Effect of Dietary vitamin E and Protein on Cadmium toxicity in Rats (카드뮴 투여 흰주의 혈청 및 간장성분에 미치는 식이성 비타민 E와 단백질의 영향)

  • This study was undertatken to investigate the effects of dietary vitamin E on the toxicity of cadmium(Cd) in rats. The two variables were the supplmentary vitamin E(400lU/kg) and the protein amount(10.5% in the low protein diet and 18.0% in the normal protein diet) In cadmium treated rats net weight gain and food intake were decreased but improved by supplementation with vitamin E in the normal protein, hematocrit values reduced by Cd were significantly increased by the addition of vitamin E to normal protein diet in Cd intoxicated rats, The supplementation with vitamin E diminished the effect of Cd on aspartate aminotransf-rase and alanine aminotransferase activities in serum In Cd treated rats fed normal protein diet with vitamin E the contents of triglyceride were decreased and total-cholesterol contents were significantly reduced in serum and both of them in liver were markedly decreased. The activity of alcohol dehydrogenase in liver was decreased by Cd however supplementation with vitamin E reduced the effects of Cd on hepatic alcohol dehydrogenase. the results of this experiment indicated that there was some interaction between vitamin E and protein levels and supplementation with vitamin E had an effect more than protein levels oncd toxicity.

Effects of the Zizyphus jujuba Seed Extract on the Lipid Components in Hyperlipidemic Rats

  • The purpose of this study was designed to observe the effects of Zizyphus jujuba seed extract on the concentrations of the lipids and blood glucose in the S.D. rats fed the experimental diets for 4 weeks. Concentrations of total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free-cholesterol, cholesteryl ester, triglyceride (TG), phospholipid (PL) and blood glucose in serum were significantly higher in the cholesterol administration groups (group 2 (cholesterol+water), group 3 (cholesterol+Zizyphus jujuba seed extract)) than those in the control group (group 1, basal diet+water). But the concentrations of total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free-cholesterol, cholesteryl ester, TG, PL and blood glucose in serum ware remarkably lower in the group 3 than those in the group 2. In the ratio of HIDL-cholesterol concentration to total cholesterol and HDL-cholesterol concentration, Zizyphus jujuba seed extract administration group was higher percentage than in the group 2. The activities of aspartate aminotransferase (AST), alanine amino-transferase (ALT), lactate dehydrogenase (LDH) and alkaline phosphatase (ALP) in serum were rather lower in the Zizyphus jujuba seed extract administration group (group 3) than in the cholesterol diet group (group 2). From the above research, Zizyphus jujuba seeds were effective on the improvement of the blood glucose, lipid compositions in serum of dietary hyperlipidemic rats. And particularly, Zizyphus jujuba seeds were more effective as a therapeutic regimen for the control of metabolic derangements in adult disease.

  • The purpose of this study was to obtain reference data of parameters for hematological health diagnosis in marine fish and also evaluate application of veterinary chemistry analyzer used to those blood tests. A blood profile of total 522 fish for black rockfish (Sebastes schlegeli), red seabream (Pagrus major), rock bream (Oplegnathus fasciatu) and black seabream (Acanthopagrus schlegeli) cultured in mari-floating netcage of Gyeongnam province was determined by hematocrit (Ht), hemoglobin (Hb) and blood chemistry tests (total protein, albumin, alkaline phosphatase, blood urea nitrogen, lactate dehydrogenase, triglyceride, total cholesterol, creatinine, aspartate aminotransferase, alanine aminotransferase, glucose). Ht was measured by microhematocrit method. Hb and plasma chemistry were analysed by establishing baseline ranges for a dry chemical system of FUJI DRI-CHEM 3000. Actually recorded values of Hb and plasma chemistry by the analyzer were notably outside from the minimum and/or maximum of the established reference value. Albumin and alanine aminotransferase were not detectable in the range of 68~66%. Lactate dehydrogenase, total protein, alkaline phosphatase and glucose were not detectable in the range of 42~21%. Total cholesterol, aspartate aminotransferase, triglyceride, hemoglobin and creatinine were not detectable in the range of 18~3%. However, the values of blood urea nitrogen were below the detectable limits of the analyzer.

