A Simple and Efficient One-Pot Three-Component Synthesis of Propargylamines Using Bismuth (III) Chloride

  • A simple highly versatile and efficient method has been developed for the three-component coupling of aldehydes, amines and alkynes to prepare propargylamines, in the presence of a catalytic amount of $BiCl_3$. The advantages of methods are high yield, mild reaction conditions, no environmental pollution and easy work up procedure.

  • Heating [Cp*Rh(η²-NO₃)(OTf) (1) and PhC≡CPh in EtOH for 3 h gave a η⁴-cyclobutadienerhodium complex, [Cp*Rh(η⁴-C₄Ph₄)] (2). Complex 1 reacted with HC=CPh in acetone at room temperature for 3 h to give a (η⁴-cyclobutadiene)-rhodium complex, [Cp*Rh(η⁴-C₄HPhC=CPh)] (3). Whereas, the reactions of 1 with HC=CCH₂Cl in acetone at room temperature for 3 h gave the triply halide-bridged dinuclear rhodium complex, [Cp*Rh(μ₂-Cl)₃RhCp*](OTf) (4). Complexes 2-4 have been structurally characterized by X-ray diffraction.

Nanocrystalline Copper Oxide(II)-Catalyzed Alkyne-Azide Cycloadditions

  • Although the use of Cu(II) salts as catalysts without reductants is limited in the cycloaddition of acetylenes with azides, the catalytic system employing average 10 nm CuO(II) nanoparticles in the absence of reductants shows good catalytic activity to form 1,4-disubstituted 1,2,3-triazoles even in wet THF as well as water. It is also noticeable that CuO(II) nanoparticle catalysts can be recycled with consistent activity. A range of alkynes and azides were subject to the optimized CuO(II) nanoparticle-catalyzed cycloaddition reaction conditions to afford the desired products in good yields.

Photoaddtion Reactions of ο-Benzoquinones to Some Olefins and Alkynes

  • Photoaddition reactions of ο-benzoquinones to some organic molecules were ivestigated to yield three types of photoadducts. Irradiation (350nm UV light) of a dichloromethane solution of tetrahal-1, 2- benzoquinones and 1, 4-diphenylbutadiene yielded 1, 3-dienes, which was found to be used to synthesize 1-phenylphenanthrenes. The 1, 3-dienes were also produced, when irradiated tetrahalo--1, 2-benzoquinones and 1,4-dipenylbut-1-en-3-yne in the similar conditions, which was applied to get 9-phenylphenanthrenes. An enolic compound come from the tautomerization of dibenzoylmethane was found to add to ο-benzoquinones to give, 1, 4-dioxenes and 1, 5-diketones as the major products. Although depenylbutadiyne did not add to ο-benzoquinones, diphenylacetylene added to ο-benzoquinones to give $\rho$-quinomethanes as well as two isomeric $\rho$-quinomethanes. One-way photoisomerism was observed for two isomeric $\rho$-quinomethanes.

Cytochrome P-450 2A6 Inhibitor Based on the Indole Moiety

  • The cytochrome P-450 enzymes (CYP 2A6) regulate many endogenous signaling molecules and drugs. Aryl alkynes such as 2-ethynylnaphthalene are important P450 inhibitors which have been extensively studied as medicines or as an effective chemical probes for profiling mouse liver microsomal P-450. Here we have synthesized indole-based novel P450 inhibitor, 5-ethynyl indole 3, and showed that it has successfully inhibited CYP 2A6 by chemical inhibition reaction. By using HPLC equipped with a photo diode array(PDA) detector, all of the peaks derived from the enzymatic reaction have been characterized.

  • Copper(I)-catalyzed Huisgen cycloaddition of terminal alkynes with unmasked azidoamines derived from amino acids is described. The reported strategy provides a new entry to highly hindered ${\beta}$-amino 1,2,3-triazole derivatives bearing a gem-diaryl group, which are potentially valuable entities as molecular catalysts for asymmetric transformations.

  • The effect of pressure on 1,3-dipolar cycloaddtion has been studied by means of FT-IR and NMR spectroscopy. Pressure accelerates 1,3-dipolar cycloaddition without solvent or catalyst. This simple and inexpensive method eliminates the need for work-up or purification. The method is expected to be applied to the synthesis of binders for solid rocket propellants.
