B13 analogues are being considered as therapeutic agents for cancer cells, since B13 is a ceramide analogue and inhibits ceramidase to promote apoptosis in cancer cells. B13 sulfonamides are assumed to have biological activity similar to B13, since they are made by bioisosterically substituting the carboxyl moiety of B13 with sulfone group. Twenty B13 sulfonamides were evaluated for their in vitro cytotoxicities against human colon cancer HT-29 and lung cancer A549 cell lines using MTT assays. Replacement of the amide group with a sulfonamide group increased cytotoxicity in both cancer cell lines. The sulfonamides with long alkyl chains exhibited activities two to three times more potent than that of B13 and compound (15) had the most potent activity with $IC_{50}$ values of 27 and $28.7{\mu}M$ for HT-29 and A549, respectively. The comparative molecular field analysis (CoMFA) and comparative molecular similarity indices analysis (CoMSIA) were used to carry out QSAR molecular modeling of these compounds. The predictive CoMSIA models for HT-29 and A549 gave cross-validated q2 values of 0.703 and 0.830, respectively. From graphical analysis of these models, we suppose that the stereochemistry of 1,3-propandiol is not important for activity and that introduction of a sulfonamide group and long alkyl chains into B13 can increase cytotoxicity.
Apicularen A (Api A) was recently isolated from Chondromyces sp. as a potent antitumor agent. Because of its unique chemical structure, a macrolide with a highly unsaturated amide side chain, and potent growth inhibitory effect in various cancer cell lines, Api A is currently in clinical trial for cancer therapy. In the present study, the effect of Api A on in vitro angiogenesis of bovine aortic endothelial cells (BAECS) was investigated. Api A potently inhibited the proliferation of BAECS in a dose-dependent manner. Treatment of the endothelial cells with up to 10 ng/ml of the compound did not show any cytotoxicity. In addition, it inhibited basic fibroblast growth factor (bFGF)-induced invasion and capillary tube formation of BAECS at concentrations of 2-5 ng/ml. These results, therefore, demonstrate that Apl A is a novel antiangiogenic agent and may suppress the growth of tumors, at least in part, by the inhibition of neovascularization.
본 연구는 희소 한우인 제주흑우의 정액 동결을 위하여 동결보호제로서 glycerol과 amide 계열의 동결보호제인 5%의 DMA, DMF 그리고 7%의 MF를 이용하여 동결 융해 후 정자의 운동성, 생존율, 정자막 온전성 및 정자의 첨체 양상의 변화 조사를 위하여 수행하였다. 제주흑우의 정액을 동결 한 결과 glycerol과 DMF를 사용하였을 때 운동성은 $64.00{\pm}9.62$와 $59.00{\pm}5.48$로 DMA와 MF에 비해 유의적으로 높았으며(p<0.05), 생존율 역시 glycero과 DMF 처리구에서 $58.25{\pm}6.63$과 $53.05{\pm}3.77$로 유의적으로 높게 나타났다(p<0.05). 그러나 glycerol과 DMF 처리구 간의 유의적 차이는 나타나지 않았다. 정자막 온전성 검사(HOST)에 있어서도 역시 glycerol과 DMF 처리에서 정자 미부의 팽창 비율 역시 $45.12{\pm}25.08$과 $44.95{\pm}8.58$로 유의적으로 높았다(p<0.05). 정자의 첨체막 양상 변화에 있어 F pattern의 비율이 MF 처리시 다른 처리구에 비하여 유의적으로 낮았으며(p<0.05), B pattern의 비율은 DMA, DMF 및 MF의 사용시 glycerol에 비하여 유의적으로 증가하였다(p<0.05). AR pattern의 비율에 있어서는 DMF를 제외한 amide의 사용은 F pattern의 감소와 조기 첨체반응에 의한 유의적인 AR pattern의 증가를 볼 수 있었다(p<0.0.5). 그러나 DMF의 사용은 glycerol 보다 AR pattern의 비율이 유의적으로 낮았다(p<0.05). 동결 융해 후 시간에 따른 정자의 생존율과 정자미부 팽창 비율에 있어서 융해 초기에는 glycerol에서 DMF 보다 높은 생존율과 정자 미부 팽창율을 나타냈으나, 1시간 이후부터는 DMF에서 glycerol 보다 높은 생존율과 정자 미부 팽창율을 나타내었다. 이러한 결과는 희소 가축의 생식세포 보존 및 유전자원 확립을 위한 중요한 자료가 될 것이며, 제주흑우 및 희소 가축의 안정적이고 효율적인 동결 정액 제조 연구에 있어 중요한 정보를 제공할 것으로 평가된다.
