토종과 중국도입종간 꾸지뽕잎의 항알레르기작용 비교 (Comparison on anti-allergic activities of leaves from Domestic and Chinese species of Cudrania tricuspidata)

  • Objectives : This study was performed to compare the effect of two Cudrania tricuspidata cultivars; Sancheong native (CT) and varieties in china(SCT) on immediate hypersensitivity of the anaphylactic type and Ova-induced allergic asthma mouse model by calculating serum cytokines and IgE. Methods : We investigated the free radical scavanging effect and quantify total phenol contents and total flavonoids of two Cudrania tricuspidata cultivars; Sancheong native(CT) and varieties in china(SCT). The sample was extracted by 80% EtOH. To induce the allergic asthma, in the control group and the CT group, mice of each group were sensitized intraperitoneally with ovalbumin (OVA) solution at the 1st, the 7th and the 14th day. After then, sensitization was performed by aerosol allergen challenges with 1% OVA solution intratracheally at the 21th, the 23th, 25th and the 27th day. At the 29th day, the mice were killed and the changes of interferon-${\gamma}$, interleukin-4, 5 and 10, total IgE and OVA-specific IgE in serum were checked. Results : CT inhibited compound 48/80-induced systemic anaphylaxis 90% with a dose of 100 mg/kg body weight at 1 hr before injection of compound 48/80. In the allergic asthma mouse model, IFN-${\gamma}$ was did not increased in the CT and SCT group than that in the control group. IL-4, IL-5, the total IgE and OVA-specific IgE were decreased in the CT group as compared with the control group and these results were statistically significant. Conclusions : Considering the above experimental results, this study showed that Sancheong native cultivar could reduce the allergic reaction.

천식 모델 마우스에서 골쇄보의 항천식 효과 (The therapeutic effect of Drynariae Rhizoma in a mouse model of allergic asthma)

  • Objective : Allergic asthma is a chronic airway disease that affects millions of people in the developed world. The disease is characterized by concurring airway inflammation, Th2 cytokine production, increased mucus secretion, airway hyperresponsiveness (AHR) to inhaled antigen, and pulmonary fibrosis. To investigate the therapeutic and anti-asthmatic effects of Drynariae Rhizoma (DR), we examined the influence of DR on the development of pulmonary eosinophilic inflammation and airway hyperresponsiveness in a mouse model of allergic asthma. Methods : In this study, BALB/c mice were systemically sensitized to ovalbumin (OVA) followed intratracheally, intraperitoneally, and by aerosol allergen challenges. We investigated the effect of DR on airway hyperresponsiveness, pulmonary eosinophilic infiltration, various immune cell phenotypes, Th2 cytokine production and OVA specific IgE production in a mouse model of asthma. Results : In asthmatic mice, we found that DR.treated groups had suppressed eosinophil infiltration, allergic airway inflammation and AHR by suppressing the production of IL-5, IL-13 and OVA specific IgE. Conclusions : Our data suggest that the therapeutic mechanism by which DR effectively treats asthma is based on reductions of Th2 cytokines (IL-5), eotaxin, OVA-specific IgE production and eosinophil infiltration.

Regulation of IgE-Mediated Food Allergy by IL-9 Producing Mucosal Mast Cells and Type 2 Innate Lymphoid Cells

  • Due to the increasing prevalence and number of life-threatening cases, food allergy has emerged as a major health concern. The classic immune response seen during food allergy is allergen-specific IgE sensitization and hypersensitivity reactions to foods occur in the effector phase with often severe and deleterious outcomes. Recent research has advanced understanding of the immunological mechanisms occurring during the effector phase of allergic reactions to ingested food. Therefore, this review will not only cover the mucosal immune system of the gastrointestinal tract and the immunological mechanisms underlying IgE-mediated food allergy, but will also introduce cells recently identified to have a role in the hypersensitivity reaction to food allergens. These include IL-9 producing mucosal mast cells (MMC9s) and type 2 innate lymphoid cells (ILC2s). The involvement of these cell types in potentiating the type 2 immune response and developing the anaphylactic response to food allergens will be discussed. In addition, it has become apparent that there is a collaboration between these cells that contributes to an individual's susceptibility to IgE-mediated food allergy.

Virus-like Particle (VLP) Mediated Antigen Delivery as a Sensitization Tool of Experimental Allergy Mouse Models

  • Antigen delivery systems play critical roles in determining the quality and quantity of Ab responses in vivo. Induction of protective antibodies by B cells is essential in the development of vaccines against infectious pathogens, whereas production of IgE antibodies is prerequisite for investigation of allergic responses, or type 1 hypersensitivity reactions. Virus-like particles (VLPs) are efficient platforms for expression of proteins of interest in highly repetitive manners, which grants strong Ab responses to target antigens. Here, we report that delivery of hen egg lysozyme (HEL), a model allergen, through VLP could provoke strong HEL specific IgE Ab responses in mice. Moreover, acute allergic responses were robustly induced in the mice sensitized with VLPs that express HEL, when challenged with recombinant HEL protein. Our data show that antigen delivery in the context of VLPs could function as a platform for sensitization of mice and for subsequent examination of allergic reactions to molecules of interest.

