Development of Capillary Columns for Ion Chromatography (이온 크로마토그래피용 모세관 컬럼의 개발)

  • In this study, capillary columns for ion chromatography were developed to analyze trace amount of ions in samples. When small I.D. capillary columns are used in ion chromatography, the typical flow rate of the mobile phase is $5{\sim}15{\mu}L/min$ and the typical column length is 50~150 mm. The capillary columns were made using RSL-300 fused silica capillary(I.D.: 0.53 mm, O.D.: 0.67 mm) and AG14 column resin(support : polystyrene-divinylbenzene, functional group : alkyl quaternary ammonium). The new conductivity cell and suppressor were also developed and made for capillary column ion chromatography. When several anions (fluoride, nitrite, nitrate, chlorate) were analyzed using these capillary columns, reproducible and good chromatograms were obtained.

Validation of analysis of urinary fluoride by ion selective electrode method (이온선택전극법에 의한 소변 중 불소 이온 분석법 검증)

  • A simple and sensitive analytical method for fluoride in urine by ion selective electrode (ISE) method was presented. Traditional buffer for fluoride determination using ISE is acetate-based one. Researchers have pointed out some drawbacks of the buffer for fluoride ISE analysis, and some other buffers including citrate-ammonium buffer and MES buffer have been studied for accurate determination of fluoride in urine here. These buffers provided promising results in environmental field, and this author focused on overcoming the interference of co-existing aluminium. The results show that MES-CyDTA buffer gave the best recovery with accuracy of 95-97.5% and precision of 1.9-7.9% for reference sample of 1.8-7.8 mg/L fluoride in urine, with smaller amount of samples and shorter analysis time compared with the traditional method which used acetate buffer. The method was applied to field samples, and which showed urinary of $0.98{\pm}0.38mg/g$ creatinine for workers in electric cable manufacturing factory (n=15) and $0.59{\pm}0.30mg/g$ creatinine for non-exposed workers (n=12).

  • This work presents the results of laboratory scale tests of the CARBOFLUOREX (CARBOnate FLUORide EXtraction) process - a novel technology for the recovery of U and Pu from the solid fluorides residue (fluorination ash) of Fluoride Volatility Method (FVM) reprocessing of spent nuclear fuel (SNF). To study the oxidative leaching of U from the fluorination ash (FA) by Na2CO3 or Na2CO3-H2O2 solutions followed by solvent extraction by methyltrioctylammonium carbonate in toluene and purification of U from the fission products (FPs) impurities we used a surrogate of FA consisting of UF4 or UO2F2, and FPs fluorides with stable isotopes of Ce, Zr, Sr, Ba, Cs, Fe, Cr, Ni, La, Nd, Pr, Sm. Purification factors of U from impurities at the solvent extraction refining stage reached the values of 104-105, and up to 106 upon the completion of the processing cycle. Obtained results showed a high efficiency of the CARBOFLUOREX process for recovery and separating of U from FPs contained in FA, which allows completing of the FVM cycle with recovery of U and Pu from hardly processed FA.

Purification and Characterization of Protease from Entomopathogenic Fungus Beauveria bassiana (곤충 병원성 곰팡이 Beauveria bassiana로부터 Protease의 정제와 특성)

  • Extracellular protease (bassiasin I), from the culture filtrate of entomopathogenic fungus Beauveria bassiana ATCC7159, was successively purified by precipitation with ammonium sulfate followed by DEAE-Sephadex A-50, CM-cellulose and Hydroxyapatite column chromatography. A typical procedure provided 41-fold purification with 13.6% yield. The molecular weight of the purified pretense (bassiasin I) was found to be approximately 32,000 by SDS-PAGE. Isoelectric-focusing analysis of the enzyme showed a pI of 9.5. $NH_2-terminal$ sequence of the pretense showed homology with those of the fungal proteases. The enzyme has an optimal pH for activity at 10.5 and is stable over pH 5.0-11.0. The maximum activity of the enzyme was at $60-65^{\circ}C$, and approximately 20% activity remained at $60^{\circ}C$ after 120 min. The pretense was inhibited by phenylmethylsulfonyl fluoride (PMSF) and diisopropyl fluorophosphate (DIPF).

