To research the effect of chemotaxis of Rhizobia toward the root exudate on nitrogen fixing ability in soybean Rhizobia symbiosis system. Root exudate from seedlings of Glycine max. L was collected aseptic conditions. B. japonicum KCTC 2422 induced the formation of symbiotic nitrogen fixing nodules on the root of soybean plant and possessed motility and chemotaxis toward the 2mM proline. LPN-100 mutant was $Nod^-$, $Che^+$, and LPN-101 was $Che^-$, $Nod^+$ strains. Physiological properties of mutants were similar to parent strain. The crude root exudate was tested for its chemotactic ability using the capillary tube method. Chemotactic responses of RCR 3407 toward crude root exudate were 2.2, 2.6, 2.9, those of KCTC 2422 were 2.3, 2.9, 3.0, respectively. The crude root exudate was fractionated into neutral, cationic and anionic fractions. Chemotactic responses of KCTC 2422 was least with anionic fraction, most with neutral and intermediate with cationic fraction. B. japonicum KCTC 2422 was attracted by carbohydrates, amino acids and carboxylic acid. Carbohydrates and amino acids were good chemoattractants and carboxylic acids were intermediate chemoattractants. The peak concentration was $10^{-3}M$ for ribose, glucose, glutamine, aspartic acid and carboxylic acids, with exception of xylose, arabinose, tryptophan, which elicited maximum responses at $10^{-4}M$. The formation of nodules and nitrogenase activity of soybean inoculated with KCTC 2422 was determined in 7days after inoculation, and those of LPN-101 was detected in 15days after inoculation, but LPN-100 didn't form of nodules in soybean plants.

This research was carried out to investigate the plant growth, inorganic ion and amino acid content characteristics in a halophyte, Suaeda glauca, under different NaCl concentrations for cultivating in the reclaimed land. S. glauca was hydroponically cultivated under 0, 50, 100, 200, 300 and 400 mM NaCl concentrations with Hogland's nutrient solution. To evaluate growth response under different NaCl concentrations, plant height and number of branches, dry weight, Fv/Fm value, and photosynthetic efficiency were investigated. To find out physiological characteristic, inorganic ion contents and amino acids in the plant were evaluated. The optimum concentration of NaCl for plant growth were 50 mM. The plant growth were gradually decreased in the concentration ranged from 100 to 400 mM. As increasing of NaCl concentration, Na ion was increased, but K, Ca, Mg ions were decreased in the plant. The amino acid contents were varied due to NaCl concentrations, but most of amino acids content in total plant was the lowest at 50 mM. Conversely, proline was exceptionally high at 50 mM of NaCl concentration. The Fv/Fm value was the highest at 50 mM of NaCl concentration. From these results, the optimum salt concentration for the growth of S. glauca was 50 mM, but the plant seems to adapt in a variety of salt environments in view of the change of ions and amino acids depending on salt concentration and the maintenance of photosynthetic efficiency even under high salt condition.
The soil management practices before transplanting the ginseng plant were studied with two organic matter sources such as a traditional organic matter (wild grass) and commercial organic fertilizer (byproducts of amino acid fermantation) during the late spring to late autumn. During the soil management practices, the soil received 40kg N/10a from five different combination treatments with above two organic matter sources, a wild grass and a commercial organic fertilizer. After the application of the treatments, the soil were ploughed regularly at the interval of 20 days and the changes of physicochemical properties during the soil management practices were investigated. The next year after soil management practices, ginseng plants were transplanted to each treatment, growth and the content of some organic components of ginseng plant were measured for comparision of the different treatments. 1. The decrease in bulk density observed during the first 40 days of management was considered to be the effect of the improved physical conditions caused by ploughing, The decrease in bulk density observed after 40 days of management was considered to be the effect of organic matter. Similar results were observed in particle density, however porosity increased with time. 2. Soil pH tended to decrease during the first 40 days of management, after which period the pH increased and was stabilized. However, CEC increased with organic matter treatment and the exchangeable $NH^+_4-N$ and $NO^-_3-N$ increased in 20 and 40 days after the management practices, respectively, and after that period it became steady. 3. The decomposition rate of treated organic matter was measured by the incubation test in laboratory conditions. The rate of decomposition was rapid during the first 20 days of management, after which period it showed slight changes. 4. The weight of ginseng root significantly increased in the treatment of 10kg N/10a organic fertilizer and 30kg N/10a wild grass. 5. The saponin content of ginseng root was highest in the 40kg N/10a wild grass treatment. The addition of organic fertilizer at the rate of more than 20kg N/10a caused the decrease in the saponin content.