Effects of Dextrin and β-cyclodextrin on Protective Effect of Hovenia dulcis Fruit Extract Against Alcohol-induced Liver Damage in vivo (Dextrin과 β-cyclodextrin이 생체 내에서 헛개나무 추출물의 알코올성 손상으로부터 간보호에 미치는 영향)

  • ${\beta}$-cyclodextrin has an ability to protect compounds from oxidative reaction by collecting them within its ring-like structure. So, In harsh condition ($40^{\circ}C$), marker compound, quercetin, was dramatically reduced in Hovenia dulcis fruit extract containing dextrin at 4 and 8 week compared to 0 week, but not that containing ${\beta}$-cyclodextrin. To evaluate the effects of dextrin and ${\beta}$-cyclodextrin on protective effect of H.dulcis fruit extract against alcohol-induced liver damage, The mice were orally injected alcohol, H. dulcis fruit extract/dextrin (HD) and H. dulcis fruit extract/${\beta}$-cyclodextrin (HCD), respectively, for 7 days. The mice orally administrated with alcohol significantly enhanced the serum concentration of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) and the activity of lactate dehydrogenase (LDH) in serum compared to the control group. HD and HCD significantly decreased the levels of serum ALT and AST and serum LDH activities compared to alcohol group. And also alcohol group significantly increased the level of total cholesterol compared to the control group, but HD and HCD significantly reduced it compared to the alcohol group. However, the levels of TG in blood were not significantly changed in all groups. The activities of alcohol dehydrogenase (ADH) were significantly increased in HD and HCD group although those of aldehyde dehydrogenase showed an increasing tendency. This data suggested that HD and HCD were able to induce alcohol degradation in the liver tissues. All together, the results showed that HCD demonstrated their ability to protect liver from alcohol-induced damage on equal terms with HD.

Effects of HgCl2 on plasma DNA content and blood biochemical values in rats (랫드에서 수은이 혈장 DNA와 혈액화학치에 미치는 영향)

  • Changes of plasma DNA contents and serum biochemical values were measured in rats administered with $HgCl_2$ to investigate the in vivo cytotoxic effects of mercury and examine the usefulness of these changes as indicators of mercury exposure and diagnosis of mercury poisoning. Rats were given once intraperitonealy $HgCl_2$(0.13. 0.32. 0.8 and 2 mg/kg b.w) and the changes of plasma DNA contents and serum biochemical values were measured at the time of 2, 4, 8, 24, 48 and 72 hours after the administration of $HgCl_2$. Plasma DNA contents began to increase from 2 hours after the administration of $HgCl_2$ in all the treatment groups significantly compared to control with dose-dependent pattern. The levels of plasma DNA reached to peak at 48 hours as 2.77, 7.60, 15.46 and 16.51 times higher than control in each treatment group of 0.13, 0.32, 0.8 and 2 mg/kgb.w, respectively and remained to be higher until 72 hours after the administration. The values of creatine kinase, aspartate aminotransferase, alanine aminotransferase, lactate dehydrogenase, blood urea nitrogen and glucose of serum were increased, however the values of alkaline phosphatase, total protein and triglyceride were decreased. These changes of increase and decrease showed dose-dependent pattern but the starting time, maintenance and magnitude of change were various and characteristic according to serum biochemical indices. Among the changes of serum biochemical values, those of aspartate aminotransferase, lactate dehydrogenase and blood urea nitrogen were apparently and significantly increased compared to control from 2 to 72 hours by the administration of 2 mg/kg $HgCl_2$. This study demonstrates that plasma DNA and serum biochemical values such as aspartate aminotransferase, lactate dehydrogenase, blood urea nitrogen and etc. are valuable as biomarkers for mercury exposure assessment and diagnosis of mercury poisoning.

The Effects of the Ultrasound and Swimming on the Changes of Serum Creatine Kinase Activities in the Nerve and Muscle Injuried Rats. (초음파와 수영이 신경근 손상 흰쥐의 혈청 CK 활성 변화에 미치는 영향)

  • This studies to investigate the effects of ultrasound and swimming on the changes of the serum creatine kinase, latate dehydrogenase, aspatate aminotransferase, alanine aminotransferase, alkaline phosphatase, in peripheral nerve and muscle injury rats. The forty Sprague-Dawley adult male rats were assigned to the 4 groups: the experimental groups(3), and the peripheral nerve and muscle injury control group(1). There was made artificial injured by ischial nerve and muscle of each rats the each experimental ultrasound group and swimming group were treated from 3 days after being injuried for the 5 minutes and 10 minutes every day during the 10 days and 16 days respectively. The results were as follows: 1. The activities of creatine kinase, lactate dehydrogenase were significantly decreased on of the ultrasound and swimming groups for 10 days. All experimental group were significantly decreased for 16 days. there were significantly decreased the swimming group and ultrasound group were to the 10 days group. 2. The activities of asparatate aminotransferase, alalnine aminotransferase on the ultrasound group were significantly increased to the injured control group, there were significantly decreased the ultrasound group for 16 days group were to the 10 days group and there were significantly increased the swimming group for 16 days group were to the 10 days group. 3. The activities of alkaline phosphatase changes were no difference all experimental groups, there were significantly increased for 16 days group to the 10 days group. From these results it may be concluded that the effects of the ultrasound and swimming, the changes of the serum activities creatine kinase , lactate dehydrogenase of (; the objective indicates) to the muscle regeneration process of the nerve and muscle injured rats.