신규 미백 소재 개발을 위하여 tyrosinase의 활성을 억제하는 천연물과, alpha-melanocyte stimulating hormone ($\alpha$-MSH)을 저해하는 펩타이드를 결합한 소재를 고체상 합성법으로 제조하였다[1,2]. Tyrosinase 활성 억제 효능이 있는 천연유래물질을 기존 연구를 바탕으로 선정하였으며, 이중에서 상업화 된 물질 17종에 대해 Mushroom Tyrosinase 활성 억제 효과를 측정하여, 그 중 caffeic acid, coumaric acid를 선별하였다. 펩타이드는 $\alpha$-MSH 분비를 억제하며, tyrosinase활성을 억제하는 것으로 알려진 melanostatin을 선정하였으며[3-5], PLG-$NH_2$ (Proline-Leucine-Glycine-$NH_2$) 중에서 유도체화 수율의 저하 원인으로 예상되는 Proline (Pro)의 서열을 다른 아미노산으로 변경하면서 선별된 천연물인 coumaric acid와 caffeic acid에 도입하였다. 또한 최종물질의 원가를 고려하여 acid-amide 형태를 acid형태로 전환한 유도체를 합성하였다. 이들 펩타이드 유도체들에 대해서 B16F1 melanoma cell에서의 멜라닌 생성 억제 효능을 평가하여 유도체화된 물질이 기존의 천연유래물질이나 펩타이드에 비하여 높은 저해효과를 가지는 것을 확인하였다.
두개의 sulphato로 다리걸친 착물 $[Cr_2(NH_2)_2(H_2O)_2(SO_4)_2]{\cdot}2H_2O$, 말단 3개의 isocyanato $[Cr(NCO)_3(H_2O)]{\cdot}3H_2O$ 화합물과 갈철광 $[FeO(OH)]{\cdot}0.2H_2O$ 화합물들은 각각의 $Cr_2(SO_4)_3{\cdot}xH_2O, Cr(CH_3COO)_3, Fe_2(SO_4)_3$과 $80^{\circ}C$, 수용액에서 요소와 반응으로 합성되었다. 생성물들의 IR 스펙트럼에서 요소(urea)의 띠가 나타나지 않지만, 결합된 아마이드(amide), 물, 연결된 sulphato와 isocyanato 그룹에 대해서 특징적인 띠를 보인다. 착물들에 대한 열무게분석(TG)과 시차열분석법(DTA) 측정을 기록하였다. 얻어진 데이터들은 예상했던 구조화 잘 일치한다. 형성에 대한 설명과 화합물의 열역학적 분해에 대한 일반적인 메커니즘을 제시하였다.
일련의 alkyl-3,4-dihydroxybenzoate (A)와 N-alkyl-3,4-dihydroxybenzamide (B) 유도체들의 치환기(R1 및 R2) 변화에 따른 피부 감작성과의 관계를 HQSTR 방법으로 분석하였다. 유도된 피부 감작성에 관한 HQSTR 모델은 매우 양호한 예측성(cross-validated $r^2_{cv}.,\;q^2=0.744$)과 적합성(non-cross-validated, $r^2_{ncv}$. =0.978)을 나타내었다. 이들 두 화합물은 멜라닌 생성 저해 활성이 클수록 피부 감작성이 낮은 반비례적인 경향을 보였으며 R1-치환기 사슬 중 C1 ${\~}$ C3 원자 부분은 피부 감작성에 기여하지 않는 경향을 나타내었다. 따라서 ester (A)는 amide (B)보다 피부 감작성이 낮으나(AB) 특징을 나타내므로 미백제의 활성 성분으로서 매우 이상적인 화합물임을 알았다.
현재 상업적으로 널리 사용 되여 지고 있는 항암제인 hydroxy urea, 6-mercaptopurine monohydrate, cytosine arabinoside, cyclophosphamide monohydrate 그리고 uracil를 3-(triethoxysilyl) propyl isocyanate 와 반응시켜 항암제가 붙어있는 3-(triethoxysilyl)propyl amide (compound I)을 합성한후 물과 가수분해 반응시켜 항암제가 결합된 silica 나노입자(10 nm~micronparticles)를 만들 수있었다. Silyl isocyanate 유도체들은 물과 반응하여 유기물질-silica 나노입자가 포함된 $-CH_2-CH_2-CH_2-NH-COOH$ 그룹이나 혹은 온도와 용매등 반응조건에 따라서 decarboxylated 된 propylamine 그룹이 생성되었다. 생체외 시험에서 항암제가 결합된 silica 나노입자는 종양 세포 제거에 효과적이고 정상세포에 거의 영향을 미치지 않는 것으로 나타났다. 그리고 악성 종양인 폐, 난소, 악성 흑색종, 중추신경계(CNS)와 결장 종양 세포가 이 연구에 사용되었다. 현재의 연구에서 전달매체 로서 silica는 silation 반응으로 손쉽게 나노입자를 얻을 수 있으므로 본 연구에 쉽게 이용 할 수 있다. 결과로부터 이 기술은 보다 부작용이 적은 생체 의약품에 적합한 carrier nanoparticles에 적용 될 수 있을 것으로 판단된다.

