  • The production of extended-spectrum ${\beta}$-lactamases ($ESBL_S$) is the main mechanism of bacterial resistance to third-generation cephalosporins and monobactams, whose prevalence varies depending on the different geographical areas. In the last years it has increased notably to the point of being considered a health problem of great importance. The characterization of the ESBLs producing Klebsiella penumoniae strains present in clinical isolates is time-consuming. I describe here the development of a new system, which consists of a multiplex PCR. I found 51 K. pneumoniae strains to be presumptive strains ESBLs producers by clinical and laboratory standards institute (CLSI) guidelines. The double disc synergy test showed 47 positive K. pneumoniae, which were K. pneumoniae isolates. All ESBLs producing K. pneumoniae strains were resistant to antibiotic amikacin, gentamicin and ciprofloxacin. By multiplex PCR analysis, $bla_{TEM}$ gene in 17 strains 44 $bla_{SHV}$ genes and $bla_{CTX}$ genes in 33 strains were identified. In this study, the multiplex polymerase chain reaction (PCR) assay was a good method to detect and differentiate ESBLs producing K. penumoniae strains in clinical isolates.

Identification and Antimicrobial Susceptibility of Bacteria Isolated from Dogs with Chronic Otitis Externa

  • Otitis externa (OE) is an inflammatory disease of the externa auditory meatus that occurs commonly in dogs. Antimicrobial susceptibility tests should be performed in case of chronic OE for successful treatment. In this study, identification and antimicrobial susceptibility test of bacteria isolated from dogs with chronic OE was performed. From 60 dogs with chronic OE, 60 bacterial species were identified. The most frequently identified species were Staphylococcus spp. (51%), followed by Pseudomonas spp. (15%) and Enterococcus spp. (14%). A single bacterial infection and multiple bacterial infections were observed in 67.5% and 32.5%, respectively. Staphylococcus spp. was susceptible to imipenum. Pseudomonas spp. was found to be susceptible to amikacin, cefepime, imipenum and piperacillin-tazobactam. Enterococcus spp. was susceptible to ampicillin-sulbactam, imipenum and piperacillin-tazobactam. Imipenum was highly susceptible antibiotic against both Gram-positive and negative bacteria whereas aztreonam and vancomycin were highly resistant. These results could suggest the optimal choice of antimicrobial agents for canine OE treatment.

병계로부터 분리된 Salmonella pullorum의 생화학적 특성과 항균성 약제에 대한 감수성 (Biological Properties and Antimicrobial Drug Susceptibilities of Salmonella pullorum Isolated from Diseased Chickens)

  • The present study was conducted to investigate biochemical properties, antimicrobial drug susceptibilities and epidemiology of 71 stranins of Salmonella pullorum isolated from about 110 diseased chickens of 23 poultry farms located in Cheongjoo, Cheongweon and Koesan county, Chungbuk province, from August 1991 to March 1993. The isolates were identified as S. pullorum by serological and biochemical means. S. pullorum were mostly isolated in chicken under 3weeks of age, and also isolated in 58, 72 days and 23 weeks of age. According to breeds, most of the cultures were isolated in colored broiler chicken (14 to 23 cases), and also variously isolated in native chicken, white broiler chicken, black bone chicken and laying hen. According to organs of diseased chickens, most of the cultures were isolated in liver (37 to 71 strains), and also variously isolated in spleen, lungs, blood, heart, oviduct and brain. According to media used for primary culture from organs, most of the cultures were isolated purely with SS and BHI medium. The majority of biochemical properties of S. pullorum isolated from diseased chickens were identical to those of the standard strains, but in the properties of rhamnose, and arabinose fermentation, some isolates were negative in spite of positive in those of standard S. pullorum. All the isolates were highly susceptible to colistin, amikacin, kanamycin, gentamicin, carbenicillin, ampicillin, sulfamethxazole, cephalothin, tetracycline and piperacillin regardless of isolated years, but no susceptible to penicillin, erythromycin, vancomycin, tylosin and novobiocin.

