The hepato-protective activity of the GODEX (Hepadif-s capsule) has been studied in the rats against $CCl_4$-ethanol induced liver toxicity. The rats were oral1y treated with $CCl_4$ (corn oil/ $CCl_4$ 1:1, 1 mg/kg). And one week passes, $CCl_4$(0.4 mg/kg) administered two times a week for 7 weeks. The drugs have been administered every two days for 4 weeks after $CCl_4$ injection. The experimental groups have consisted of the GODEX (250 mg/kg), Hepadif (200 mg/kg), DDB complex (DDB 50 mg/kg and garlic oil powder 50 mg/kg), DDB (50 mg/kg), and vehicle control respectively. There was a significant decrement on the serum level of aspartate aminotransferase (AST), alanine aminotransferase (ALT) and total bilirubin in all treated groups. Specially, ALT level of GODEX and Hepadif only treated groups was decreased c1early. Also, serum albumin level was significantly enhanced in GODEX treated group compared with control and DDB treated groups. In histological results, hepatocellular vacuolar degeneration, lobular restructure and necrosis of bile duct were severely showed in control. But other treated groups showed centerilobular degeneration and mild hyper-plasia. Hepadif or DDB has a effects of the recovery on serum parameters and structure ill liver injury. When it was compared GODEX to Hepadif alone or DDB complex or DDB, it suggested to have the best activity of the liver recovery.
Carbon tetrachloride (CCI$_4$) induces the hepatotoxicity due to the reactive free radicals generated by cytochrome P-450 (CYP-450) enzyme, which result in destabilization of cellular membrane. Diltiazem, a calcium channel blocking agent, has been known to suppress the CYP-450 enzyme activities. To study the effect of diltiazem in $CCl_4$-treated rats, we measured the activities of alanine aminotransferase (ALT), aspartate aminotransferase (AST) and glutathione S-transferase (GST), contents of bilirubin, albumin, total protein, cholesterol, triglyceride, blood urea nitrogen, creatinine malondialdehyde and calcium. Also we conducted liver histopathologic examinations. Diltiazem, when administered 1 hour prior to CCI$_4$ treaeent, significantly reduced the activities of ALT and AST, the contents of microsomal malondialdehyde and calcium in liver and microsome as compared with those of $CCl_4$-treated rats. In addition, histopathologic examination showed that diltiazem prevented the development of centrilobular necrosis induced by CCI$_4$ in liver tissue. Our results suggested that diltiazem could inhibit the formation of free radicals and the influx of calcium. Therefore diltiazem may be applied to suppress the liver damage caused by $CCl_4$.
This study was conducted to examine the effects of Opuntia ficus-indica complex (OF) on the lipid metabolism, bile acid in feces, alanine aminotransferase (ALT) activity, aspatate aminotransferase (AST) activity, composition of urine and expression of cholesterol related mRNA in streptozotocin (STZ)-induced diabetic rats. Thirty two male Sprague-Dawley rats were randomly divided into non-diabetic control (NC), diabetic control (DC), diabetic OF of 2% (OF-2) and diabetic OF of 5% (OF-5), then each group was fed for 3 weeks. Plasma total cholesterol, non-esterified fatty acids (NEFA), total cholesterol, low density lipoprotein (LDL), very low density lipoprotein (VLDL) were decreased significantly (p<0.05) in OF-5 group compared to DC, but high density lipoprotein (HDL) was not changed. AST and ALT were also reduced and bile acid excretion was improved. Composition of urine in OF-5 was almost same in NC. The expression of cholesterol $7{\alpha}$-hydroxylase (CYP7A1), 3-hydroxy-3-methylglutaryl-coenzyme A reductase (HMG-CoA-R), Low density lipoprotein receptor (LDL-R) mRNA indicated that feeding OF have the effects of cholesterol decreation in plasma by synthesis of bile acid from cholesterol. These results provide experimental evidence about improved lipid metabolism of the OF feeding in the STZ-induced diabetic rats.
Excessive alcohol use causes oxidative stress in the liver, and antioxidant therapy has been an attractive approach for the treatment of ethanol-induced liver damage. The present study examined the hepatoprotective effect of Sasa quelpaertensis Nakai (Korean name, Jeju-Joritdae) in C57BL/6 mice intoxicated with ethanol. Mice were intraperitoneally administered with ethanol alone, or together with test materials three times at 12-h intervals. At 3 h after the last dosing, hepatotoxicity was assessed based on serum activities of aspartate aminotransferase and alanine aminotransferase, and hepatic contents of thiobarbituric acid-reactive substances and glutathione. Sasa quelpaertensis extract mitigated the acute ethanol hepatotoxicity as effectively as silymarin. Its n-butanol fraction was more active than methylene chloride or aqueous fraction. p-Coumaric acid, a major constituent of S. quelpaertensis, was found to effectively prevent the ethanol-induced hepatotoxicity. These data suggest that S. quelpaertensis and p-coumaric acid could be useful for the prevention of liver disease caused by alcohol abuse.
Five kinds of allylthiopyridazine derivatives were synthesized and their chemoprotective activities examined in rats exposed to aflatoxin $B_1$-toxicant. Rats were pretreated with five allylthiopyridazine derivatives at daily oral doses of 50 mg/kg for 10 consecutive days, and during this period with one or three repeated doses of the potent hepatotoxin, aflatoxin $B_1$. The hepatoprotective effects of the allylthiopyridazine derivatives against aflatoxin $B_1$ (1 mg/kg, three times at intervals of 3 days, i.p., or at 3 mg/kg, once at final days, i.p.) administration were showed the significantly normal as compared with control in body and liver weights. Aspartate aminotransferase and alanine aminotransferase levels were markedly elevated after aflatoxin $B_1$ administration, and pretreatment with allylthiopyridazine derivatives, before aflatoxin $B_1$ administration, resulted in decreased levels of these enzymes. In addition, the allylthiopyridazine derivatives, K6 (3-methoxy-), K8 (3-chloro-), K16 (3-ethoxy-) and K17 (3-n-propoxy), induced elevated hepatic GSH levels. Four kinds of allylthiopyridazine derivatives investigated were effective against aflatoxin $B_1$ -induced hepatotoxicity.