The reaction of titanium tetraisopropoxide with 2 equiv of N,N-diethyl acetoacetamide affords Ti($O^iPr)_2(CH_3COCHCONEt_2)_2$ (1) as colorless crystals in 80% yield. Compound 1 is characterized by spectroscopic (Mass and $^1H/^{13}C$ NMR) and microanalytical data. Molecular structure of 1 has been determined by a single crystal X-ray diffraction study, which reveals that it is a monomeric, cis-diisopropoxide and contains a six coordinate Ti(IV) atom with a cis($CONEt_2$), trans($COCH_3$) configuration (1a) in a distorted octahedral environment. Variable-temperature $^1H$ NMR spectra of 1 indicate that it exists as an equilibrium mixture of cis, trans (1a) and cis, cis (1b) isomers in a 0.57 : 0.43 ratio at -20$^{\circ}C$ in toluene-$d_8$ solution. Thermal properties of 1 as a MOCVD precursor for titanium dioxide films have been evaluated by thermal gravimetric analysis and vapor pressure measurement. Thin films of pure anatase titanium dioxide (after annealing above 500$^{\circ}C$ under oxygen) have been grown on Si(100) with precursor 1 in the substrate temperature range of 350- 500$^{\circ}$ using a bubbler-based MOCVD method.
생분해성 고분자와 수화젤의 약물 전달 시스템과 조직 공학을 포함하는 다양한 생의학적인 응용이 점차 확대되고 있다. 하이드록실 pendant group을 갖고 있는 폴리아미노산의 하나인 PHEA는 뛰어난 생분해성과 우수한 생체적합성을 갖는 고분자로 알려져 있다 PHEA 및 그 유도고분자를 제조하고 이를 다양한 생의학적인 응용에 적용하는 연구가 진행되어 왔다. 본 연구에서는 유기용매에서 가교제인 HMDI를 사용하여 PHEA 수화젤을 제조하였다. PHEA는 아스팔트산의 열 축중합에 의해 합성한 polysuccinimide와 ethanolamine과의 고분자 반응으로부터 제조하였다. 제조한 수화젤에 대한 서로 다른 수용액과 pH용액에서의 팽윤 거동을 조사하였고, SEM을 통한 수화젤의 모폴로지와 가수분해 거동을 관찰하였다.
본 연구에서는 UV-VIS spectrophotometer를 이용한 total carotenoids, flavonoids, phenolics 함량 데이터와 FT-IR 스펙트럼 데이터를 다변량통계분석법을 통하여 기능성 성분 함량이 높은 아프리칸 얌 고속 선발 시스템을 구축하였다. 62개 아프리칸 얌의 total carotenoids 함량은 $0.01-0.91{\mu}g{\cdot}g^{-1}$ dry wt 나타냈다. Total flavonoids와 phenolics 함량은 $12.9-229.0{\mu}g{\cdot}g^{-1}$ dry wt와 $0.29-5.2mg{\cdot}g^{-1}$ dry wt로 각각 나타났다. 아프리칸 얌은 FT-IR 스펙트럼상의 1700-1500, 1500-1300, $1,100-950cm^{-1}$, 부위에서 중요한 스펙트럼 변화가 나타났다. 이 부위는 각각 amide I과 II을 포함하는 아미노산 및 단백질계열의 화합물, phosphodiester group을 포함한 핵산 및 인지질 그리고 단당류나 복합 다당류를 포함하는 carbohydrates 계열의 화합물들의 질적, 양적 정보를 반영하는 부위이다. PCA 분석과 PLS-DA 분석에서 62개 아프리칸 얌은 유연성이 높은 종으로 3개의 그룹을 형성하였다. 아프리칸 얌의 FT-IR 스펙트럼 데이터와 UV-VIS spectrophotometer을 이용한 total carotenoids, flavonoids, phenolics 함량 데이터 간에 PLS regression 분석하였다. Total carotenoids, flavonoids, phenolics 함량 성분의 실측 값과 예측 값간에 상관계수($R^2$)가 각각 0.83, 0.86, 0.72로 나타났다. 이 결과, 아프리칸 얌으로부터 FT-IR 스펙트럼을 이용한 total carotenoids, flavonoids, phenolics 함량 예측이 가능하였다. 본 연구에서 확립된 대사체 수준에서 아프리칸 얌의 유용 기능성 성분 함량 예측 모델링을 통해 품종, 계통의 신속한 선발 수단으로 활용이 가능할 것으로 예상된다.
