EXPERIMENTAL STUDY OF THE EFFECTS ON THE GROWTH OF MANDIBULAR CONDYLE FOLLOWING CYCLOPHOSPHAMIDE ADMINISTRATION IN RAT (Cyclophosphamide가 백서하악과두의 성장에 미치는 영향에 관한 실험적 연구)

  • In this study, effects of cyclophosphamide on the growth of the mandibular condyle head were investigated with Spraque-Dawley rats of the 28 days of age. Rats were devided into four groups. Three were used as experimental groups, and one as control. Each rat in experimental group was injected intraperitoneally with cyclophosphamide repeatedly three times, 20mg/kg for the first group, 40mg/kg for the second, and 60mg/kg for the third each time. Rats in control group were injected with physiological saline in the same method. Rats in each group were sacrificed at 5, 10, and 15 days following the last injection. The specimens were stained with H-E, toluidine blue, PAS, and alcian blue. The results were as follows; 1. In experimental group, with increasing the injection doses, the thickness of the condylar cartilage from the transitional zone to the hypertrophic zone became thinner than in control group. 2. Weaker metachromasia to toluidine blue and less positive reaction to PAS were seen. 3. In primary marrow cavity the fewer trabecular was formed, The direction of trabecular formation became obscuerer, and the lower density of bone was resulted in.

A case of splenic extraskeletal mesenchymal chondrosarcoma in a Yorkshire Terrier dog

  • A 7-year-old male Yorkshire Terrier dog was present to an animal clinic and a large soft mass was found in the spleen by radiological examination, and total splenorectomy was performed. Grossly, a large protruded splenic mass was soft to moderately firm and multilobulated. On the cut surface, the mass was off-white to tan, dark red, and rust colored with many cavitation and had gelatinous areas. Histologically, the tumor cells were characterized by coexistence of the primitive mesenchymal tissues and mature or immature cartilage tissues. Primitive mesenchymal areas were composed of round/oval or spindle shaped immature cells with high mitosis. The tumor cells of the cartilage areas were located in basophilic cartilaginous matrix. Intercellular matrix in the cartilaginous areas was stained blue with Masson's trichrome and deep blue with alcian blue, respectively. Immunohistochemically, the cartilaginous tumor cells demonstrated positive reactions for vimentin and S-100, and surrounding mesenchymal tumor cells are immunopositive for vimentin. This case was diagnosed as splenic extraskeletal mesenchymal chondrosarcoma of a Yorkshire Terrier dog, a toy breed.

Distribution of Mouse Uterine Mast Cells during Estrous Cycle (발정주기에 따른 생쥐 자궁조직 내 비만세포의 분포)

  • We examined the distribution of uterine mast cells in cycling mice of different ages between 7 weeks and 38 weeks by toluidine blue staining. Mast cell density increased gradually depending on the age and culminated at 30 weeks after birth. Of the estrous stages, uteruses at metestrus showed markedly higher density of mast cells than those at the other stages in cycling mice of all ages analysed in this study and the majority of the mast cells were found in myometrium. We also discriminated types of the uterine mast cells in cycling mice at 10 weeks old by Alcian blue-safranin double staining. Mucosal type was most abundant among three types, including connective-tissue types and mixed types, through the entire estrous cycles. The portion of mixed types increased slightly after estrous. The abundance of uterine collagen protein determined by Masson trichrome staining was exactly consistent with the density distribution of the uterine mast cells during estrous cycles. Taken together, these results may imply that uterine mucosal mast cells, together with collagen proteins, are heavily involved in tissue remodeling of cycling mouse uterus.

  • The ultrastructure of the digestive organ of Korean Planaria (Dugesia japonica) is studied by light microscope and transmission electron microscope. 1. Pharynx The epithelium surrounding pharyngeal lumen has a number of microvilli on the free surface. The epithelial cells contain PAS-positive granules which are 0.4 to 0.6 $\\mum$ in size. They also contain hundreds of vesicles and vacuoles. The pharyngeal epithelium of the external surface surrounded by pharyngeal cavity possesses a number of cilia and microvilli on the free surface. A number of muscle bundles are found in the pharyngeal tissue. The parietal epithelium surrounding pharyngeal cavity have microvilli and electron-dense secretory granules. 2. Caeca The cells which constitute the cecal epithelium are divided into four kinds of cells. 1) Phagocytic cell : These cells are characterized by presence of a number of lysosomes. These cells have highly developed mitochondria, polyribosomes and granular endoplasmic reticulum of which cisternae are distended. 2) Granular club cell : These cells contain round granules 5 $\\mum$ in diameter which show strong PAS-positivity and weak eosinophilia. The cells have highly developed granular endoplasmic reticulum. 3) Storage cell : These cells include thousands of glycogen granules in the cytoplasm. These cells also have second kind of round granules which are 1.4 to 3 $\\mum$ in size and exhibit PAS-positive reaction. 4) Immature storage cell : These cells have a large nucleus and contain a small number of granules which have PAS-positive granules and a few lipid droplets. Several chromatoid bodies are found in the cytoplasm around the nucleus.

Histochemical Analysis of the Cutaneous Wound Healing in the Amphibian (양서류 피부 상처회복과정에 대한 조직화학적 분석)

  • The wound healing is very complex biological processing including inflammatory, reepithelialization and matrix construction. According to the biological systematic category, the ability of the healing is very different. Generally healing ability of the lower animal group has been known more excellent compared to its higher group. Therefore, lower animals have been used as the experimental model to explore the mechanism of the wound healing or repair. To verify histochemical characteristics of the wound healing, we have used skin of the frog (Bombina orientalis) as known common amphibian. At day 1, 10, and 16, the mucous substance was very actively synthesized and strong positive by PAS and Alcian blue (pH 2.5). Day 10 after wounding, margin of the wound was gradually strong positive by PTAH staining for detection of collagen synthesis. At 3 to 6 hour and day 23 to 27, we have found the cell division was active through the MG-P staining, in which the concentration and division of DNA in nucleus was green to deep blue color.