Eliminatory Effect of Mixture including Hot Water Extract of Dendropanax morbifera Lev. on Alcohol-induced Blood Alcohol Concentration and Hangover in Rat (흰쥐에서 황칠나무 열수 추출물을 포함한 혼합물의 혈중 알코올 농도와 숙취 해소 효과)

  • This study was performed to investigate the ameliorating effect of a hangover beverage mixture (SBJ) that contains Dendropanax morbifera Lev. and several medicinal plant extracts, on hepatoprotection and alcohol-metabolizing enzymes in alcohol-induced hangover in both in vitro and in vivo models. In human hepatoma cell line, HepG2, 300 mM of ethanol-induced hepatotoxicity was significantly improved by pretreatment of SBJ by dose-dependent manner. In the in vivo study, administration of alcohol to rats raised to the concentration of blood alcohol and lactate dehydrogenase (LDH). Blood alcohol and LDH levels in SBJ-treated rats significantly decreased at 0.5 h and 8 h after acute ethanol administration (40%, 4.6 g/kg body weight) as compared to alcohol-treated rats. Hepatic alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH) activity were significantly higher in SBJ-treated rats than in alcohol-treated rats. SBJ supplementation reduced formation of malondialdehyde (MDA), and inhibited reductions of hepatic superoxide dismutase (SOD), hepatic glutathione (GSH), glutathione-S-transferase (GST), glutathione reductase (GR) and glutathione peroxidase (GPx) levels, compared with rats administered alcohol. Plasma catalase (CAT), aspartate aminotransferase (AST) and alanine aminotransferase (ALT) levels showed unaltered resulted in all experimental groups compared with the control group. These results suggest that SBJ exhibit hepatoprotective properties by enhancing ADH, ALDH activity and stimulating the antioxidant defense system in alcohol-induced hangover.

Effect of Elsholtzia splendens Extracts on the Blood Lipid Profile and Hepatotoxicity of the Mice

  • Effects of extracts obtained from the flowers of Elsholtzia splendens on the serum lipid profile and hepatotoxicity in mice were investigated. Female ICR mice were given E. splendens ethanolic extract (ESEs) orally at a dose of 10 or 50 mg/kg BW for 50 days. Significant dose-dependent decreases in triglyceride and low-density lipoprotein (LDL)-cholesterol of serum were observed. In addition, ESEs prolonged the lag-time of LDL oxidation in vitro. In the serum of ICE mice given ESEs orally at 10 and 50 mg/kg BW, the serum levels of aspartate aminotransferase (AST) and lactic dehydrogenase (LDH) increased significantly, while total protein, albumin, creatinine, alanine aminotransferase (ALT), and total bilirubin did not change. Therefore, ESEs may be beneficial to human health, although it has some hepatotoxicity.

Effects of Hwangryeun Extract on the Recovery of Liver Function in $CCl_4$-exposed Rats (황련추출물이 $CCl_4$중독 Rat의 간기능 회복에 미치는 영향)

  • Effects of Hwangryeun(Copitidis rhizoma) extract on recovery of liver function were researched in $CCl_4$ intoxicated rats. Concentration ${\alpha}$-fetoprotein, total protein in plasma showed a tendency to decrease in Hwangryeun(Copitidis rhizoma) extract applied groups while that of albumin showed a increase. Activities of plasma aspartate aminotransferase and alanine aminotransferase in Hwangryeun(Copitidis rhizoma) extract applied groups showed a lower value than those of controlled groups. Alkaline phosphatase, lactate dehydrogenase and ${\gamma}$-glutamyl transpeptidase activities showed a tendency to decrease in Hwangryeun(Copitidis rhizoma) extract applied groups. Concentration of plasma triglyceride and total cholesterol showed a lower values than that of control group.

Studies on the Blood Chemical Properties in Racehorses: I. Serum Enzyme Activities (경주마(競走馬)의 혈액화학성상(血液化學性狀)에 관한 연구: I. 혈청효소(血淸酵素) 활성치(活性値))

  • The present study was carried out to investigate the serum enzyme activities of racehorses. The enzymes investigated were aspartate and alanine aminotransferase (GOT, GPT), ${\gamma}$-glutamyl transpeptidase(${\gamma}$-GTP), Lactic dehydrogenase(LDH), creatine phosphokinase(CPK), and alkaline phosphatase(ALP). Animals used were 30 healthy racehorses(♀17, ♂13) average weighing 435kg and were from 2 to 8 years of age. LDH activity was progressively decreased with age and next in the order of GOT and CPK activities. GOT and ${\gamma}$-GPT activities were not changed with age but ALP activity tended to be decreased wish age. Activities of GOT, GPT, ${\gamma}$-GTP and ALP were higher in female than in male. There was no difference in CPK activity by sex, and no difference was found out among breeds.
