Quantitative Analysis and Preformulation of Extracts from Alnus Japonica

  • Alnus japonica has been known to exert antioxidative, anti-inflammatory, anti-cancer and immune response inhibitory effects. The aim of this study was to figure out the characteristics of extracts obtained with different extraction solvent such as water, 100% ethanol, 70% ethanol or 70% methanol because characteristic components such as oregonin and hirsutanone extracted from Alnus japonica might be essential for the biological activity. For this purpose, oregonin and hirsutanon of four extracts, index ingredient of Alnus japonica, were analyzed with HPLC and physicochemical studies such as SEM, particle size and zeta potential were conducted. In cell cytotoxicity study, extract of water showed the highest cytotoxicity among four extracts. In case of oregonin, 70% MeOH and water extracts showed high contents and in case of hirsutanone, all extracts showed similar contents except 70% EtOH extracts. The extract of 70% MeOH from Alnus japonica for both oregonin and hirsutanone appeared to have the highest content. Both oregonin and hirsutanone extracted from Alnus japonica using 70% methanol showed stability in pH 1.2.

  • Genus Alnus are deciduous board-leaved trees or shrubs found in the damp place and mountain and more than 17 species are growing in Korea. The bark of Alnus japonica has been used for the treatment of fever, hemorrhage and diarrehea in oriental traditional medicine. Quantitative analysis of diarylheptanoids which were characteristic components of Alnus species (A. japonica, A. hirsuta, and A. hirsuta var. sibirica), has been conducted by HPLC. The results showed that these plants were rich source of diarylheptanoids and the amount of oregonin and hirsutanonol which were major diarylheptanoids showed seasonal variation.

  • Soil contamination by metals is of particular interest, given that their retention times within the profile can be indefinite. Thus, phytostabilization can be viewed as a means of limiting metal toxicity in soils. Due to their ability to grow on contaminated soils, alders have repeatedly been used as key species in phytostabilization efforts. Alder ability to grow on contaminated sites stems, in part, from its association with microbial endophytes. This work emphasizes the fungal endophytes populations associated with Alnus incana ssp. rugosa and Alnus alnobetula ssp. crispa (previously A. viridis ssp. crispa) under a phytostabilization angle. Fungal endophytes were isolated from alder trees that were growing on or near disturbed environments; their tolerances to Cu, Ni, Zn, and As, and acidic pH (4.3, 3, and 2) were subsequently assessed. Cryptosporiopsis spp. and Rhizoscyphus spp. were identified as fungal endophytes of Alnus for the first time. When used as inoculants for alder, some isolates promoted plant growth, while others apparently presented antagonistic relationships with the host plant. This study reports the first step in finding the right fungal endophytic partners for two species of alder used in phytostabilization of metal-contaminated mining sites.

Chemotaxonomic Significance of Oregonin in Alnus japonica Native to Baekdudaegan Mountain Range in Korea

  • Alnus japonica is indigenous to korea and usually grows in wet lowlands and damp regions in the mountains. Oregonin, is known as a representative compound of the diarylheptanoid mainly found in Alnus species and has been reported to have anti-inflammatory and antioxidant effects. In this experiment, we conducted a study on the presence of orogonin in the native Alnus japonica from the Baekdudaegan mountain range in Korea. We collected a total of 30 samples from Chuncheon, Yangyang, and Jeongseon and conducted experiments. In the HPLC analysis, we confirmed that oregonin was detected in all samples. After the quantitative analysis, samples with the highest content from each region were selected and LC-MS/MS experiments were performed. As a result, the same molecular weight as oregonin, 477.2 m/z, was confirmed in all samples.

물오리나무의 뿌리혹으로부터 Frankia 공생균주의 분이 (Isolation of Symbiotic Frankia Strain from the Root Nodule of Alnus hirsuta)

  • An endophyte was isolated from the root nodule of alnus hirsuta. The isolated endophyte was identified as a Frankia sp. through morphological characteristics. Their infectivity and effectivity were confirmed by nitrogen-fixing root nodules induced on inoculated Alnus seedlings. Reisolated endophyte from the induced nodule showed identical morphological characteristics as the first isolate, showing the nodule was induced by the first isolate. Consequently, the first isolate was confirmed as a true symbiont of Almus hirsuta root nodule. The isolate was designated as a Frankia SNU 014201 strain.

