Combined Application Effects of Arbuscular Mycorrhizal Fungi and Biochar on the Rhizosphere Fungal Community of Allium fistulosum L.

  • Arbuscular mycorrhizal fungi (AMF) are widespread soil endophytic fungi, forming mutualistic relationships with the vast majority of land plants. Biochar (BC) has been reported to improve soil fertility and promote plant growth. However, limited studies are available concerning the combined effects of AMF and BC on soil community structure and plant growth. In this work, a pot experiment was designed to investigate the effects of AMF and BC on the rhizosphere microbial community of Allium fistulosum L. Using Illumina high-throughput sequencing, we showed that inoculation of AMF and BC had a significant impact on soil microbial community composition, diversity, and versatility. Increases were observed in both plant growth (the plant height by 8.6%, shoot fresh weight by 12.1%) and root morphological traits (average diameter by 20.5%). The phylogenetic tree also showed differences in the fungal community composition in A. fistulosum. In addition, Linear discriminant analysis (LDA) effect size (LEfSe) analysis revealed that 16 biomarkers were detected in the control (CK) and AMF treatment, while only 3 were detected in the AMF + BC treatment. Molecular ecological network analysis showed that the AMF + BC treatment group had a more complex network of fungal communities, as evidenced by higher average connectivity. The functional composition spectrum showed significant differences in the functional distribution of soil microbial communities among different fungal genera. The structural equation model (SEM) confirmed that AMF could improve the microbial multifunctionality by regulating the rhizosphere fungal diversity and soil properties. Our findings provide new information on the effects of AMF and biochar on plants and soil microbial communities.

  • Water extracts of six plants, such as Allium sativum, A. cepa, Zingiber officinale, Platycodon grandiflorum, Oenanthe javanica, and Capsella brusapastoris, were tested in vitro for inhibitory activity against mycelial growth of anthracnose fungi, Colletotrichum gloeosporioides, C. dematium, and C. coccodes. Among the plant extracts, an Allium sativum extract has good inhibitory effects in all the fungi. Four phytohormones namely, IAA(indole-3-acetic acid), NAA(a-Naphthyl acetic acid), 2,4-D(2,4-Dichloro phenoxy acetic acid) and BAP(Benzyl adenine purine) were used to find out the role over mycelial growth of these fungi. All the concentrations of BAP have good inhibitory effect against mycelial growth of these fungi than that of other tested plant hormones.

  • 양파 중간모본 웅성불임 계통 '원예30001'은 2000년 Ginque 자식 집단에서 웅성불임개체를 선발하여 고정종 자식계통 YG-1-1을 교배하여 2002년에 유지계통을 선발하고 2009년까지 4회 여교잡을 하여 계통의 안정성을 확인한 후 MMS17로 명명하여 2009년에 직무육성신품종 심사를 거쳐 2010년에 국립종자원에 '원예30001'로 명명하여 품종보호 출원하였다. '원예30001'은 웅성불임(CMS-S)계통으로 초형은 반개장형이며 도복기는 5월 20일이다. 초장은 57 cm, 엽수 8매, 엽초경은 15.9 cm로 지상부의 생육이 왕성하며 구중은 260 g, 구형지수 89인 원형 황색 양파계통이다. 노균병에 중간 정도의 저항성이 있으나 내한성과 저장성은 강하다. 모구의 채종 특성으로 개화기는 5월 23일이며 구당 화경수는 6개, 화경 장은 135 cm이며 화구 폭은 84 mm이며 100% 웅성불임 이다.

Sequence Homologies of GTP-binding Domains of Rab and Rho between Plants and Yeast/Animals Suggest Structural and Functional Similarities

  • Small GTP-binding proteins are divided into three major group: Ras, Rho and Ypt/Rab. They have the conserved regions designed G1 to G5 that are critical in GDP/GTP exchange, GTP-induced conformational change and GTP hydrolysis. We isolated and characterized genomic DNA or cDNAfragments encoding G1 to G3 domains of small GTP-binding protein Rab and Rho from several plant species using two different PCR-based cloning strategies. Seven rab DNA fragments were isolated from 4 different plants, mung-bean, tobacco, rice and pepper using two degenerate primers corresponding to the GTP-binding domain G1 and G3 in small GTP-binding proteins. The amino acid sequences among these rab DNA fragments and other known small GTP-binding proteins shows that they belong to the Ypt/Rab family. Six rho DNA fragments were isolated from 5 different plants, mung-bean, rice, Arabidopsis, Allium and Gonyaulax using the nested PCR method that involves four degenerate primers corresponding to the GTP-binding domain G1, G3 and G4. The rho DNA fragments cloned show more than 90% homology to each other. Sequence comparison between plant and other known Rho family genes suggests that they are closely related (67 to 82% amino acid identity). Sequence analysis and southern blot analysis of rab and rho in mung-bean suggest than thses genes are encoded by multigene family in mung-bean.