닭에서 분리한 Salmonella Gallinarum의 생화학적 특성, 약제감수성과 polymerase chain reaction을 이용한 신속진단 (Biochemical characteristics, antimicrobial susceptibility of Salmonella Gallinarum detection in chickens and rapid diagnosis by polymerase chain reaction)

  • Salmonella enterica serovar Gallinarum is the causative agent of fowl typhoid (FT) and Salmonella enterica serovar Pullorum is pullorum disease (PD), a severe systemic disease of chick and it has the same antigenic fomula, the close relation but distinct pathogen. The traditional bacteriologic and serologic methods routinely used but tedious, time consuming. some of biochemical differences are helpful in differentiating the two organisms, however variation in the characteristics of some strains can be observed. During 2006 to 2008, there was isolated 30 strains. The biochemical characteristics of S. Gallinarum was nonmotile, fermentation of dulcitol, maltose but positive arginine (6.6%), lysine (83.3%) and arabinose (20.0%). The antimicrobial susceptibility test showed 100% sensitive to amikacin, ampicillin, amoxicillin/clavulanic acid and florfenicol, but resistant to penicillin (100%) and erythromycin (60.0%). This PCR method can be applied in the diagnosis between S. Gallinarum and S. Pullorum.

유통되는 쇠고기에서 분리한 대장균의 항생제 내성 조사.연구 (2) (Study on antimicrobial resistance of Escherichia coli isolated from domestic beef on sale (2))

  • In this study, antimicrobial resistance of E. coli isolated from domestic beef on sale in Busan and Gyeongnam province was investigated from March to October 2008. A total of 400 beef samples were collected for the monitoring of antimicrobial resistance, and 39 (9.8%) strains of E. coli were isolated. Antimicrobial resistance test was carried out by agar disc diffusion method with 17 antimicrobials. In general, E. coli isolates showed the highest antimicrobial resistance to tetracycline (85.3%), followed by doxycycline (76.5%), streptomycin (61.8%) and sulfamethoxazole/trimethoprim (61.8%). Then they showed higher resistance to several antimicrobials like kanamycin and neomycin (55.9%). However, They had low antimicrobial resistance to amikacin (8.8%), amoxicillin/clavulanic acid (2.9%). Of 39 isolates, 31 (79.5%) were resistant to more than 2 antimicrobials. Among 17 antimicrobials examined, tetracyclines were the most resistant, followed by aminoglycosides, sulfonamides. The resistance was seemed to be correlated to amounts of antimicrobial use. In the result of this study, we suggest that there be need to regulate the abuse of antimicrobial on food-producing animals in Korea because the concern on antimicrobial resistant is gradually increased worldwide.

전북지역 소 설사유래 병원성대장균 감염실태 조사 (Prevalence of enterovirulent Escherichia coli from diarrhea of cattles in Jeonbuk, Korea)

  • Enterovirulent Escherichia coli are among the most important causes of diarrhea in cattles. Between January and December, 2017, a total of 150 stool specimens from cattles were investigated for enterohaemorrhagic E. coli (EHEC), enteropathogenic E. coli (EPEC), enteroaggregative E. coli (EAEC), enterotoxigenic E. coli (ETEC) and enteroinvasive E. coli (EIEC) using real-time PCR. 131 E. coli were isolated from feces. The most frequently isolated pathotype in feces was EHEC (37 isolates). EPEC, ETEC and EAEC were detected in feces with 14, 7 and 3 respectively. EIEC was not detected. Antimicrobial resistance test was performed by agar disc diffusion method with 14 antimicrobials. Enterovirulent E. coli isolates showed the highest antimicrobial resistance to ampicillin 61.3%, followed by tetracycline 54.5% and streptomycin 45.5%. They had low resistance to amikacin 11.4%. Of 44 isolates, 37 (84.1%) were resistant to more than 2 antimicrobials. futher study a highest antimicrobial susceptibility to trimethoprim/sulfamethoxazole 50.0% and florofenicol 47.7%.

Serovars distribution and antimicrobial resistance patterns of Salmonella spp. isolated from the swine farms and slaughter houses

  • Salmonella spp. is an important pathogen to both public and swine industry. The aim of this study was to investigate the distribution of Salmonella serovar and antibiotics susceptibility of the isolates from Korean swine producing systems. A total of 63 (5.28%) Salmonella spp. was isolated from 1,194 samples (977 fecal materials and 67 organ samples). The predominant Salmonella (S.) enterica serotype and serovar was group B (69.8%) and S. Typhimurium (47.6%), S. Derby (20.6%) and S. Heidelberg (1.6%). But S. Cholerasuis which is characterized host specific by septicemia and enteritis to pigs was not isolated. Antimicrobial susceptibility of the isolates varies as follows: Norfloxacine (75%), Ciprofloxacin (67.5%), Amikacin (60%), Colistin (60%), Enrofloxacin (55%). All of isolates were resistant to Erythromycin, Penicillin, Tetracycline and Lincomycin. The results of this study provided useful information regarding antimicrobial susceptibility and resistance patterns to treat salmonellosis and to prevent emergence of multidrug resistance Salmonella.

