The Study of DNA markers to identify of Allium sativum L. (한약재 마늘(Allium sativum L.)의 식별을 위한 유전자 감식연구)

  • Objectives : This study was carried out to identify DNA markers of "Allium sativum" be circulated from Korea and China, which is difficult to discriminate from morphological characters because of fragmental materials of bulb. That is, all these studies focused on the discrimination of Allium sativum L. But these day, Chinese A. sativum was in circulated Korean A. sativum in Korean medicine markets. Therefore, the purpose of our study was to develop molecular markers for discrimination between Korean A. sativum and imports from China. Methods : Materials were collected randomly from a markets in Korea and China and be analyzed with matK, ndhF and trnL-F regions of chloroplast DNA (cpDNA). We collected 45 A. sativum individuals from Korean and Chinese medicine markets, in 2013. Results : As a results, matK and ndhF regions of cpDNA was shown to be identify, Species that grow from warm place and cold place can divide as five SNP (Single nucleotide polymorphisms) markers in matK and ndhF genes. Also, in trnL-F regions, found one SNP that can divide Korean A. sativum and Chinese A. sativum. Conclusions : From the analysis of matK and ndhF regions of cpDNA, we presumed that three markers of cpDNA were found by useful marker that can distinguish Korean, Chinese, Warm place type, and Cold place type. Individual differences of Korean and Chinese was thought that appear in geographical difference and genetic difference by environment for long hour even if same species.

Antimycotic Activity of Allium Sativum Against Beauveria Bassiana, Pathogenic Fungus of White Muscardine Disease in Silkworm, Bombyx mori L. (Lepidoptera: Bombycidae)

  • White Muscardine is the most common fungal disease of silkworm, Bombyx mori L. caused by the pathogenic fungus, Beauveria bassiana (Balsamo) Vuillemin. In the present investigation, an attempt has been made to screen locally available medicinal/ weed plants against Beauveria bassiana. Among the plant extracts (PE) tested, 5% aqueous crude extract of the bulb of Allium sativum (Garlic) has been found to be most effective against Beauveria bassiana. The radial growth of Beauveria bassiana in vitro was inhibited to the tune of 54.9% in aqueous extract and 54.4% in ethanolic extract of Allium sativum and correspondingly mycelial dry weight gave rise to 110.7 mg and 108.7 mg against 201.7 mg in control 15 days post treatment. Similarly, silkworm larvae topically inoculated with the Beauveria bassiana conidia ($1.8{\times}10^6/ml$) registered survival up to 53.0% against 0.0% in control after treatment with aqueous extract of Allium sativum. Simultaneously, as a preventive measure, silkworm larvae were put to rear in conidia contaminated seat paper instantly treated with aqueous extract of Allium sativum that also increased survival up to 61.0% against 4.6% in control. It is also observed that the plant extract is absolutely innocuous to silkworm.

Cloning and Characterization of Highly Repetitive Sequences in the Genome of Allium sativum L. (마늘(Allium sativum L.) 게놈의 고반복서열의 분이와 특성 조사)

  • We have studied the DNA of Allium sativum L. with respect to highly repetitive sequences. Fast reassociated DNA fragments expected to be highly repetitive sequences based on $C_{o}t$ curve were isolated and characterized. Their copy numbers were approximately $10^{5}~10^{7}$ per haploid genome. Nucleotide sequences analysis of six candidates reveals that their G/C content were low, 25-40% and typical patterns of repeating sequences exist. Repeat sequences were used as probes to access restriction fragment length polymorphism (RFLP) of genomic DNAs of four local clones, Tanyang, Mungyong, So san, and Uisong. The hybridization pattern were very similar among these four local clones.clones.

  • Exposure of freshly laid eggs of Exorista bombycis (Louis) to volatiles emanating from bulbs of Allium sativum L. for different durations resulted in signigicant reduction in their hatchability. Maggots hatched from the eggs exposured for 64 h and 72 h were failed to emerge from host larvae. The duration of development stages of E. bombycis was prolonged besides reduction in rate of pupation and adult emergence as the egg exposure period increased. The findings are interpreted as the chronic effects of volatiles of garlic affecting maggots following developmental defects sustainable during embryonic development. The known major chemical components of A. sativum such as allicin, thioacrolein, ajoune, 2-propene sulfenic acid, 2-propene thiol and propylene were presumed to be responsible for the adverse consequences reported in this paper.