A field experiment was conducted to evaluate the effect of slow-release fertilizer on the yield and quality of the third-harvest tea leaves. The yield of the third harvested tea leaves was decreased to 5.8-14.4% in slow-release fertilizer block, except to the N $50kg\;10a^{-1}$ ($316kg\;10a^{-1}$), compared to traditional urea treatment ($313kg\;10a^{-1}$). Nitrogen uptake and nitrogen uptake efficiency of slow-release fertilizer was reduced as nitrogen application level increased. The contents of chemical components related to the tea quality such as total-nitrogen, total amino acid, chlorophyll and theanine were somewhat lower in the slow-release fertilizer treatments, except to the treatment of N $50kg\;10a^{-1}$, than those in the traditional urea application, but those of tannin, caffeine and vitamin C were not different among the treatments. In scoring test, apparence and quality of green tea of the slow-release fertility treatments were not different, except to the N $40kg\;10a^{-1}$ treatment, compared to those in the treatment of urea. In conclusion, slow-release fertilizer and conventional urea treatments showed not different in both yield and quality of green tea.
Intensity and stability of the blue color of phospho-molybdate complex in P analysis are known to be influenced by the matrix, reducing regent and acidity of the extractants. Objective of this research was to compare the efficiency of the color-developing reagents for P concentrations in distilled water and extracts of Lancaster, Mehlich II, and Mehlich III methods. Efficiencies on which to base this study were evaluated by the optimum ranges of P, reproducibility of calibration curve and stability of the developed color. Color-developing reagents employed were ammonium molybdate-1,2,4- aminonaphtholsulfonic acid (ANS), ammonium molybdate-ascorbic acid-bismuth subcarbonate (AB), and ammonium molybdate-ascorbic acid-antimony potassium tartarate (AA). The ANS revealed the lowest sensitivity but the widest ranges for P concentrations in color development. On the other hand, the AA bore the narrowest color-developing ranges and its sensitivity was similar to AB. However, at P concentrations lower than $0.5mg\;L^{-1}$, AA was more sensitive than AB. Based on the data on the ranges of calibration curve, stability of color and reproducibility of analytical data. AA reagent was considered to be suitable for the determination of P in distilled water and AB reagent was practically recommendable for soil P analysis in extracts by Lancaster, Mehlich II, and Mehlich III procedures.

A phosphate solubilizing bacterium. strain 60-2G, possessing a strong ability to solubilize insoluble phosphate was isolated from the rhizosphere of grass. On the basis of GC-FAME profile, carbon utilization pattern, and the DNA sequence of a conserved partial 16S rRNA gene, the 60-2G was identified as Enterobacter intermedium. The analysis by HPLC revealed that the strain 60-2G produced mainly gluconic and 2-ketogluconic acids with small amounts of lactic acid in broth culture medium containing hydroxyapatite. During the incubation period of the strain 60-2G in broth culture, pH of the medium decreased upto 3.8 while the soluble phosphate concentration increased. The reversed correlation between pH and soluble phosphate concentration indicated that the solubility of P was due to the produced organic acids. The sequence homology of the deduced amino acids suggested that E. intermedium 60-2G synthesized PQQ which is essential for the oxidation of glucose by glucose dehydrogenase.
