본 논문에서는 디지털라디오와 관련하여 동일계열 기술인 DRM과 DRM+ 전송방식을 기반으로 AM과 FM의 디지털방송 추진의 가능성과 필요성을 제시한다. 주파수 활용에서 IN-BAND와 OUT-OF-BAND 양쪽 모두 이용이 가능한 DRM+의 특징을 이용하여 FM밴드와 인접한 LOW-VHF 대역 TV채널 5번과 6번 주파수를 OUT-OF-BAND방식으로 디지털라디오방송을 추진할 때 기존 FM방송 신호와 충돌과 혼신이 없고, 새로운 라디오영역으로 확보 가능하며, 우수한 신호전달 능력이 있는지 확인하고자 하였다. 특히 IN-BAND방식 Hybrid 모드에서 SFN을 이용한 다수의 송신기 사용이 어려운 문제점을 언급하였다. 그와 반대로 DRM+에 의한 OUT-OF-BAND 방식은 다수의 송신기 배치가 가능하며, 기존 FM송신기 및 안테나와 콤바인이 쉽고, 사업자별로 출력지정과 권역설정이 자유로우며, AM/FM 통합 디지털라디오 제작과 보급에 유리하여 디지털라디오 추진에 적합한 방안이 될 수 있음을 제안하였다.
Ballast water is widely recognized as a serious environmental problem due to the risk of introducing non-indigenous aquatic species. In this study we aimed to investigate measures which can minimize the transfer of aquatic organisms from ballast water. Securing more reliable technologies to determine the viability of aquatic organisms is an important initiative in ballast water management systems. To evaluate the viability of marine phytoplankton, we designed the staining methods of fluorescein diacetate (FDA) and Calcein-AM assay on each target species belonging to different groups, such as bacillariphyceae, dinophyceae, raphidophyceae, chrysophyceae, haptophyceae and chlorophyceae. The FDA method, which is based on measurements of cell esterase activity using a fluorimetric stain, was the best dye for determining live cells of almost all phytoplankton species, except several diatoms tested in this study. On the other hand, although fluorescence of Calcein-AM was very clear for a comparatively longer time, green fluorescence per cell volume was lacking in most of the tested species. According to the Flow CAM method, which is a continuous imaging technique designed to characterize particles, green fluorescence values of stained cells by FDA were significantly higher than those of Calcein-AM treatments and control, implying that the Flow CAM using FDA assay could be adapted as an important tool for distinguishing living cells from dead cells. Our results suggest that the FDA and Calcein-AM methods can be adapted for use on phytoplankton, though species-specific characters are greatly different from one organism to another.
Daily milk yields on test days were estimated using morning or afternoon partial milk yields collected by official agencies and the accuracy of the estimates was determined. Test-day data for milk yields consisted of 3,156,734 records of AM/PM partial milking measurements of 255,437 milking Holstein cows from 3,708 farms collected from December 2008 to April 2013. A linear regression model (LRM) was applied to estimate daily milk yields using alternate AM/PM milk yield records within lactation stages, milking intervals, and parities on every daily milk yield. The alternate statistical approach was a non-linear hierarchical model (NHM) in which Brody's growth function was implemented by reflecting an animal's physiological milk production cycle. When compared with LRM, daily milk yields predicted by the NHM were assumed to be functionally related to day in milk (or lactation) stage, milking intervals, and partial milk yields. Since the results were in terms of accuracies based on comparisons of different statistical models, accuracies of estimates of daily milk yields by NHM were close to those determined by the LRM. The average of these accuracies was 0.94 for AM partial milk yields and 0.93 for PM partial milk yields for first calving cows. However, the accuracies of AM/PM milk yield estimations from cows under a calving stage higher than the first parity were 0.96 and 0.95, respectively. Correlations between the estimated daily milk yields and the actual daily milk yields ranged from 0.96~0.98. These accuracies were lower for unbalanced AM/PM milking intervals and the first calving cows. Overall, prediction of daily milk yields by NHM would be more appropriate than by LRM due to its flexibility under different milk yield-related circumstances, which provides an idea of the functional relationship between milking intervals and days in milk with daily milk yields from statistical viewpoints.
Objectives : The purpose of this study is to find parameters to compare the effects of Sa-Am acupuncture with large intestine tonifying and sedating on radial pulse in healthy subjects. Methods : Sixty healthy subjects participated in this study, divided into large intestine tonifying acupuncture group, large intestine sedating acupuncture group and control group. Radial pulse was measured by 3 dimensional pulse imaging system(DMP-3000) before, right after, 30 minutes after and 60 minutes after acupuncture at Cun, Guan, and Chi in the acupuncture group and in the control group at the same time points. Results : 1. Angle of main peak and magnitude of fourier component significantly changed by integrated analysis of Cun, Guan and Chi. 2. Amplitude of H1, Pulse Power volume / min, elasticity, AIx / HR, Frequency and magnitude of fourier component significantly changed by analysis of Cun, Guan, and Chi. Conclusions : The effect of Sa-Am acupuncture with large intestine tonifying and sedating in healthy human may be observed on time, amplitude, pulse area, augmentation index and fourier components parameters. The parameters analysed in this study may be used to differentiate the effects between Sa-Am acupuncture with large intestine tonifying and sedating on radial pulse. Further studies on the effects of Sa-Am acupuncture using radial pulse are needed.
