Broad Spectrum Antibacterial Activity of Allium cepa, Allium roseum, Trigonella foenum graecum and Curcuma domestica

  • In many parts of the world Allium cepa, Allium roseum, Trigonella foenum graecum and Curcuma domestica are extensively used as food and are popular in herbal medicine. The four were screened against 26 pathogens and all exhibited broad-spectrum anti-bacterial activity. The aqueous as well as fractionated methanol extract of Allium cepa and A. roseum demonstrated broader level of activity against most of the organisms. On the other hand the unfractionated methanol extracts as well as the fractions of both Trigonella foenum graecum and Curcuma domestica showed broad spectrum of activity. Fractionation was found to improve their level of activity. In both cases the ethyl acetate fractions exhibited higher level of activity. All the materials tested were inactive against any of the four moulds. Allium cepa, Allium roseum, Trigonella foenum graecum and Curcuma domestica are proposed as non toxic, safe, broad spectrum antibacterial agents.

  • Identification of compatible parental line is of great importance in introduction of useful characters to onion breeding program, beyond the severe hybridization barrier. Phylogenic analysis of Allium section Cepa was conducted through PCR by URPs, repeated sequences of A. fistulosum, and microsatellite markers. Totally 76 accessions originated from 21 countries were clustered into five groups at a 0.84-similarity level: group I;A. cepa and its wild relatives and A. cepa ssp. ascalonicum, group II; A. cepa ssp. wakegii, A. cepa ssp. proliferum and Samcheung-pa group III; A. fistulosum and A. altaicum, group IV; A. galanthum, group V; Soeckkori-pa. Samcheung-pa and Soekkori-pa, Korean local varieties, shared band type of both Cepa group and Altaicum group, indicating that those are derived from interspecific hybridization between A. fistulosum and A. cepa.

A Study of Glutathione S-transferase Inhibitors obtained from Allium cepa var. cepa Extract (양파 추출물에서의 글루타티온 전달효소 활성 저해제에 관한 연구)

  • In this study, an ethanol extract being obtained from Allium cepa var. cepa examins the inhibitory effects on the glutathione S-transferase and the separation had been done by silica-gel column chromatography using various eluents, such as ethyl acetate, methanol, and 50% methanol. A volume of column fraction was 50ml and evaporation has been performde by the rotary evaporator under reduced pressure. Each fraction is being examined by thin layer chromatography and the UV spectrum at 365 nm was used to investigate separation patterns of spots on thin layer chromatography. When the eluent was changed, the spot patterns showed another different pattern on thin layer chromatography, so on. Fractions showing similar pattern are combined and eventually, three fractions are obtained. Each fraction is testified to examine the inhibition effects on glutathione S-transferase. All of these showed inhibition effects on glutathione S-transferase. The GC-MS shows that each fraction contains more than 2 compounds.

  • The investigation was aimed to reduce the period of bed log through the protection of harmful fungi and to know the effects of physiological activating substances on the yield increase of Lentinus edodes. Mycelial growth was evaluated both the bed logs and physiological activating substances, respectively. I percent extract of Allium cepa and malt as a physiological activating substance was highly effective in mycelial growth of Lentinus edodes. Better effects among the bed logs by the application fo physiological activating substances observed in both Quercus serrata and Carpinus laxiflora. This results suggested that supplement of physiological activating substances to the bed log would be beneficial for the production of Lentinus edodes.

Wound Healing Effects of Allium Cepa L. in Dogs (개에서 Allium cepa L.의 창상치유효과)

  • This study was performed to investigate the wound healing effects of Allium cepa L. on Staphylococcus intermedius contaminated wound. 4 full-thickness skin wounds ($2\;cm{\times}2\;cm$) were made in the back and Staphylococcus intermedius was inoculated in seven dogs. In each dog, the wounds were treated with onion extract (experimental group) and normal saline (control group), respectively. The wound contraction rate of the experimental group was higher than that of the control group during the experimental period. The occupation rate showed that the wounds of the experimental groups contracted more vigorously than that of the control group in the early period of wound healing. The mean value of tensile strength on the 22 day after wounding was $4,741{\pm}848\;g/cm^2$ in the experimental group and $2,400{\pm}397\;g/cm^2$ in the control group. The number of S. intermedius in the experimental groups was decreased more rapidly than that of the control group. These results suggest that application of activated onion extract in like full-thickness skin wounds promote wound healing.

  • To use onion(Allium cepa L.) residue as raw materials of food product, yields of onion-juice according to various extracting methods and optimal extracting condition of quercetin and its related glycosides were carried out. Onion juices gained by the methods of pressing, rotary crushing, freeze pressing and enzyme treatment. The yield by the method of enzyme treatment was higher than others. The yields of juice from fresh onion and heat-treated onion(8$0^{\circ}C$/10min) by pressing were 66% and 83%, respectively. Ethanol extraction of onion was efficient at 75$^{\circ}C$ and acetic acid extraction was proper at 3% concentration for 3 hours at 11$0^{\circ}C$. The onion extract was fractionated in the order of hexane, chloroform, ethylacetate and butanol to test flavonoids. The highest abundant flavonoids were found in ethylacetate and butanol fraction.