The purpose of this experiment is for investigating how much the amino acid liquid fertilizer and composted liquid manure, culture microorganism effect on the breeding of grasses and knowing the extent of the thatch content through an analysis of the soil. For testing about soil chemical, the quality of grasses, and the extent of the thatch content in the soil, we cultivated 6 kinds of microorganisms having the effect of thatch dissolution and sprayed these 6 microorganisms with composted liquid manure and the amino acid liquid fertilizer on the place Creeping bentgrass have planted. This conduction started from June to October, 2012 and 2013 (The interval: 2 weeks). In the result of the turf growth, there is no big difference between soil chemical and trace element. And we can know Leaf Color Index, Chlorophyll Index and Root Length are almost same as among treatment. In conclusion, the mixing fertilization of culture microorganism and composted liquid manure is better effective than the traditional fertilization. And it can be expected the effect of the quality of grass and Thatch decomposition in soil.
BACKGROUND: Caused by cultural environment, the fruit skin stain results in serious damages to pear fruit. Particularly susceptible to this damage, 'Niitaka' pear accounts for 82% of pear cultivation in Korea and many farmers growing the pear trees have suffered economic losses due to fruit skin stain. This study investigated the effect of different treatments of 'Niitaka' pear during growing period on the occurrence of fruit skin stain. METHODS AND RESULTS: The treatments in the field included gibberellin (GA) paste, spraying with amino acid tree fertilizer, functional bagging, and coating of the inner paper bag with agents. The relationships between tree vigor, mineral nutrition concentration and fruit skin stain occurrence were also investigated. The fruit skin stain symptoms occurred from young fruit (May 25) until harvest. There was no exposed fruit flesh. The occurrence of fruit skin stain was significantly reduced in normal tree (shoot length 110 cm), as well as using GA paste treatment, and bagging in calcium and lime sulfur coated bags. However, spraying with amino acid tree fertilizer made no difference in comparison to control. In addition, bags in which the inner paper was coated with lime sulfur and soybean oil resulted in chemical injury to the fruit skin caused by bagging. The K concentration of shoot wood and fruit skin were higher than those of the control. Also, there were lower T-N, K concentration of leaf. CONCLUSION: These results suggest that occurrence of fruit skin stain in 'Niitaka' pear fruits during the growing period can be reduced by GA paste and bagging in calcium and lime sulfur coated bags. The symptoms of chemical injury to the fruit skin caused by bagging in lime sulfur and soybean oil coated inner paper were different compared to skin stain occurring in fruit during the growing period.
In order to investigate the inter-relation with the growth of the barley root and GOT and GPT activities the growth of root and GOT, GPT activities were measured those which have been supplied various nitrogen compounds ($NO_3-N$, $NH_4-N$, Urea, and Amino acid). The results obtained are summarized as follows: 1. Growth of barley root supplied with $NH_4-N$ is generally increased in length and weight compared with that of the root fertilized by $NH_4-N$. 2. The above-mentioned root with $NH_4-N$ is not only decreased in its weight and length but also is apt to inhibited its growth, in compared with the root provided with $NO_3-N$. 3. The activities of GOT and GPT for the root fertilized by $NH_4-N$, the badly grown root is generally increased, while of the root supplied with $NO_3-N$ is decreased compared with that of the root fertilized by $NH_4-N$. 4. The activities of GOT and GPT for the root provided with amino acid known as the considerable growth inhibiting compound for rice is generally decreased, while that of the badly known-grown root is increased. 5. The activities of GOT and GPT in the supernatant fraction of the barley is for the most part, high and low in the mitocondrial fraction.

To examine of possibility protein recycling of shaving scraps contained chrome generated from manufacturing process of leather, the characteristics of hydrolyzed protein that differently treated with MgO as alkaline agent were investigated. In alkaline hydrolysis of saving scraps treated with MgO, MgO had to be treated over 5.0% to maintain over pH 8.0 that is insoluble of chrome. Under the condition of alkaline treated with MgO, the solubility of chrome is low with about 60%. The average molecular weight of hydrolyzed proteins from shaving scraps treated with MgO was about 80~100 KD. The amino acid contents of that were largely collagen proteins such as glycine, alanine and proline, and acidic amino acids such as aspartic acid and glutatamic acid. The contents of Mg, Ca and Na in hydrolyzed protein were too much as liquid fertilizer, and chrome contents was 30~40 ppm that largely decreased in comparing with raw materials (40,000~42,000 ppm).