Allergenicity of Hot Peppers Cultivated in Korea

  • The proteins from 15 types of cultivar of hot peppers cultivated in Korea were extracted and its allergenicity was investigated by immunoblotting and enzyme-linked immunosorbent assay (ELISA). The immunoblotting of hot pepper proteins extracts (HPEs) against serum of hot pepper sensitized patients revealed dominant IgE binding to 14, 37, and 40 kDa molecules. The specific levels of IgE to HPEs sample No. 1, 3, and 7 were much higher than the other samples in patients. Also, IgE binding capacity of HPEs were not reduced by thermal processing and digestion in ELISA using human IgE antibody acquired from hot pepper sensitized patients. By means of Western blotting using anti-thaumatin IgY, thaumatin-like protein (TLP) acting as allergen in several plants and fruits was detected in tested hot peppers. This study demonstrates that the antigenic protein in hot peppers are present but are differently contained according to cultivars.

Effects of coffee intake on airway hypersensitivity and immunomodulation: an in vivo murine study

  • BACKGROUND/OBJECTIVES: Coffee is a complex chemical mixture, with caffeine being the most well-known bioactive substance. The immunomodulatory and anti-inflammatory properties of coffee and caffeine impact health in various aspects, including the respiratory system. The objective is to investigate the effects of coffee and caffeine on airway hyperresponsiveness and allergic reactions, as well as to analyze and compare associated cytokine profiles. MATERIALS/METHODS: BALB/c mice were intraperitoneally sensitized with ovalbumin (OVA) and given OVA inhalation to induce airway hypersensitivity. Two weeks after sensitization, they were intragastrically gavaged with coffee or caffeine, both containing 0.3125 mg caffeine, daily for 4 weeks. Control mice were fed with double-distilled water. Serum OVA-specific antibody levels were measured beforehand and 5 weeks after the first gavage. Airway hyperresponsiveness was detected by whole body plethysmography after gavage. Cytokine levels of bronchoalveolar lavage and cultured splenocytes were analyzed. RESULTS: Coffee effectively suppressed T helper 2-mediated specific antibody response. Airway responsiveness was reduced in mice treated with either coffee or caffeine. Compared to the control, coffee significantly reduced OVA-specific immunoglobulin (Ig) G, IgG1 and IgE antibody responses (P < 0.05). Caffeine also attenuated specific IgG and IgG1 levels, though IgE level was unaffected. Coffee significantly reduced interleukin (IL)-4 and increased IL-10 concentration in spleen cells and bronchoalveolar lavage fluid (P < 0.05). CONCLUSIONS: Coffee effectively attenuated airway hyperresponsiveness and systemic allergic responses induced by OVA food allergen in mice. As a complex composition of bioactive substances, coffee displayed enhanced immunomodulatory and anti-inflammatory effects than caffeine.

순천향대학교 천안병원에 내원한 알레르기 환자의 특성 (Characteristics of Allergic Patients in Soonchunhyang University Cheonan Hospital)

  • The purpose of this study was to evaluate the general features of allergic patients in northwestern Chungcheongnamdo who visited Soonchunhyang University Cheonan Hospital. The subjects in this study were 1692 suspected allergic patients. After their allergic symptoms were checked and a MAST-CLA test was conducted, the following results were obtained: 1. The mean age of the subjects was 23.1 years old. The male patients represented 56.2% and the female patients accounted for 43.8%. 1387 (82.0%) patients of those investigated suffered from allergic disease. 2. Among the 1387 patients, 1022 (73.7%) patients showed an increased total IgE level. The positive rate of those who were in their 40s (87.0%) was the highest, but their age made no difference to their positive rate of total IgE. 3. Concerning the positive rate for allergen specific antibody by age, those who were in their teens (73.5%) topped the list and similar in all age group except in their 40s. By gender, the positive rate of the male and female were 56.3% and 43.9% respectively. Regarding the positive rates by allergic disease, those who suffered from allergic rhinitis (60.4%) were most vulnerable, followed by the patients with allergic dermatitis (47.4%), with bronchial asthma (47.2%) and with urticaria (39.4%). 4. As for seasonal positive rates, they were most susceptible in April (77.2%) and May (71.1%). We discovered a significant difference according to seasons; Spring (60.1%), Winter (45.4%) and Summer (39.2%). 5. In case of Korean inhalent panel, the most dominant allergen-specific antibodies were "Cockroach mix" (31.1%), followed by "D. pteronyssius" (23.8%) and "Dog" (14.3%). In the event of food panel, the most popular allergen-specific antibodies were "D. farinae" (25.0%), followed by "D. pteronyssinus" (19.8%) and "Hose dust" (12.0%). 6. The residential type made no difference to the positive rates of "House dust", "Cockroach mix" and "Dog" as major antigen but compared with others, positive rates for "Tick" were somewhat higher in apartment buildings.