Intestinal hypoganglionosis is a rare innervation disorder that provides numerous nutritional, medical and surgical challenges. In this case report, we present a case of a newborn with intestinal hypoganglionosis leading to intestinal failure and intestinal failure-associated liver disease who responded to $Omegaven^{TM}$, a fat emulsion comprised of omega-3 fatty acids. $Omegaven^{TM}$ has been shown to be beneficial in the management of cholestatic liver injury. Clinical success with $Omegaven^{TM}$ was seen in this patient with a clear decrease in aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase and complete resolution of cholestasis with a direct bilirubin of zero within two weeks of initiation of $Omegaven^{TM}$. No current guidelines for the diagnosis and management of hypoganglionosis are available. We recommend a multidisciplinary approach and the use of novel therapies such as fat emulsions composed of omega-3 fatty acids for improved patient outcomes. Appropriate compassionate use protocols should be obtained from the Food and Drug Administration prior to initiation of $Omegaven^{TM}$.
Electronic cigarettes (e-cigarettes) are becoming increasingly popular worldwide and their cellular effects warrant further evaluation. In this study, we investigated the effects of an e-cigarette cartridge solution on allergen related asthmatic airway inflammation (AI) and airway hyperresponsiveness (AHR), when it is delivered by intratracheal route in mice. Asthmatic AI and AHR were induced by systemic sensitization to ovalbumin (OVA) followed by intratracheal, intraperitoneal, and aerosol allergen challenges in BALB/c mice. The cartridge solution of e-cigarette (containing 16 mg/ml nicotine) was diluted 50 times and $100{\mu}l$ of the diluted solution was intratracheally instilled to OVA-sensitized (OVA-S) mice two times a week for 10 weeks. Long-term e-cigarette inhalation elicited no remarkable changes in the activities of alanine aminotransferase (ALT), aspartate aminotransferase (AST), lactate dehydrogenase enzymes in serum, however, increased infiltration of inflammatory cells including eosinophils, into airways from blood, aggravated the asthmatic AI and AHR, and stimulated the production of cytokines such as interleukin (IL)-4, IL-5 and IL-13, and OVA-specific IgE production. Our data suggest that the inhalation of e-cigarette solutions can function as an important factor to exacerbate the allergy-induced asthma symptoms. Further studies are needed to address the effects of e-cigarette solutions on human health.
There was a mass outbreak of death caused by transport stress and malnutrition in Japanese macaques imported from Japan at a farm stock in Korea, which were diagnosed on the ground of history taking, clinical finding, hematological examination and necropsy. Twenty eight Japanese macaques imported from Japan died suddenly from the 4th day after arrival in a farm. These animals were confined very small cages individually and lose the appetite for 10 days from shipping. Main clinical findings observed from alive Japanese macaques consisted of hypothermia and dehydration. On hematological observation for typical 4 cases showed increased erythrocytes and packed cell volume. On serum biochemical examination for 3 cases showed hypoglycemia, whereas blood urea nitrogen, aspartate aminotransferase and alanine aminotransferase were increased. On necropsy for 3 cases, hemorrhage and congestion were observed on the lung and the walls of stomach and intestine were thinned. Histologically, hemorrhagic pneumonia and enteritis with loss of villi were seen. The death of these animal was caused by poor management during transport, which was resulted in transport stress and malnutrition.
This study was conducted to investigate effects of honeybee venom (Apis mellifera L. BV) on the calving, the growth performance and blood chemistry occurrence of Hanwoo calves. A total of twenty, a 3 day old male experimental calves were allocated into four groups, BV treated (0.05 mg/kg, 0.1 mg/kg and 0.2 mg/kg BV, n=5, respectively) and non-treated (n=5, Control) of Hanwoo calves in Icheon, Kyonggi province. Treatment of BV in calves increased body weight and weight gain during 60 days compared with control group. The concentrations of IgG in blood were significantly increased in BV groups compared with control at 60 days after treatment with BV. Cholesterol and glucose concentrations in BV group were significantly lower with control at 60 days. There were no differences in plasma biochemical components including aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphate (AKLP), total protein, albumin, globuline, total bilirubin, creatinine, blood urea nitrogen (BUN) and $Ca^+$. In conclusion, the BV treatment had effect on growth, the score of the calf scours and blood biochemical profile.
This study was performed to investigate the pathphysiological side effects of chloramphenicol (CAP) on olive flounder, Paralichthys olivaceus. 150 mg/kg/bw/day of CAP was orally administered forcedly to flounder for 4 days and then the flounder was monitored for 11 days. The level of hematocrit was significantly decreased from 2 day to 3 day of CAP treatment in CAP-treated group. The levels of aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP) and gamma glutamyl transferase (GGT) were slightly increased during the treatment of CAP and gradually decreased after the treatment of CAP. The levels of blood urea nitrogen (BUN) and creatinine kinase (CK), although not significant, were slightly increased during the treatment of CAP. The level of lactate dehydrogenase (LDH) was not changed during the treatment of CAP but slightly increased after the treatment of CAP. However, no histopathological changes were observed in various organ after the treatment. From these results, it is suggest that CAP can evoke several side effects on haematopoietic organs in olive flounder.