Determination of Cytoplasmic Male Sterile Factors in Onion Plants (Allium cepa L.) Using PCR-RFLP and SNP Markers

  • We have developed a polymerase chain reactionrestriction fragment length polymorphism (PCR-RFLP) marker that can distinguish male-fertile (N) and male-sterile (S) cytoplasm in onions. The PCR-RFLP marker was located in a chloroplast psbA gene amplicon. Digesting the amplicons from different cytoplasm-containing varieties with the restriction enzyme MspI revealed that N-cytoplasm plants have a functional MspI site (CCGG), whereas the S-cytoplasm plants has a substitution in that site (CTGG), and thus no MspI target. The results obtained using this PCR-RFLP marker to distinguish between cytoplasmic male sterile factors in 35 onion varieties corresponded with those using a CMS-specific sequence-characterized amplified region (SCAR) marker. Moreover, the PCR-RFLP marker can identify N- ot S-cytoplasms in DNA sample mixtures in which they are in up to a 10-fold minority, indicating that use of the marker has high diagnostic precision. We also demonstrated the usefulness of the SNP detected in the psbA gene for high-throughput discrimination of CMS factors using Real-time PCR and a TaqMan probe assay.

국내 자생식물 20종의 혈관이완 효능에 대한 실험연구 (Research on vasorelaxant effects of 20 Korean native plants)

  • Objectives : The objective of present study was to investigate the vasorelaxant effects of 20 Korean native plants on isolated rat thoracic aorta precontracted with phenylephrine (PE). Methods : Dried 20 plant materials were extracted 3 times with water, ethanol, or methanol for 3h in the reflux apparatus at 70 ± 5℃. Male SD rats were anesthetized by ether inhalation, and their aorta rings were isolated and placed in 10 ㎖ Krebs Henseleit (KH) buffer. While using an isolated organ-chamber technique, the aorta rings were maintained by bubbling with a gas mixture of 95% O2-5% CO2 at 37℃. Changes in isometric tension of aorta rings were recorded via isometric transducers connected to a Powerlab Data Acquisition System. Results : Among the 20 native plants, Chrysanthemum indicum L. flower, Nelumbo nucifera Gaertn. rhizome, Schisandra chinensis (Turcz.) Baill. fruit, Anthriscus sylvestris (L.) Hoffm. root, Corydalis turtschaninovii Besser tuber, Corydalis decumbens (Thunb.) Pers. tuber, and Dolichos lablab L. seed showed significant vasorelaxant effect on the contraction of aorta rings induced by PE. In contrast, Mertensia maritima subsp. asiatica Takeda whole plant, Ajuga decumbens Thunb. whole plant, Trigonotis peduncularis (Trevis.) Benth. ex Baker & S.Moore whole plant, Dioscorea quinquelobate Thunb. rhizome, Allium microdictyon Prokh aerial part, Momordica charantia L. fruit, Carthamus tinctorius L. flower, and Clematis terniflora DC. root constricted more the aorta rings precontracted by PE Conclusion : These results suggest that the possibility as useful herbal resources for the development of functional foods or medicines for hypertension treatment.

  • 본 실험은 양액내 NO$_3$$^{-}$-N : NH$_4$$^{+}$-N 비율이 잎파(Allium fistulosum L.) 생육 및 품질에 미치는 영향을 알아보고자 실시하였으며, 그 결과는 다음과 같다. 양액내 pH는 NO$_3$$^{-}$-N : NH$_4$$^{+}$-N 비율이 9:1인 처리구에서는 상승하였고, NO$_3$$^{-}$-N : NH$_4$$^{+}$-N 비율이 1:1, 1:3인 처리구에서는 하강하는 경향이 있었으나, 3:1 처리구에서는 안정적인 PH를 유지하였다. 외관상 생육은 NO$_3$$^{-}$-N : NH$_4$$^{+}$-N 비율이 9:1인 처리구에서 가장 양호하였으며, NO$_3$$^{-}$-N : NH$_4$$^{+}$-N 비율이 1 :3인 처리구에서 가장 저조하였다. 식물체내의 NO$_3$$^{-}$-N의 함량은 양액내의 NO$_3$$^{-}$-N 비율이 높을수록 증가하였다. Pyruvic acid 함량은 생육이 양호한 NO$_3$$^{-}$-N : NH$_4$$^{+}$-N 비율이 9:1, 3:1인 처리구에서 높았으며, 1:3처리구에서는 낮았다.