The multicellular layers(MCL) of human cancer cells is a three dimensional(3D) in vitro model for human solid tumors which has been used primarily for the assessment of avascular penetration of anti-cancer drugs. For anti-cancer drugs with penetration problem, MCL represents a good experimental model that can provide clinically relevant data. Calcein-AM is a fluorescent dye that demonstrates the cellular vitality in a graded manner in cancer cell culture system. In the present study, we evaluated the use of calcein-AM for determination of anti-proliferative activity of anti-cancer agents in MCL model of DLD-1 human colorectal cancer cells. Optical sectioning of confocal imaging was compromised with photonic attenuation and penetration barrier in the deep layers of MCL. By contrast, fluorescent measurement on the cryo-sections provided a feasible alternative. Cold pre-incubation did not enhance the calcein-AM distribution to a significant degree in MCL of DLD-1 cells. However, the simultaneous determination of drug penetration and cellular vitality appeared to be possible in drug treated MCL. In conclusion, these data suggest that calcein-AM can be used for the simultaneous determination of drug-induced anti-proliferative effect and drug penetration in MCL model.
Objectives: The purpose of this study is to investigate the anti-cancer effects of different fractions of Astragalus membranaceus (AM) in human non-small cell lung cancer (NSCLC) cells. Methods: We isolated hexane, ethyl acetate, and butanol fractions from crude ethanol extract of AM. The cell death was examined by MTT assay and trypan blue exclusion assay. Apoptosis was detected by DAPI staining, annexin V-PI double staining and cell cycle analysis. The expression of apoptosis-related proteins and mitogen-activated protein kinases (MAPKs) was examined by western blot. Results: Among various fractions of AM, the ethyl acetate fraction of AM (EAM) showed the strongest cytotoxic effect in NSCLC cells. EAM reduced the cell proliferation in a time- and dose-dependent manner in NSCLC cells. In addition, EAM induced the chromatin condensation, and increased the population of sub-G1 phase and annexin V-positive cells in a time-dependent manner, indicating that EAM induced apoptosis in NSCLC cells. Consistently, EAM enhanced the expression of cleaved caspase-8 and -9, and induced the accumulation of cleaved- poly (ADP-ribose) polymerase (PARP). Among MAPK proteins, only ERK was dephosphorylated by EAM, suggesting that ERK might be related with EAM-induced apoptosis. Conclusion: Our results clearly demonstrate that EAM exhibited anti-cancer effects in NSCLC cells by induction of apoptosis. We provide a valuable evidence which suggests that AM could be a desirable therapeutic option for treatment of NSCLC.
본 논문은 진행파관 증폭기의 비선형성을 줄이기 위해 새로운 전치 왜곡 방법을 제안하였다. 진행파관 증폭기의 비선형 전달 특성은 carrier complex power series를 이용하여 해석하였고, 이를 선형화하기 위한 전치 왜곡 선형화기의 전달 특성을 inverse complex power series로 해석하였다. Schottky 다이오드와 전송 선로 종단에 부착된 부하 저항을 반사형 구조로 만듦으로 전치 왜곡기의 비선형 왜곡 특성을 얻을 수 있었다. 제안된 선형화기를 적용하여 K-대역용 진행파관 증폭기의 AM-to-AM 및 AM-to-PM 특성 을 각각 -5.825 dB과 $-37.321^{\circ}$에서 0.786 dB과 $6.742^{\circ}$로 개선시켰다.
In this study we developed the transposon-mediated shuttle vector 'Hanpvid', which composed of HaNPV (Heliothis armigera nuclear polyhedrosis virus) genomic DNA and a transposon cassette from Bacmid of Bac-to-Bac system. Hanpvid replicates in E. coli in the same way as Bacmid and retains infective function in cotton bollworm cells (Hz-AM1). Using Hanpvid we constructed a recombinant virus, which could infect Hz-AM1 cells and generate recombinant HaNPV (rHa-Bar) containing the barnase gene, a ribonuclease gene from Bacillus amyloliquefaciens. Since the expression vector carrying barnase gene cannot replicate in the absence of barstar, a specific inhibitor of barnase, we constructed a new cotton bollworm cell line (AM1-NB) using the marker rescue method. In AM1-NB barstar was integrated into the cellular chromosome to sustain the replication of rHa-Bar. To screen out recombinant HaNPV for potential use as biopesticide, Hz-AM1 and AM1-NB cell lines were infected with rHa-Bar, respectively. The results obtained indicate that Viral progenies in AM1-NB were 23 and 160 times greater than those in Hz-AM1 48 h and 72 h after infection, respectively. With additional insertion of the polyhedron gene from AcNPV (Autographa californica nuclear polyhedrosis virus) into the Hanpvid genome, rHa-Bar regained the polyhedron phenotype and its pest-killing rate greatly improved. Toxic analysis showed that the lethal dosages ($LD_{50}$) and the lethal time(s) ($LT_{50}$) of rHa-Bar were reduced by 20% and 30%, respectively, compared to wt-HaNPV in the third instar larvae of cotton bollworm. This study shows that in AM1-NB barnase can be effectively produced and used as pest-killing agent for the biological control of cotton pests.