항원감작식품과 어린이 알레르기 질환과의 관계 (The Relationship between Food Allergen Sensitization and Allergic Disease in Childhood)

  • 2005년 3월부터 2006년 4월까지 어린이 74명(남 47, 여 27, 1-4세)을 대상으로 식품에 대한 IgE 특이 항체량을 검사하였다. 알레르기 증상을 나타낸 어린이의 연령은 영 유아(0-3세)가 34명, 4-6세가 24명, 7-9세가 8명, 10세 이상이 8명으로, 나이가 어릴수록 알레르기 증상을 보이는 어린이가 많은 것으로 나타났다. 검사한 어린이가 21종류의 식품 중 3종류, 4종류, 5종류의 식품에 반응을 보인 어린이는 각각 10명, 21명, 15명으로 나타났다. IgE 항체반응에서 class 2 미만으로 나타난 어린이의 식품은 콩 28명, 우유 29명, 치즈 21명, 난백 7명, 돼지고기 18명으로 나타났고 class 2 이상은 콩 20명, 우유 17명, 치즈 24명, 난백 20명, 돼지고기 21명으로 나타나 이러한 식품들이 주로 식품 알레르기와 관련이 있는 것으로 판단된다. 가족력에서는 알레르기 증상을 나타낸 비율이 남아는 45.5%, 여아는 50%로 절반 정도로 나타났고, 가족 중 알레르기 질병의 병력은 아토피 피부염은 26%, 아토피 비염은 10.5%,아토피 피부염과 아토피 비염은 31.5%, 두드러기 21.0%, 천식 10.5%이었다. 섭식력은 모유와 분유를 혼합하여 섭취한 어린이가 분유만 섭취한 어린이, 모유만 섭취한 어린이보다 높았다. 알레르기 유발식품으로 제한하는 식품은 계란과 우유 12.5%, 우유 2.5%, 계란 10.0%등이었다.

반복성 천명을 가진 소아에서 천식 발생 예측 지표의 적용 (Asthma predictive index in children with recurrent wheezing)

  • 목 적 : 소아 천식이 급격히 증가하고 있다. 영유아 시기의 천명을 보이는 환아들에서 천식 발생을 예측할 수 있는 고위험군을 선별하는 지표들이 알려져 있다. 저자들은 Tucson Children's Respiratory Study Group에서 제시한 천식 예측 지표 API와 변형한 천식 예측 지표 mAPI를 한국 영유아 천명 환아들에게 적용하여 의미있는 예측 지표를 규명하고자 하였다. 방 법 : 반복성 천명을 주소로 서울아산병원 소아과를 방문한 262명의 환아들을 대상으로 API와 mAPI를 적용하여 고위험군과 저위험군으로 나누었다. 각 군에서 알레르기 지표 뿐만 아니라 환아와 가족들의 알레르기 질환의 병력을 조사하였다. 결 과 : API 고위험군 환아(n=131)는 저위험군 환아(n=131) 보다 성별은 차이가 없었고 연령, 총호산구수, 호산구 비율, 혈중 ECP, 총 IgE가 높았으며, 흔한 항원에 대한 감작 여부가 포함되지 않았지만 흡입항원과 식품항원에 대한 감작률이 더 높게 나타났다. mAPI 고위험군 환아(n=141)는 저위험군 환아(n=121) 보다 총호산구수, 호산구 비율, 혈중 ECP, 총 IgE, egg, milk, D.p-, D.f-특이 IgE 농도가 높았으며 성별과 연령의 차이는 없었다. 위험군별 다변량 분석에서는 API군에서는 흡입항원에 대한 감작 여부가 고위험군을 결정하는 유의한 지표로 나타났으며 mAPI군에서는 혈중 ECP 농도가 유의한 지표로 나타났다. 대상군 중 6세 이상 추적 관찰된 환아는 모두 29명이었으며 최근 1년간 천명의 병력이 확인되고 임상적으로 천식으로 진단된 환아는 15명이었다. API의 민감도, 특이도, 양성예측률, 음성예측률은 66.7%, 50.0%, 58.8%, 58.3%이었으며, mAPI는 93.3%, 57.1%, 70.0%, 88.9%로서 mAPI가 더 높았다. 결 론 : API와 mAPI를 적용한 고위험군 환아에서 흔한 흡입 항원과 식품항원에 대한 감작률이 높아서, 영유아 천명 환아에서 주관적인 증상에 의한 진단보다는 객관적인 흡입항원과 식품항원에 대한 감작 여부를 지표로 사용하는 것이 더 의미 있을 것으로 생각한다. 또한 한국 영유아 천명 환아에서는 mAPI가 소아 천식의 발생을 예측하는데 더 도움이 될 것으로 생각한다. 그러나 본 연구는 29명의 소수의 환아들만 추적 관찰된 결과이므로 이후로 보다 더 많은 환아들을 추적 관찰하는 것이 필요할 것이다.