봉래산(부산)의 식물상 (The Flora of Mt. Bongrae)

  • The purpose of this study was to investigate the flora of Mt. Bongrae in Busan from February 2004 to May 2005. The flora in the surveyed area were identified 291 taxa that belonged to 82 families, 201 genera, 251 species, 35 varieties, and 5 forms. Among them, evergreen broad trees were identified 21 taxa, naturalized plants 30 taxa, especial plants decided by Ministry of Environment 24 taxa. By the vegetation of each slope, in northeast slope, it was dominated such as Pinus thunbergii community, Chamaecyparis obtusa community, Amorhpa fruticosa community, Trachelospermum asiaticum var. intermedium-Hedera rhombea community, north-northeast slope, it was distributed both Pinus thunbergii community and board-leaved tree community. In the southwest slope of coast site, it was presented plantation plants, according to altitude distributed such as natural vegetation, evergreen broad trees, warm-temperate plants. In southeast slope, it was to represent secondary vegetation such as Alnus firma community as a forest fire. Underside was mainly to search herbs such as naturalized plants and cruciferous family as a vegetable garden. In northwest slope, it was searched trees such as Pinus thunbergii community, Lindera obtusiloba, Styrax japonca, Prunus sargentii, Aralia elata, Zanthoxylum piperitum, Akebia quinata and herb layer such as Osmunda japonica, Allium thunbergii, Liriope platyphylla, Corydalis ochotensis, Scilla scilloides, Viola orientalis. Around the top, it was distributed such as Rhododendron mucronulatum-Rhododendron schlippenbachii community, Carpinus coreana, Viburnum erosum, Sorbus alnifolia.

산채류로부터 혈소판응집 억제물질의 검색 (Screening of Inhibitors of Platelet Aggregation from Edible Plants)

  • 산채류를 비롯한 식물자원으로부터 항혈전 성분을 탐색하기 위하여 전국일원의 농산시장 및 농촌지도소, 평창산채시험장과 한국인삼연초연구원의 실험포장으로부터 약 160점의 시료를 수집하였으며, modified smear method를 적용하여 그 methanol 추출물의 혈소판응집 저해활성을 비교하였다. Platelet rich plasma 즉 혈소판 혈장을 이용하는 modified smear method는 응집유도물질로서 ADP와 collagen을 사용하였을때 electrical impedence method와 높은 상관성을 나타내었고, 재현성도 비교적 우수하여 정제하지 않은 식물체의 조추출물의 항혈전 활성을 효율적으로 검색할 수 있었다. 참취, 개미취, 곤달비, 산마늘, 산도라지, 영양부추, 산뽕, 쇠비름, 생열귀등이 ADP와 collagen 모두 혈소판응집 저해활성을 나타내었으며, 곰취, 사철쑥, 씀바귀, 민들레, 산지치, 갯완두, 참대등은 collagen을 사용한 경우에는 저해활성이 다소 낮게 검출되었다. 반면에 참나물, 옥잠, 냉이, 메밀, 매실, 복분자, 해란초등은 오히려 혈소판응집 촉진효과를 나타내었다. 한편 DPPH법으로 활성산소 억제율을 비교한 결과 산채류중에는 고사리, 참취, 곰취, 구절초, 사철쑥, 고려엉겅퀴, 달개비, 냉이 그리고 생약재중에서는 적작약, 음양곽, 복분자등이 항산화활성이 높았다.

청량산(봉화군, 안동시)의 관속식물상 (A Flora of Vascular Plants in Mt. Cheongnyangsan (Bonghwa-gun, Andong-si))

  • 연구는 경북 봉화군 명호면과 재산면, 안동시 도산면과 예안면에 걸쳐 위치하는 청량산(870 m)에 자생하는 관속식물을 밝히고 주요 자원식물들의 분포를 파악하였다. 2006년 3월부터 2014년 10월까지 총 16회에 걸쳐 조사된 식물은 총 97과 330속 541종 3아종 61변종 9품종 614분류군으로 확인되었다. 이 중 한국특산식물은 21분류군, 산림청 지정 희귀식물 중 취약종은 8분류군, 약관심종은 5분류군, 환경부 지정 식물구계학적 특정식물 중 V등급이 2분류군, IV등급이 10분류군, III등급이 18분류군으로 확인되었다. 또한 청량산에 분포하는 분포특이종으로 세뿔투구꽃, 산토끼꽃 등 4종이 조사되었다. 이들은 유용성에 따라 구분하면 식용 234종류, 사료용 213종류, 약용 174종류, 관상용 62종류, 목재용 16종류, 염료용 12종류, 섬유용 11종류의 순으로 나타났고, 이 외에 용도를 알 수 없는 식물이 167종(27.2%)로 확인되었다. 귀화식물은 32분류군으로 귀화율은 5.3%, 도시화지수는 10.0%로 나타났다.