Effect of Topdressing Methods of Nitrogen Fertilizer on Growth, Yield and Storage of Onion(Allium cepa L.) in Mulch-Cropping System (양파(Allium cepa L,) 멀칭재배시 질소비료 추비방법이 생육, 수량 및 저장성에 미치는 영향)

  • In order to fad out an efficient way of topdressing nitrogen fertilizer in mulch-cropping system of onion(Allium cepa L.), solid, slow-release, and liquid forms of nitrogen fertilizers were allied to cv. 'Changnyungdaego' various number of times at different time, with 5 topdress applications of solid fertilizer serving as a control. Whole basal application of conventional solid fertilizer and 2 slow-release fertilizers were labor-saving and showed improved storage quality of bulbs, but resulted in poor plant growth and considerably low yield due to fertilizer shortage from early April. This suggests that topdress application is necessary. Liquid form of nitrogen fertilizer was more effective for plant growth and yield and saving labor than the solid form. Early applications was effective for increasing yield and storage quality of onion bulbs harvested. Thus two applications of liquid form of nitrogen fertilizer in February and March at rome month interval are recommended in mulch crowing system of onion.

  • The optimized RAPD-PCR conditions, which can be utilized as a basic information for the analysis of the genetic characteristics were investigated with four onion varieties, named Changryungdaego, Yeoeuijuhwang, Yakwangju, and Dabonghwang using Operon primers, OPR01 (TGCGGGTCCT) and OPZ20 (ACTTTGGCGG). We tested several concentrations of DNA, primer, and MgCl2, annealing temperature, number of PCR cycle, and presence/absence of pre-heating time at the begining of PCR reation in the 25${mu}ell$ volume. The best RAPD profiles were obtained using 50ng of DNA, 5mM of primer, 1.5mM of MgCl2, 45$^{\circ}C$ of annealing temperature and an absence of pre-heating time. An establishment of the stable and reproducible RAPD-PCR conditions are expected to be useful for the subsequent RAPD-related investigation, such as genetic characterization of the onion strains, re-establishment of phylogenetic relationships and development of new varieties.

Effect of Allium cepa (red) and Angelica gigas Nakai on Hair-growth Promotion in C57BL/6 Mice (C57BL/6 마우스에서 모발성장 촉진에 대한 Allium cepa (red)와 Angelica gigas Nakai의 효과)

  • This study was conducted to evaluate the hair-growth promotion effect in C57BL/6 mice of a new substance mixed with Allium cepa (red)-bioconversion extract and Angelica gigas Nakai. The ethanol extract of Allium cepa (red) was bioconverted through the use of the Bacillus subtilis KJ-3 (BS3) strain, which was named Red-BCQ. The quercetin content of Red-BCQ increased by about 7.4-fold after bioconversion. Angelica gigas Nakai extract (Agnex) contains a large amount of coumarins such as decursin (D) and decursinol angelate (DA). A 1 mg portion of Agnex contained 0.4146 mg of D and 0.3659 mg of DA. Minoxidil has been known to promote hair growth. In this study, the hair-growth promotion effects of Red-BCQ, Agnex, and a mixture of both Red-BCQ and Agnex were compared with 5% minoxidil. Twenty-five mice were divided into five experimental groups including saline (CON), 5% minoxidil (PCON), Red-BCQ (RA), Agnex (AG), and a Red-BCQ-Agnex mixture (RAG)-treated group. Samples were administered orally once a day at a fixed time for 4 weeks. Hair growth was monitored by photograph at 7, 14, 21, and 28 days. We also observed 5α-reductase, alkaline phosphatase (ALP), γ-glutamyl transpeptidase (γ-GT), insulin-like growth factor (IGF)-1, transforming growth factor (TGF)-β1, antioxidant enzyme, and the hair follicles of the skin tissue. In all the results, the RAG-administered group showed greater antioxidative and hair-growth promotion effects than the other groups. These data suggest that RAG has potent stimulating activity on hair growth in C57BL/6 mice.

Variation of Nuclear DNA Content in Interspecific Allium cepa L.×A. fistulosum L. hybrids and Their Successive Backcross Lines (양파와 파간의 종간잡종 F1과 여교잡계통의 핵 DNA 함량)

  • Interspecific hybrid plants between Allium cepa L. (2n=2X=16) and A. fistulosum L. (2n=2X=16)and their backcross lines were developed by artificial pollination in order to introduce new desirable characters of A, cepa to A. fistulosum. The 2C nuclear DNA content has been estimated by flow cytometry in 5 Allium fistulosum inbreed lines, 2 interspecific hybrid lines of A. cepa${\times}$A. fistulosum and 34 their backcross lines $BC_1F_1$ to $BC_2F_2$, using propidium iodide (PI) as a fluorescence dye. Estimated 2C DNA values ranged from 22.2 pg to 23.7 pg in 5 A. fistulosum inbreed lines, 37.9 pg in F1 hybrid between A. cepa and A. fistulosum, 24.3 pg to 27.3 pg in 7 backcross lines in $BC_1F_1$, 21.9 pg to 24.4 pg in 9 $BC_1F_2$, 22.9 pg to 25.1 pg in 14 $BC_2F_1$, 22.6 pg to 23.4 pg in 4 $BC_2F_2$. This study showed mean 2C nuclear DNA content of $F_1$ hybrid was higher than their backcross progeny lines, while it was lower than female parental line, A. cepa (2C DNA=33.2 pg). Mean 2C DNA content of backcross lines, $BC_1F_1$ to $BC_2F_2$ was not significantly different but their 2C DNA contents in the more progress generation from $BC_1F_1$ to $BC_2F_2$ were reduced.
