH 1.70 and ammonium thiocyanate solution, 3.95M in the ferrothiocyanate method. 2. When hydrogen peroxide, cumene hydroperoxide, and oxidized linoleic acid were added to serum, and extracted them by Bligh-Dyer method to examine the extraction efficiency, we confirmed that cumene hydroperoxide and oxidized linoleic acid were extracted in $CHCI_3$ phase, and hydrogen peroxide in $MeOH-H_2O$ phase, respectively. 3. The concentration of lipid hydroperoxide of total lipids extracted from normal adult serum was $2.0{\times}10^{-5}M$, and increased proportionally the concentration of lipid hydroperoxide by increasing the amount of serum. 4. When we compared the total lipids extracted by Bligh-Dyer method and total lipids extracted after lipoprotein is precipitated by Yagi method in human blood serum, the concentration of lipid hydroperoxide was showed nearly the same value. From our results, we concluded that the concentration of lipid hydroperoxide in human blood serum could be determined quantitatively by ferrothiocyanate method.

IN VITRO DETERMINATION & QUANTIFICATION OF HYDROGEN PEROXIDE PENETRATION DURING NONVITLAL BLEACHING (무수치 표백시술시 치경부를 통한 표백제 누출량의 정량적 측정)

  • It has been demonstrated that intracoronal bleaching of pulpless teeth may result in cervical root resorption. Several authors postulated that bleaching agents such as hydrogen peroxide penetrated through the dentinal tubules to damage the surrounding tissues that cause cervical root resorption. The purpose of this study was to suggest on in vitro model for direct determination of hydrogen peroxide penetration through CEJ during nonvital bleaching. In addition, this model permit the quantification of the amount of hydrogen peroxide penetrated during the procedure. Freshly extracted intact premolars, removed for orthodontic reasons were used. Root canal treatment was performed in each tooth. And then the outer surface and crown portion of the teeth was sealed with wax leaving the CEJ. The prepared teeth mounted on the wax laminates were placed in plastic assay tubes containing 1.5ml bidistilled water with their entire root, including the CEJ, submerged in the solution. The teeth were dividied into four groups. Thermo group : thermocatalytic bleaching with superoxol Walk group: walking bleaching with sodium perborate & superoxol Combi group : combination of thermocatalytic & walking bleaching Dw group : walking bleaching with sodium perborate & water The bleaching procedure was performed three times. The bleaching intervals were at 3 days. The hydrogen peroxide present in the assay system was added to ferrous ammonium sulfate resulting in ferric ion release. Upon the addition of potassium thiocyanate a ferrithiocyanate complex results, which absorbs light at the wavelength of 467nm. The radicular penetration of hydrogen peroxide in the four groups was assessed directly using spectrophotometer. The amount of hydrogen peroxide in the samples tested is determined by comparing them with a standard curve generated by known amounts of hydrogen peroxide. The results were obtained as follows : 1. In all experimental groups except the Dw group showed lower penetration amount in day 4 than day 1, there was statistical importance in the difference (P<0.05). 2. After 3rd treatment, Thermo group showed slightly increased value and narrow distribution. Walk group showed much more penetration amount and widely dispersed value. Value of Combi group showed wide distribution without regard to treatment time, but value of Dw group evenly distributed. 3. Thermo group, Walk group and Dw group showed a tendency of increasing penetration amount with increasing treatment times(P<0.01), but Combi group revealed no statistically important differences. 4. Combi group showed the highest degree of penetration. Walk group showed lower penetration than Combi group. Thermo group & Dw group showed lower than Walk group. 5. Cervical root permeability to hydrogen peroxide varied from 0 to 35 %.