Solution-Processed Fluorine-Doped Indium Gallium Zinc Oxide Channel Layers for Thin-Film Transistors (용액공정용 불소 도핑된 인듐 갈륨 징크 산화물 반도체의 박막 트랜지스터 적용 연구)

  • In this study, we have developed solution-processed, F-doped In-Ga-Zn-O semiconductors and investigated their applications to thin-film transistors. In order for forming the appropriate channel layer, precursor solutions were formulated by dissolving the metal salts in the designated solvent and an additive, ammonium fluoride, was incorporated additionally as a chemical modifier. We have studied thermal and chemical contributions by a thermal annealing and an incorporation of chemical modifier, from which it was revealed that electrical performances of the thin-film transistors comprising the channel layer annealed at a low temperature can be improved significantly along with an addition of ammonium fluoride. As a result, when the 20 mol% fluorine was incorporated into the semiconductor layer, electrical characteristics were accomplished with a field-effect mobility of $1.2cm^2/V{\cdot}sec$ and an $I_{on}/_{off}$ of $7{\times}10^6$.

Synthesis and $^{18}F$ Labelling of Organic Ammonium Salts to New Cardiac Flow Tracer for PET and Their Biodistribution (양전자단층촬영에 이용 가능한 새로운 심근 혈류 추적자 개발; F-18이 표지된 유기암모늄염의 합성과 체내분포에 관한 연구)

  • In order to develop a $^{18}F$-labelled myocardial perfusion agent(flow tracer) for PET, $^{18}F$-labelled organic ammonium cations were synthesized and evaluated in relation to their biodistribution. Five quaternary organic ammonium compounds were labelled with $^{18}F$ in a side chain with moderate to good yields by direct introduction of $^{18}F$-fluoride. Radiochemical yields have been achieved in 30-40min by the precursors (tosylates) in dimethylsulfoxide 15-60% (decay corrected). The reaction was found to be autocatalyzed. A remote controlled procedure was developed in these synthesis. $^{18}F$-Labelling and HPLC-purification of com-pounds needed about 60 min(Yield; 7-20%). Up to now the two compounds N-4-[$^{18}F$]fluorobutyl-pyridinium cation(1) and N, N dibenzyl-4(2-[$^{18}F$]fluoroethyl)piperidinium cation(2) were investigated in relation to their biodistribution in mice. Compound 1 showed at 1 min post injection the high uptake of 19.22% ID/g organ in the myocardium but a following fast decline to 1.12% ID/g organ after 40min. Uptake of compound 2 was after 1min in the heart 5.90% ID/g organ but after 40min at the relative high value of 4.33% ID/g organ. Heart:blood ratio for compound(1) at 1 min was 8.3, at 40 min 2.6 for compound II 2.0(1min) and 15.0(40 min). As data of compound 2 showed greater heart uptake, slower myocardial release, and higher heart: blood ratios, compound 2 is a good candidate for further evaluation.

  • Azeotropic drying process is routinely applied to enhance nucleophilicity of $[^{18}F]$fluoride ion during the nucleophilic production of PET radiotracers; however, the drying process requires usually 15-25 min. Due to the high demand of employing fluorine-18 ($t_{1/2}=109.8min$) in PET radiopharmaceutical production, several research groups have focused on the method development, obviating tedious removal process of the residual target water ($[^{18}O]H_2O$) for $[^{18}F]$fluoride ion complex to be used in radiofluorination. Some development in radiofluorination in a mixed organic solvent system was demonstrated with various aliphatic substrates, but only kryptand as a phase transfer agent was utilized in the reported method. Here, we extend to investigate the development scope of applicability with basic alkyl ammonium salt as a phase transfer agent through the extensive elution efficiency study and radiofluorination outcome for aliphatic radiofluorination.

Biochemical Characterization of a Protease with Fibrinolytic Activity from Maggots of Protaetia brevitarsis (Protaetia brevitarsis의 maggot로부터 fibrinolytic activity을 가진 protease의 생화학적 특성 연구)

  • Fibrin clots remained in blood vessels can be one of the serious factor caused cardiovascular disease, such as ischemia, infarction and necrosis The development of an antithrombotic and thrombolysis solvent is necessary to prevent and treat these diseases. In this study, the fibirinolytic protease was prepared from the maggots of Protaetia brevitarsis using ammonium sulfate fractionation and desalting column. The optimum pH and temperature for the enzyme activity were pH 9.0 and $50^{\circ}C$, respectively. The enzyme activity was relatively stable at pH 7.0-9.0 and temperature below $60^{\circ}C$. The activity of the enzyme was strongly inhibited by phenylmethanesulfonyl fluoride. And the activity of the enzyme was inhibited by $Ca^{2+}\;and\;Zn^{2+}$, but it was not by $Mg^{2+}\;and\;Fe^{2+}$ ions. In these experimental results, we have speculated that the enzyme derived from maggots of Protaetia hrevitarsis is a serine protease with a strong fibrinolytic activity.