A case of polypoid cystitis in a dog

  • Among benign proliferation of the urinary bladder, polypoid cystitis is a rare disease in dogs. It is characterized by epithelial proliferation, chronic inflammation in lamina propria, and development of a polypoid mass or masses without evidence of neoplasia. This report describes histopathologic features of polypoid cystitis in dog. A 10-year-old spayed female shihtzu-dog was presented with two-month history of hematuria. Abdominal ultrasonography confirmed the thickened bladder wall and calculi in both kidneys. Surgical biopsy sample was taken from the thickened bladder mucosa for the histopathologic examination. The mass was covered with irregular hyperplastic transitional epithelium with the projection into the lumen in multifocal areas as well as many Brunn's nests in lamina propria. Many inflammatory cells such as lymphocyte, plasma cell, and macrophage and few neutrophils were occupied in lamina propria and submucosa. Proliferated fibrous tissues in lamina propria were clarified by using special staining methods. These collagens were stained blue with Masson's trichrome and red with van Gieson, but negative for alcian blue. Based on the clinical, gross, and histopathologic examinations, this case was diagnosed as polypoid cystitis in a dog. In our best knowledge, this is the first report of polypoid cystitis in dog in Korea.

Surface Characteristics and Adhesive Properties of Pichia farinosa SKM-1, Pichia anomala SKM-T, and Galactomyces geotrichum SJM-59 for Preparation of Probiotics

  • Probiotics are generally excreted within a few days if their ingestion in feces at the same rate as or even more quickly than a transit marker (meaning not clear). Ability of probiotics to adhere to intestine prolongs their persistence in gastrointestinal tract, allowing them to exert healthful effects longer. Hydrophobicities, zeta potentials, Alcian blue-binding capacities, and sedimentation profiles of Pichia farinosa SKM-1, P. anomala SKM-T, and Galactomyces geotrichum SJM-59 were determined to evaluate characteristic properties of cell surfaces responsible for adhesion. Results of intestinal Caco-2 cell line in vitro and murine intestine in vivo studies revealed these strains exhibit adhesive properties regardless of their cell surface hydrophobicity.

Ultrastructure of the Integumentary System of the Amberjack, Seriola dumerili (Teleostei: Carangidae), from Korea (잿방어, Seriola dumerili 피부계의 전자현미경적 구조)

  • Integumentary structure of the amberjack, Seriola dumerili, was examined by light and transmission electron microscopy. Integument of the amberjack consists of epidermal and dermal layers. The epidermal thickness is about $28.32{\mu}m$. The epidermal layer could be classified into superficial, intermediate, and basal layers by morphology and position of the supporting cells. Secretory cells of the epidermal layer were classified into mucous cells and club cells in the amberjack. Mucous cells contained acidic mucous and were alcian-blue positive with AB-PAS (pH 2.5) reaction. The club cells contained well-developed rough endoplasmic reticula and large central vacuole. Also, filament-rich cells were identified in the epidermal layer. The dermal layer consists mainly of collagenous fiber, and it contains fibrocytes and pigment cells. The cytoplasm of fibrocyte had a well-developed rough endoplasmic reticulum. The pigment cells contained electron-dense melanin granules.

Histochemical Properties Study on the Mucosubstances of the Intestinal Mucosa in the Raja kenojei (홍어 장점막 점액의 조직화학적 성상에 관한 연구)

  • This study is performed in order to clarify the histochemical structure, the distribution of mucous cell and goblet cell, and the histochemical properties of the mucosubstances in the middle region of intestinal mucosa and rectum of Raja kenojei. In the H&E stain, distribution of the mucous cells and acidophilic cells were a more compacted middle portion than other regions of intestine, but the former was more than the latter in the number of mucous cells to rectum. The mucosubstances were stained with aldehyde fuchsin pH 1.7-alcian blue(pH 2.5) stain and then compared to distribution of the mucosubstances used in image and microscope technology(IMT-Size5). The middle intestine of Raja kenojei was composed of mucous cells having only large amounts of mucosubstances in the distal region was much more than that of proximal region. It was two types of mucous cells to rectum, one type was the same as proximal intestine while the other had small amounts of weakly sulfated and large amounts of carboxylated mucins.

Effect of Yam Yogurt on Colon Mucosal Tissue of Rats with Loperamide-induced Constipation

  • The effects of lactic acid fermented yam yogurt (Yam/YG) on colon mucosal tissue were investigated in a loperamide-induced constipation rat models. Sprague-Dawley rats were fed for 6 weeks with 3 types of diets (normal, supplemented with lactic acid bacteria, and supplemented with Yam/YG), and were then administered loperamide intraperitoneally twice daily for 5 days. Administration of loperamide decreased fecal excretion and the moisture content of feces with increasing of numbers of pellets in the colon. On the histopathologic findings from hematoxylin and eosin (H& E) and alcian blue stainings, supplementation with Yam/YG resulted in the recovery of depleted goblet cells and mucin, and increased the numbers of Ki-67 positive cells, indicating restoration of colonic mucosa through cell proliferation and crypt regeneration against damages observed in crypt epithelial cells of loperamide-induced rats. These results indicate that Yam/YG improves evacuation and mucus production in the gastrointestinal tracts of constipated-induced rats.