  • Phytochemical examination of the leaves of Alnus hirsuta, one of the indigenous species grows in Korea, has led to the isolation of one flavan 3-ol and two flavonol glycosides. Structures of these compounds are identified as (-)-epicatechin (1), quercetin $3-O-{\beta}-D-galactopyranoside$ (2), quercetin $3-O-{\beta}-D-galactopyranoside$ (3) by means of physico-chemical and spectral evidences.

방크스소나무 및 산오리나무의 발아와 묘목의 생장에 미치는 Mn과 Al의 영향 (The Effect of Manganese and Aluminium on the Germination of the Seeds of Pinus banksiana and Alnus tinctoria)

  • The aim of present study is to elucidate the effects of manganese and aluminium ions on the germination and seedling growth of Pinus banksiana and Alnus tinctoria and to estimate the limiting level of those ions in the cultivated soils. The rate of germination of Pinus banksiana was increased under the relatively low concentration of Mn and Al (100 p.p.m), however, the germinating activity was not affected. High concentration over 110 p.p.m. of Mn and Al was related to the reduced percentage of germination and seedling growth of Pinus banksiana and Alnus tinctoria. Soils sampled from various parts of Korea contained Mn as much as high level that would be harmful for the seedling growth of Pinus banksiana and Alnus tinctoria.

오리나무 수피엑스의 위염 및 위궤양에 대한 효과 (The Effect of Alnus japonica Cortex Extract on Gastric Lesion and Ulcer of Rats)

  • Alnus japonica cortex has been used as antidiarrhea, antihemorrhage and the remedy of indigestion. This study was performed to investigate the effectiveness of the methanol extracts of the Alnus japonica cortex on the gastric lesion and ulcer. The methanol extract was fractionated with hexane, chloroform and butanol, followed by bioassay on antigastritic and antiulcer activity. The methanol extract showed low acute toxicity with minimum lethal dose of more than 5000 mg/kg, p.o. in mice. The chloroform and the butanol fraction reduced gastric lesion in HCI. ethanol induced gastritic model. On gastric secretion in pylorus ligated rat, the hexane and chloroform fraction decreased the volume and acidity. The butanol fraction had significant inhibitory effects on aspirin and Shay's ulcer. The butanol fraction showed a tendency to inhibit the decrease of mucin secretion due to ingestion of absolute ethanol.

  • In this study, when the forest fire occurred, in order to estimate greenhouse gas emissions, tree glow in Gangwon Youndong area, Juglans mandshurica, Alnus japonica, Acer palmatum of carbon dioxide and carbon monoxide emissions were about. Water content were measured before the experiment, Juglans mandshurica 196.24%, Alnus japonica 169.17% Acer palmatum 210.10% moisture content showed a big difference, Living leaves of Acer palmatum were a lot of moisture. Also, 50g weight of carbon dioxide on the Juglans mandshurica 53.3644g, Alnus japonica 49.4256g, was released about Acer palmatum 51.3394g, Juglans mandshurica living leaves were the most carbon dioxide emissions. Carbon monoxide emissions result, About weight 50g Juglans mandshurica 1.5329g, Alnus japonica 1.7189g, 2.5002g about Acer palmatum was released, Acer palmatum living leaves were the most carbon monoxide emissions.

  • Alnus sibirica Fisch. ex Turcz.(AS) geographically distributes in Korea, Japan, Northeast China and Russia. The bark of Alnus species have been used as traditional medicine for the treatment of allergic skin diseases. The content evaluation of compounds which were isolated from the barks of A. sibirica processed enzymatic hydrolysis(EAS); hirsutanonol(1), 5-O-methylhirsutanonol(2), rubranol(3), hirsutenone(4) and muricarpone B(5) was conducted using HPLC. Consequently, in EAS, the content of glycoside was decreased considerably. But the aglycones(1, 3 and 4) were increased highly and 2, 5 were formed newly.