Purification and Properties of $\beta$-Fructofuranosidase (마늘(Allium sativum L.)로부터 추출한 $\beta$-Frucdtofuranosidase의 정제 및 성질)

  • A $\beta$-fructofuranosidase of garlic (Allium sativum L. ; Seosan) was purified by ammonium sulfate fractionation and gel filtration chromatography with a recovery of 8.3%. The molecular mass of the purified enzyme was estimated to be 79kDa by SDS-PAGE, which revealed two subunits of 41kDa and 38kDa. Maximum enzyme activity was observed at pH 5.0 and 40$^\circ C$ and the enzyme was stable over the pH range of 4.0-6.0 and below 50$^\circ C$. The Km value for sucrose was 15.5mM and the enzyme activity was significantly inhibited by bivalent metal ions(Hg$^2+$, Cu$^2+$, Cd$^2+$) and EDTA.

The garlic is a kind of vegetable propagated vegetatively through cloves.   • Gartic bulblets were planted to investigate the effect of some different cultivating conditions on the growth and bulb formation of the garlic plants (Allium sativum L.) Two different conditions namely perfect and imperect aerobic condition, and 3 different fertilizer levels was made. The split plot design was adopted for this experiment. 1) For the growth rate, under the imperfect aerobic condition, the plant height was more increased than that of perfect aerobic condition no relation to the fertilizer levels. 2) With respect to the fresh weight of garlic, the similar tendency to the growth rate was observed, but dry weight was did not. 3) The uptake of phosphorus was found to be increased in the imperfect condition. It could there be concluded that imperfect aerobic condition seems to be much favorable condition than the perfect aerobic condition to the development of garlic bulbs.

Determination of Dimethyl Disulfide, Diallyl Disulfide, and Diallyl Trisulfide in Biopesticides Containing Allium Sativum Extract by Gas Chromatography

  • BACKGROUND: Garlic (Allium sativum) extract has been allowed as commercial biopesticide material for pesticidal activity in the Environmentally-friendly Agriculture Promotion Act. Nine commercial biopesticides containing A. sativum extract have been marketed in Korea. However, the analytical method of the active substances in these materials has not been studied. METHODS AND RESULTS: Cartridge clean-up method for the determination of dimethyl disulfide(DMDS), diallyl disulfide(DADS), and diallyl trisulfide(DATS) in biopesticides containing A. sativum extract was developed and validated by gas chromatography(GC). The clean-up method was optimized using hydrophilic lipophilic balance (HLB) solid phase extraction(SPE) cartridges for the bioactive sulfides in biopesticides containing A. sativum extract, and the eluate was analyzed to quantify the DMDS, DADS, and DATS using the GC. The developed method was validated, and the LOQ and recovery rates of DMDS, DADS, and DATS were 0.226, 0.063, and $0.051mg\;L^{-1}$ and 80.6, 84.8, and 73.1%, respectively. From the nine commercial biopesticide samples, contents of DMDS, DADS, and DATS were analyzed using the developed method and results showed $2.3mg\;L^{-1}$, respectively. CONCLUSION: The developed method could be used in determining the quality of biopesticides for the manufacture of commercial biopesticides containing A. sativum extract.

  • Cultivated garlic, Allium sativum L. is economically important for leaves and bulbs, which historically were used in Korea for spices and condiments of Korean food as well as medicine crops. This experiment was carried out to investigate the effect of development and differentiation on culture of A. sativum (cv: white 6) by explant position, hormone composition and sucrose concentration in culture media. Culture method was investigated to induce shoot primordium. Culture efficiency was better with lower tissue of foliage leaf in explant position and on the medium with NAA 0.02 + BAP 1.0 mg/l in hormone composition than any other. Precocious shoot and callus were induced from shoot apex. Shoot was efficiently differentiated on 4,000 mg/l sucrose with increasing concentration of BAP. Shoot primordium was also induced with liquid rotary culture by histological observation. Rhizoid was induced from callus tissue cluster on medium with NAA 0.02 + BAP 2.0 mg/l.

Stem Rot of Garlic (Allium sativum) Caused by Sclerotium rolfsii

  • Stem rot disease was found in garlic (Allium sativum L.) cultivated from 2008 to 2010 in the vegetable gardens of some farmers in Geumsan-myon, Jinju City, Gyeongnam province in Korea. The initial symptoms of the disease were typical water-soaked spots, which progressed to rotting, wilting, blighting, and eventually death. White mycelial mats had spread over the lesions near the soil line, and sclerotia had formed over the mycelial mats on the stem. The sclerotia were globoid in shape, 1~3 mm in size, and tan to brown in color. The optimum temperature for growth and sclerotia formation on potato dextrose agar (PDA) medium was $30^{\circ}C$. The diameter of the hyphae ranged from approximately 4 to $8\;{\mu}m$. Typical clamp connection structures were observed in the hyphae of the fungus, which was grown on PDA medium for 4 days. On the basis of the mycological characteristics and pathogenicity of the fungus on the host plants, the causal agent was identified as Sclerotium rolfsii Saccardo. This is the first report of stem rot disease in garlic caused by S. rolfsii in Korea.