Vibrio damsela의 분리연구 (Studies on the Isolation of Vibrio damsela)

  • Authors studied on the isolation of V. damsela from sea water, fish and shellfish at the Keoje Hae keumkang on the southern sea and at Hongdo island and Heucksan island on the western sea of Korea from May to September in 1986. Authors investigated for the isolated strains to bacteriological identification, hemolysis about various erythrocytes and antibiotic susceptibilities. The results obtained were as follows: 1. V. damsela was isolated 14 strains from total 383 specimens; 233 cases of sea water, 40 cases of fish and 110 cases of shellfish, respectively. Eight strains were isolated from sea water and 6 strains were isolated from shellfish. 2. The biochemical characteristics which differentiate it from other Vibrio species were indole negative, ornithine negative, Voges-Proskauer positive, arginine positive, galactose positive, glucose positive, maltose positive, mannose positive, trehalose positive, and growth in nutrient broth with 1% to 6% NaCl. 3. On hemolysis reaction on blood agar media using human, rabbit and guinea pig erythrocytes, human erythrocytes were 11 strain positive, rabbit erythrocytes were 12 strain positive and guinea pig erythrocytes were 13 strain positive. 4. Senistivity test using with chemotherapeutic agents of "BioLab" Microbial Sensitivity Test Discs were generally sensitived to amikacin, ampicillin, cephalothin, chloramphenicol, clindamycin, erythromycin, gentamycin, kanamycin, methicillin, penicillin, streptomycin, tetracycline and tobramycin, respectively, but were resistant to lincomycin.

Prevalence of Listerial spp. over Commerical Frozen and Refrigerated Foods at the Supermarket Level

  • As a part of investigation for listeriosis, we attmepted iolationof Listeria spp. from commerical frozen and refrigerated foods at the supermarket level. Seven strains of Listeria spp. were isolate from 6 samples (7.15) among 84 samples of frozen foods, and ten strains of Listeria spp. were also isolated from 8 samples (7.6%)_ among 105 samples of refrigerated foods at the supermarkets in Pusan area. From a total of 189 commerical foods, the number of isolated Listeria spp. and ratio were 6 strains(3.25) of L. grayi, one strain (0.5%) of L. welshimeri, 6 strains (3.2%) of L. innocua and 4 strains (2.15) of L monocytogenes. Listeria spp. isolates except L. monocytogenes did not show $\beta$-hemolysis on blood agar and positive reaction in CAMP test with Staphyloccccus aureus. In the antibiotic susceptibility,most isolates of Listeria spp. were susceptible to 12 antibiotics such as ampicillin , cephalotin, penicillin, amikacin, gentamicin, erythromycin, kanamycin, vancomycin, tobramycin, carbenicillin , tetracycline and trimethoprim /sulfamethoxiazole. Four strains of L. monocytogenes were susceptible to ntibiotics used in this study except nitrofurantio. The serotype of 3 strains and one strain of L. monocytogenes were classified into Listeria O serotype 1 and 4, respectively.

  • Identification of antibiotic resistant bacteria isolated from animals is necessary for seeking a proper treatment and for preventing the spread of the bacteria among animals. Methicillin-resistant Staphylococcus pseudintermedius (MRSP) is of worldwide concern in veterinary medicine. This study was conducted to investigate the antimicrobial susceptibility of Staphylococcus pseudintermedius (S. pseudintermedius) isolated from dogs and cats. Out of the total number of 150 specimens, 35 isolates (23.3%) were identified as S. pseudintermedius when tested by MALDI-TOF MS (Matrix Assisted Laser Desorption Ionization-Time of Flight Mass Spectroscopy). Among them, 5 isolates (14.3%) were the case of MRSP. In the results of the antimicrobial susceptibility test, the isolates of S. pseudintermedius were susceptible to amikacin (100%), cephalothin (100%), vancomycin (100%), amoxicillin (85.7%), oxacillin (85.7%), enrofloxacin (82.8%), chloramphenicol (80%). On the other hand, they were resistant to penicillin (71.4%), tetracycline (48.6%), ampicillin (37.1%), kanamycin (31.5%), erythromycin (31.4%), respectively.
